The expression of pro-apoptotic molecules, p53 and Bax, in the spinal cord of rats with experimental autoimmune encephalomyelitis (EAE) was examined. Apoptosis was confirmed by the terminal deoxynucleotidyl transferasemediated dUTP nick end-labeling (TUNEL) method. TUSEL (+) apoptotic cells were mainly either ED1 (+) macrophages or T-cells in the parenchyma of E N . Western blot analysis showed that both p53 and Bax expression significantly ( p < 0.01) increased in the spinal cords of EAE rats at the peak stage, and thereafter declined. An immunohistochemical study showed that inflammatory cells (notably T cells) in the parenchyma express p53 and Bax, while brain cells, includng neurons and glia, were devoid of these nuclear staining of these molecules. The nuclear expression of p53 largely matches apoptotic cells in the parenchyma of EAE. These finchngs suggest that pro-apoptotic molecules, p53 and Bax, may play an important role in eliminating T cells in the parenchyma in EAE.

We investigated metabolism and carcinogenesis in livers of Sprague-Dawley rats fed juices and pelleted diets containing Korean native plants petasites japonicus Maxim by evaluating cell localization and expression of cytochrome P450s and GST-P. Anti-cytochrome P450s application in liver sections revealed three to four times increased expression of cytochrome P450E1 immunoreactivity in degenerative hepatocytes when compared to histologically normal hepatocytes. Anti-GST-P in showed positive pren plastic foci as well as in individual hepatocytes randomly scattered throughout all liver sections examined. Additionally GST-P was evident in proliferative endothelial cells and biliary epithelial cells in exposed rat livers. These results suggested that the increased level of cytochrome P4502E1 in affected hepatocytes was a direct consequence of Petasites japonicus toxicity. Further immunoreactivity to anti-GST-P in hepatocytes endothelial cells and biliary epithelial cells indicated a possible preneoplastic effects of Petasites japonicus in Sprague-Dawley rat.

To elucidate the molecular mechanisms of autoimmune inflammation in the central nervous system, we examined the expression and localization of STAT1, STAT3, STAT4 and STAT6 molecules during experimental autoimmune encephalomyelitis (EAE) by competitive PCR. In the present study, we quantitated IL-4 and IL-12 p40 mRNA by competitive PCR in the CNS during EAE. IL-4 mRNA was found at early and peak stages. On the other hand, the IL-12 p40 mRNA level reached maximal levels at the peak stage and still found at the recovery stage of the disease. We examined the kinetics of STAT mRNA in the CNS during EAE and demonstrated that STAT1 and STAT4 mRNA reached a maximal level at the peak stage of EAE, whereas STAT3 mRNA level increased gradually to the recovery stage. STAT6 mRNA increased rapidly at the early stage followed by gradual decrease till the recovery stage. Taken together, these findings suggest that STAT4 which was probably activated by IL-12 plays a pro-inflammatory role and that STAT3 which was activated throughout the disease course seems to serve as a transducer of anti-inflammatory signals.
A case of facial pseudocystic nodule caused by hyaluronic acid filler

Injectable hyaluronic acid(HA) fillers are widely used in aesthetic procedures for facial soft-tissue augmentation. Given with its low immunogenicity, hyaluronic acid fillers generally do not cause many long-term side effects. Although hyaluronic acid filler shows a high degree of safety, as it is the leading injectable fillers worldwide, the appearance of adverse effects is becoming significant for patients and doctors. Here we describe a case of a 33-year-old woman with multiple facial nodules located at filler injection sites. We performed MRI and excisional biopsy for the exact diagnosis and treatment. In histopathologic findings, there were several encapsulated hyaluronic acids with collagenous pseudocystic wall. To the best of our knowledge, this is the remarkable Korean case of pseudocytic nodules caused by monophasic hyaluronic acid filler injection. Therefore, we present this facial pseudocytic nodule case with a review of literature.
Choroidal Ischemia and Serous Macular Detachment Associated with Severe Postoperative Pain

To report the association of a unilateral serous macular detachment with severe postoperative pain. A 71-year-old woman presented with a sudden decrease in vision in the right eye, seven days after a total knee replacement arthroplasty. The patient`s history was unremarkable except for a severe pain greater than the visual analog scale of 8 points for about 2 days after surgery. Retinal examination showed a well differentiated serous detachment that was about 3.5 disc diameter in size and located in the macular area. Fluorecein angiography and indocyanine green angiography showed delayed perfusion of the choriocapillaris without leakage points in the early phase and persistent hypofluorescence with pooling of dye in the subretinal space in the late phase. There was a spontaneous resolution of the serous detachment and the choroidal changes with residual pigment epithelial changes. Severe postoperative pain may influence the sympathetic activity and introduce an ischemic injury with a focal, choroidal vascular compromise and secondary dysfunction of overlying RPE cells in select patients.
