        Cytokine-cytokine receptor interactions in the highly pathogenic avian influenza H5N1 virus-infected lungs of genetically disparate Ri chicken lines

        Objective: The highly pathogenic avian influenza virus (HPAIV) is a threat to the poultry industry as well as the economy and remains a potential source of pandemic infection in humans. Antiviral genes are considered a potential factor for HPAIV resistance. Therefore, in this study, we investigated gene expression related to cytokine-cytokine receptor interactions by comparing resistant and susceptible Ri chicken lines for avian influenza virus infection. Methods: Ri chickens of resistant (Mx/A; BF2/B21) and susceptible (Mx/G; BF2/B13) lines were selected by genotyping the Mx dynamin like GTPase (Mx) and major histocompatibility complex class I antigen BF2 genes. These chickens were then infected with influenza A virus subtype H5N1, and their lung tissues were collected for RNA sequencing. Results: In total, 972 differentially expressed genes (DEGs) were observed between resistant and susceptible Ri chickens, according to the gene ontology and Kyoto encyclopedia of genes and genomes pathways. In particular, DEGs associated with cytokine-cytokine receptor interactions were most abundant. The expression levels of cytokines (interleukin-1β [IL-1β], IL-6, IL-8, and IL-18), chemokines (C-C Motif chemokine ligand 4 [CCL4] and CCL17), interferons (IFN-γ), and IFN-stimulated genes (Mx1, CCL19, 2’-5’-oligoadenylate synthaselike, and protein kinase R) were higher in H5N1-resistant chickens than in H5N1-susceptible chickens. Conclusion: Resistant chickens show stronger immune responses and antiviral activity (cytokines, chemokines, and IFN-stimulated genes) than those of susceptible chickens against HPAIV infection.         Nutritional and Tissue Specificity of IGF-I and IGFBP-2 Gene Expression in Growing Chickens - A Review -

        Nutritional regulation of gene expression associated with growth and feeding behavior in avian species can become an important technique to improve poultry production according to the supply of nutrients in the diet. Insulin-like growth factor-I (IGF-I) found in chickens has been characterized to be a 70 amino acid polypeptide and plays an important role in growth and metabolism. Although it is been well known that IGF-I is highly associated with embryonic development and post-hatching growth, changes in the distribution of IGF-I gene expression throughout early- to late-embryogenesis have not been studied so far. We revealed that the developmental pattern of IGF-I gene expression during embryogenesis differed among various tissues. No bands of IGF-I mRNA were detected in embryonic liver at 7 days of incubation, and thereafter the amount of hepatic IGF-I mRNA was increased from 14 to 20 days of incubation. In eyes, a peak in IGF-I mRNA levels occurred at mid-embryogenesis, but by contrast, IGF-I mRNA was barely detectable in the heart throughout all incubation periods. In the muscle, no significant difference in IGF-I gene expression was observed during different stages of embryogenesis. After hatching, hepatic IGF-I gene expression as well as plasma IGF-I concentration increases rapidly with age, reaches a peak before sexual maturity, and then declines. The IGF-I gene expression is very sensitive to changes in nutritional conditions. Food-restriction and fasting decreased hepatic IGF-I gene expression and refeeding restored IGF-I gene expression to the level of fed chickens. Dietary protein is also a very strong factor in changing hepatic IGF-I gene expression. Refeeding with dietary protein alone successfully restored hepatic IGF-I gene expression of fasted chickens to the level of fed controls. In most circumstances, IGF-I makes a complex with specific high-affinity IGF-binding proteins (IGFBPs). So far, four different IGFBPs have been identified in avian species and the major IGFBP in chicken plasma has been reported to be IGFBP-2. We studied the relationship between nutritional status and IGFBP-2 gene expression in various tissues of young chickens. In the liver of fed chickens, almost no IGFBP-2 mRNA was detected. However, fasting markedly increased hepatic IGFBP-2 gene expression, and the level was reduced after refeeding. In the gizzard of well-fed young chickens, IGFBP-2 gene expression was detected and fasting significantly elevated gizzard IGFBP-2 mRNA levels to about double that of fed controls. After refeeding, gizzard IGFBP-2 gene expression decreased similar to hepatic IGFBP-2 gene expression. In the brain, IGFBP-2 mRNA was observed in fed chickens and had significantly decreased by fasting. In the kidney, IGFBP-2 gene expression was observed but not influenced by fasting and refeeding. Recently, we have demonstrated in vivo that gizzard and hepatic IGFBP-2 gene expression in fasted chickens was rapidly reduced by intravenous administration of insulin, as indicated that in young chickens the reduction in gizzard and hepatic IGFBP-2 gene expression in vivo stimulated by malnutrition may be, in part, regulated by means of the increase in plasma insulin concentration via an insulin-response element. The influence of dietary protein source (isolated soybean protein vs. casein) and the supplementation of essential amino acids on gizzard IGFBP-2 gene expression was examined. In both soybean protein and casein diet groups, the deficiency of essential amino acids stimulated chickens to increase gizzard IGFBP-2 gene expression. Although amino acid supplementation of a soybean protein diet significantly decreased gizzard IGFBP-2 mRNA levels, a similar reduction was not observed in chickens fed a casein diet supplemented with amino acids.         Analysis of Consumers` Preferences and Price Sen-sitivity to Native Chickens

