        Genome-wide identification, evolution and expression analysis of the FtsH gene during fruit development in pear (Pyrus bretschneideri)

        Filtration temperature-sensitive H (FtsH) is an ATP-dependent protease family that plays important roles in many aspects of cellular processes and environmental adaption. However, little information is available about the FtsH genes in pear (Pyrus bretschneideri). In this study, 19 PbrFtsH genes were identified and classified into eight groups with several subgroups. The FtsH genes in pear show high sequence and structural conservation in the coding regions, and the majority of the FtsH members were located on the chromosome 3 (three members), chromosome 9 (four members), and chromosome 15 (three members). The WGD/segmenta, dispersed duplication played a major driving force in pear FtsH evolution. A Synteny analy- sis provided deep insight into the evolutionary relationships of PbrFtsH genes. The subcellular localization of the PbrFtsH genes showed them in the mitochondrion or chloroplast. Expression profiles derived from transcriptome data and RT-qPCR analysis revealed distinct expression patterns of PbrFtsH genes in different fruit developmental stages. These results provided valuable information that extends our understanding of the function, evolution and expression profile of the FtsH family during fruit development in pear, and a foundation for the further functional characterization of PbrFtsH genes, which can be applied to pear crop improvement.

        Nonlinear Analysis of Single Reinforced Concrete Piles Subjected to Lateral Loading

        This paper proposes a method for the nonlinear analysis of laterally loaded single reinforced concrete piles based on the beam-onnonlinear- Winkler-foundation approach. A nonlinear fiber beam-column element is used to model the nonlinear behavior of a pile. The pile is divided into a series of segments, of which the cross section is assumed to be plane and normal to the longitudinal axis. The internal force of a segment is derived by integrating the nonlinear stress-strain relationships of all steel and concrete fibers within the cross section of the segment. The substructure technique is introduced to calculate the stiffness matrix of the segments. The nonlinear behavior of soils surrounding the pile is characterized by a modified strain wedge model. The results show that (1) the predicted results using the proposed method are consistent with the measurements for all three full-scale tested piles, and (2) updating the neutral axis of segments has a significant effect on the calculated lateral deflection; however, it has a slight effect on the calculated bending moment. Moreover, an empirical equation is derived from the numerical analyses for estimating the cracked flexural rigidity of bored piles subjected to lateral loading.

        Auricular acupressure promotes uterine involution after cesarean section: A randomized controlled trial

        Background: Postpartum subinvolution of the uterus is a more common condition after cesarean section. Auricular acupressure (AA) is widely used for the treatment of postpartum diseases. However, few studies have explored the effects of AA as a treatment of uterine involution following cesarean section to date. This study aimed to assess the efficacy and safety of AA for uterine involution after cesarean section. Methods: A total of 109 women who underwent cesarean section participated in this study. They were randomly allocated to either real AA or sham AA in a 1:1 ratio by a computer program. For 3 days, the real AA and sham AA groups received treatment 3 times daily. A series of assessments at 42 days after cesarean section, namely on the uterine size, the incidence of hydrometra, the first anal exsufflation time, bleeding volume at 6 hours, bleeding volume at 6–24 hours along with other general assessments were carried out. Results: A total of 89 women completed the study. The uterine size at 42 days after a cesarean section was 6.3 cm smaller in the real AA group than in the sham AA group (P < 0.01). The incidence of hydrometra on day 42 postpartum was lower in the real AA group than in the sham AA group (P < 0.01). The lochia duration and the first anal exsufflation time after cesarean section were shorter in the real AA group than in the sham AA group (P < 0.05). Conclusion: AA improves uterine involution after cesarean section. Trial registration: ChiCTR1800015569.

        Experimental and theoretical studies of drug-polymer interactions to control the drug distributions in dissolving microneedles

        Dissolving microneedles (DMNs) have attracted extensive attention for transdermal drug delivery inrecent years. Generally, the skin elasticity impedes DMNs to fully insert into skin thus optimizedfabrication methods are needed to improve delivery efficiency. Here, we investigated the formulations ofDMNs as the main influencing factors during the microneedle preparation process. Through the selectionof the matrix materials and the optimization of the ratio and formulae, the DMNs can meet therequirements of convenient and effective drug delivery. For DMNs prepared with polyvinyl alcohol (PVA)as matrix material, the 25 wt% concentration of PVA was beneficial to prepare MNs, which MNs was ableto focus drug on the part of MN tips. However, for DMNs fabricated with hyaluronic acid (HA), theincreasing material concentration had a weaker ability to concentrate drug in MNs and increasedthe difficulty of preparing MNs. The computer simulation analysis was consistent with the results of theexperiment, indicating the feasibility of modelling to investigate the preparation technology of DMNsthat offer appropriate, convenient and highly efficient drug delivery.

