        Causes, Features, and Outcomes of Drug-Induced Liver Injury in 69 Children from China

        Background/Aims: Drug-induced liver injury (DILI) is a frequent cause of pediatric liver disease; however, the data on DILI are remarkably limited. Methods: All 69 children hospitalized with DILI between January 2009 and December 2011 were retrospectively studied. Results: A total of 37.7% of the children had medical histories of respiratory infection. The clinical injury patterns were as follows: hepatocellular 89.9%, cholestatic 2.9%, and mixed 7.2%. Liver biopsies from 55 children most frequently demonstrated chronic (47.3%) and acute (27.3%) hepatitis. Hypersensitivity features, namely, fever (31.9%), rash (21.7%), and eosinophilia (1.4%), were found. Twenty-four children (34.8%) developed chronic DILI. Antibiotics (26.1%) were the most common Western medicines (WMs) causing DILI, and the major implicated herbs were Ephedra sinica and Polygonum multiflorum. Compared with WM, the children whose injuries were caused by Chinese herbal medicine (CHM) showed a higher level of total bilirubin (1.4 mg/dL vs 16.6 mg/dL, p=0.004) and a longer prothrombin time (11.8 seconds vs 17.3 seconds, p=0.012), but they exhibited less chronic DILI (2/15 vs 18/39, p=0.031). Conclusions: Most cases of DILI in children are caused by antibiotics or CHM used to treat respiratory infection and present with hepatocellular injury. Compared with WM, CHM is more likely to cause severe liver injury, but liver injury caused by CHM is curable. (Gut Liver 2015;9:525-533)

        Analysis on temperature field of friction pair of aviation friction clutch based on different groove shapes of friction disk

        To examine the influence of the groove shape of a friction disk on the transient temperature field in the engagement process of aviation wet friction clutch, a transient thermal analysis model of friction clutch was established, and the application method of thermal boundary conditions, including the calculation of convective heat transfer coefficient and heat flux density, was studied. The transient heat loading process was established by the partition method of friction disk and steel disk. The temperature characteristics of wet clutch friction pairs with different groove shapes during the engagement process were analyzed and tested on an SAE#2 testing machine. Results show that when the clutch is engaged, the temperature of the friction pair composed of two-way parallel groove, three-way parallel groove, and waffle groove friction disk increases continuously at first and finally decreases slightly; the highest temperature of the friction disk and steel disk is at the ring near the outer ring of the friction disk;and the waffle groove friction disk is more suitable for aviation wet friction clutch.

        Analysis of the Genome Sequence of Strain GiC-126 of Gloeostereum incarnatum with Genetic Linkage Map

        Gloeostereum incarnatum has edible and medicinal value and was first cultivated and domesticated in China. We sequenced the G. incarnatum monokaryotic strain GiC-126 on an Illumina HiSeq X Ten system and obtained a 34.52-Mb genome assembly sequence that encoded 16,895 predicted genes. We combined the GiC-126 genome with the published genome of G. incarnatum strain CCMJ2665 to construct a genetic linkage map (GiC-126 genome) that had 10 linkage groups (LGs), and the 15 assembly sequences of CCMJ2665 were integrated into 8 LGs. We identified 1912 simple sequence repeat (SSR) loci and detected 700 genes containing 768 SSRs in the genome; 65 and 100 of them were annotated with gene ontology (GO) terms and KEGG pathways, respectively. Carbohydrate-active enzymes (CAZymes) were identified in 20 fungal genomes and annotated; among them, 144 CAZymes were annotated in the GiC-126 genome. The A mating-type locus (MAT-A) of G. incarnatum was located on scaffold885 at 38.9 cM of LG1 and was flanked by two homeodomain (HD1) genes, mip and beta-fg. Fourteen segregation distortion markers were detected in the genetic linkage map, all of which were skewed toward the parent GiC-126. They formed three segregation distortion regions (SDR1-SDR3), and 22 predictive genes were found in scaffold1920 where three segregation distortion markers were located in SDR1. In this study, we corrected and updated the genomic information of G. incarnatum. Our results will provide a theoretical basis for fine gene mapping, functional gene cloning, and genetic breeding the follow-up of G. incarnatum.

