        Streptozotocin으로 유발된 당뇨백서에서 수종의 한약재가 혈당강하에 미치는 영향

        N/A Objectives : This study was aimed to verify the hypoglycemic effects of the herbs which appear frequently as the effects of antipyresis & putting out the fire(淸熱○火)-Phellodendri Cortex(黃柏), Scrophulariae Radix(玄蔘), Radicis Lycii Cortex(地骨皮), Gardeniae Fructus(○子), Aloe(○○), Rehmanniae Radix(生地黃), Anemarrhenae Rhizoma(知母)-which were chosen among the herbs that were regarded as holding the hypoglycemic effects. Methods : To verify the hypoglycemic effects through the animal test, after the rats for test were treated 1 times with %mg/kg i.v. injection of streptozotodSigma, USA) which had been dissolved in the pH4.3 citrate buffer and were induced experimental hyperglycemia, the streptozotocin(ST2) induced hyperglycemic rats were prescribed with extract liquid 7.58 of each herb produced for test. And then the component such as glucose, insuline, and glucagon in serum and the change of body weight were measured and analyzed. Through that course, the significance for cure of DM were verified. Results : According to the booklet investigation, seven herbs which were useful for cure of DM were chosen and tested per 0s. As the result of that, It became evident that .Scrophulariae Radix, Anemarrhenae Rhizoma were significant for the hypoglycemic effects on DM. The insulin measurement showed the significant increase on both 3rd day and 7th day in the group of Scrophulariae Radix, Gardeniae Fructus, Aloe, Rehmanniae Radix and only showed the significant increase on 7th day ipl the group of Phellodendri Cortex, Radicis Lycii Cortex, Anemarrhenae Rhizoma. Besides, the glucagon measurement showed the significant decrease in the group of Radicis Lycii Cortex, Gardeniae Fructus, Rehmanniae Radix, Anemarrhenae Rhizoma. And the measurement of weight change showed the significant increase in all the sample herb groups. Conclusions : According to above result, Phellodendri Cortex, Scrophulariae Radix, Radicis Lycii Cortex, Gardeniae Frutus, Aloe, Rehmanniae Radix, Anemarrhenae Rhizoma were expected to be applied to the treatment of DM.

        Hypoglycemic and Hypolipemic Effects of Ixeris dentata in Diabetic Rats

        Blood glucose and total lipid levels in rats with streptozotocin-induced diabetes were determined after intraperitoneal administration of methanolic extract of Ixeris dentat whole plants and its main component, cynaroside (=luteolin 7-O--$\beta$-D-glucoside). From the data obtained, it was concluded that intraperitoneal administration of the methanolic extract produced a significant hypoglycemic effect. Total blood lipids were also decreased. Cynaroside in the streptozotocin-diabetic rats failed to exhibit hypoglycemic effect but a significant hypolipemic activity was observed. Thus, it is suggested that this methanolic extract may contain one or more hypoglycemic and hypolipemic principles including the main flavone glucoside, cynaroside, which can significantly reduce the levels triglyceride and total cholesterol in streptozotocin-diabetic rats.

        Physiological Effect of Hamcho Yogurt on Streptozotocin-Induced Diabetic Rats

        함초 추출물 분말을 첨가하여 유산 발효시킨 함초 요구르트를 streptozotocin-유발 당뇨쥐에 투여하여 항당뇨효과 및 콜레스테롤 저하 효과를 규명하기 위하여 3주간 실험하였다. 당뇨 대조군에 비해 함초 요구르트 투여군에서 혈당치 농도 및 식이 섭취량이 현저히 감소하였다. 경구당부하 실험에서도 당뇨 대조군에 비해 함초 요구르트 투여군에서 현저한 혈당상승 억제작용을 나타내었다. 당뇨쥐에 함초 요구르트와 일반 요구르트 섭취에 의해 혈중 총 콜레스테롤 농도 감소와 더불어 동맥경화지수도 감소하였다. 따라서, 함초 요구르트에 의한 혈당치 및 콜레스테롤 농도 감소효과는 고혈당증 및 고 콜레스테롤 혈중에도 유익한 작용을 할 수 있는 기능성식품 개발 가능성이 있음을 시사하였다. The present study was undertaken to investigate the hypoglycemic effect of Hamcho yogurt fermented by lactic acid bacteria containing Hamcho (Salicornia herbacea) extract powder in streptozotocin(STZ)-induced diabetic rats for 3 week period. The Hamcho yogurt given to the STZ-diabetic rats had significant effect in lowering the serum glucose concentrations and food intakes compared to the STZ-diabetic rats. The Hamcho yogurt administration increased the tolerance to oral glucose loading in the STZ-diabetic rats. The Hamcho yogurt and yogurt given to the STZ-diabetic rats were significantly decreased the serum total cholesterol concentration and the atherogenic index. Based on the beneficial effect against the blood glucose and cholesterol levels, we suggest the Hamcho yogurt can be used as functional food for the hypoglycemic and hypocholesterolemic effects.

