        Bacillus subtilis로 제조한 검정콩 청국장의 항돌연변이 및 in Vitro 항암활성 증진효과

        Black soybean (Seoritae) chungkukjang was fermented with different kind of starters (Bacillus subtilis KCCM 11315, Bacillus licheniformis CN-115 and Bacillus megaterium SMY-212). Antimutagenic and in vitro anticancer effects of the black soybean chungkukjangs were studied by Ames test using Salmonella enterica serovar Typhimurium TA100 and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay on AGS human gastric adenocarcinoma cells and HT-29 human colon adenocarcinoma cells, respectively. The NH2-N and NH3-N contents in the black soybean chungkukjang fermented with Bacillus subtilis (BS-C) were similar levels in the soybean chungkukjang fermented with Bacillus licheniformis (BL-C). Methanol extracts from the black soybean chungkukjangs showed higher antimutagenicities against aflatoxin B1 (AFB1) and N-methyl-N'-nitro-N-nitrosoguanidine (MNNG). At the 2.5 mg/plate concentration, the BS-C exhibited 79% antimutagenicity against AFB1, while the black soybean chungkukjang fermented with Bacillus megaterium (BM-C) revealed 33% antimutagenicity. The BS-C and BL-C also increased antimutagenic effect against MNNG than BM-C. The BS-C and BL-C showed higher inhibitory effects on the growth of AGS human gastric adenocarcinoma cells than the BM-C at the same concentration. And the BS-C exhibited the highest inhibitory effects (76%) on the growth of HT-29 human colon adenocarcinoma cells among the fermented chungkukjangs (p<0.05). In this study, black soybean chungkukjang fermented with different kind of starters exerted differently on the antimutagenic and anticancer activities of the chungkukjangs. B. subtilis-fermented chungkukjang showed the highest chemopreventive effect compared to the chungkukjangs with other starters. (Cancer Prev Res 12, 120-127, 2007)

        Effects of hydrophilic compounds in purple rice husk on AFB1-induced mutagenesis

        Rice husk has been shown to possess cancer chemopreventive activity. This study focused on mutagenicity and antimutagenicity of the crude acidified methanolic extract of purple rice husk and its solvent- partitioned fractions using a Salmonella mutation assay and rat liver micronucleus test. The purple rice husk extracts were not mutagenic in S. typhimurium strain TA98, but were mutagenic in TA100 (crude extract and its hydrophilic fractions). There was no significant mutagenicity of purple rice husk extracts in rat liver. The rice husks possessed antimutagenicity against AFB1-induced mutagenesis in both bacterial and animal models. The dichloromethane-partitioned fraction (DPF) showed the highest antimutagenicity. Vanillic acid, a major phenolic compound found in DPF, exhibited antimutagenicity against AFB1 in both bacterial and rat models. The inhibitory mechanism was associated with the induction of detoxifying enzymes in AFB1 metabolism of rat liver. These findings suggested that vanillic acid may be a principal antimutagenic compound in purple rice husk.

        In vivo Antimutagenicity of Dadih Probiotic Bacteria towards Trp-P1

        In vitro acid- and bile-tolerant lactic acid bacteria isolated and identified from Indonesian traditional fermented milk dadih might be considered as potential probiotic strains after further characterization with animal models, especially for their therapeutic properties. Five dadih lactic bacteria isolates each had moderate survival rate for 2 h at pH 2.0, as well as bile tolerance. The aim of this research was to identify candidate probiotic lactic bacteria among indigenous dadih lactic isolates originated from Bukit Tinggi, West Sumatra, especially their in vivo antimutagenic property. Milk cultured with Enterococcus faecium IS-27526 significantly lowered fecal mutagenicity of rats as compared to the control group, skim milk, and milk cultured with L. plantarum IS-20506. These results suggest that Enterococcus faecium IS-27526 may serve as a potential probiotic strain with its antimutagenicity.

