We hypothesized that supplementing finishing diets with palm oil would promote adipogenic gene expression and stearoyl-CoA desaturase (SCD) gene expression in subcutaneous (s.c.) and intramuscular (i.m.) adipose tissues of feedlot steers. Eighteen Angus and Angus crossbred steers were assigned to three groups of 6 steers and fed a basal diet (control), with 3% palm oil, or with 3% soybean oil, for 70 d, top-dressed daily. Tailhead s.c. adipose tissue was obtained by biopsy at 14 d before the initiation of dietary treatments and at 35 d of dietary treatments. At slaughter, after 70 d of dietary treatment, tailhead s.c. adipose tissue and i.m. adipose tissue were obtained from the longissimus thoracis muscle. Palm oil increased plasma palmitic acid and soybean oil increased plasma linoleic acid and ${\alpha}$-linolenic acid relative to the initial sampling time. Expression of AMP-activated protein kinase alpha ($AMPK{\alpha}$) and peroxisome proliferator-activated receptor gamma ($PPAR{\gamma}$) increased between the initial and intermediate biopsies and declined thereafter (p<0.03). SCD gene expression did not change between the initial and intermediate biopsies but declined by over 75% by the final period (p = 0.04), and G-coupled protein receptor 43 (GPR43) gene expression was unaffected by diet or time on trial. Soybean oil decreased (p = 0.01) $PPAR{\gamma}$ gene expression at the intermediate sample time. At the terminal sample time, $PPAR{\gamma}$ and SCD gene expression was less in i.m. adipose tissue than in s.c. adipose tissue (p<0.05). $AMPK{\alpha}$ gene expression was less in s.c. adipose tissue of palm oil-fed steers than in control steers (p = 0.04) and CCAAT enhancer binding protein-beta ($CEBP{\beta}$) gene expression was less in s.c. and i.m. adipose tissues of palm oil-fed steers than in soybean oil-fed steers (p<0.03). Soybean oil decreased SCD gene expression in s.c. adipose tissue (p = 0.05); SCD gene expression in palm oil-fed steers was intermediate between control and soybean oil-fed steers. Contrary to our original hypothesis, palm oil did not promote adipogenic gene expression in s.c. and i.m. adipose tissue.

Objectives: HIF-1β (ARNT) formed dimerization of HIF-1α, involved in various aspects of cancer biology, including proliferation and survival under hypoxic condition. This is an in vitro study that investigated the mechanism by which small interference HIF-1β (siHIF-1β) inhibits HIF-1α dimerization and thus it lead to suppress tumor cell growth. Method: Three HCC cell lines (HepG2, Huh 7 and Hep3B) were transfected with HIF-1β coding siRNA and cultured under hypoxic conditions (1% O2, 24 hours). Following transfection, the expression levels of HIF-1β , HIF-1α, and growth factors were examined through immuno-blotting. In addition, tumor growth was measured by MTT assay and tumor activity was measured by TUNEL, tumor invasion and migration assay. Results: The inhibition of HIF-1β by small interference RNA (siHIF-1β) led to suppression of tumor cell growth through inhibition of HIF dimerization under hypoxia condition. This effect was increased and maximized at 48 hrs after treatment of HIF-1β coding siRNA. Inhibition of HIF dimerization through treatment of siHIF-1β also regulated the expression of tumor growth related factors (VEGF, EGF and HGF). Inhibition of HIF- dimerization precluded tumor invasion and migration in variable HCC cell lines. Conclusions: Inhibition of HIF dimerization by siHIF-1β may lead to anti-tumor effects in a hypoxic condition of tumor environment.

Pulmonary pneumatoceles are air-filled thin-walled spaces within the lung and are rare in adult cases of pneumonia. We report the case of a 74-year-old male who was admitted with a cough and sputum production. He had been treated with oral dexamethasone since a brain tumorectomy 6 months prior. Contrast-enhanced computed tomography (CT) of the chest revealed a large pneumatocele in the right middle lobe and peripheral pneumonic consolidation. Bronchoalveolar lavage was performed; cultures identified extended-spectrum β-lactamase (ESBL) producing Proteus mirabilis. A 4-week course of intravenous ertapenem was administered, and the pneumatocele with pneumonia resolved on follow-up chest CT. To the best of our knowledge, this is the first reported case of pulmonary pneumatocele caused by ESBL-producing P. mirabilis associated with pneumonia.

Background: Acne is a inflammatory skin disorders caused by inflammatory biomarkers. Magnesium ascorbyl phosphate (MAP) is a stable precursor of vitamin C. It achieves a constant delivery of vitamin C into the skin and has antioxidative effects. Objectives: To evaluate changes in the expression of inflammatory biomarkers after treatment of cultured sebocytes with MAP Methods: RT-PCR, ELISA and western blotting were performed for inflammatory cytokines, matrix metalloproteinases (MMPs), antimicrobial peptides (AMPs) and keratinocyte differentiation markers before and after treatment of cultured sebocytes with MAP (10-2 mol), lipopolysaccharide (LPS) and a combination of MAP and LPS. RT-PCR and western blotting were also done for Toll-like receptor-4 (TLR-4) and NF-ĸB before and after treatment of cultured sebocytes with MAP, LPS and a combination of MAP and LPS. Results: The expressions of inflammatory cytokines (e.g., IL-1 β and IL-8) and MMPs (e.g., MMP-9) were decreased after treatment with MAP and a combination of MAP and LPS, and increased after treatment with LPS. AMPs (e.g., LL-37) were decreased after treatment with MAP and a combination of MAP and LPS, and increased after treatment with LPS. The expression of TLR-4 and NF-ĸB was decreased after treatment with MAP and a combination of MAP and LPS, and increased after treatment with LPS. Conclusion: MAP, which is a stable precursor of vitamin C, may be a good effective and alternative agent to improve inflammatory reaction in acne.
Clinical outcome of high-dose-rate interstitial brachytherapy in patients with oral cavity cancer