        This study analyzed consumers` preferences and price sensitivity to native chickens. A survey was conducted from Jan 6 to 17, 2014, and data were collected from consumers (n=500) liv-ing in Korea. Statistical analyses evaluated the consumption patterns of native chickens, pref-erence marketing for native chicken breeds which will be newly developed, and price sensitivity measurement (PSM). Of the subjects who preferred broilers, 24.3% do not pur-chase native chickens because of the dryness and tough texture, while those who preferred native chickens liked their chewy texture (38.2%). Of the total subjects, 38.2% preferred fried native chickens (38.2%) for processed food, 38.4% preferred direct sales for native chicken distribution, 51.0% preferred native chickens to be slaughtered in specialty stores, and 32.4% wanted easy access to native chickens. Additionally, the price stress range (PSR) was 50 won and the point of marginal cheapness (PMC) and point of marginal expensiveness (PME) were 6,980 won and 12,300 won, respectively. Evaluation of the segmentation market revealed that consumers who prefer broiler to native chicken breeds were more sensitive to the chicken price. To accelerate the consumption of newly developed native chicken meat, it is necessary to develop a texture that each consumer needs, to increase the accessibility of native chickens, and to have diverse menus and recipes as well as reasonable pricing for native chickens.

        Effect of Bacillus Subtilis-based Direct-fed Microbials on Immune Status in Broiler Chickens Raised on Fresh or Used Litter

        Type of dietary direct-fed microbials (DFMs) or poultry litter could directly influence the composition of gut microbiota. Gut microbiota plays an important role in shaping the developing immune system and maintaining the homeostasis of the mature immune system in mammal and chickens. The present study was carried out to investigate the interaction among litter, DFMs and immunity in broiler chickens exposed to a field-simulated environment. Immune status of broiler chickens was assessed by serum antibodies against Eimeria spp. and Clostridium spp. and intestinal cytokine mRNA expression. The current experimental design had a $3{\times}2$ factorial arrangement of treatments with three types of litter, i.e., fresh litter or used litter that was obtained from a farm with no disease outbreak (used litter) or a farm with history of a gangrenous dermatitis outbreak (GD litter), and two dietary treatments with or without DFMs. It was found that either DFM addition or type of litter significantly affected anticoccidial antibody levels of broiler chickens at d 42. In general, dietary DFMs increased the anticoccidial antibodies in the fresh-litter raised chickens, but lowered the levels in the GD-litter raised chickens. Serum antibodies against Clostridium perfringens ${\alpha}$-toxin were significantly (p<0.05) higher in chickens raised on GD litter compared with those raised on fresh litter. Cytokine mRNA expression was significantly (p<0.05) altered by either the type of litter or DFMs. Of interest, dietary DFMs lowered interferon-${\gamma}$, interleukin 1beta, and CXCLi2 cytokine mRNA expression in chickens raised on fresh litter but increased them in GD-litter raised chickens. In conclusion, dietary DFMs modulate various immune parameters of broiler chickens, but the DFM-mediated effects were dependent upon the type of litter on which chickens were raised.

