        Hydrogen sulfide ameliorates abdominal aorta coarctation-induced myocardial fibrosis by inhibiting pyroptosis through regulating eukaryotic translation initiation factor 2α phosphorylation and activating PI3K/AKT1 pathway

        This study aimed to assess the effects of exogenous hydrogen sulfide (H<sub>2</sub>S) on abdominal aorta coarctation (AAC) induced myocardial fibrosis (MF) and autophagy in rats. Forty-four Sprague-Dawley rats were randomly divided into control group, AAC group, AAC + H<sub>2</sub>S group, and H<sub>2</sub>S control group. After a model of rats with AAC was built surgically, AAC + H<sub>2</sub>S group and H<sub>2</sub>S group were injected intraperitoneally with H<sub>2</sub>S (100 µmol/kg) daily. The rats in the control group and the AAC group were injected with the same amount of PBS. We observed that H<sub>2</sub>S can improve left ventricular function and the deposition of myocardial collagen fibers, inhibit pyroptosis, down-regulate the expression of P-eif2α in myocardial tissue, and inhibit cell autophagy by activating the phosphatidylinositol 3-kinase (PI3K)/AKT1 signaling pathway (p < 0.05). In addition, angiotensin II (1 µM) H9c2 cardiomyocytes were injured in vitro experiments, and it was also observed that pyroptosis was inhibited after H<sub>2</sub>S (400 µmol/kg) intervention, the expression of P-eif2α in cardiomyocytes was significantly down-regulated, and the PI3K/AKT1 signaling pathway was activated at the same time. Therefore, increasing the expression of P-eif2α reverses the activation of the PI3K/AKT1 signaling pathway by H<sub>2</sub>S. In conclusion, these findings suggest that exogenous H<sub>2</sub>S can ameliorate MF in rats with AAC by inhibiting pyroptosis, and the mechanism may be associated with inhibiting the phosphorylation of eif2α and activating the PI3K/AKT1 signaling pathway to inhibit excessive cell autophagy.

        Recent advance of single atom-based photocatalysts for energy conversion and environmental purification

        Developing the high-performance semiconductor photocatalytic materials is an eternal topic under the background of the current energy and environment requirements. In recent years, single-atom photocatalysts (SAPCs) have been brought a lot of attention in energy conversion and environmental purification because of their unique characteristics and properties, including the unique coordination patterns, outstanding atomic utilization, quantum confinement effects, high catalytic activity, etc. Hence, this critical review focuses on the summarized various synthetic methods and the recent important applications of SAPCs, including photocatalytic H2 evolution (PHE) from water splitting, photocatalytic CO2 reduction, photodegradation of organic pollutants, etc. The prospects and challenges for future research topics of SAPCs with excellent activity and stability for various photocatalytic applications are prospected at the end of this review. We sincerely expect that this critical review can promote deep-level insight into the SAPCs subject for the future significant applications in other fields.

        Cloning and analysis of the molecularly characterized chitinase genes of Daphnia carinata and Simocephalus vetulus

        Daphniidae plays an important role in maintaining the integrity and sustainability of food chains of freshwater ecosystems. However, studies on the intact genes of chitinase in Daphniidae have not been carried out till date. In this study, we obtained the full-length complementary DNA (cDNA) from Daphnia carinata and Simocephalus vetulus, members of the Family Daphniidae, by rapid amplification of cDNA ends and polymerase chain reaction. The two cDNAs were named DcChi and SvChi, respectively. The result showed that DcChi and SvChi were 1404 and 1319 bp in length and they encoded 383 and 382 amino acids, respectively. Based on their cDNA sequences, the genomic structures of the two chitinases were characterized. Sequence analysis revealed that DcChi was composed of three exons and two introns, while SvChi had four exons and three introns. Based on the presence of conserved catalytic domain sequences, the two chitinases could be clustered within the same phylogenetic group. Homology analysis showed that the two deduced proteins had a high similarity (65–88%) to those from published species in Family Daphniidae and a low similarity to those from the species in Class Malacostraca Subclass Copepoda (38–42%) and those from the species in Class Insecta Order Diptera (33–38%). Based on the results of multiple alignments of chitinases, we designed peptides that were specific to chitinases from the Family Daphniidae. The quantitative real time PCR analysis indicated that the transcript level of the chitinase gene increased with the growth of the two species.

