        Suppression of long non-coding RNA PCAT19 inhibits glioma cell proliferation and invasion, and increases cell apoptosis through regulation of MELK targeted by miR-142-5p

        Background Glioma has the chief type of primary brain tumors worldwide. The glioma may be controlled by regulatorsincluding some lncRNAs, miRNAs, and proteins. Objective Our study aims to discover the underlying mechanism for lncPCAT19/miR-142-5p/MELK axis in gliomaprogression. Methods The clinical samples were from patients with gliomas in our Hospital. Hematoxylin–eosin staining (H&E) wasapplied to determine the clinical pathological changes. Real time PCR was performed to measure the levels of lncPCAT19,miR-142-5p, MELK, and expression of other genes. Western blot was conducted to detect the protein level of MELK. RIPassay was performed to analyze the interaction between lncPCAT19 and miR-142-5p, and dual-luciferase reporter assay wasused to determine the binding site between lncPCAT19 and miR-142-5p. CCK-8, colony formation assay, flow cytometry,and trans-well assay were carried out to confirm cell proliferation, colony formation, apoptosis, and invasion, respectively. Results LncPCAT19 was increased in cancer tissues. Then, lncPCAT19 could interact with and down-regulate miR-142-5p. Knockdown of lncPCAT19 distinctly inhibited tumor growth in vivo. Interfering lncPCAT19/overexpression of miR-142-5pdecreased glioma cell proliferation, colony formation and invasion, and promoted cell apoptosis by down-regulating expressionof Cyclin B1, CDK2, N-cadherin, Bcl-2, and by up-regulating expression of Bax and E-cadherin. Moreover, overexpressionof lncPCAT19 overturned tumor-suppressing role of miR-142-5p in cells. Additionally, lncPCAT19 and miR-142-5psynergistically regulated expression of MELK. In conclusion, lncPCAT19 enhanced glioma development via increasingMELK by performing as a sponge of miR-142-5p. Conclusions LncPCAT19 promotes glioma progression by sponging miR-142-5p to upregulate MELK levels. Thus, lncPCAT19/miR-142-5p/MELK signaling would be a potential target for glioma treatment.

        Taguchi Approach for Anti-heat Stress Prescription Compatibility in Mice Spleen Lymphocytes In Vitro

        Heat stress (HS) may induce immunosuppression as well as inhibit the proliferation of lymphocytes. This study evaluated the effects on immune function of our prescription on splenic lymphocytes under HS as well as its compatibility. The effects of four herbal extracts from Agastache rugosa, Atractylodes lancea, Cortex Phellodendri, and Gypsum Fibrosum on heat treated splenic lymphocytes were investigated and the compatibility of the prescription was also explored by using the Taguchi method. This study revealed changes in proliferation by traditional Chinese medicines of splenic lymphocytes after HS. Proliferation in the HS group was significantly lower than the control group. Under HS, the effects of higher concentrations of Agastache rugosa (100 and 200 μg/mL), Atractylodes lancea (100 and 200 μg/mL), Cortex Phellodendri (50 and 100 μg/mL) and Gypsum Fibrosum (100 and 200 μg/mL) caused a significant increase on ConA/LPS-induced proliferation of lymphocytes than lower concentrations. We, therefore, conclude that the prescription of traditional Chinese medicines may recover splenic lymphocytes from the immunosuppression induced by HS. The Taguchi design, which allows rapid and high efficiency for the selection of the best conditions for our prescription on HS-treated splenic lymphocytes, demonstrated that Agastache rugosa (200 μg/mL), Atractylodes lancea (200 μg/mL), Cortex Phellodendri (100 μg/mL) and Gypsum Fibrosum (100 μg/mL)were the optimal conditions for the prescription. The validation experiment confirmed that our composition in optimum extraction conditions enhanced effects on ConA or LPS-stimulated lymphocytes under HS. The results showed that the Taguchi optimization approach is a suitable method for optimization of the composition of prescription.