        토종실용닭의 암수 간 산육 능력 및 스트레스 반응 정도 비교

        This study compared the production and physiological characteristics of male and female Korean native commercial chickens. We investigated the growth performance, vitality, and stress response of 479 male and 608 female chickens from 1 d to 12 weeks of age. The body weight, feed consumption and feed conversion ratio were measured as growth performance. The survival rate, amount of telomeric DNA, DNA damage rate, heterophil-lymphocyte ratio (H/L ratio), and heat shock protein (HSP)-70, HSP-90α, and HSP-90β gene expression levels were analyzed as indicators of vitality and stress response. Body weight was significantly higher in male chickens than in female chickens after 2 weeks of age (P<0.01). Feed intake was higher in male chickens than in female chickens, whereas the feed conversion ratio showed the opposite trend. The survival rate was significantly higher in female chickens than in male chickens (82.8% vs. 73.8%, P<0.05). Stress response analysis revealed no differences between male and female chickens in terms of telomeric DNA content, DNA damage rate, H/L ratio, and HSP gene expression levels. Taken together, it was concluded that there was a significant difference in the growth performance and survival rate between male and female Korean native commercial chickens, whereas there was no difference in the degree of stress response between them. Therefore, in terms of the productivity of Korean native commercial chickens, it is judged that separate-sex rearing is much more advantageous than mixed rearing, regardless of the physiological characteristics of males and females. 본 연구는 토종실용닭 암수 간의 생산 특성 및 생리적 특성을 비교하고자 이들의 산육 능력, 강건성 및 스트레스 반응 정도를 살펴보았다. 시험은 토종실용닭 암컷 608수와 수컷 479수를 대상으로 1일령부터 12주간 실시하였다. 산육 능력의 평가는 체중, 사료 섭취량, 사료요구율을 조사하였고, 강건성 및 스트레스 반응 정도의 측정은 생존율, 텔로미어 함유율, DNA 손상률, heterophil-lymphocyte 비율, HSP-70, HSP-90α 및 HSP-90β 유전자 발현율을 분석하였다. 시험 결과, 암수 간의 체중은 2주령부터 시험 종료 시까지 수컷이 암컷보다 유의하게 높았고(P<0.01), 사료 이용성은 시험 전 기간 수컷이 암컷보다 많은 사료를 섭취한 반면 낮은 사료요구율을 보였다. 시험 전 기간 생존율은 암컷이 82.8%, 수컷이 73.8%로 암컷이 수컷보다 유의하게 높았다(P<0.05). 스트레스 반응 지표 분석 결과, 텔로미어 함유율, DNA 손상률, heterophil-lymphocyte 비율 및 HSP 유전자 발현율 모두에서 암컷과 수컷 간의 차이는 없는 것으로 나타났다. 이상의 결과를 종합할 때, 토종실용닭 암수 간의 산육 능력과 생존율의 차이는 있으나 스트레스 반응 정도의 차이는 없는 것으로 판단된다. 따라서 토종실용닭의 생산성 측면에서 암수의 생리적 특성과 무관하게 암컷과 수컷을 분리하여 사육하는 것이 바람직한 것으로 사료된다.

        Effects of Water Restriction on the Growth Performance, Carcass Characteristics and Organ Weights of Naked Neck and Ovambo Chickens of Southern Africa

