        Aerodynamic force characteristics and galloping analysis of iced bundled conductors

        Aerodynamic characteristics of crescent and D-shape bundled conductors were measured by high frequency force balance technique in the wind tunnel. The drag and lift coefficients of each sub-conductor and the whole bundled conductors were presented under various attack angles of wind. The galloping possibility of bundled conductors is discussed based on the Den Hartog criterion. The influence of icing thickness, initial ice accretion angle and sub-conductor on the aerodynamic properties were investigated. Based on the measured aerodynamic force coefficients, a computationally efficient finite element method is also implemented to analyze galloping of iced bundled conductors. The analysis results show that each sub-conductor of the bundled conductor has its own galloping feature due to the use of aerodynamic forces measured separately for every single sub-conductors.

        The effects of dantrolene and 2-aminoethoxydiphenyl borate (2-APB) on arsenic-induced osteoporosis

        Background Arsenic can increase the risk of osteoporosis among the population in arsenic–endemic areas compared with the general population in non-endemic areas. And calcium overload is the related mechanisms of arsenic-induced osteoporosis However, there is a relative lack of clinical research on the treatment of arsenic-induced osteoporosis. Dantrolene and 2-APB, as Ca 2+ infl ux inhibitor, can reduce calcium overload and play an important role in the excitation–contraction coupling of skeletal muscle. Therefore, we study the eff ects of dantrolene and 2-APB on arsenic-induced osteoporosis in this study. Objective To study the eff ects of arsenic on the trabeculae of rats, and observe the eff ects of dantrolene and 2-aminoethoxydiphenyl borate (2-APB) on the trabeculae. Results Compared with the control group, some indices of micro-CT (BV/TV, Tb.Th, Tb.N, BMD, and DA) and the alkaline phosphatase (ALP) activity of bone marrow stromal cells (BMSCs) decreased, while some indices of micro-CT (Tb.Sp, Tb.Pf, and SMI), biochemical indicators (ALT, AST, ALP, and GGT), and the Ca 2+ concentration of BMSCs increased in the Sodium arsenite (NaAsO 2 ) group ( P < 0.05). Compared with the NaAsO 2 group, the indices of micro-CT and biochemical indicators improved, the Ca 2+ concentration of BMSCs decreased, and the ALP activity of BMSCs increased in the dantrolene group, 2-APB group and dantrolene + 2-APB group ( P < 0.05), especially dantrolene + 2-APB group. Conclusion NaAsO 2 can lead to osteoporosis in rats, dantrolene and 2-APB can reduce the degree of arsenic-induced osteoporosis.

        NEDD8-activating enzyme inhibitor, MLN4924 (Pevonedistat) induces NOXA-dependent apoptosis through up-regulation of ATF-4

        <P><B>Abstract</B></P> <P>It has been reported that MLN4924 can inhibit cell growth and metastasis in various kinds of cancer. We have reported that MLN4924 is able to inhibit angiogenesis through the induction of cell apoptosis both in vitro and in vivo models. Moreover, Neddylation inhibition using MLN4924 triggered the accumulation of pro-apoptotic protein NOXA in Human umbilical vein endothelial cells (HUVECs). However, the mechanism of MLN4924-induced NOXA up-regulation has not been addressed in HUVECs yet. In this study, we investigated how MLN4924 induced NOXA expression and cellular apoptosis in HUVECs treated with MLN4924 at indicated concentrations. MLN4924-induced apoptosis was evaluated by Annexin V-FITC/PI analysis and expression of genes associated with apoptosis was assessed by Quantitative RT-PCR and western blotting. As a result, MLN4924 triggered NOXA-dependent apoptosis in a dose-dependent manner in HUVECs. Mechanistically, inactivation of Neddylation pathway caused up-regulation of activating transcription factor 4 (ATF-4), a substrate of Cullin-Ring E3 ubiquitin ligases (CRL). NOXA was subsequently transactivated by ATF-4 and further induced apoptosis. More importantly, knockdown of ATF-4 by siRNA significantly decreased NOXA expression and apoptotic induction in HUVECs. In summary, our study reveals a new mechanism underlying MLN4924-induced NOXA accumulation in HUVECs, which may help extend further study of MLN4924 for angiogenesis inhibition treatment.</P> <P><B>Highlights</B></P> <P> <UL> <LI> MLN4924 triggered NOXA-dependent apoptosis in a dose-dependent manner in HUVECs. </LI> <LI> Inactivation of neddylation caused up-regulation of ATF4, a substrate of CRL. </LI> <LI> NOXA was transactivated by ATF4 and further induced apoptosis. </LI> <LI> Knockdown of ATF-4 significantly decreased NOXA expression and apoptotic induction. </LI> <LI> We revealed a new mechanism underlying MLN4924-induced NOXA accumulation in HUVECs. </LI> </UL> </P>

        Metabolic reprogramming and epigenetic modifications in cancer: from the impacts and mechanisms to the treatment potential

