        In Vitro Antiviral Activity of Ribavirin Against Severe Fever with Thrombocytopenia Syndrome Virus Myung Jin LEE1, Kye-Hyung KIM1, Jongyoun YI2, SuJin CHOI1, Chung-Jong KIM1, Nak- Hyun KIM1, Kyoung-Ho SONG1, Pyoeng Gyun CHOI1, Ji-Hwan BANG1, Wan Beom PARK1, Eu Suk KIM1, Sang-Won PARK1, Hong Bin KIM1, Nam Joong KIM1, Myoung- Don OH1 Seoul National University College of Medicine, Korea1, Pusan National University School of Medicine, Korea2 Background: Severe fever with thrombocytopenia syndrome (SFTS) is an emerging infectious disease caused by a novel Bunyavirus, severe fever with thrombocytopenia syndrome virus (SFTSV). No effective antiviral therapy is proven yet, but clinical use of ribavirin (RBV) has been tried. We investigated the antiviral effect of RBV against SFTSV in vitro. Methods: To test for cytotoxicity of RBV, Vero cells were treated with different concentrations of RBV (3.90 to 500 μg/mL, two-fold dilution) and analyzed by cell viability MTS assay 48h post-infection. To determine antiviral activity of RBV against SFTSV, Vero cells were infected with SFTSV strain Gangwon/Korea/2012 at 100 TCID50 (50% tissue culture infective dose) per well in a 96-well plate, and RBV was added at the concentrations showing no or minimal cytotoxicity. Viral RNAs were extracted from the culture supernatants and quantifi ed using one-step real-time reverse transcription- PCR to amplify the partial large segment of SFTSV. Statistical analysis was done by one-way ANOVA with Tukey`s post hoc test. Results: Cytotoxicity due to RBV was not observed at RBV concentration =31.3 μg/ mL. Viral RNAs at 24h post-RBV treatment were reduced with increasing RBV concentrations (1-32 μg/mL), compared with those of mock-treated cells (P <0.01, Figure). Half maximal inhibitory concentration (IC50) of RBV was 3.69 μg/mL at 24h post-RBV treatment. Conclusions: Our study shows that RBV has antiviral effect against SFTSV in a dose-dependent manner. Further studies are required to evaluate the effi cacy of RBV in SFTS.

      • 榮山姜水系의 魚類相과 榮山潮內의 魚類相의 變化

        1987年 10月과 1988年 4月부터 8月까지 榮山江本流와 支流 그리고 榮山湖內의 49個 調査地点에서 魚類相과 榮山湖의 淡水化에 따른 魚類相의 變化를 調査하였다. 本 調査에서 採集된 魚類는 總 20科 44屬 64種 및 亞種 17,728個體로 그 가운데 Cyprinidae 31종, Gobiidae 7종, Cobitidae 4종, Bagridae 3종 등 이었으며, 본 조사에서 새로이 서식이 확인된 種은 Squalidus chankaensis tsuchigae, Liobagrus obesus, Oncorhynchus masou, Hemiramphus kurumens, Gasteroteus aculeatus, Mugil cephalus, M . haematocheila, Rhinogobius giurinus, Acanthogobius flavimanus, A . hasta, Chaenogobius annularis, Tridentiger obscurus등 12종이었으며, 韓國特産種은 15種이었다. 榮山江本流 및 榮山湖內에서는 총 59種의 魚類가 채집되었고 이들 水系로 流入되는 小河川들에서는 34種의 魚類가 그리고 支流인 黃龍江과 砥石江에서는 各各 43種과 29種의 魚類가 채집되었다. 榮山湖內 水質의 淡水化에 따라 1次淡水魚 뿐만 아니라 汽水性 魚類의 出現種數가 전체적으로 增加하는 傾向을 보였다. The freshwater fish fauna of Yongsan River stream and the change of fish fauna in Yongsan Lake were investigated from October, 1987, to August, 1988. total collected fishes were 20 families, 44 genera, 64 species and subspecies, 17,728 individuals including 31 species of Cyprinidae, 7 species of Gobiidae, 4 species of Cobitidae and 3 species of Bagridae, and among them 15 species were known as Korean endemic species. Squalidus chankaensis tsuchigae, Liobagrus obesus, Oncorhynchus masou, Hemiramphus kurumens, Gasteroteus aculeatus, Mugil cephalus, M. haematocheila, Rhinogobius giurinus, Acanthogobius flavimanus, A. hasta, Chaenogobius annularis, Tridentiger obscurus were observed in the present studied areas for the first time. In Yongsan River system and the total number of species including primary freshwater fishes and peripheral fishes increased as the water quality of Yongsan Lake changed into feshwater.