        In semi-arid areas of Southern Africa, dehydration can compromise the performance and welfare of local chickens, particularly during the growing period when confinement is curtailed and birds are left to scavenge for feed and water. The effect of water restriction on the growth performance was compared in Naked Neck (NNK) and Ovambo (OVB) chickens that are predominant in Southern Africa. A total of 54 eight-wk-old pullets each of NNK and OVB chickens with an initial average weight of $641{\pm}10g/bird$ were randomly assigned to three water intake treatments, each having six birds for 8 wk. The water restriction treatments were ad libitum, 70% of ad libitum and 40% of ad libitum intake. Nine experimental pens with a floor space of $3.3m^2$ per strain were used. The pens were housed in an open-sided house with cement floor deep littered with a 20 cm layer of untreated wood shavings. Feed was provided ad libitum. Average daily water intake (ADWI), BW at 16 weeks of age (FBW), ADG, ADFI, feed conversion ratio (FCR) and water to feed ratios (WFR) were determined. Ovambo chickens had superior (p<0.05) FBW, ADG and ADWI than NNK chickens. Body weight of birds at 16 weeks of age, ADG, ADFI, ADWI, and WFR declined progressively (p<0.05) with increasing severity of water restriction while FCR values increased (p<0.05) as the severity of water restriction increased. Naked Neck chickens had better FCR at the 40% of ad libitum water intake level than Ovambo chickens. The dressing percentage per bird was higher in water restricted birds than those on ad libitum water consumption, irrespective of strain. Heart weight was significantly lower in birds on 40% of ad libitum water intake than those on ad libitum and 70% of ad libitum water intake, respectively. In conclusion, NNK chickens performed better than OVB chickens under conditions of water restriction and would be ideal to raise for meat and egg production in locations where water shortages are a major challenge.

        Effect of feeding fermented soybean meal on broiler chickens’ performance: a meta-analysis

        Objective: The present study aimed to quantify the effects of fermented soybean meal (FSBM) on broiler chickens’ performance by employing a meta-analysis approach. Methods: A total of 16 studies were included in the database after being systematically selected using a PRISMA protocol. Hedges’ g effect size was used to quantify pooled standardized mean difference (SMD) using random-effects models at 95% confidence intervals (95% CI). Publication bias among studies was computed with Egger’s test and visualized using funnel plots. Results: Results indicated that dietary FSBM inclusion increased final body weight (BW) (SMD = 0.586, 95% CI: 0.221 to 0.951, p = 0.002) of broiler chickens, particularly in starter period (SMD = 0.691, 95% CL: 0.149 to 1.233, p = 0.013) while in the finisher period, the effect was weaker (SMD = 0.509, 95% CI: 0.015 to 1.004, p = 0.043). Average daily gain (ADG), feed intake (FI), and feed conversion ratio (FCR) were not affected with FSBM inclusion when compared to control. Subgroup analysis revealed that FI increased in starter period (SMD = 0.582, 95% CI: 0.037 to 1.128, p = 0.036). When considering types of microorganism as moderating variables in the subgroup analysis, we found that Aspergillus oryzae, mixed probiotics+bromelain protease, Bacillus subtilis, and Lactobacillus bacteria significantly increased ADG and FI (p<0.01). Additionally, either Bacillus subtilis+protease or Bacillus subtilis alone decreased FCR (p<0.001). However, meta-regression analysis showed that levels of FSBM inclusion had no effects on final BW (p = 0.502), ADG (p = 0.588), FI (p = 0.861), and FCR (p = 0.462). Conclusion: Substituting SBM in broiler chickens’ diet with FSBM improved BW of broiler chickens, especially in the starter period whereas the effects on ADG, FI, and FCR were mostly dependent on microbial strains used for fermentation.         Indigenous chicken production in Fiji Islands: knowledge, constraints and opportunities

        Objective: The objective of the study was to understand and document socio-economic characteristics, production parameters, challenges and management practices used by Fijian households which keep indigenous chickens. Methods: A survey involving 200 households was carried out in coastal and inland communities of Fiji's wet and semi-dry ecoregions. Data on the influence of ecoregion and location of households relative to the sea on management practices, challenges and productivity of indigenous chickens were analyzed using logistic regression and general linear model of SAS software. Results: Irrespective of location relative to the sea and ecoregion, households indicated that they kept indigenous chickens for food and income generation. The Welsummer was the most (p>0.05) preferred breed. Households in the semi-dry inland communities had the largest (p<0.05) flocks compared to those in semi-dry coastal communities and the wet region. Chickens in the semi-dry region performed better (p<0.05) than those in the wet region in terms of number of clutches per year and mature live weight. Predators and feed shortages were the biggest challenges faced by households in all areas. The mongoose was ranked as the most (p>0.05) common predator followed by domestic dogs. Most households in the wet ecoregion's coastal communities housed their chickens at night, whereas communities in semi-dry ecoregion housed their chickens most of the time (p<0.05). In all regions, no households sold their chickens to commercial markets (p>0.05). Households in semi-dry ecoregion were more likely (p>0.05) to sell their chickens at the local market place. Conclusion: The productivity of local chickens in Fiji is low because of feed shortage, predators such as the mongoose and lack of market linkages.