      • Glucose-derived acetate and ACSS2 as key players in cisplatin resistance in bladder cancer

        <P><B>Abstract</B></P> <P>Cisplatin is an important chemotherapeutic agent against metastatic bladder cancer, but resistance often limits its usage. With the recent recognition of lipid metabolic alterations in bladder cancers, we studied the metabolic implications of cisplatin resistance using cisplatin-sensitive (T24S) and resistant (T24R) bladder cancer cells. Real-time live metabolomics revealed that T24R cells consume more glucose, leading to higher production of glucose-derived acetate and fatty acids. Along with the activation of general metabolic regulators, enzymes involved in acetate usage (ACSS2) and fatty acid synthesis (ACC) and a precursor for fatty acid synthesis (acetyl-CoA) were elevated in T24R cells. Consistently, metabolic analysis with <SUP>13</SUP>C isotope revealed that T24R cells preferred glucose to acetate as the exogenous carbon source for the increased fatty acid synthesis, contrary to T24S cells. In addition, ACSS2, rather than the well-established ACLY, was the key enzyme that supplies acetyl-CoA in T24R cells through glucose-derived endogenous acetate. The relevance of ACSS2 in cisplatin resistance was further confirmed by the abrogation of resistance by an ACSS2 inhibitor and, finally, by the higher expression of ACSS2 in the patient tissues with cisplatin resistance.       • Influences of guideway geometry parameters and track irregularity on dynamic performances of suspended monorail vehicle-guideway system

        This work elaborately investigates the influences of the guideway geometry parameters and track irregularity on the dynamic performances of the suspended monorail vehicle-guideway system (SMVGS). Firstly, a spatial dynamic analysis model of the SMVGS is established by adopting ANSYS parameter design language. Then, the dynamic interaction between a vehicle with maximum design load and guideway is investigated by numerical simulation and field tests, revealing the vehicle-guideway dynamic features. Subsequently, the influences of the guideway geometry parameters and track irregularity on the dynamic performances of the SMVGS are analyzed and discussed in detail, and the reasonable ranges of several key geometry parameters of the guideway are also obtained. Results show that the vehicle-guideway dynamic responses change nonlinearly with an increase of the guideway span, and especially the guideway dynamic performances can be effectively improved by reducing the guideway span; based on a comprehensive consideration of all performance indices of the SMVGS, the deflectionspan ratio of the suspended monorail guideway is finally recommended to be 1/1054~1/868. The train load could cause a large bending deformation of the pier, which would intensify the car-body lateral displacement and decrease the vehicle riding comfort; to well limit the bending deformation of the pier, its cross-section dimension is suggested to be more than 0.8 m×0.8 m. The addition of the track irregularity amplitude has small influences on the displacements and stress of the guideway; however, it would significantly increase the vehicle-guideway vibrations and rate of load reduction of the driving tyre.

        Carotid stiffening predicts cardiovascular risk stratification in mid-life: non-invasive quantification with ultrafast ultrasound imaging

        Purpose: The present study investigated the association between Systematic COronary Risk Evaluation (SCORE)-estimated cardiovascular risk and carotid stiffening in a middle-aged population using ultrafast pulse wave velocity (ufPWV).Methods: This study enrolled 683 participants without known cardiovascular disease or diabetes mellitus who underwent ufPWV measurements. Clinical interviews, physical examinations, laboratory findings, carotid intima-media thickness (cIMT), pulse wave velocity (PWV) at the beginning of systole (PWV-BS), and PWV at the end of systole (PWV-ES) were assessed. Each participant underwent an assessment of SCORE risk based on major cardiovascular risk factors (CVRFs), including age, sex, smoking, systolic blood pressure (SBP), and total cholesterol (TC). Crude and adjusted odds ratios (ORs) with 95% confidence intervals and ordinal logistic regression were used. Overall CVRFs were adjusted to assess ORs.Results: cIMT and carotid stiffening in PWV-BS and PWV-ES were significantly different between sex subgroups (all P<0.05), but only PWV-ES increased gradually in age and SCORE-estimated risk subgroups (all P<0.05). Compared with cIMT (r=0.388, P<0.001) and PWV-BS (r=0.159, P<0.001), PWV-ES was more strongly correlated with SCORE categories (r=0.405, P<0.001). Higher PWV-ES values were associated with SCORE categories independently of sex, SBP, TC, and smoking in moderate-risk and high-risk subgroups (OR, 1.63; P<0.001 and OR, 2.12; P=0.024, respectively), but were not independent of age in all risk subgroups (all P>0.05).Conclusion: Carotid stiffening quantified by ufPWV is linked to SCORE categories, and elevated PWV-ES may aid in cardiovascular risk stratification.

