        Adaptive Fault-Tolerant Neural Control for Large-scale Systems with Actuator Faults

        The active adaptive fault-tolerant neural control problem is discussed for large-scale uncertain systems against actuator faults. The unknown interconnections among subsystems are assumed to be nonlinear, not traditional linear. A general actuator fault model is proposed, which integrates bias and gain time-varying faults. Then, based on Lyapunov stability theory, a novel fault diagnostic algorithm and accommodation scheme are proposed, where the assumptions in the existing works are removed and fault-tolerant controller singularity problem is avoided. Finally, simulation results of near space vehicle show the efficiency of the presented control approach.

        Transcriptional regulation of Glis2 in hepatic fibrosis

        The role of Gli-similar 2 (Glis2) in hepatic fibrosis (HF) is controversial. In this study, we focused on the functional and molecular mechanisms involved in the Glis2-mediated activation of hepatic stellate cells (HSCs)—a milestone event leading to HF. The expression levels of Glis2 mRNA and protein were significantly decreased in the liver tissues of patients with severe HF and in mouse fibrotic liver tissues as well as HSCs activated by TGFβ1. Functional studies indicated that upregulated Glis2 significantly inhibited HSC activation and alleviated BDL-induced HF in mice. Downregulation of Glis2 was found to correlate significantly with DNA methylation of the Glis2 promoter mediated by methyltransferase 1 (DNMT1), which restricted the binding of hepatic nuclear factor 1-α (HNF1-α), a liver-specific transcription factor, to Glis2 promoters. In addition, the enrichment of DNMT1 in the Glis2 promoter region was mediated by metastasis-associated lung adenocarcinoma transcriptor-1 (MALAT1) lncRNA, leading to transcriptional silencing of Glis2 and activation of HSCs. In conclusion, our findings reveal that the upregulation of Glis2 can maintain the resting state of HSCs. The decreased expression of Glis2 under pathological conditions may lead to the occurrence and development of HF with the expression silencing of DNA methylation mediated by MALAT1 and DNMT1.

        LncRNA KIAA0087 suppresses the progression of osteosarcoma by mediating the SOCS1/JAK2/STAT3 signaling pathway

        Long noncoding RNAs (lncRNAs), widely expressed in mammalian cells, play pivotal roles in osteosarcoma (OS) progression. Nevertheless, the detailed molecular mechanisms of lncRNA KIAA0087 in OS remain obscure. Here, the roles of KIAA0087 in OS tumorigenesis were investigated. KIAA0087 and miR-411-3p levels were detected by RT-qPCR. Malignant properties were assessed by CCK-8, colony formation, flow cytometry, wound healing, and transwell assays. SOCS1, EMT, and JAK2/STAT3 pathway-related protein levels were measured by western blotting. Direct binding between miR-411-3p and KIAA0087/SOCS1 was validated by a dual-luciferase reporter, RIP, and FISH assays. In vivo growth and lung metastasis were evaluated in nude mice. The expression levels of SOCS1, Ki-67, E-cadherin, and N-cadherin in tumor tissues were measured by immunohistochemical staining. Downregulation of KIAA0087 and SOCS1 and upregulation of miR-411-3p were found in OS tissues and cells. Low expression of KIAA0087 was associated with a poor survival rate. Forced expression of KIAA0087 or miR-411-3p inhibition repressed the growth, migration, invasion, EMT, and activation of the JAK2/STAT3 pathway and triggered apoptosis of OS cells. However, the opposite results were found with KIAA0087 knockdown or miR-411-3p overexpression. Mechanistic experiments indicated that KIAA0087 enhanced SOCS1 expression to inactivate the JAK2/STAT3 pathway by sponging miR-411-3p. Rescue experiments revealed that the antitumor effects of KIAA0087 overexpression or miR-411-3p suppression were counteracted by miR-411-3p mimics or SOCS1 inhibition, respectively. Finally, in vivo tumor growth and lung metastasis were inhibited in KIAA0087-overexpressing or miR-411-3p-inhibited OS cells. In summary, the downregulation of KIAA0087 promotes the growth, metastasis, and EMT of OS by targeting the miR-411-3p-mediated SOCS1/JAK2/STAT3 pathway.

