Branched polymeric prodrug/programmed cell death 4 complexes for combinational cancer therapy

Here, we demonstrate a co-delivery system constructed by integrating chemotherapeutic molecules into branched polymeric prodrug which can condense nucleic acids. Demethylcantharidate (DCA) was chosen as a model drug and premodified through nucleophilic substitution reaction by its two carboxylic groups with allyl chloride. The synthesized intermediate (DCA-dially) was then used to polymerize with tris (2-aminoethyl) amine (TAEA) through progressively ammonolysis reaction. The obtained poly (DCA-alt-TAEA) (DCAT) was used to pack PDCD4 into spherical-like nanoparticles through electrostatic interaction. Gel retardation assays implied that DCAT could integrate DNA at the weight ratio of 1 and protect it from digestion by nuclease. Acid-base titration experiments showed that DCAT obtained preferable buffering capability which was beneficial for the endosomal escape of DCAT/PDCD4 complexes. Cellular tests involving gene transfection efficiency and cytotoxicity indicated that DCA and PDCD4 co-delivered by the complexes significantly and synergistically suppressed the viability of SMMC-7721 cells. These results suggest that integrating chemotherapeutic molecules into nucleic acid-packing polymeric prodrug as cationic polymer/PDCD4 complexes is a highly efficient way to co-deliver chemotherapeutic drugs and nucleic acids for cancer therapy.
Infrared Image Edge and Texture Analysis Method based on Visual Habit

The general edge extraction algorithm is not ideal to process infrared images, which is low contrast and blurred edge. In this paper, we used the multi-fractal spectrum to edge of infrared image. We extracted the edge information of the image and calculate the measure and fractal spectrum with multiple singular values of each pixel. Analysis of the similarities and differences of multiple measure, the function in edge extraction, meanwhile, analyzed the fiction of fractal characteristics to edge image extraction. This method differs from the traditional gradient algorithm,It determines whether the edge or not just according to the local extreme points , but according to the pixels in the local and global relationships to determine whether the pixel is a real edge. It can neglect important edge pixel and texture pixel,which is more in line with the human visual mental. It provide a good reference for recognition of infrared image and further processing.

Quantitative real-time RT-PCR is considered to valuable method for the analysis of gene expression. But for the data accuracy of gene expression, the normalization of data using reference genes is essential. In this study, we focus on the analysis of the relative expression of nine commonly used candidate reference genes in the different developmental stages and female and male pupae/adults of Chilo suppressalis, and expression stabilities were analyzed by the ΔCt method, geNorm, NormFinder, and RefFinder. The results showed that H3 was the most stable reference genes in the different developmental stages while EF1 was the optimal genes in pupae/adult of different sexes of C. suppressalis. To validate, the expression profile of the gene encoding heat shock protein 60 (Hsp60) was investigated. The results showed that the transcription of target gene Hsp60 varied depending on the reference gene selected for normalization in different developmental stages, but the trend of expression of Hsp60 between the most stable reference gene combination and the least stable reference gene in pupae/adults of different sexes was the same. Therefore, it is imperative to select the reference genes to estimate target gene expression accurately in the specific experiment. This study provides scientific basis on functional analyses of the important genes contributed to the development and sex differentiation of C. suppressalis.

This research is devoted to explore the nonlinear vibration characteristics of smart nanoshells under multi-phyisical magneto-electric fields. The nano-scale shell has been treated as a thin shell with prescribed curvature which is modeled by nonlocal elasticity theory. The material composition of the smart nanoshell has been considered as a two phase composite for which the effective properties depend on the percentage of each phase. The discretization of governing equations has been carried out based on differential quadrature method (DQM). It has been exhibited that nonlinear vibration properties of curved nanoshells rely on nonlocality coefficient, piezoelectric phase percentage, radius of curvature, and electrical/magnetic potential.
Genome-wide identification of the ARRs gene family in tobacco (Nicotiana tabacum)