        Dimethyl Cardamonin Exhibits Anti-inflammatory Effects via Interfering with the PI3K-PDK1-PKCα Signaling Pathway

        Consumption of herbal tea [flower buds of Cleistocalyx operculatus (Roxb.) Merr. et Perry (Myrtaceae)] is associated with health beneficial effects against multiple diseases including diabetes, asthma, and inflammatory bowel disease. Emerging evidences have reported that High mobility group box 1 (HMGB1) is considered as a key late proinflammatory factor by its unique secretion pattern in aforementioned diseases. Dimethyl cardamonin (2′,4′-dihydroxy-6′-methoxy-3′,5′-dimethylchalcone, DMC) is a major ingredient of C. operculatus flower buds. In this study, the anti-inflammatory effects of DMC and its underlying molecular mechanisms were investigated on lipopolysaccharide (LPS)-induced macrophages. DMC notably suppressed the mRNA expressions of TNF-α, IL-1β, IL-6, and HMGB1, and also markedly decreased their productions in a time- and dose-dependent manner. Intriguingly, DMC could notably reduce LPS-stimulated HMGB1 secretion and its nucleo-cytoplasmic translocation. Furthermore, DMC dose-dependently inhibited the activation of phosphatidylinositol 3-kinase (PI3K), phosphoinositide-dependent kinase 1 (PDK1), and protein kinase C alpha (PKCα). All these data demonstrated that DMC had anti-inflammatory effects through reducing both early (TNF-α, IL-1β, and IL-6) and late (HMGB1) cytokines expressions via interfering with the PI3K-PDK1-PKCα signaling pathway.

        Insecticidal activities and biochemical properties of Pinellia ternata extracts against the beet armyworm Spodoptera exigua

        The beet armyworm, Spodoptera exigua Hübner (Lepidoptera: Noctuidae), is a key pest of various agricultural crops in many countries throughout the world. The pest requires extensive use of pesticides and field-evolved resistances to conventional insecticides in China and other countries. Pinellia ternata Breit is native to the eastern part of Asia and foundmainly in China,which has been used in traditional Chinese medicines for N1000 years, but few studies have focused on the insecticidal activity of the P. ternate. In order to find natural products that could be used to control the pest in an safe, efficient and ecofriendlymanner,we first time analyzed the components of the anhydrous ethanolic extracts fromthe tubers of Pinellia ternate by using GC–MS method, and then investigated the insecticidal activities and biochemical mechanisms of the extracts against S. exigua. The result of GC–MS showed that 2-Methoxy-4-vinylphenolmay be an insecticidal active component, and we also found the extracts had notable insecticidal activity and disturbed the regular metabolismof S. exiguamainly through altering the activities of detoxification enzymes, digestive enzymes and protective enzymes. These properties suggest that the anhydrous ethanolic extracts from P. ternate can serve as a potential, efficient and ecofriendly S. exigua-control biopesticide.

        Nonlinear tribo-dynamic model of helicopter mid-reducer system under light loss of lubrication

        When the helicopter tail transmission system loses oil due to damage, the system needs to persist for 30 minutes. Therefore, the nonlinear tribo-dynamics model of the helicopter intermediate reducer (HIR) is established, which is coupled with the loss of oil lubrication. To simulate condition of the HIR system under the loss of lubrication (LOL), this study begins by establishing a complete HIR system CFD model, and analyse the lubrication of the gear tooth surface and the oil churning/windage under different lubrication conditions. Then, a coupled model based on the result of the CFD is proposed to calculate the coefficient of friction (COF) under the condition of light loss of lubrication (LLOL). The results show that COF gradually decreases first and then increases rapidly.
