LncRNA-IMAT1 Promotes Invasion of Meningiomas by Suppressing KLF4/hsa-miR22-3p/Snai1 Pathway

Malignant meningiomas often show invasive growth that makes complete tumor resection challenging, and they are more prone to recur after radical resection. Invasive meningioma associated transcript 1 (IMAT1) is a long noncoding RNA located on Homo sapiens chromosome 17 that was identified by our team based on absolute expression differences in invasive and non-invasive meningiomas. Our studies indicated that IMAT1 was highly expressed in invasive meningiomas compared with non-invasive meningiomas. In vitro studies showed that IMAT1 promoted meningioma cell invasion through the inactivation of the Krüppel-like factor 4 (KLF4)/hsa-miR22-3p/Snai1 pathway by acting as a sponge for hsa-miR22-3p, and IMAT1 knockdown effectively restored the tumor suppressive properties of KLF4 by preserving its tumor suppressor pathway. In vivo experiments confirmed that IMAT1 silencing could significantly inhibit the growth of subcutaneous tumors and prolong the survival period of tumor-bearing mice. Our findings demonstrated that the high expression of IMAT1 is the inherent reason for the loss of the tumor suppressive properties of KLF4 during meningioma progression. Therefore, we believe that IMAT1 may be a potential biological marker and treatment target for meningiomas.

Somatic embryogenesis (SE) is a key regeneration process in plant. AcSERK1 is a gene specifi cally expressed in the early stage of SE in pineapple ( Ananas comosus ), suggesting that the promoter of SERK1 might contain specifi c cis acting element regulating SE. To identify embryonic cell-specifi c element in the SERK1 promoter, a series of binary plant transformation vectors with GUS (β-glucuronidase) reporter gene were systematically analyzed by transient gene expression system in wild-type and transgenic pineapple embryogenic callus. Histochemical and quantitative GUS assays demonstrated that the activity of the AcSERK1 upstream regulatory sequence lacking − 921 to -911 or -910 to -880 was signifi cantly reduced in the embryonic callus of the pineapple, and these two regions were needed for the embryonic cell-specifi c. Besides, a promoter lacking − 943 to -922 was shown to signifi cantly increase GUS activity in embryogenic callus, suggesting repressive elements exist in this region. Our data of stable transformation assays confi rmed again the 5’ upstream regulatory sequence (-921 to -880) of the AcSERK1 gene is an essential functional region. Our fi ndings lay the basis for better understanding of the molecular mechanisms of AcSERK1 gene in the regulation in early stage of SE.

The strain energy adjustment processes and rock failure modes corresponding to different excavation methods, such as Tunnel Boring Machine (TBM) or blasting, are quite different during construction of deep tunnel. Based on the diversion tunnel excavation of Jin-Ping II hydropower station (JPII) in southwestern China, the distribution characteristics of damage zones and adjustment process of rock strain energy under different excavation methods are analyzed and discussed, and the occurrences of rock bursts in the diversion tunnels are also monitored and analyzed. Research reveals that, the adjustment process of rock strain energy and the distribution of damage zones are obviously different under different excavation methods, and the depth and distribution of damage zone are positively correlated with the accumulation depth of rock strain energy. For blasting excavation, due to the combined effects of blast loading and in situ stress transient unloading, the surrounding rock is damaged seriously. The accumulation depth of rock strain energy is significantly larger than that by TBM excavation, while the accumulation peak of rock strain energy is smaller. For TBM excavation, the strain energy releases smoothly and slowly, and much more strain energy is accumulated in the vicinity of excavation face. Under similar geological conditions in the JPII, the rock bursts of intensive and mediate grades can be more frequently observed after blasting for the impact of severe excavation disturbance, and the strain energy transient adjustment may be the main disturbance contributor. While during TBM excavation, due to the smooth adjustment process of rock strain energy, the disturbance to surrounding rock is limited, and the accumulation peak of rock strain energy is higher and closer to the excavation face, which may result in more spalling events or minor rock bursts.