        Harm Avoidance is Correlated with the Reward System in Adult Patients with Attention Deficit Hyperactivity Disorder: A Functional Magnetic Resonance Imaging Study

        Objective: Hypoactivity in the reward system among patients with attention deficit hyperactivity disorder (ADHD) is a well-known phenomenon. Whether the activity in the reward pathway is related to harm avoidance, such as in sensitivity to punishment, is unclear. Evidence regarding the potential difference between ADHD patients and controls in terms of this association is scarce. Methods: Event-related functional magnetic resonance imaging was conducted on subjects performing the Iowa gambling test. Fourteen adults with ADHD and 14 controls were enrolled in the study. Results: Harm avoidance was found to be positively correlated with the activities of the bilateral orbitofrontal cortex and right insula in individuals with ADHD. A group difference was also confirmed. Conclusion: Understanding the roles of harm avoidance and brain activation during risk tasks is important.

        RNA sequencing reveals that Prx II gene knockout can down-regulate the allograft rejection of dermal mesenchymal stem cells

        In this study, we used RNA sequencing (RNA-seq) to analyze and compare bulk cell samples from wild-type (WT) dermal mesenchymal stem cells (DMSCs) (n = 3) and Prx II knockout DMSCs (n = 3). The purpose of the study was to elucidate the role of Prx II on allogeneic immune rejection of transplanted DMSCs. The results revealed differential expression of 472 genes (176 up-regulated and 296 down-regulated; p ≤ 0.05) between the PrxII+/+ (WT) and PrxII−/− sample groups. When highly regulated genes were categorized according to the Gene Ontology (GO) molecular function classification and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis, the PrxII−/− samples showed a robust downward trend in allograft rejection. The study identified 43 all immunologically rejected differentially expressed genes, of which 41 showed lower expression in the PrxII−/− vs. PrxII+/+ (WT) samples. These findings suggest that Prx II gene knockout may down-regulate the allograft rejection that occurs during DMSCs transplantation and improve the survival rate of DMSCs in the host. This study provides a new perspective on the clinical treatment of stem cell transplantation.

        Effects of a traditional Chinese medicine formula and its extraction on muscle fiber characteristics in finishing pigs, porcine cell proliferation and isoforms of myosin heavy chain gene expression in myocytes

