      • The Variation of Winter Buds among 10 Selected Populations of Kalopanax septemlobus Koidz. in Korea

        The objective of this study was to understand the conservation of gene resources and provide information for mass selection of winter bud characters among the selected populations of Kalopanax septemlobus Koidz using analysis of variance(ANOVA) tests. The obtained results are shown below; 1. Ten populations of K. septemlobus were selected for the study of the variation of winter bud characters in Korea. The results of the analysis of variance(ANOVA) tests shows that there were statistically significant differences in all of the winter bud characters among those populations. 2. Correlation analysis shows that width between Height and DBH(Diameter at breast height) characters have negative relationship with all of the characters, as ABL(Apical branch length), ABW(Apical branch width), AWBL(Apical branch winter bud length), AWBW(Apical branch winter bud width), ABT(Apical branch No. of thorns), ABLB(Apical branch No. of lateral bud) and LBL(Lateral branch length), LBW(Lateral branch width), LBT(Lateral branch No. of thorns), LBLB(Lateral branch No. of lateral bud). 3. The result of principal component analysis(PCA) for winter buds showed that the first principal components(PC s) to the fourth principal component explains about 78% of the total variation. The first principal component(PC) was correlated with AWBW, LWBW, and LBL and the ratio of ABL/ABW and LBL/LBW out of 16 winter bud characters. The second principal component correlated with ABL, ABW, ABLB, LWBL(Lateral branch winter bud length), and LBW and the ratio of AWBL/AWBW. The third principal component correlated with ABL, ABW, LWBL, LBL, and the ratio of LBL/LBW. The fourth principal component correlated with LBL and the ratio of LWBL/LWBW(Lateral branch winter bud width), LBL/LBW. Therefore, these characters were important to analysis of the variation for winter bud characters among selected populations of K. septemlobus in Korea. 4. Cluster analysis using the average linkage method based on 10 selected populations for the 16 winter bud characters of K. septemlobus in Korea showed a clustering into two groups by level of distance 1.1(Fig. 3). As can be seen in Fig. 3, Group I consisted of three areas(Mt. Sori, Mt. Balwang and Mt. Worak) and Group Ⅱ contisted of seven areas(Suwon, Mt. Chuwang, Mt. Kyeryong, Mt. Kaji, Mt. Jiri, Muan, and Mt. Halla). The result of cluster analysis for winter bud characters corresponded well with principal component analysis, as is shown in Fig. 2.

        Effects of Phenytoin and Diazepam on the Seizure Activity in the Cortical Dysplasia Animal Models

        Dysplasia-associated seizure disorders are markedly resistant to pharmacological intervention. Relatively little research has been conducted studying the effects of antiepileptic drugs(AEDs) on seizure activity in a rat model of dysplasia. We have used rats exposed to methylazoxymethanol acetate(MAM) in utero, an animal model featuring nodular heterotopia, to investigate the effects of AEDs in the dysplastic brain. Pilocarpine was used to induce acute seizure in MAM-exposed and age-matched vehicle-injucted control animals. Field potential recordings were used to monitor amplitude and numbers population spikes, and paired pulse inhibition in response to stimulation of commissural pathway. Two commonly used AEDs were tested: diazepam 5, 2.5mg/kg;phenytoin 40, 60mg/kg. Diazepam(DZP) and phenytoin(PHT) reduced the amplitude of population spike in control and MAM-exposed rats. However, the amplitude of population spike was nearly eliminated in control rats as compared to the MAM-exposed rats. Pharmaco-resistance was tested by measuring seizure latencies in awake rats after pilocarpine administration(320mg/kg, i.p.) with and without pretreatment with AEDs. Pre-treatment with PHT 60 mg prolonged seizure latency in control rats, but not in MAM-exposed animals. The main findings of this study are that acute seizures initiated in MAM-exposed rats are relatively resistant to standard AEDs assessed in vivo. These data suggest that animal model with cortical dysplasia can be used to screen the effects of potential AEDs.