        β-glucan 함량과 관능검사를 통한 상황버섯 품질기준 모색

        Objective : Phellinus spp. mushroom is an object of interest because it has excellent anticancer effect. Owing to the similalarities in the morphology, Phellinus linteus and Phellinus baumii are often used as same Sang Hwang Mushroom in the Korean market.. The quality control for mushrooms is needed because there are many differences in the efficacy according to cultivation method and cultivation area. Therefore, a reliable authentication method of these herbal medicine is necessary to compare and measure the amount of beta-glucan which is known to have a hypoglycemic effect, from the mushrooms collected in various regions Methods : 7 samples of medicinal mushrooms supplying phellinus spp. were collected in Korea, China and Cambodia. We investigated the hardness, colors, extract ratio, α-amylase and α-glucosidase inhibitory activities, glucose transporter 4 (GLUT-4) expressions of water extracts from Phellinus spp and also MTT assay were examined for cell toxicity. Results : The results revealed that Phellinus spp.water Ext.inhibited α-glucosidase activity. glucose transporter 4 (GLUT-4), the key insulin signaling pathway transcription factor, was remarkably increased by the Phellinus baumii water extract Conclusions : These results suggest that The more yellowish the mushroom is, the lower the hardness, the more the content of β-glucan is proportional. Because the more β-glucan, the greater the effect of hypoglycemia. compared to the hypoglycemic effect, Phellinus Baumii grown at hanging on selves for 7 month in the green house is the best.

        Enzymatic Production of Soluble Dietary Fiber from the Cellulose Fraction of Chinese Cabbage Waste and Potential Use as a Functional Food Source

        Use of Chinese cabbage waste as a raw material for production of soluble dietary fiber was investigated. Chinese cabbage was treated with a mixture of 1M NaOH and hydrolyzed using Celluclast 1.5 L. A supernatant was then treated using 85% ethanol, and separated into alcoholsoluble dietary fiber (ASF) and alcohol-insoluble dietary fiber (AIF) fractions. AIF effectively hindered diffusion of glucose and bile acid from dialysis membranes, and exerted a significantly (p<0.05) greater effect on retardation of bile acid activity than both carboxymethylcellulose and pectin. Water-soluble dietary fiber obtained from the cellulose fraction of Chinese cabbage using enzymatic hydrolysis is a potential source of dietary fiber with prebiotic, hypoglycemic, and hypolipidemic effects.

        합병증을 동반하지 않은 2형 당뇨병 환자에 대한 백호가인삼탕과 혈당강하제 병행치료의 혈당 강하 효과 및 안전성 연구 : 체계적 문헌 고찰과 메타분석

        Objectives: The purpose of this study is to assess the efficacy and safety of Baekhogainsam-tang for type 2 diabetes without complications by systemic review and meta-analysis of randomized controlled trials (RCTs). Methods: For a systematic review and meta-analysis, we set a key question in accordance with PICOT-SD. We searched the following up to March 31. 2021: PubMed, EMBASE, The Cochrane Library, CNKI, CiNii, KISS, KMBASE, OASIS, and ScienceON. A meta-analysis was conducted by synthesizing the results, including fasting plasma glucose level, postprandial 2-hour plasma glucose level, and Glycated Hemoglobin (HbA1c). Results: A total of five trials are included in this systemic review. The treatment group (Baekhogainsam-tang plus conventional treatment) showed more statistically significant effect than did the control group (conventional treatment only) in fasting plasma glucose level, postprandial 2-hour plasma glucose level, and glycated hemoglobin (HbA1c). Conclusions: Baekhogainsam-tang showed statistically significant effects in hypoglycemic effect and in improving insulin resistance. However, the number of studies included in the meta-analysis was insufficient, and the BIT used in the included studies was not standardized. This topic requires further attention and more clinical research.