        Objective: This study evaluated the effects of a traditional Chinese medicine formula (TCMF) on muscle fiber characteristics in finishing pigs and the effects of the formula's extract (distilled water, ethyl acetate and petroleum ether extraction) on porcine cell proliferation and isoforms of myosin heavy chain (MyHC) gene expression in myocytes. Methods: In a completely randomized design, ninety pigs were assigned to three diets with five replications per treatment and six pigs per pen. The diets included the basal diet (control group), TCMF1 (basal diet+2.5 g/kg TCMF) and TCMF2 (basal diet+5 g/kg TCMF). The psoas major muscle was obtained from pigs at the end of the experiment. Muscle fiber characteristics in the psoas major muscle were analyzed using myosin ATPase staining. Cell proliferation was measured using 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) dye and cytometry. Isoforms of MyHC gene expression were detected by real-time quantitative polymerase chain reaction. Results: The final body weight and carcass weight of finishing pigs were increased by TCMF1 (p<0.05), while the psoas major muscle cross-sectional area was increased by TCMF (p<0.05). The cross-sectional area and diameter of psoas major muscle fiber Ι, IIA, and IIB were increased by TCMF2 (p<0.05). The cross-sectional area and fiber diameter of psoas major muscle fiber IIA and IIB were increased by diet supplementation with TCMF1 (p<0.05). Psoas major muscle fiber IIA and IIB fiber density from the pigs fed the TCMF1 diet and the type IIB fiber density from the pigs fed the TCMF2 diet were lower compared to pigs fed the control diet (p<0.05). Pigs fed TCMF2 had a higher composition of type Ι fiber and a lower percentage of type IIB fiber in the psoas major muscle (p<0.05). The expression levels of MyHC Ι, MyHC IIa, and MyHC IIx mRNA increased and the amount of MyHC IIb mRNA decreased in the psoas major muscle from TCMF2, whereas MyHC Ι and MyHC IIx mRNA increased in the psoas major muscle from TCMF1 (p<0.05). Peroxisome proliferator-activated receptor ${\gamma}$ $coactivator-1{\alpha}$ and CaN mRNA expression in the psoas major muscle were up-regulated by TCMF (p<0.05). Porcine skeletal muscle satellite cell proliferation was promoted by $4{\mu}g/mL$ and $20{\mu}g/mL$ TCMF water extraction (p<0.05). Both $1{\mu}g/mL$ and $5{\mu}g/mL$ of TCMF water extraction increased MyHC IIa, MyHC IIb, and MyHC IIx mRNA expression in porcine myocytes (p<0.05), while MyHC Ι mRNA expression in porcine myocytes was decreased by $5{\mu}g/mL$ TCMF water extraction (p<0.05). Porcine myocyte MyHC Ι and MyHC IIx mRNA expression were increased, and MyHC IIa and MyHC IIb mRNA expression were down-regulated by $5{\mu}g/mL$ TCMF ethyl acetate extraction (p<0.05). MyHC Ι and MyHC IIa mRNA expression in porcine myocytes were increased, and the MyHC IIb mRNA expression was decreased by $1{\mu}g/mL$ TCMF ethyl acetate extraction (p<0.05). Four isoforms of MyHC mRNA expression in porcine myocytes were reduced by $5{\mu}g/mL$ TCMF petroleum ether extraction (p<0.05). MyHC IIa mRNA expression in porcine myocytes increased and MyHC IIb mRNA expression decreased by $1{\mu}g/mL$ in a TCMF petroleum ether extraction (p<0.05). Conclusion: These results indicated that TCMF amplified the psoas major muscle cross-sectional area through changing muscle fiber characteristics in finishing pigs. This effect was confirmed as TCMF extraction promoted porcine cell proliferation and affected isoforms of MyHC gene expression in myocytes.

      • Clinical outcomes on tension-releasing suture appendage on single-incision sling devices for postoperative voiding dysfunction involving undue tape tension

        Objective: To determine the clinical outcomes of adding tension releasing suture(TRS) appendage for manipulation of over-tensioned single-incision slings(SIS) as a means to relieve post-operative voiding dysfunction. Material and Method: This was a retrospective case series study conducted in the in a tertiary referral center from January 2010 to July 2017 after gaining Institutional Review Board approval. The records of patients with urodynamic stress incontinence(USI) without needing concurrent procedures that underwent anti-incontinence surgery using MiniArc, Solyx, and Ajust with voiding dysfunction were collated and analyzed. The primary outcome measure was the recovery of normal post-void residual urine(PVR) after TRS manipulation. The secondary outcome measures were the pain intensity noted during manipulation (quantified through Visual Analog Scale), and the continence rate (assessed through 1. Objective cure- 1-hour pad test weight <2g and absence of USI; and 2. Subjective cure- index score of <1 on question #3 on UDI-6: "Urine leakage related to physical activity, coughing, or sneezing?"). Results: There were 73 patients with high post-void residual (PVR) urine. The forty-two(9.5%) patients with over-tensioned sling were managed with TRS manipulation while the 31 patients(7%) with high PVR and no sling over-tension were managed with intermittent catheterization. All patients in both groups regained normal PVR. The TRS manipulated group demonstrated an objective cure of 92.9%(39/42) and subjective cure of 91%(38/42). Pain experienced during TRS manipulation significantly increased with the Ajust system (p=0.018). There were 3 patients who had persistence of USI, 2 from MiniArc and 1 from Solyx. Conclusion: The TRS manipulation is a well-tolerated procedure that can effectively relieve voiding dysfunction for over-tensioned SIS without affecting continence cure rates.