The protein encoded by SLC27A2 gene is an isozyme of long-chain fatty-acid-coenzyme A ligase family, and it converts free long-chain fatty acids into fatty acyl-CoA esters, and thereby plays a key role in lipid biosynthesis and fatty acid degradation. In the present study, SLC27A2 located on human chromosome 15 was selected as candidate gene and we isolated and cloned partial fragments of mRNA sequence and genomic fragments of porcine SLC27A2 gene. The coding region of the gene as determined by alignments shared 90% and 82% identity with human and mouse cDNAs, respectively. Detection in LargeWhite and Meishan breeds showed that a single nucleotide polymorphism (SNP) ($A{\rightarrow}G$) existed in exon 7, which caused corresponding amino acid changed for encoding. In LargeWhite pigs it encoded for Val while in Meishan pigs it encoded for Ile, so we developed the PCR-RFLP genotype method for detection of this polymorphism. Association study in 135 $F_2$ reference family indicated that significant correlation existed between the polymorphism and growth and carcass traits.

Emodin, a natural anthraquinone isolated from the traditional Chinese medicine Radix rhizoma Rhei, can induce apoptosis in many kinds of cancer cells. This study demonstrated that emodin induces apoptosis in human colon cancer HCT116 cells by provoking oxidative stress, which subsequently triggers a p53-mitochondrial apoptotic pathway. Emodin induced mitochondrial transmembrane potential loss, increase in Bax and decrease in Bcl-2 expression and mitochondrial translocation and release of cytochrome c to cytosol in HCT116 cells. In response to emodin-treatment, ROS increased rapidly, and subsequently p53 was overexpressed. Pretreatment with the antioxidant NAC diminished apoptosis and p53 overexpression induced by emodin. Transfecting p53 siRNA also attenuated apoptosis induced by emodin, Bax expression and mitochondrial translocation being reduced compared to treatment with emodin alone. Taken together, these results indicate that ROS is a trigger of emodin-induced apoptosis in HCT116 cells, and p53 expression increases under oxidative stress, leading to Bax-mediated mitochondrial apoptosis.

Reproduction is a complex physiological process that is regulated by multiple genes and pathways. Compared with studies of common livestock, fewer studies of genes related to the fertility of rabbits (Oryctolagus cuniculus) have been reported, and the molecular mechanism of their high productivity is still poorly understood. To identify candidate genes associated with development and prolificacy in rabbits, we analyzed gene expression differences among the ovaries of mature Californian rabbit (LC), and mature (HH) and immature Harbin white rabbit (IH) using digital gene expression technology. We detected 885 and 321 genes that were significantly differentially expressed in comparisons between HH/IH and HH/LC, respectively. The functions of the differentially expressed genes (DEGs) were determined by GO classification and KEGG pathway analysis. The results suggest that most of the DEGs between the mature and immature developmental stages were predominantly associated with DNA replication, cell cycle, and progesterone-mediated oocyte maturation, and most were up-regulated in the IH group compared with the HH group. The DEGs involved in disparate fecundities between HH and LC were associated with reproduction, fructose and mannose metabolism, steroid hormone biosynthesis, and pyruvate metabolism. Our results will contribute to a better understanding of changes in the regulatory network in ovary at different developmental stages and in different fertility of rabbit.

<P><B>Abstract</B></P> <P>A new small molecule acceptor of IDTCN with indaceno[2,1-b:6,5-b’]dithiophene as electron-donating core and CPTCN (2-(6-oxo-5,6-dihydro-4H-cyclopenta[<I>c</I>]thiophen-4-lidene)malononitrile) as electron-withdrawing end-capping group was synthesized and investigated in organic solar cells. The IDTCN shows strong and broad absorption with a high extinction coefficient and appropriate energy levels matching with wide bandgap donor polymer. The best device based on PBT1-EH:IDTCN exhibits an open circuit voltage (<I>V</I> <SUB>oc</SUB>) of 0.93 V, a short circuit current (<I>J</I> <SUB>sc</SUB>) of 13.31 mA/cm<SUP>2</SUP>, a FF of 0.702, achieving a PCE of 8.69%.</P> <P><B>Highlights</B></P> <P> <UL> <LI> A thiophene-fused electron-withdrawing group is used as end group for small molecule acceptors. </LI> <LI> The OPV devices achieve high power conversion efficiencies of up to 8.69% with a high fill factor of 0.702. </LI> <LI> IDTCN in neat film tend to form unusual isotropic orientation instead of face-on orientation. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>