        Assessment of In Vitro Assay System for Thyroid HormoneDisruptors Using Rat Pituitary GH3 Cells

        The development of in vitro assays has been recommended to screening and test-ing the potential endocrine disruptors (EDs). These assay systems focus only on identifying thethe thyroid hormone (TH) disruptors. The aim of this study was to evaluate a test system to detectTH disruptors using rat pituitary tumor GH3 cells. The test system is based on the TH-dependentincrease in growth rate. As expected, L-3,5,3-triiodothyronine (T3) markedly induced a morphologicalchange in GH 3 cells from flattened fibroblastic types to rounded or spindle-shaped types. T3 stimu-lated GH3 cell growth in a dose-dependent manner with the maximum growth-stimulating effect9 M. In addition, T3 increased the release of growth hor-mone and prolactin into the medium of the GH3 cells culture. Using this assay system, the TH-dis-rupting activities of bisphenol A (BPA) and its related compounds were examined. BPA,dimethylbisphenol A (DMBPA), and TCI-EP significantly enhanced the growth of GH3 cells in therange of 1 × 10-5M to 1 × 10-6M concentrations. In conclusion, this in vitro assay system might bestandardization before it can be used as a broad-based screening tool.

        In this paper, it has been proposed an automatic vegetable bundling mechanism that makes use of heat melt sticking. The proposed mechanism is consisted of three modules for the rationality of manufacture. Design specification has been determined by the bundling mechanism that was optimized mechanically. And, it has been manufactured the prototype of the automatic bundling machine. It has been carried out directly the field test for verification of performance in farmhouse which produces vegetable. In field test, this bundling machine showed efficiency of about 3.6 times than the manual working by manpower, and other performance it got satisfied result. This automatic vegetable-bundling machine was already registered as the agricultural machine. and This machine will be commercialized soon.

        An Allium-odorum is difficult material to handling, because it is soft and weak as mechanical. For bundling work of allium-odorum, we must grip in the bundle shape of unit weight, however, it is difficult that grip allium-odorum by hand because the bundle bulk is very big. Especially, in packing work, the bundle shape of allium-odorum package than other vegetables is very important, because difference of a price becomes much according to the bundle shape. In this paper, we propose bundling mechanism for a quadrangle shape in bundle shape, and semi-automatic bundling mechanism that makes use of a eccentric roller and a triangularity link. We carry out mechanical modeling and analysing respectively using the CATIA V5 and SimDesigner. We manufactured the prototype of the semi-automatic bundling machine, and got satisfied result through field test. And this machine will be commercialized and announced soon.

        An Analysis of the Changes in Bone Mineral Density in Long-Stay Patients of a Geriatric Hospital in Relation to Physical Therapy

        The purpose of this study is to examine the changes in bone mineral density in long-stay patients of a geriatric hospital in relation to physical therapy, thereby providing basic data for preventing the onset or deterioration of osteoporosis in inpatients of geriatric hospitals or relevant facilities. The subjects of this study were 133 elderly patients who had been in H geriatric hospital in Yongin-si for more than four years. Their bone mineral density T-scores at admission and those after four years were measured to compare and analyze the changes in bone mineral density in relation to physical therapy. According to the results, the decrease in T-score for the group without physical therapy was greater by .40 than that for the group with physical therapy, indicating a statistically significant difference between the two groups(p<.01). Accordingly, physical therapy proved to be effective in preventing bone mineral density loss and maintaining the current density.