        Metabolic reprogramming and epigenetic modifications are hallmarks of cancer cells. In cancer cells, metabolic pathway activity varies during tumorigenesis and cancer progression, indicating regulated metabolic plasticity. Metabolic changes are often closely related to epigenetic changes, such as alterations in the expression or activity of epigenetically modified enzymes, which may exert a direct or an indirect influence on cellular metabolism. Therefore, exploring the mechanisms underlying epigenetic modifications regulating the reprogramming of tumor cell metabolism is important for further understanding tumor pathogenesis. Here, we mainly focus on the latest studies on epigenetic modifications related to cancer cell metabolism regulations, including changes in glucose, lipid and amino acid metabolism in the cancer context, and then emphasize the mechanisms related to tumor cell epigenetic modifications. Specifically, we discuss the role played by DNA methylation, chromatin remodeling, noncoding RNAs and histone lactylation in tumor growth and progression. Finally, we summarize the prospects of potential cancer therapeutic strategies based on metabolic reprogramming and epigenetic changes in tumor cells.

        Traffic Flow Sensing Using Wireless Signals

        As an essential part of the urban transportation system, precise perception of the traffic flow parameters at the traffic signal intersection ensures traffic safety and fully improves the intersection's capacity. Traditional detection methods of road traffic flow parameter can be divided into the micro and the macro. The microscopic detection methods include geomagnetic induction coil technology, aerial detection technology based on the unmanned aerial vehicles (UAV) and camera video detection technology based on the fixed scene. The macroscopic detection methods include floating car data analysis technology. All the above methods have their advantages and disadvantages. Recently, indoor location methods based on wireless signals have attracted wide attention due to their applicability and low cost. This paper extends the wireless signal indoor location method to the outdoor intersection scene for traffic flow parameter estimation. In this paper, the detection scene is constructed at the intersection based on the received signal strength indication (RSSI) ranging technology extracted from the wireless signal. We extracted the RSSI data from the wireless signals sent to the road side unit (RSU) by the vehicle nodes, calibrated the RSSI ranging model, and finally obtained the traffic flow parameters of the intersection entrance road. We measured the average speed of traffic flow through multiple simulation experiments, the trajectory of traffic flow, and the spatio-temporal map at a single intersection inlet. Finally, we obtained the queue length of the inlet lane at the intersection. The simulation results of the experiment show that the RSSI ranging positioning method based on wireless signals can accurately estimate the traffic flow parameters at the intersection, which also provides a foundation for accurately estimating the traffic flow state in the future era of the Internet of Vehicles.

        Effects of rosmarinic acid on immunoregulatory activity and hepatocellular carcinoma cell apoptosis in H22 tumor-bearing mice

        Rosmarinic acid (RA) is a natural polyphenolic compound that exists in many medicinal species of Boraginaceae and Lamiaceae. The previous studies have revealed that RA had therapeutic effects on hepatocellular carcinoma (HCC) in the H22-xenograft models by inhibiting the inflammatory cytokines and $NF-{\kappa}B$ p65 pathway in the tumor microenvironment. However, its molecular mechanisms of immunoregulation and pro-apoptotic effect in HCC have not been fully explored. In the present study, RA at 75, 150, and 300 mg/kg was given to H22 tumor-bearing mice via gavage once a day for 10 days. The results showed that RA can effectively inhibit the tumor growth through regulating the ratio of $CD4^+/CD8^+$ and the secretion of interleukin (IL)-2 and interferon-${\gamma}$, inhibiting the expressions of IL-6, IL-10 and signal transducer and activator of transcription 3, thereby up-regulating Bax and Caspase-3 and down-regulating Bcl-2. The underlying mechanisms involved regulation of immune response and induction of HCC cell apoptosis. These results may provide a more comprehensive perspective to clarify the anti-tumor mechanism of RA in HCC.

        20(S)-ginsenoside Rh2 ameliorates ATRA resistance in APL by modulating lactylation-driven METTL3

        Background: 20(S)-ginsenoside Rh2(GRh2), an effective natural histone deacetylase inhibitor, can inhibit acute myeloid leukemia (AML) cell proliferation. Lactate regulated histone lactylation, which has different temporal dynamics from acetylation. However, whether the high level of lactylation modification that we first detected in acute promyelocytic leukemia (APL) is associated with all-trans retinoic acid (ATRA) resistance has not been reported. Furthermore, Whether GRh2 can regulate lactylation modification in ATRA-resistant APL remains unknown. Methods: Lactylation and METTL3 expression levels in ATRA-sensitive and ATRA-resistant APL cells were detected by Western blot analysis, qRT-PCR and CO-IP. Flow cytometry (FCM) and APL xenograft mouse models were used to determine the effect of METTL3 and GRh2 on ATRA-resistance. Results: Histone lactylation and METTL3 expression levels were considerably upregulated in ATRA-resistant APL cells. METTL3 was regulated by histone lactylation and direct lactylation modification. Overexpression of METTL3 promoted ATRA-resistance. GRh2 ameliorated ATRA-resistance by downregulated lactylation level and directly inhibiting METTL3. Conclusions: This study suggests that lactylation-modified METTL3 could provide a promising strategy for ameliorating ATRA-resistance in APL, and GRh2 could act as a potential lactylation-modified METTL3 inhibitor to ameliorate ATRA-resistance in APL.