        Streptococcus mutans의 우식 활성에 미치는 Propolis의 효과

        Propolis, a natural resinous product collected by honey bees, is known to exhibit various biological activities including anti-microbial, anti-inflammatory, and anti-tumor activities. The present study was designed to investigate the effects of ethanol extracted propolis(EEP) on the growth, glucosyltransferase(GTase) activity, and adherence to the saliva-coated hydroxyapatite(S-HA) beads of Streptococcus mutans JC-2(serotype c), S.mutans LMM-7(serotype e), and S. mutans OMZ-175(serotype f) in vitro. We found that the EEP had minimal inhibitory concentrations of 500-1000㎍/㎖ against S.mutans strains when these strains were cultured for 48 hours, and inhibited both water-insoluble and water-soluble glucan synthesizing GTase activities of S. mutans strains to the 93-100% at the concentration of 1600㎍/㎖, and also we found that the EEP inhibited the adherence of S. mutans strains to S-HA beads to the 76-96% at the concentrations of 200㎍/㎖. These results indicate that EEP has anti-cariogenic activity in vitro.

        Helicobacter pylori의 생육에 영향을 미치는 성분과 식품첨가물에 관한 연구

        본 연구에서는 일반적으로 섭취하고 있는 식품재료나 음식 중에 공통적으로 함유한 성분 또는 가공중에 많이 첨가되는 성분을 선정하여 이들이 Helicobacter pylori 생육에 어떤 영향을 미치는지를 검토하였다. 첫째, 일반식품의 주요 성분인 탄소원, 질소원, 무기질을 대상으로 Helicobacter pylori 생육에 미치는 영향을 검토하였다. 그 결과 식품성분 중 탄소원의 종류에 따라 균일 생육활성은 다르게 나타났다. 우선 단당류가 이당류나 다당류에 비교하여 생육을 촉진하였고 특히 단당류 중 포도당은 가장 균의 생육에 큰 영향을 미치는 것으로 나타났다. 그러나 만노오스는 균의 생육을 오히려 억제시키는 것으로 나타났다. 당알코올류인 솔비톨, 만니톨, 말티톨, 크리시톨 등은 균의 생육에 아무런 영향을 미치지 않은 것으로 나타났다. 질소원과 Helicobacter pylori의 생육과의 관계는 평소 식생활에서 많이 섭취된다고 생각되는 일반 음식에 많이 함유되어 있는 유기형 아미노산 성분고 무기형 질소화합물을 대상으로 균의 생육 정도를 비교하였다. 리신, 메티오닌, 시스테인, 트립토판 등은 균의 생육을 촉진하고 아르기닌, 황산암모늄, 염화암모늄 등은 균의 생육에 도움이 되지 않고 암노니아, 요소등은 균의 생육에 도움이 되지 않고 암모니아, 요소 등은 균의 생육에 조금 영향을 미침을 관찰할 수 있었다. 특히 리신과 메티오닌은 가장 균의 생육에 영향을 미치는 것으로 나타났다. 무기염류의 영향에서는 황산마그네슘, KH-2PO_4, 염화마그네슘 등은 큰 영향을 미치지 못하였고 황산철, 염화제일철, 염화제이철은 생육을 억제하는 것으로 나타났다. 식품첨가물의 Helicobacter pylori의 생육 정도를 검토한 것으로, 그 다음이 질화나트륨, 염화나트륨 순이었으며 토코페롤은 전혀 생육에 영향을 미치지 못하였다. The purpose of this study was to choose principal food components contained in diet foods and food additives used for manufacturing processed foods, and elucidate their in vivo effects on the growth pattern of Helicobacter pylori. To do this, the antibacterial effects of various sources of carbon, nitrogen and mineral as an effect agent on Helicobacter pylori were first assessed based upon bacterial growth degree. Results show that the source of carbon tested had different effects on bacterial growth of Helicobacter pylori. It was revealed that a promotional effect of monosaccharides resulted in enhanced growth of Helicobacter pylori compared with disaccharides and polysaccharides. In particular, glucose was observed to be most effective in growth of Helicobacter pylori among monosaccharides tested, whereas mannose to hinder the growth of Helicobacter pylori. Polyols such as sorbitol, mannitol, maltitol and xylitol was, however, observed to show no promotion or suppression effect on growth of Helicobacter pylori. Apart from these, the sources of amino acid and inorganic nitrogen were chosen and tested to assess the promotion or suppression effect of nitrogen sources on growth of Helicobacter pylori. It was found that amino acid such as lysine showed its promotion effect on the growth of Helicobacter pylori, while arginine, (NH_1)_2SO_4 and NH_4Cl showed no effect on its growth. Ammonia and urea were, however, observed to have a positive effect on the growth of Helicobacter pylori. Among these effect agents, lysine and methionine were revealed to show the most positive effect on growth of Helicobacter pylori. Minerals such as MgSO_4, KH_2PO_4, and MgCl_2 appeared to exert their positive growth effects, whereas CaCl_2, and CaSO_4 had a little effect. In addition FeSO_4, FeCl_2, and FeCl_3 brought suppression on the growth of Helicobacter Pylori. In studies of the growth of Helicobacter pylori by food additives, ascorbic acid showed extreme suppression on its growth. Sodium nitrate and sodium chloride were also found to be of negative effect on the growth of Helicobacter pylori in order of degree, whereas tocopherol had nothing to do with microbial growth.