      • Molecular Propulsion: Chemical Sensing and Chemotaxis of DNA Driven by RNA Polymerase

        <P>Living cells sense extracellular signals and direct their movements in response to stimuli in environment. Such autonomous movement allows these machines to sample chemical change over a distance, leading to chemotaxis. Synthetic catalytic rods have been reported to chemotax toward hydrogen peroxide fuel. Nevertheless individualized autonomous control of movement of a population of biomolecules under physiological conditions has not been demonstrated. Here we show the first experimental evidence that a molecular complex consisting of a DNA template and associating RNA polymerases (RNAPs) displays chemokinetic motion driven by transcription substrates nucleoside triphosphates (NTPs). Furthermore this molecular complex exhibits a biased migration into a concentration gradient of NTPs, resembling chemotaxis. We describe this behavior as 'Molecular Propulsion', in which RNAP transcriptional actions deform DNA template conformation engendering measurable enhancement of motility. Our results provide new opportunities for designing and directing nanomachines by imposing external triggers within an experimental system.</P>

        Relational Benefits and Visual Arts Outcomes via WeChat Business

        The purpose of this study was to understand the effect of WeChat relational benefits on customer satisfaction and purchase intention. Due to the rapid development in the technological field, the use of WeChat has expanded drastically in recent years. In visual arts industry, WeChat business occupies a significant proportion of emerging industries. The recruited participants that were recruited via an online survey website were 335 (www.sojump.com). SPSS 22.0 statistical system was used to analyze the descriptive statistics, reliability, and validity. This study conceptualizes the positive relationship among relational benefits, customer satisfaction, and purchase intention of visual arts products. The findings indicated that WeChat users’ relational benefits can be categorized into two distinct benefit types: confidence, and social benefits. The more relational benefits WeChat users perceived, the higher the level of satisfaction. In contrast, the positive influence of customer satisfaction on purchase intention is weaker in consumers with a higher level of perceived risk. The results of this study will help WeChat business to retain its customers, thus, gaining a long-term value for visual arts industry. Theoretical and managerial implication for WeChat business are provided.

        Who Wants Checks and Balances? Endogeneity of the Balancing Perspective

        The premise of the intentional model of split-ticket voting is that some voters split their tickets simply because they prefer divided government and believe in constant "checks and balances." This article examines whether this premise stands firm in an emerging democracy like Taiwan. That is, by using survey data in Taiwan, we explore whether one's attitude toward divided or unified government is "real." We hypothesize that a citizen's attitude toward "checks and balances" is subject to change, and conditional on whether her preferred party is in power. Specifically, we speculate that a citizen would tend to hold the balancing perspective or favor divided government, if her preferred party is in opposition. However, if her preferred party becomes the ruling party, she would be more likely to oppose (hold) the balancing (non-balancing) perspective or favor unified government. We then utilize panel survey data embedded in Taiwan's Election and Democratization Studies (TEDS) to verify our hypothesis.

        DEVELOPMENT OF CHINA LIGHT-DUTY VEHICLE TEST CYCLE

        Driving cycles provide a basis for vehicle development and calibration and also serves as the foundation for energy consumption and emissions certification of vehicles. This paper presents the China Light-Duty Vehicle Test Cycle (CLTC) developed by the China Automotive Technology & Research Center (CATARC). First, the important steps and technical routes toward the CLTC development process are summarized. Second, the specific CLTC development process is presented in detail, including the data acquisition and data analysis procedures, weighting factor development and driving cycle construction. Then, the main driving characteristics of the New European Driving Cycle (NEDC), the Worldwide Harmonized Light-Duty Vehicles Test Cycle (WLTC), the Federal Test Procedure (FTP-75), the CLTC and the actual collected data are compared. The CLTC has low average speed, a high idle speed ratio and more frequent acceleration and deceleration characteristics. Finally, 70 vehicles are t ested based on the NEDC, WLTC, and CLTC according to their legislative procedures in the vehicle emission laboratories of the CATARC and the manufacturers. The results show that the CLTC’s fuel consumption is much higher than that of the NEDC and WLTC, and CLTC can effectively reflect the actual fuel consumption of users.