        연구논문 : 지유 에탄올 추출물의 식품 위해성 세균에 대한 항균 활성 및 Salmonella serotype Typhimurium TA100에 대한 항돌연변이 활성 효과

        This study was performed to analyze the antibacterial activity against food hazardous microorganisms and antimutagenic effects of Sanguisorba officinalis L. ethanol extracts on Salmonella Typhimurium TA100. The antibacterial activity was evaluated by paper disc diffusion assay, minimum inhibition concentration (MIC), and optical density of the culture with the ethanol extract for 24 hr. Antibacterial activity was tested with seven microorganisms including Escherichia coli, Escherichia coli O157:H7, Pseudomonas aeruginosa, Salmonella Typhimurium, Listeria monocytogenes, Bacillus cereus, and Staphylococcus aureus. The paper disc diffusion assay showed distinct clear inhibition zones around the discs treated with the extract for five microorganisms, except Escherichia coli and Escherichia coli O157:H7. MIC values were 0.625-2.5 mg/mL for these five strains that showed clear zones. The time-kill assay was consistent with the results from the paper disc diffusion assay and MIC test. Additionally, antimutagenicity of the extract was determined using the Ames test. The ethanol extract at 5 mg/plate inhibited 72.42% and 89.85% of mutagenicity induced by 4-nitroquinoline 1-oxide and sodium azide, respectively. These results demonstrate that the ethanol extract from S. officinalis L. has remarkable antibacterial activity and antimutagenicity.

        각종 변이원에 대한 쇠비름 추출물의 돌연변이 억제 효과

        This study was performed to determine the effects of antimutagenicity of Porturaca oleracea L. in Korea. In Ames test, the ethanol extract of Poturaca oleracea L. inhibited mutagenic activity of N-methyl N'-nitro N-nitrosoguanidine(MNNG), 4-nitroquinoline-l-oxide (4NQO), benzo(α)pyrene(B(α) P) and 3 amino-1.4-dimethyl-5H-pyrido-(4,3-b)indole (Trp-P-1) in Salmonella typhimurium TA98 and TA100. But hot-water extract Porturaca oleracea L. only inhibited mutagenic activity of MNNG in Saltmonella typhimurium TA100. On 4NQO, the ethanol extract 100~1,600㎍ /plate of Porturaca oleracea L. showed a slight inhibitory effect of 13~48%, 4~47% in TA98 and TA100, respectively, but on MNNG, it showed higher inhibitory effect of 6~86% in TA100. And the treatment of 1,600㎍/plate of ethanol extract of Porturaca oleracea L. had strong antimutagenicity with 74-87% inhibition against TA98 and TA100 induced by B(a)P and with 85~93% inhibition against TA98 and TA100 induced by Trp-P-1. The ethanol extract was fractionated with ether, chloroform, ethylacetate, butanol and water. Among them, most of the fraction except water fraction showed strong antimutagenicity effects against mutation induced by 4-NQO, MNNG, B(a)P and Trp-P-1. Chloroform fraction had strong antimutagenicity with 91% inhibition against TA100 induced by MNNG, diethyl etherfraction had strong antimutagenicity with 92%, 98% inhibition against TA98 and TAI00 induced by 4NQO, Chloroform fraction had strong antimutagenicity with 97% inhibition against TA100 induced by B(a)P and diethyl etherfraction had strong antimutagenicity with 98% inhibition against both strain induced by Trp-P-1, respectively.

        Ames 실험계에서 어성초 물분획물의 항돌연변이 효과

        The antimutagenic effects of methanol extracts of sun dried and freeze dried Houttuynia cordata (HC) using Salmonella typhimurium TA100 in the Ames test system were evaluated. The methanol extract of freeze dried HC showed higher antimutagenic effect against aflatoxin B1 (AFB1) than that of sun-dried HC by approx. 10%. Whereas, the inhibitory activity of the methanol extracts of the HC on N-methyl- N'-nitro-N-nitrosoguanidine (MNNG) was somewhat lower compared to AFB1. The separated fractions from the methanol extract showed strong antimutagenic activities both in chloroform and aqueous fractions against AFB1. However, in case of MNNG, the aqueous fraction exhibited antimutagenic effect while the chloroform fraction did not show the antimutagenic activities. From this results, the lipid soluble (chloroform) fraction from the methanol extract of HC exhibited strong antimutagenic effect on AFB1 which is an indirect mutagen, but not on MNNG which is a direct mutagen, however, the aqueous fraction from the methanol extract of HC showed strong antimutagenic effects both on AFB1 and MNNG. The aqueous fraction of HC which generally be prepared as a folk medicine or tea proved to have the antimutagenic activity in this study. (Cancer Prev Res 10, 67-72, 2005)