        A Coumarin-based Fluorescent Sensor for Selective Detection of Copper (II)

        Cu (II) detection is of great importance owing to its significant function in various biological processes. In this report, we developed a novel coumarin-based chemosensor bearing the salicylaldimine unit (2) for $Cu^{2+}$ selective detection. The results from fluorescence spectra demonstrated that the sensor could selectively recognize $Cu^{2+}$ over other metal cations and the detection limit is as low as $0.2{\mu}M$. Moreover, the confocal fluorescence imaging in HepG2 cells illustrated its potential for biological applications.

      • Improved kinetics of LiNi<sub>1/3</sub>Mn<sub>1/3</sub>Co<sub>1/3</sub>O<sub>2</sub> cathode material through reduced graphene oxide networks

        <P>An electronically conducting 3D network of reduced graphene oxide (RGO) was introduced into LiNi<SUB>1/3</SUB>Mn<SUB>1/3</SUB>Co<SUB>1/3</SUB>O<SUB>2</SUB> (LNMC) cathode material in a special nano/micro hierarchical structure. The rate test and cycling measurement showed that the hierarchical networks remarkably improve the high rate performance of LNMC electrode for lithium-ion batteries. The effect of RGO conducting networks on kinetic property was investigated by electrochemical impedance spectroscopy (EIS) and potentiostatic intermittent titration (PITT). The EIS results reveal that the RGO network greatly decreases the resistance of lithium batteries, especially the charge transfer resistance which can be attributed to the significantly improved conducting networks. The enhancement of apparent diffusion coefficient by the RGO conducting networks is shown by PITT. The power performance was found to be limited by the electrical conduction in the two-phase region, which can be greatly facilitated by the hierarchical RGO network together with carbon black.       • Construction and Differential Screening of a cDNA Library Specific to Osmotic Stress of Haloxylon ammodendron Seedlings

        A subtracted cDNA library specific to osmotic stress of Haloxylon ammodendron (Mey.) Bge seedlings was constructed by suppression subtractive hybridization (SSH) and T/A cloning. SSH was performed between two groups of H. ammodendron seedlings, one was cultivated in Hoagland (H) solution as a driver and the other group was treated with osmotic stress of the Hoagland solution by the addition of 400 mM mannitol (M), as a tester. The library consisted of about 400 recombinant clones, with the average size being of 500 bp, ranging from 300 bp to 1500 bp. Using a PCR-select differential screening kit, 100 recombinant clones were randomly chosen from the subtracted cDNA library and hybridized with forward,reverse subtracted and unsubtracted probes for two rounds. As a result, 21 positive clones specific to osmotic stress were obtained and some of them were verified by Northern blot analysis. The sequencing analysis of 6 positive clones and the following homology comparison to GenBank [blastx] non-redundant databases characterized that two sequences obtained in this experiment may contribute to novel drought-related genes.

        Rapid separation and identification of 31 major saponins in Shizhu ginseng by ultra-high performance liquid chromatography-electron spray ionization-MS/MS

        Background: Among the various ginseng strains, Shizhu ginseng is endemic to China, mainly distributed in Kuandian Manchu Autonomous County (Liaoning Province, China); however, not much is known about the compounds (especially saponins) in Shizhu ginseng. Methods: A rapid, sensitive, and reliable ultra-high performance liquid chromatography coupled with MS/MS (UHPLCeMS/MS) method was developed to separate and identify saponins in Shizhu ginseng. Results: The separation was carried out on a Waters ACQUITY UPLC BEH C18 column (100 mm × 2.1 mm, 1.7 ㎛) with acetonitrile and 0.1% formic acid aqueous solution as the mobile phase under a gradient elution at 40℃. The detection was performed on a Micromass Quattro Micro API mass spectrometer equipped with electrospray ionization source in both positive and negative modes. Under the optimized conditions, a total of 31 saponins were identified or tentatively characterized by comparing retention time and MS data with related literatures and reference substances. Conclusion: The developed UHPLCeMS/MS method was suitable for identifying and characterizing the chemical constituents in Shizhu ginseng, which provided a helpful chemical basis for further research on Shizhu ginseng.
