Chicken RNA-binding protein T-cell internal antigen-1 contributes to stress granule formation in chicken cells and tissues

        Yingjie Sun,Pin Zhang,Hang Zheng,Luna Dong,Lei Tan,Cuiping Song,Xusheng Qiu,Ying Liao,Chunchun Meng,Shengqing Yu,Chan Ding 대한수의학회 2018 Journal of Veterinary Science Vol.19 No.1

T-cell internal antigen-1 (TIA-1) has roles in regulating alternative pre-mRNA splicing, mRNA translation, and stress granule (SG) formation in human cells. As an evolutionarily conserved response to environmental stress, SGs have been reported in various species. However, SG formation in chicken cells and the role of chicken TIA-1 (cTIA-1) in SG assembly has not been elucidated. In the present study, we cloned cTIA-1 and showed that it facilitates the assembly of canonical SGs in both human and chicken cells. Overexpression of the chicken prion-related domain (cPRD) of cTIA-1 that bore an N-terminal green fluorescent protein (GFP) tag (pntGFP-cPRD) or Flag tag (pFlag-cPRD) induced the production of typical SGs. However, C-terminal GFP-tagged cPRD induced notably large cytoplasmic granules that were devoid of endogenous G3BP1 and remained stable when exposed to cycloheximide, indicating that these were not typical SGs, and that the pntGFP tag influences cPRD localization. Finally, endogenous cTIA-1 was recruited to SGs in chicken cells and tissues under environmental stress. Taken together, our study provide evidence that cTIA-1 has a role in canonical SG formation in chicken cells and tissues. Our results also indicate that cPRD is necessary for SG aggregation.

      • Class Φ₂ DC-DC converter with PWM on-off control

        Bo Song,Xu Yang,Yingjie He 전력전자학회 2011 ICPE(ISPE)논문집 Vol.2011 No.5

This paper presents a very high frequency dc-dc converter based on class Φ₂ inverter and PWM on-off control is selected as the control strategy. Class Φ₂ resonant inverter can provide a lower switch voltage stress than class E inverter. Besides, a two-stage resonant gate driver is also introduced, which provides low gating losses and is suited for very high frequency applications. In addition, PWM on-off control is used to modulate the converter at a fixed frequency so that there is only designed frequency content at output voltage. The prototype dc-dc converter operating at 30㎒ provides 107W and closed-loop peak efficiency is 70% during input voltage range of 35V-47V when output voltage is regulated at 73V.

Optimal Location of Piles in Stabilizing Slopes Based on a Simplified Double-row Piles Model

        Changdong Li,Wenqiang Chen,Yingjie Song,Wenping Gong,Qihua Zhao 대한토목학회 2020 KSCE JOURNAL OF CIVIL ENGINEERING Vol.24 No.2

Embedding single-row piles is often adopted to stabilize slopes in engineering practice. However, for large-scale and complex slopes, single-row piles might not be able to stabilize the slope; rather, double-row piles, even multirow piles, should be adopted. Currently, the optimal locations of double-row stabilizing piles considering local instability of the slope have rarely been studied. In this paper, a simplified analytical model used to analyze double-row pile stabilized slopes is proposed, where the local failure of the slope above the first row of piles is considered. Through the kinematic approach of limit analysis combined with the strength reduction technique, the required resistance forces provided by double-row piles respectively are derived for different pile locations denoted by the rotational angle. Moreover, a framework is developed for analyzing the optimal locations of multirow piles considering multistage potential slip surfaces. The results of an illustrative example are presented, and the reasonableness of the proposed method is verified. It is concluded that the optimal locations of double-row piles lie within middle-lower part of the corresponding stabilized part of the slope. Finally, discussion illustrates the influences of the seismic effects and soil shear strength parameters on the derived optimal pile locations. This study provides novel scientific insight into the optimized design of stabilizing pile locations in engineering practice.

The Formation of Magnetite Nanoparticle in Ordered System of the Soybean Lecithin

Li, Tiefu,Deng, Yingjie,Song, Xiaoping,Jin, Zhixiong,Zhang, Ying Korean Chemical Society 2003 Bulletin of the Korean Chemical Society Vol.24 No.7

A method of preparation of magnetite nanoparticles in ordered systems, as in vesicle and microemulsion, consisting of soybean lecithin and water has been introduced. The size of magnetite grain was controlled by the content of soybean lecithin and size of liposomes in the systems. It was found by experiment that magnetite nanoparticles were formed inside the double layer vesicles. The magnetite nanoparticles were separated by magnetic separation and centrifugation and the dispersion of the magnetite nanoparticles prepared at 10% (w/w) soybean lecithin was particularly stable. The formation of pure magnetite nanoparticles was confirmed by analyses of XRD and electron diffraction pattern.

