        ISG15: leading a double life as a secreted molecule

        ISG15 is a well-known intracellular ubiquitin-like molecule involved in ISGylation. However, a recent study has revived the notion first put forward two decades ago that ISG15 is also a secreted molecule. Human neutrophils, monocytes and lymphocytes can release ISG15, even though this protein has no detectable signal peptide sequence. ISG15 has also been found in the secretory granules of granulocytes. The mechanism underlying ISG15 secretion is unknown. Secreted ISG15 acts on at least T and natural killer (NK) lymphocytes, in which it induces interferon (IFN)-c production. However, the mechanism by which ISG15 stimulates these cells also remains unclear. ISG15 and IFN-c seem to define an innate circuit that operates preferentially, but not exclusively, between granulocytes and NK cells. Inherited ISG15 deficiency is associated with severe mycobacterial disease in both mice and humans. This infectious phenotype probably results from the lack of secreted ISG15,because patients and mice with other inborn errors of IFN-c immunity also display mycobacterial diseases. In addition to raising mechanistic issues, the studies described here pave the way for clinical studies of various aspects, ranging from the use of recombinant ISG15 in patients with infectious diseases to the use of ISG15-blocking agents in patients with inflammatory diseases.

        A major daidzin metabolite 7,8,4′-trihydroxyisoflavone found in the plasma of soybean extract-fed rats attenuates monocyte-endothelial cell adhesion

        <P><B>Abstract</B></P> <P>Among many functional foods and their phytochemicals, ingestion of soybean (<I>Glycine max</I>) is highly correlated to reduced risk of cardiovascular diseases. Validation of potential health benefits of functional foods requires information about the bioavailability and metabolism of bioactive compounds. In this context, several phase I and II metabolites of isoflavones were target-analyzed in the plasma of rats acutely supplemented with soybean embryo extract. A daidzein metabolite, 7,8,4′-trihydroxyisoflavone (7,8,4′-THI), was found to have the highest average area under curve value (574.3±112.8). Therefore, its potential prevention effect on atherosclerosis was investigated using monocyte-endothelial cell adhesion assay. Different from its precursor daidzein or daidzin, 7,8,4′-THI attenuated adhesion of THP-1 monocytes to tumor necrosis factor-alpha (TNF-α) stimulated human umbilical vein endothelial cells (HUVECs). In addition, 7,8,4′-THI significantly downregulated TNF-α stimulated the expression of vascular cell adhesion molecule-1 and monocyte chemotactic protein-1 and phosphorylation of IκB kinase and IκBα involved in the initiation of atherosclerosis in HUVECs. Therefore, 7,8,4′-THI, a highly bioavailable hydroxylated isoflavone metabolite, has potential anti-atherosclerotic effect via inhibiting monocyte-endothelial adhesion.</P> <P><B>Highlights</B></P> <P> <UL> <LI> 7,8,4′-Trihydroxyisoflavone was found in the rat plasma after soybean intake. </LI> <LI> The highest average AUC of 7,8,4′-THI was 10 times higher than its precursor daidzein. </LI> <LI> 7,8,4′-THI inhibited TNF-α stimulated monocyte-endothelial cell adhesion. </LI> </UL> </P>

      • Comprehensive phenolic composition analysis and evaluation of Yak-Kong soybean (<i>Glycine max</i>) for the prevention of atherosclerosis

        <P><B>Abstract</B></P> <P>Yak-Kong (YK) (<I>Glycine max</I>), a small black soybean cultivar with a green embryo, was evaluated for functional constituents with a focus on atherosclerosis prevention. In comparison to common yellow and black soybean cultivars, YK contains significantly higher concentrations of antioxidants, particularly in its seed coat. A comprehensive phenolic composition analysis revealed that proanthocyanidins were the major phenolic group in YK. In contrast to other proanthocyanidin-rich foods, YK was rich in bioavailable proanthocyanidins (with a degree of polymerization ≤3) specifically with A-type dimers. Significant concentrations of phloridzin and coumestrol were also exclusively found in YK seed coat and the embryo, respectively. Extracts of both the proanthocyanidin-rich seed coat and isoflavonoid-rich embryo of YK attenuated adhesion of THP-1 to LPS-stimulated human umbilical vascular endothelial cells, suggesting that they are important sources of coronary heart disease-preventive phenolics.         Analysis of the variability of deflection of a prestressed composite bridge deck

        Nearly 400 composite railway bridge decks of a new kind belonging to the trough type with Ushaped cross section have been constructed in Belgium over the last fifteen years. The construction of these bridge decks is rather complex with the preflexion of precambered steel girders, the prestressing of a concrete slab and the addition of a 2nd phase concrete. Until now, they have been designed with a classical computation method using a pseudo-elastic analysis with modular ratios. Globally, they perform according to the expectations but variability has been observed between the measured and the computed camber of these bridge decks just after the transfer of prestressing and also at long-term. A statistical analysis of the variability of the relative difference between the measured camber and the computed camber is made for a sample of 36 bridge decks using no less than 10 variables. The most significant variables to explain this variability at prestressing are the ratio between the maximum tensile stress reached in the steel girders during the preflexion and the yield strength and the type of steel girder. For the same sample, the long-term camber under permanent loading is computed by two methods and compared with measurements taken one or two years after the construction. The camber computed by the step-by-step method shows a better agreement with the measured camber than the camber computed by the classical method. The purpose of the paper is to report on the statistical analysis which was used to determine the most significant parameters to consider in the modeling in order to improve the prediction of the behaviour of these composite railway bridge decks.