        Physiological functions and Ca 2+ signal transduction pathway exploring during molting of silkworm, Bombyx mori

        Calcium ions are an important second messagers in the life of organisms, such as in silkworm growth and their developmental processes; these ions can trigger various effects, including the muscle excitation-contraction coupling mechanism, secretion of endocrine and exocrine glands, neuronal activities, midgut nutrients metabolisms, sensory system, enzymatic systems, and acceleration of metabolism activities. This paper reviewed the ecdysis process and mechanisms, where calcium plays an important role in larvae-to-larvae ecdysis and larvaeto-pupae and pupae-to-moth metamorphosis of silkworms; the paper focuses on the advanced achievements in the field of calcium actions during larvae-to-larvae and larvae-to-pupae changes. Calcium ion channels are shown to be the action mechanism of phospholipase C in the G protein cascade of the calcium signal transduction pathway, and the activators and inhibitors of these calcium channels are further discussed.

        Response of sheep rumen fermentation and microbial communities to feed infected with the endophyte Epichloë gansuensis as evaluated with rumen-simulating technology

        Achnatherum inebrians, a perennial grass, is widely distributed in China. When infected by the endophyte Epichloë gansuensis, A. inebrians produces an abundance of alkaloids that enhance plant survival but are toxic to animals. Here we used in vitro fermentation to study the impact of endophyte- infected A. inebrians (E+) addition on rumen fermentation characteristics and on microbial community and diversity as assessed with amplicon sequencing technology. We examined E+ addition at five levels, E0, E25, E50, E75, and E100, corresponding to 0%, 25%, 50%, 75%, and 100% of the fermentation substrate, respectively. Both the fermentation characteristics and rumen microbial community structure differed significantly among treatments. E100 resulted in the highest values for pH, the Shannon index, Kiritimatiellaeota, and Lentisphaerae levels relative to the other treatments. In contrast, E25 was associated with higher levels of ammonia nitrogen, total volatile fatty acid, propionate, butyrate, isobutyrate, valerate, of the phyla Bacteroidetes and Firmicutes, and of the genus Prevotella_1, Succiniclasticum, Family_XIII_AD3011_group, Rikenellaceae_RC9_gut_group, Prevotellaceae_UCG-001, and Pyramidobacter as compared with other treatments. E50 resulted in the greatest values for the abundance-based coverage estimator (ACE) and the Chao1 index as compared with other treatments. E0 resulted in the greatest values for digestibility of dry matter, gas production, acetate, and Ruminobacter as compared with other treatments. This approach avoided animal toxicity experiments and confirmed that rumen fermentation characteristics and rumen microbiota were affected by E+ toxin. Therefore, E25 showed higher abundance in Prevotella_1, Prevotellaceae_ UCG-001, and Lachnospiraceae_XPB1014_group that implied they should play significant roles in E+ alkaloids degradation. And then, we can infer that rumen microorganisms should function as an antidote with respect to this poisoning reaction at moderate dietary percentages of E+.

        Define of Optimal Addition Period of Osteogenic Peptide to Accelerate the Osteogenic Differentiation of Human Pluripotent Stem Cells

        Background: The addition of growth factiors is commonly applied to improve the osteogenic differentiation of stem cells. However, for human pluripotent stem cells (hPSCs), their complex differentiation processes result in the unknown effect at different stages. In this study, we focused on the widely used bone forming peptide-1 (BFP-1) and investigated the effect and mechanisms of its addition on the osteogenic induction of hPSCs as a function of the supplementation period. Methods: Monolayer-cultured hPSCs were cultured in osteogenic induction medium for 28 days, and the effect of BFP-1 peptide addition at varying weeks was examined. After differentiation for varying days (0, 7, 14, 21 and 28), the differentiation efficiency was determined by RT–PCR, flow cytometry, immunofluorescence, and alizarin red staining assays. Moreover, the expression of marker genes related to germ layers and epithelial-mesenchymal transition (EMT) was investigated at day 7. Results: Peptide treatment during the first week promoted the generation of mesoderm cells and mesenchymal-like cells from hiPSCs. Then, the upregulated expression of osteogenesis marker genes/proteins was detected in both hESCs and hiPSCs during subsequent inductions with BFP-1 peptide treatment. Fortunately, further experimental design confirmed that treating the BFP-1 peptide during 7–21 days showed even better performance for hESCs but was ineffective for hiPSCs. Conclusion: The differentiation efficiency of cells could be improved by determining the optimal treatment period. Our study has great value in maximizing the differentiation of hPSCs by adding osteogenesis peptides based on the revealed mechanisms and promoting the application of hPSCs in bone tissue regeneration.         Construction of Customer Appeal Classification Model Based on Speech Recognition