        Investigation of Antioxidant, Hypoglycemic and Anti-Obesity Effects of Euphorbia Resinifera L.

        Objectives: The aim of this work is to evaluate the in vitro antioxidant, hypoglycemic, and antiobesity effects of Euphorbia resinifera extracts and investigate the phenolic constituents and the toxicity of these extracts. Methods: Phytochemical screening was performed to detect polyphenols and flavonoids. Antioxidant activity was evaluated by four methods (DPPH, ABTS, H 2 O 2 , and xanthine oxidase inhibition). The hypoglycemic effect was determined by the inhibition of α-amylase and α-glucosidase enzymes in vitro and via a starch tolerance study in normal rats. The antiobesity effect was estimated by in vitro inhibition of lipase. Results: Phytochemical screening revealed that the ethanolic extract was rich in polyphenols (99 ± 0.56 mg GEA/g extract) and tannins (55.22 ± 0.17 mg RE/g extract). Moreover, this extract showed higher antioxidant activity in different tests: the DPPH assay (IC 50 = 53.81 ± 1.83 µg/mL), ABTS assay (111.4 ± 2.64 mg TE/g extract), H 2 O 2 (IC 50 = 98.15 ± 0.68 µg/mL), and xanthine oxidase (IC 50 = 10.26 ± 0.6 µg/mL). With respect to hypoglycemic effect, the aqueous and ethanolic extracts showed IC 50 values of 119.7 ± 2.15 µg/mL and 102 ± 3.63 µg/mL for α-amylase and 121.4 ± 1.88 and 56.6 ± 1.12 µg/mL for α-glucosidase, respectively, and the extracts lowered blood glucose levels in normal starch-loaded rats. Additionally, lipase inhibition was observed with aqueous (IC 50 = 25.3 ± 1.53 µg/mL) and ethanolic (IC 50 = 13.7 ± 3.03 µg/mL) extracts. Conclusion: These findings show the antioxidant, hypoglycemic, and hyperlipidemic effects of E. resinifera extracts, which should be investigated further to validate their medicinal uses and their pharmaceutical applications.

        버섯 균사체 및 배양곡물의 혈당강하 효과

        혈당강하 효능을 갖는 버섯배양곡물을 개발하기 위하여 수종의 버섯균과 버섯배양곡물의 식후 혈당상승 억제 효과를 조사하였다. 균사체의 포도당 투과지연 효과를 측정한 결과 120분 동안 투석후 전체 포도당 중 13.1±3.6∼41.8±8.0%의 포도당이 투석막 내액에 잔존하는 것으로 나타나 대부분의 균사체가 pectin보다 포도당의 투석막 밖으로의 투과를 지연시키는 효과가 컸다. 당뇨쥐에 균사체를 투여한 후 혈당변화를 조사한 결과 맥아당 섭취후 30분 경과시 389.4±43.8∼426.3±49.4 ㎎/dL로 대조구의 535.3±78.6 ㎎/dL에 비해 상당히 낮은 값을 나타내어 버섯균사체의 식후 혈당상승 억제 효과가 우수한 것으로 나타났으며, 특히 Phellinus linteus와 Paecilomyces japonica의 효과가 우수하였다. Paecilomyces japonica를 배양한 버섯배양곡물을 당뇨쥐에 투여한 후 혈당변화를 조사한 결과 버섯 균사를 배양하지 않은 곡물에 비해 현저히 낮은 식후 혈당상승을 나타내어 버섯배양곡물이 우수한 혈당강하 효능을 나타냄을 알 수 있었고 특히 버섯배양 보리의 효과가 큰 것으로 나타났다. To develop basidiomycetes-fermented cereals with hypoglycemic property, inhibitory effects of basidiomycetes mycelia and basidiomycetes-fermented cereals on postprandial glucose were investigated. In vitro effect of basidiomycetes mycelia on retarding the membrane transport of glucose was compared with pectin. For basidiomycetes mycelia, 13.1±3.6∼41.8±8.0% of total glucose remained in inner solution of dialysis membrane after dialysis for 120 min, indicating that most of basidiomycetes mycelia might effectively retard membrane transport of glucose. Glucose tolerance of basidiomycetes mycelia and basidiomycetes-fermented cereals was tested on streptozotocin-induced diabetic rats administrated with maltose. Postprandial glucose levels of basidiomycetes mycelia, 389.4±43.8∼426.3±49.4 ㎎/dL, were considerably lower than that of control, 535.3±78.6 ㎎/dL, at 30 minutes after maltose administration. Namely, basidiomycetes mycelia showed better postprandial glucose lowering effect than pectin. Brown rice and barley fermented with Paecilomyces japonica showed much lower postprandial glucose level than raw brown rice and barley, especially hypoglycemic effect of barley fermented with Paecilomyces japonica was significant.