Background The growth of axillary buds determines the shoot branching and morphology of plants, and its initiation and development are regulated by a series of hormonal signals, such as cytokinin. Arabidopsis response regulators (ARRs) can regulate the growth and development, disease resistance and stress resistance of plants by participating in cytokinin signaling. Objective To explore the distribution and expression pattern of ARR members in tobacco. Methods The identifcation, isoelectric points, molecular weights, protein subcellular localization prediction, multiple sequence alignment, phylogenetic analysis, protein motifs and structures, chromosome distributions of deduced ARR proteins were conducted. The gene expression profling of various tissues in response to topping, low temperature and drought were analyzed by RNA-seq and qRT-PCR. Results 59 ARR genes from cultivated tobacco (Nicotiana tabacum) were identifed, namely NtARRs, including 21 type A NtARRs and 38 type B NtARRs. The 59 NtARRs were expressed mainly in all organs except the fruits. Some representative NtARRs may participate in axillary bud initiation and development, as well as in stress resistance through cytokinin signal transduction. Conclusion Understanding the roles of NtARRs in the molecular mechanisms responsible for axillary bud growth and stress tolerance could aid in targeted breeding in crops.

Based on the exact analytical method, this paper calculates the no-load back electromotive force, air gap fux density, cogging torque, and electromagnetic torque of outer rotor wheel-hub permanent magnet synchronous motor (PMSM); the calculation results are in good agreement with those of the fnite element method, which verifes the accuracy of the exact analytical method for solving the outer rotor PMSM. On this basis, the second-generation non-dominated sorting genetic algorithm (NSGA-II) is applied to optimize the multiple structural parameters and performance of the motor more quickly. Compared with the initial scheme of the motor, the torque ripple and permanent magnet mass of the optimized scheme is decreased by a quarter; the average electromagnetic torque, efciency is also improved. Finally, the simulation and experimental results show that the proposed method is reasonable and efective.

This paper concerns on the control problem of a class of nonlinear systems with input delay and dynamic uncertainties using multi-dimensional Taylor Network (MTN) control method. Firstly, a new variable is introduced to eliminate the effect of input delay by combining Padé approximation with Laplace transformation. Secondly, a MTN-backstepping-based control strategy is constructively designed by introducing a new coordinate transformation, and the proposed controller has the advantages of simple structure and good real-time performance. Finally, the effectiveness of the proposed MTN-based control approach is demonstrated by three examples.
Chemical constituents of Lotus Seed Epicarp

Lotus (Nelumbo nucifera Gaertn.) is an important aquatic economic crop in China, and it has been widely cultivated in Asia. Almost every part of lotus (roots, leaves, flowers, and seeds) not only can be used as a food-stuff, but also have multiple medicinal functions [1]. In China, lotus leaves and seeds are publicly identified as both food and medicine resources. There is much research concerned with lotus leaves and seeds. It has been reported that the extract from lotus leaves and seeds have multiple bioactivities, such as being anti-oxidant, anti-inflammatory, immuno-modulatory, and anti-obesity [3-5] etc. The bioactivities of lotus are due to the functional components in lotus including polysaccharides, polyphenols, flavonoids etc. As an inedible part of lotus, lotus seed epicarp also has attracted additional attention. Lotus seed epicarp, the main by-product of lotus seed processing, is also abundant in polyphenols and worthy to be used as a functional food. Lotus seed epicarp extract has been used as a potential antioxidant and anti-obesity additive in Chinese Cantonese Sausage, which was the first use of lotus seed epicarp extract in meat product. However, little is known about the purification and identification of the key compounds from lotus seed epicarp. The aim of this study on the titled plant led to the isolation and purification of eleven known compounds. Eleven compounds ethyl linoleate (1), (e)-9-octadecenoic acid ethyl ester (2), 7β-hydroxy betulinic acid (3), Betulinic acid (4), α-amyrin (5), β-amyrin (6), Kaempferol-3-rutinoside (7), Isorhamnetin-3-O-β-gentiobioside (8), Quercetin (9), Rutin (10), Quercetin-3-O-β-Dgalactopyranoside (11) were isolated from the 95% ethanol extract of lotus seed epicarp. Their structures were elucidated spectroscopically. To the best of our knowledge, It is worth nothing that compounds 1, 2, 3, 4, 5, 6, 7, 8 and 11 were never isolated from this plant.