Exosome-mediated lnc-ABCA12-3 promotes proliferation and glycolysis but inhibits apoptosis by regulating the toll-like receptor 4/nuclear factor kappa-B signaling pathway in esophageal squamous cell carcinoma

        Junliang Ma,Yijun Luo,Yingjie Liu,Cheng Chen,Anping Chen,Lubiao Liang,Wenxiang Wang,Yongxiang Song 대한약리학회 2023 The Korean Journal of Physiology & Pharmacology Vol.27 No.1

Esophageal squamous cell carcinoma (ESCC) is a kind of malignant tumor with high incidence and mortality in the digestive system. The aim of this study is to explore the function of lnc-ABCA12-3 in the development of ESCC and its unique mechanisms. RT-PCR was applied to detect gene transcription levels in tissues or cell lines like TE-1, EC9706, and HEEC cells. Western blot was conducted to identify protein expression levels of mitochondrial apoptosis and toll-like receptor 4 (TLR4)/nuclear factor kappa-B (NF-κB) signaling pathway. CCK-8 and EdU assays were carried out to measure cell proliferation, and cell apoptosis was examined by flow cytometry. ELISA was used for checking the changes in glycolysis-related indicators. Lnc-ABCA12-3 was highly expressed in ESCC tissues and cells, which preferred it to be a candidate target. The TE-1 and EC9706 cells proliferation and glycolysis were obviously inhibited with the downregulation of lnc-ABCA12-3, while apoptosis was promoted. TLR4 activator could largely reverse the apoptosis acceleration and relieved the proliferation and glycolysis suppression caused by lnc-ABCA12-3 downregulation. Moreover, the effect of lnc-ABCA12-3 on ESCC cells was actualized by activating the TLR4/NF-κB signaling pathway under the mediation of exosome. Taken together, the lnc-ABCA12-3 could promote the proliferation and glycolysis of ESCC, while repressing its apoptosis probably by regulating the TLR4/NF-κB signaling pathway under the mediation of exosome.

A study on the wake structure of the double vortex tubes in a ventilated supercavity

Wei Wang,Cong Wang,Yingjie Wei,Wuchao Song 대한기계학회 2018 JOURNAL OF MECHANICAL SCIENCE AND TECHNOLOGY Vol.32 No.4

To study the wake structure of the double vortex tubes in a ventilated supercavity, the computational fluid dynamics (CFD) method based on the finite volume method and the volume of fluids (VOF) multiphase flow model were used to solve the Reynolds-averaged Navier-Stokes (RANS) equations for performing numerical simulation analysis on the wake structure of a supercavitating vehicle. By analyzing the reasons for the upward drift in the tail section of the supercavity and the formation of the double vortex tubes, the effects of gravity and the attack angle of the vehicle on the formation of gas leakage in the double vortex tubes of the supercavity were explained. The pressure and vorticity inside the vortex tubes were thereby analyzed as well. The results showed that, in addition to the gravitational condition, the wetted area of the vehicle with an attack angle also caused the supercavitating tail to form gas leakage in the form of double vortex tubes. Moreover, the wetted area played a dominant role compared to the role of gravity. The supercavity closure to the inside tail section was a high-pressure area, and this area separated downstream into two parts, resulting in the generation of the double vortex tubes. The vorticity and pressure in the vicinity of the vortex tubes attenuated downstream in the direction of the vortex tubes. When the tail section of the cavity leaked gas via three vortex tubes, the values of the vorticity inside the upper part of the vortex tubes became very small, and the air flow inside the upper part of the vortex tubes increased with the attack angle of the vehicle.

The role of exosomal miR-181b in the crosstalk between NSCLC cells and tumor-associated macrophages

        Ma Junliang,Chen Shaolin,Liu Yingjie,Han Hao,Gong Ming,Song Yongxiang 한국유전학회 2022 Genes & Genomics Vol.44 No.10

Background: It has been reported that tumor-associated macrophages (TAMs) participate in modulating the progression of cancer in the tumor microenvironment. However, the crosstalk between TAMs and non-small cell lung cancer (NSCLC) is still unclear. Objective: We investigated whether NSCLC-derived exosomes could affect TAMs, which feedback modulated progression of NSCLC. Methods: MiR-181b expression was measured by RT-PCR. Human THP-1 monocyte was differentiated into macrophages with phorbol myristate acetate, which were further identified by transmission electron microscopy and western blot. Macrophage M1 and M2 polarizations were detected by flow cytometry, RT-PCR and western blot. Proliferation, migration, and invasion of NSCLC cells treated with conditioned mediums were detected by EdU and Transwell assays. Results: We demonstrated that miR-181b was up-regulated in exosomes derived from NSCLC patients' serum and NSCLC cells. MiR-181b could be transferred to macrophages via exosomes in the co-culture system of macrophages and NSCLC cells, which promoted macrophage M2 polarization. Further examinations revealed that exosomes derived from NSCLC cells could enhanced macrophage M2 polarizations by regulating miR-181b/JAK2/STAT3 axis, and silencing miR-181b in NSCLC cells and JAK2 inhibitor used in macrophages could reverse the effects. Importantly, the conditioned medium of macrophages treated with NSCLC cell-derived exosomes could promote NSCLC cell proliferation, migration, and invasion. Silencing miR-181b in NSCLC cells and JAK2 inhibitor used in macrophages could block the effects. Conclusions: All of these results indicated that exosomal miR-181b participated in the crosstalk between NSCLC cells and TAMs, providing potential therapeutic targets for NSCLC.