Purpose: To evaluate the clinical outcome of high-dose-rate (HDR) interstitial brachytherapy (IBT) in patients with oral cavity cancer. Materials and Methods: Sixteen patients with oral cavity cancer treated with HDR remote-control afterloading brachytherapy using 192Ir between 2001 and 2013 were analyzed retrospectively. Brachytherapy was administered in 11 patients as the primary treatment and in five patients as salvage treatment for recurrence after the initial surgery. In 12 patients, external beam radiotherapy (50–55 Gy/25 fractions) was combined with IBT of 21 Gy/7 fractions. In addition, IBT was administered as the sole treatment in three patients with a total dose of 50 Gy/10 fractions and as postoperative adjuvant treatment in one patient with a total of 35 Gy/7 fractions. Results: The 5-year overall survival of the entire group was 70%. The actuarial local control rate after 3 years was 84%. All five recurrent cases after initial surgery were successfully salvaged using IBT ± external beam radiotherapy. Two patients developed local recurrence at 3 and 5 months, respectively, after IBT. The acute complications were acceptable (≤grade 2). Three patients developed major late complications, such as radio-osteonecrosis, in which one patient was treated by conservative therapy and two required surgical intervention. Conclusion: HDR IBT for oral cavity cancer was effective and acceptable in diverse clinical settings, such as in the cases of primary or salvage treatment.

Background/Aims: Hypoxia-inducible factor-1α (HIF-1α) is a central transcriptional factor involved in the cellular responses related to various aspects of cancer biology, including proliferation, survival, and angiogenesis, and the metabolism of the extracellular matrix in hypoxia. This study evaluated whether adenovirus-mediated small hairpin RNA (shRNA) against HIF-1α (shHIF-1α) inhibits cell proliferation and angiogenesis in hepatocellular carcinoma (HCC) cell lines. Methods: Knockdown of HIF-1α expression was constructed by adenovirus-mediated RNA interference tools, and HCC cell lines infected with shHIF-1α coding virus were cultured under a hypoxia condition (1% O2) for 24 hours. Following infection, the expression levels of HIF-1α, angiogenesis factors, and matrix metalloproteinase (MMP) were examined using Western blotting. Cell proliferation and angiogenesis were measured by a cell proliferation assay (MTT assay) and an angiogenesis-related assay (invasion and tube-formation assay), respectively. Results: Adenovirus mediated inhibition of HIF-1α induced suppression of tumor growth in HCC cell lines. It also down-regulated the expression of angiogenesis factor and MMP proteins. Angiogenesis as well as mobility of vascular cells to tumor was suppressed by adenovirus-mediated shHIF-1α-infected groups in human umbilical vein endothelial cells (HUVECs). Conclusions: These data suggest that adenovirus-mediated inhibition of HIF-1α inhibits the invasion, tube formation, and cell growth in HUVECs and HCC cells. (Korean J Hepatol 2010;16:280-287)
Growth and Yield Variation of Clay-coated Rice Seeds in Direct Seeding Culture on Dry Paddy

Clay-coated rice seeds (clay-coated seeds A and B) were directly sown on dry paddy and their growth and yield were compared with the normal drill-sown seeds on dry flat paddy. In clay-coated seeds, germination was 1 day earlier and the emergence rate was higher up to 5% than that of normal drill-sown seeds. But the apparent number of seedling stand per m2 was lower than that of normal drill-sown seeds, which is due to the smaller amount of seeding in clay-coated seeds. At the early growth stage, the plant height of clay-coated seeds A was taller than that of drill-sown seeds, while the plant height of clay-coated seeds B was 0.7 cm shorter than that of drill-sown seeds. At the late growth stage, however, the difference was insignificant in both cases. The maximum tillering stage was 10 days earlier in drill-sown seeds. Lodging index was the lowest in clay-coated seeds B and there was no difference between clay-coated seeds A and drill-sown seeds. The ratio of stem base weight, culm diameter and culm wall thickness were higher in clay-coated seeds, while the lower internodes (4th, 5th and 6th) length was shorter in claycoated seeds than in drill-sown seeds. In clay-coated seeds, the number of panicle per m2 was smaller, while the number of spikelet per panicle was a little larger than in drill-sown seeds. The rate of ripened grain and brown rice 1,000 grain weight were lower in the clay-coated seeds, thus the yield was 98~99% level of drill-sown seeds. Considering that the amount of seeding in clay-coated seeds was two-thirds of that in drill-sown seeds, it is expected that clay coating method could become an additional technique for direct seeding cultivation.