        Outcome of Multipair Donor Kidney Exchange by a Web-Based Algorithm

        <P>Donor kidney exchange is an established method to overcome incompatibility of donor-recipient pairs (DRP). A computerized algorithm was devised to exchange donor kidney and was tested in a multicenter setting. The algorithm was made according to the consensus of participating centers. It makes all possible exchange combinations not only between two incompatible DRP but also circularly among three DRP and selects an optimum set of exchange combinations, considering several factors that can affect the outcome of the exchanged transplant. The algorithm was implemented as a web-based program, and matching was performed five times. Fifty-three DRP were enrolled from five transplant centers. The numbers of DRP that were enrolled in each matching were 38 (25:13), 39 (34:5), 33 (31:2), 32 (28:4), and 34 (30:4) (carryover:newcomer). The numbers of generated exchange combinations were 4:11, 3:17, 2:12, 2:3, and 2:3 (two-pair exchange:three-pair exchange), and the numbers of DRP in selected exchange combinations were six, 12, six, five, and four in each matching. The numbers of DRP with blood type O recipient or AB donor were five and one, respectively, in selected exchange combinations. Six DRP of two-pair exchange combinations and six DRP of three-pair exchange combinations underwent transplantation successfully. Computerized algorithm of donor kidney exchange was tried not only between two incompatible DRP but also circularly among three DRP. It showed that the algorithm has potential to improve the outcome of donor kidney exchange, especially for disadvantaged DRP with blood type O recipients or AB donors.</P>

        The Effects of NEES on PARP Expression and Cell Death in Rat Cerebral Cortex After Ischemic Injury

        The majority of strokes are caused by ischemia and result in brain tissue damage, leading to problems of the central nervous system including hemiparesis, dysfunction of language and consciousness, and dysfunction of perception. The purpose of this study was to investigate the effects of Poly(ADP-ribose) polymerase(PARP) on necrosis in neuronal cells that have undergone needle electrode electrical stimulation(NEES) prior to induction of ischemia. Ischemia was induced in male SD rats(body weight 300g) by occlusion of the common carotid artery for 5 min, after which the blood was reperfused. After induction of brain ischemia, NEES was applied to Zusanli(ST 36), at 12, 24 and 48 hours. Protein expression was investigated using immuno-reactive cells, which react to PARP antibodies in cerebral nerve cells, and Western blotting. The results were as follows: In the cerebral cortex, the number of PARP reactive cells after 24 hours significantly decreased(p<.05) in the NEES group compared to the GI group. PARP expression after 24 hours significantly decreased(p<.05) in the NEES group compared to the GI group. As a result, NEES showed the greatest effect on necrosis- related PARP immuno-reactive cells 24 hours after ischemia, indicating necrosis inhibition, blocking of neural cell death, and protection of neural cells. Based on the results of this study, NEES can be an effective method of treating dysfunction and improving function of neuronal cells in brain damage caused by ischemia.

        Hepatoprotective and Antioxidant Capacities of Paecilomyces japonica and Cordyceps sinensis in Rats with CCl₄-Induced Hepatic Injury

        Antioxidant and hepatoprotective activities of the fruiting bodies of Paecilomyces japonica (PJ) grown on silk-worm larvae and of Cordyceps sinensis (CS), a wild form of Cordyceps, were investigated. The 70% ethanol extracts of both fungi, PJ and CS, were found to exhibit free radical scavenging effects on 1,1-diphenyl-2-picrylhydrazyl (DPPH). The inhibitory potency of PJ (IC?? = 163 ㎍ㆍ㎖?¹) was approximately 4.5 folds that of CS (IC?? = 731 ㎍ㆍ㎖?¹). They exhibited a moderate decrease in serum transaminase activities elevated by hepatic damage induced by carbon tetrachloride (CCl₄)-intoxication in rats. Both fungal extracts also exhibited not only significant increases in hepatic superoxide dismutase (SOD) and catalase at a dose of 300 ㎍ㆍ㎏?¹ in CCl₄-intoxicated rats, but also a significant decrease in malondialdehyde (MDA) production in a thiobarbituric acid (TBA) reactant assay in rats. Although no significant glutathione peroxidase (GSH-px) activity was detected, SOD and catalase elevations were obtained.