      • 全南 海南郡 數個 小河川의 魚類相

        1998년 5월과 1989년 3월부터 7월까지 全南 海南郡의 小河川에 대한 魚類相을 調査하였다. 12개의 調査地点에서 채집된 魚類는 총 14科 36屬 44種 8,204個體로 그 중 1차 淡水 魚가 26種이었으며, 韓國特産種은 Squalidus gracilis majimae, Acanthorhodeus gracilis, Microphysogobio yaluensis, Rhodeus uyekii, Cobitis longicorpus, Liobagrus mediadiposalis, Odontobutis platycephala 등 7종이었다. 계곡천에서 채집된 어류는 13종, 옥천천에서는 24종, 삼산천에서는 23종, 고현천에서는 17종, 현산천에서는 15종, 구산천에서는 27종 그리고 송지천에서는 12종의 어류가 채집되었다. 해남군의 소하천에서는 Zacco platypus, Rhodeus uyekii, Zacco temmincki, Squalidus gracilis majimae, Cobitis longicorpus 등이 풍부하게 분포한 종으로 나타났다. 삼산천에서 Lefua costata, 송지천에서 Gasterosteus aculeatus aculeatus가 채집된 것이 특기할 점이었다. The freshwater fish fauna was investigated from May, 1988 to July, 1989 in streams of Haenam-gun, Chollanam-do, Korea. The collected fishes were 14 families, 36 genera, 44 species, 8,204 individuals, and among them 7 species were known as Korean endemic species and 26 species were primary freshwater fisher. The collected fishes were 13, 24, 23, 17, 15, 27 and 12 species in Kyegok-ch'on, Okch'on-ch'on, Samsan-ch'on Kohyon-ch'on, Hy0nsan-ch'on, Kusan-ch'on and Songji-ch'on respectively. In these studied area, Zacco platypus, Rhodeus uyekii, Zacco temmincki, Squalidus gracilis majimae, Cobitis longicorpus were collected abudantly. It was remarkable that Lefua costata was collected in Samsan-ch'on and Gasterosteus aculeatus aculeatus was collected in Songji-ch'on.

        The freshwater fish fauna was investigated from May, 1988 to July, 1989 at eleven stations in Tamjingang. The collected fishes were 12 families, 28 genera, 36 species, 5,633 individuals, and among them 9 species were known as Korean endemic species and 29 species were primary freshwater fishes. In these studied area, Zacco platypus, Z. temmincki, Microphysogobio yaluensis, Rhodeus uyekii. Cobitis longicorpus, C. lutheri were collected abundantly but Pseudorasbora parva, Abbottina rivularis, Acheilognathus limbata, A. yamatsutae, Pseudobagrus fulvidraco, Liobagrus mediadiposalis were rare species.

      • 全南地方 4個 小河川의 漁類相

        The freshwater fish fauna was investigated from April, 1987 to August, 1988 in Watan chon, Pulgap chon, Ch'ong chon and Ch'onggye chon of Chol-lanamdo. The collected fish were 6 families, 19 genera, 22 species in Watan chon 9 families, 20 genera, 25 species in Pulgap chon, 7 families, 17 genera, 17 species in Ch'ong chon and 7 families, 13 genera, 15 species in Ch'onggye chon. It was remarkable point in the distributional study of freshwater fishes of Korea that two individuals of Cobitis koreensis were collected in Pulgap chon for the first time and the morphology of them was very similar to the Tongjin River population. This indicated that Pulgap chon was found to be the southern limit of distribution of C. koreensis koreensis. Growth rate and age composition were analyzed by total length of 6 species such as Squalidus gracilis majimae, Abbottina springeri, A. rivularis, Moroco oxycephalus, Cobitis lutheri and Zacco temmincki.