        Hypoglycemic Effect of Mushroom Fermented Milk in Streptozotocin-Induced Diabetic Rats

        버섯 추출물을 첨가하여 유산 발효시킨 버섯발효유와 이때 사용된 버섯 추출물 및 버섯발효유 상등액의 항당뇨 효과를 규명하고자 Sprague-Dawley 수컷에 streptozotocin을 50 mg/kg body weight씩 복강내 주사하여 당뇨를 유발시켜 검토하였다. 버섯발효유, 상등액 및 버섯 추출물을 식이 중에 10% (v/w)씩 동량 첨가 한 식이를 3주간 급여한 후 혈당치, 인슐린 농도 및 경구당부하실험을 실시하였다. 버섯발효유의 이화학적 성분을 분석 한 결과 단백질 2.87%, 지방 0.09%, 탄수화물 6.0%, 식이섬유 0.3%, 락토스 2.01%, 슈크로스 1.23% 및 칼슘과 철 성분을 각각 95.9 및 0.08 mg/l00 g함유하고 있었다. Streptozotocin-유발 대조군 당뇨쥐에 비해 버섯발효유 투여군에서 현저한 혈당강하 효과가 있었으며, 이러한 효과는 인슐린 농도증가에 의한 것으로 나타났다. 또한, 버섯발효유 상등액 및 버섯 추출물에도 혈당강하 효과가 있는 것으로 나타났다. 실험 종료시점에 실시한 경구당부하실험에서도 버섯발효유, 상등액 및 추출물 순으로 현저한 효과를 보였다. 이상의 결과로 볼 때 혈당강하 효과가 있는 버섯과 유산균을 접목한 버섯발효유 제조는 이들 상호간의 시너지 효과에 의해 당뇨성 흰쥐에서 현저한 항당뇨 효과를 발휘하였다. Nutritional concentrations by chemical analyses of mushroom fermented milk were protein 2.87%, fat 0.09%, carbohydrates 6.0%, dietary fiber 0.3%, lactose 2.01%, sucrose 1.23%, calcium 95.9 mg/100 g and iron 0.08 mg/100 g. The present study was undertaken to investigate the hypoglycemic effects of the equal volume of either water (streptozotocin (STZ)rontrol rats), mushrooms water-extract (STZ-extrart fed rats), mushroom fermented milk product (STZ-mushroom yogurt fed rats) or mushroom fermented milk supernatant (STZ-supernatant fed rats) (10%, v/w), in STZ-induced diabetic rats for 3 week period. The mushroom fermented milk given to the STZ-diabetic rats decreased the blood glucose significantly and increased the blood insulin, compared with the STZ-control rats. The supernatant and mushroom water extract also slightly retarded the development of hyperglycemia in the STZ-diabetic rats. Taken together the results, the mushroom yogurt may have a potential for the hypoglycemic effect in the STZ-diabetic rats.

        Hypoglycemic Effects of Glycosaminoglycan from Urechis unicinctus in Diabetic Mice

        To further utilize glycosaminoglycan from Urechis unicinctus, the hypoglycemic effect and possible mechanism of glycosaminoglycan on diabetic mice were evaluated. Diabetes was induced in mice by intraperitoneal injections of streptozotocin for 3 consecutive days and fed with high-sugar and high-lipid fodder. After diabetes was confirmed, the hypoglycemic effect of glycosaminoglycan from U. unicinctus was investigated in the diabetic mice. Results demonstrated that glycosaminoglycan could significantly decrease blood glucose concentrations, HOMA-IR, AUG, and liver MDA content in diabetic mice. In addition, it significantly enhanced liver SOD and GSH-Px activity, as well as liver GCK activity and hepatic glycogen levels. Glycosaminoglycan from U. unicinctus exhibited efficacy against diabetes, suggesting its potential use as a natural intervention against diabetes.