        Experimental Observations of Boiling and Flow Evolution in a Coiled Tube

        A sequence of visually experimental observations was conducted to investigate the flow boiling and two-phase flow in a coiled tube. Different boiling modes and bubble dynamical evolutions were identified for better recognizing the phenomena and understanding the two-phase flow evolution and heat transfer mechanisms. The dissolved gases and remained vapor would serve as foreign nucleation sites, and together with the effect of buoyancy, centrifugal force and liquid flow, these also induce very different flow boiling nucleation, boiling modes, bubble dynamical behavior, and further the boiling heat transfer performance. Bubbly flow, plug flow, slug flow, stratified/wavy flow and annular flow were observed during the boiling process in the coiled tube. Particularly the effects of flow reconstructing and thermal non-equilibrium release in the bends were noted and discussed with the physical understanding. Coupled with the effects of the buoyancy, centrifugal force and inertia or momentum ratio of the two fluids, the flow reconstructing and thermal non-equilibrium release effects have critical importance for flow pattern in the bends and flow evolution in next straight sections.

        INTERACTION OF WATER-DISPERSIBLE, LIGAND-FREE NaYF 4 :Yb/Er UPCONVERSION NANOPARTICLES WITH BOVINE SERUM ALBUMIN

        The oleate capped NaYF 4 :Yb/Er upconversion nanoparticles (OA-UCNPs) were ¯rstlysynthesized by the thermal decomposition method and subsequently converted to water-dis-persible, ligand-free UCNPs (LF-UCNPs) via an easy acid treatment process. The interactionof LF-UCNPs with bovine serum albumin (BSA) was then investigated. The binding of LF-UCNPs with BSA was con¯rmed by the static quenching of intrinsic °uorescence of BSA uponaddition of LF-UCNPs. The spectra of synchronous °uorescence and circular dichroism (CD) ofBSA in the absence and presence of LF-UCNPs revealed that the presence of LF-UCNPs ledto a slight conformational change of BSA, suggesting a minor nanotoxicity of LF-UCNPstoward BSA.

        Development and validation of risk score for predicting spontaneous rupture of hepatocellular carcinoma

        Purpose: Spontaneous rupture is a potentially serious complication of liver cancer. A risk score was developed and validated for predicting spontaneous rupture based on a retrospective study. Methods: Multiple logistic regression analysis was used to study the relationship between clinical variables and spontaneous rupture. The independent rupture predictors were converted into a score based on the odds ratio. Predicted attributes of the developed scores were then verified using a dataset in 2019. Results: The incidence of spontaneous rupture was 5.5% from 2002 to 2019. A 10-point score (α-FP of ≥400 μg/L, 1; protrusion from liver surface, 2; ascites, 3; tumor size of >5 cm, 4) was derived for prediction of rupture and area under the receiver-operating characteristic curve was 0.9 (95% confidence interval, 0.87–0.92). When applying a cutoff value of 5 points or more, the specificity was 0.87 and the sensitivity was 0.84. A validation cohort consisting of 202 hepatocellular carcinoma patients reproduces the predictive, identification, and calibration characteristics. The observed rate of spontaneous rupture according to risk stratification of the score was 0.6% for those with a score of 0–4, 21.6% for a score of 5–7, and 36.4% for a score of 8–10 in the validation cohort. Conclusion: Here, based on routine clinical data, we determine the factors that affect prognosis and propose an effective tool for predicting spontaneous rupture, which may be useful in guiding priority treatment of high-risk patients or clinical routine preventive treatment.

        Pre-harvest ethylene control affects vase life of cut rose ‘Carola’ by regulating energy metabolism and antioxidant enzyme activity

        We studied the role of ethylene control in regulating energy metabolism, antioxidant enzyme activity, and vase life of cutrose Rosa hybrida ‘Carola’. Rose fl owers at stage II were sprayed with one of the following solutions: water (control), 10μL L −1 1-methylcyclopropene (1-MCP), or 0.5 g L −1 2-chloroethanephosphonic acid (ethephon). After harvest, ethyleneproduction rate, respiration intensity, energy charge (EC), activities of energy metabolism-related and antioxidant enzymes,and malondialdehyde (MDA) content were measured. Results showed that 1-MCP enhanced the activities of superoxidedismutase, H + -adenosine triphosphatase, Ca 2+ -adenosine triphosphatase, succinic dehydrogenase, and cytochrome c oxidase,increased adenosine triphosphate (ATP) content, maintained high EC levels, inhibited respiration intensity, reduced peroxidase(POD) and polyphenol oxidase (PPO) activity and MDA accumulation, and prolonged vase life. Ethephon promotedethylene production and respiration intensity, increased POD and PPO activity, reduced ATP content and EC levels, andaccelerated senescence. Our results support a novel role for ethylene control in regulating senescence of ‘Carola’.