        Antimutagenic Effects of Lycopene and Tomato Purée

        Several health benefits, including protection from tumors at various anatomic sites, such as the lungs, stomach, and prostate gland, have been attributed to tomatoes and tomato-based products. Among tomato carotenoids, lycopene is the most active antioxidant, although it has many other biological effects, but data on its antimutagenic effects are scarce and often discrepant. The aim of our work was to determine the protective effects of lycopene, with regard to mutagenicity, via two indirect mutagens/carcinogens—2-amino-3-methylimidazo[4,5-f]quinoline (IQ) and aflatoxin B1 (AFB1) and the direct mutagen/carcinogen N-nitroso-N-methylurea (MNU)—using the Ames and micronucleus tests. The significant, dose-dependent, antimutagenic effects of two concentrations of lycopene (30μg and 300μg per plate) were demonstrated at various concentrations of both AFB1 and IQ in two strains of Salmonella typhimurium (TA98 and TA100). The protective effects of lycopene relative to MNU were lower in comparison to its protective effects relative to AFB1 and IQ. Mice treated for 3 days with different doses of lycopene (either 25 or 50mg/kg of body weight) prior to administration of individual mutagens resulted in a significant reduction of micronuclei numbers in the micronucleus test. Tomato purée (tested using the Ames test and AFB1) revealed a much stronger, dose-dependent, antimutagenic effect compared with corresponding doses of pure lycopene. Results indicate that lycopene has antimutagenic effects, although the effects are lower than that of tomato purée, which contains a complex mixture of bioactive phytochemicals. The antimutagenic effect is connected with the chemoprotective role of lycopene, tomatoes, and tomato products in the prevention of carcinogenesis.

        Sodium Azide와 2-AF에 대한 마(Dioscorea batatas Decene)의 항변이원성

        The objective of this study was to screen the antimutagenicity of yam enzymatic browning reaction product(YEBRP), mucopolysaccharide and dietary fiber from yam to the mutagen of sodium azide and 2-aminoflourene(2-AF). Antimutagenicity of YEBRP on the mutagenicity of sodium azide showed no difference compared to control without YEBRP but that of 2-AF was high in all substrate.(P<0.01) On the mutagenicity of sodium azide and 2-AF, antimutagenicity of mucopolysaccharide and dietary fiber were high (p<0.01) in α-cellulose and hemicellulose. Antimutagenicity of α-cellulose on the mutagenicity of 2-AF was high at 5 hours reaction time but that was decreased as the reaction time increased.

        Antimutagenic and Cancer Cell Growth Inhibitory Effects of Seaweeds

        The antimutagenic and cancer cell growth inhibitory effects of methanol extracts from 9 kinds of seaweed were studied in the Ames assay and cell culture systems, respectively. The methanol extracts from the seaweeds of sea lettuce, chlorella, sea tangle, sea mustard, sporophyll of sea mustard, fusiforme, seaweed papulosa, purple laver and ceylon moss showed antimutagenicities against aflatoxin B₁(AFB₁) and N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) in the Salmonella typhimurium TA100. These extracts revealed relatively higher antimutagenicity against AFB₁(indirect mutagen) than MNNG(direct mutagen). Sporophyll of sea mustard and seaweed papulosa exhibited strong antimutagenic activity against AFB₁, and sporophyll of sea mustard, sea tangle and ceylon moss also reduced the mutagenicity induced by MNNG. The sporophyll fo sea mustard exerted the highest antimutagenic activity among the samples treated. The methanol extracts from 9 kinds of seaweed inhibited the growth of two cancer cell lines, AGS human gastric adenocarcinoma cells and HT-29 human colon carcinoma cells. Sea tangle, sea mustard and sporophyll of sea mustard inhibited the growth of cancer cells significantly. These results suggest that various seaweeds show not only antimutagenic activity but also growth inhibitory effect of some cancer cells.