        Isolation and characterization of avian metapneumovirus from chickens in Korea

        Avian metapneumovirus (aMPV) causes upper respiratory tract infections in chickens and turkeys. Although the swollen head syndrome (SHS) associated with aMPV in chickens has been reported in Korea since 1992, this is the study isolating aMPV from chickens in this country. We examined 780 oropharyngeal swab or nasal turbinate samples collected from 130 chicken flocks to investigate the prevalence of aMPV and to isolate aMPV from chickens from 2004-2008. Twelve aMPV subtype A and 13 subtype B strains were detected from clinical samples by the aMPV subtype A and B multiplex real-time reverse transcription polymerase chain reaction (RRT-PCR). Partial sequence analysis of the G glycoprotein gene confirmed that the detected aMPVs belonged to subtypes A and B. Two aMPVs subtype A out of the 25 detected aMPVs were isolated by Vero cell passage. In animal experiments with an aMPV isolate, viral RNA was detected in nasal discharge, although no clinical signs of SHS were observed in chickens. In contrast to chickens, turkeys showed severe nasal discharge and a relatively higher titer of viral excretion than chickens. Here, we reveal the co-circulation of aMPV subtypes A and B, and isolate aMPVs from chicken flocks in Korea.

        병아리의 성 감별은 양계 산업에 매우 중요한 요소이다. 현재, 산업적으로 병아리의 성 감별은 반성유전형질을 이용한 깃털감별법이 가장 널리 이용되고 있다. 그러나 깃털 감별로 생산된 병아리는 암컷은 모두 조우성, 수컷은 모두 만우성이므로 깃털의 조만성이 생산능력에 미치는 영향을 아는 것이 매우 중요하다. 따라서 본 연구에서는 실용 토종닭을 대상으로 조우성 닭과 만우성 닭의 생산능력을 비교 분석하고자 하였다. 분석 결과, 발생 후 12주령까지의 생존율에서 조우성 닭이 만우성 닭에 비해 유의하게 높은 생존율을 보이고 특히 암컷에서 이의 차이가 두드러졌다(P<0.05). 체중은 거의 모든 주령에서 조우성 닭과 만우성 닭 간의 차이는 없는 것으로 나타났다. 사료 섭취량 및 사료요구율에 있어서도 모든 조사 기간내 두 집단 간의 차이가 없는 것으로 나타났다. 결론적으로 실용 토종닭의 경우 조우성 닭이 만우성 닭에 비해 생존율은 우수하나 산육 능력 및 사료 이용성에는 차이가 없는 것으로 보여진다. 이는 토종닭의 깃털 감별 종계 조성과 생산체계를 구축함에 있어 깃털 조만성에 의해 생산능력이 영향을 받지 않음을 시사하는 것으로써 산업적으로 깃털 감별을 실용화하는데 매우 바람직한 결과로 사료된다. Sex identification in day-old chicks is essential in the poultry industry. Currently, the feather-sexing using a sex-linked inheritance is a chick sexing method used extensively in the poultry industry. For chicks produced by feather-sexing, all females are early-feathering (EF) chicks and all males are late-feathering (LF) chicks. Therefore, investigating the effects of EF and LF phenotypes on production traits of chickens is critical. The purpose of this study was to analyze and compare the production performance between EF and LF chickens in Korean native commercial chickens. The results showed that the survival rate of the EF chickens was significantly higher than that of the LF chickens, from hatching to 12 weeks of age (P<0.05), with the highest difference observed in females. However, no significant difference was observed in body weight between the EF and LF chickens at almost all ages. In addition, no significant difference was observed between in feed utility, such as average daily feed intake and feed conversion ratio, between the EF and LF groups. In conclusion, although the survival rate of early-feathering chickens was superior to that of late-feathering chickens, no significant difference was observed in growth performance and feed utility among Korean native chickens. The results suggest that production capacity is not influenced by feathering type in the establishment of a breeder structure and production system for feather-sexing Korean native chickens, which is valuable for the practical application of feather-sexing in the poultry industry.