        Aiming at the problems of poor customer satisfaction and poor accuracy of customer classification, this paperproposes a customer classification model based on speech recognition. First, this paper analyzes the temporaldata characteristics of customer demand data, identifies the influencing factors of customer demand behavior,and determines the process of feature extraction of customer voice signals. Then, the emotional associationrules of customer demands are designed, and the classification model of customer demands is constructedthrough cluster analysis. Next, the Euclidean distance method is used to preprocess customer behavior data. The fuzzy clustering characteristics of customer demands are obtained by the fuzzy clustering method. Finally,on the basis of naive Bayesian algorithm, a customer demand classification model based on speech recognitionis completed. Experimental results show that the proposed method improves the accuracy of the customerdemand classification to more than 80%, and improves customer satisfaction to more than 90%. It solves theproblems of poor customer satisfaction and low customer classification accuracy of the existing classificationmethods, which have practical application value.

        How to improve the survival rate of implants after radiotherapy for head and neck cancer?

        Implants have been widely used in restorative treatment for patients who have undergone head and neck cancer surgery. With the development of combination treatment of head and neck cancer, radiotherapy has been a common means of therapy. However, it could induce various changes in hard and soft tissues and reduce the success and survival rate of the implants. Some research, using either animal models or clinical studies, have shown that certain strategies could be used for improving the survival rate of implants. In this review, we discussed the changes in both hard and soft tissues, which may reduce the survival rate of the implants, and the proposed methods for improving the survival rate of patients after radiotherapy.

        A potential red-emitting phosphor with high color-purity for near-UV light emitting diodes

        New red tungstates phosphors, Na5La1-xLnx(WO4)4 (Ln = Eu, Sm) and Na5Eu1-xSmx(WO4)4, were prepared by solid-state reaction technique. And their structure and photo-luminescent properties were investigated. The introduction of Sm3+ broadened the excitation band around 400 nm of the phosphors,and strengthened the red emission. And the possible energy transfer process from Sm3þ to Eu3+ is discussed. The single red LED was fabricated by combining InGaN chip with Na5Eu0.94Sm0.06(WO4)4 as red phosphor, intense red light can be observed by naked eyes. Then the phosphor Na5Eu0.94Sm0.06(WO4)4 may be a good candidate for red component of near-UV InGaN-based W-LEDs,because of efficient red-emitting with broadened absorption around 400 nm and appropriate CIE chromaticity coordinates (x = 0.65, y = 0.34).

        “One-Time” versus Staged Multivessel Intervention in Intermediate to Very High-Risk Patients with Non-ST-Segment Elevation Acute Coronary Syndromes

        Background and Objectives: To compare clinical outcomes of staged versus “one-time” percutaneous coronary intervention (PCI) in intermediate to very high-risk patients with non–ST-segment elevation acute coronary syndromes (NSTE-ACS) and multivessel coronary disease (MVD). Subjects and Methods: 1531 NSTE-ACS patients with multivessel PCI and meeting the criteria of intermediate to very high risk were screened from a prospectively registered database obtained from General Hospital of Shenyang Military Region between 2008 and 2012. They were categorized into “one-time” PCI (n=859) and staged PCI (n=672) according to intervention strategy. The primary outcomes included a 3-year major adverse cardiac event (MACE), a composite of cardiac death, myocardial infarction (MI), and target vessel revascularization. Results: At 3 years, no significant differences in MACE (20.8% vs. 19.7%, p=0.608) and cardiac death/MI (7.1% vs. 9.1%, p=0.129) were observed between the two groups. After propensity score matching, there was no statistical significance in MACE (18.9% vs. 21.8%, p=0.249); whereas cardiac death/MI was significantly lower in the staged PCI group (7.0% vs.11.1%, p=0.033). Ninety-day landmark analysis showed that the staged PCI group had a lower 90-day incidence of MACE (1.2% vs. 3.3%, p= 0.037) and cardiac death/MI (0.7% vs. 2.6%, p=0.031). For the 90-day to 3-year follow-up period, the incidences of MACE (17.9% vs. 19.1%, p=0.641) and cardiac death/MI (6.3% vs. 8.7%, p=0.191) were similar in both groups. Conclusion: In intermediate- to very high-risk NSTE-ACS patients with MVD, staged PCI is superior to “one-time” PCI in terms of cardiac death/MI.