        Utility of FDG-PET/CT for the Detection and Characterization of Sternal Wound Infection Following Sternotomy

        Purpose FDG-PET/CT has the potential to play an important role in the diagnosis of sternal wound infections (SWI). The purpose of this study was to analyze the diagnostic accuracy of FDG-PET/CT for SWI in patients following sternotomy. Methods We performed a single-center, retrospective analysis of patients who had undergone median sternotomy and FDG-PET/ CT imaging. The gold standard consisted of positive bacterial culture and/or the presence of purulent material at surgery. Qualitative patterns of sternal FDG uptake, SUVmax, and associated CT findings were determined, and an imaging scoring system was developed. The diagnostic performances were studied in both the recent (≤ 6 months between sternotomy and imaging) and remote surgery phase (> 6 months). Results A total of 40 subjects were identified with 11 confirmed SWI cases. Consensus interpretation was associated with a sensitivity of 91%and specificity of 97%. Combination of uptake patterns yielded an AUC of 0.96 while use of SUVmax yielded an AUC of 0.82. Conclusions Results suggest that FDG-PET/CT may be useful for the diagnosis of SWI with optimal diagnostic accuracy achieved by identifying specific patterns of uptake. SUVmax can be helpful in assessing subjects with remote surgery, but its use is limited in the context of recent surgery. Further studies are required to confirm these results.

        Accompanying Learning in Dance Classes in Quebec Schools through Movement Observation-Analysis (MOA)

        As a school discipline in Quebec, dance is heir to the Modern Educational Dance (MED) movement, founded by Rudolf Laban (1948, 1976), better known in Quebec as creative dance (Raymond, 2014, p. 22). About ten years ago, Movement Observation-Analysis (MOA) (Harbonnier, Dussault, Ferri, 2021) was introduced into the artistic and pedagogical training of school dance teachers in Quebec. With the aim of revising ministerial programs for teaching dance in Quebec schools, which are over twenty years old, we wondered how MOA concepts were useful to dance teachers in Quebec schools. To this end, we asked ten school dance teachers to describe their use of OAM in their teaching, using the technique of the explicitation interview (Vermersch, 1994). With regard to the pedagogical function of school dance teachers, the results of our research, through the case study of teacher Suzie, show that MOA provides an enriched and clarified conceptual framework and lexicon to refine movement observation activities (diagnostic function), improve pedagogical communication (physical demonstration and verbal instructions), and provide knowledge about movement to share with students, fostering their autonomy in learning (learning support function).

        SiRENE: A new generation of engineering simulator for real-time simulators at EDF

        For Safety Assisted Engineering works, real-time simulators have emerged as a mandatory tool among all the key actors involved in the nuclear industry (utilities, designers and safety authorities). EDF, Electricit´e de France, as the leading worldwide nuclear power plant operator, has a crucial need for efficient and updated simulation tools for training, operating and safety analysis support. This paper will present the work performed at EDF/DT to develop a new generation of engineering simulator to fulfil these tasks. The project is called SiRENE, which is the acronym of Re-hosted Engineering Simulator in French. The project has been economically challenging. Therefore, to benefit from existing tools and experience, the SiRENE project combines: - A part of the process issued from the operating fleet training full-scope simulator. - An improvement of the simulator prediction reliability with the integration of High-Fidelity models, used in Safety Analysis. These High-Fidelity models address Nuclear Steam Supply System code, with CATHARE thermalhydraulics system code and neutronics, with COCCINELLE code. - And taking advantage of the last generation and improvements of instructor station. The intensive and challenging uses of the new SiRENE engineering simulator are also discussed. The SiRENE simulator has to address different topics such as verification and validation of operating procedures, identification of safety paths, tests of I&C developments or modifications, tests on hydraulics system components (pump, valve etc.), support studies for Probabilistic Safety Analysis (PSA). etc. It also emerges that SiRENE simulator is a valuable tool for self-training of the newcomers in EDF nuclear engineering centers. As a modifiable tool and thanks to a skillful team managing the SiRENE project, specific and adapted modifications can be taken into account very quickly, in order to provide the best answers for our users’ specific issues. Finally, the SiRENE simulator, and the associated configurations, has been distributed among the different engineering centers at EDF (DT in Lyon, DIPDE in Marseille and CNEPE in Tours). This distribution highlights a strong synergy and complementarity of the different engineering institutes at EDF, working together for a safer and a more profitable operating fleet.
