        The purpose of this study is to determine how a Taekwondo player's athletic satisfaction affects his/her psychological health and overall personal happiness. A questionnaire survey was carried out on a sample group of 200 students attending colleges located in Seoul and Gyeonggi Province. Then, 23 questionaries deemed not trustworthy were eliminated, leaving 178 questionaries for analysis. The questionnaire, which contained total 128 questions, showed the confidence level falling between .5241 and .8945 (Cronbach's alpha). The study, which relied on frequency analysis, t-test, One-way ANOVA, and Scheffe test, produced the following results. First, athletic satisfaction showed a statistically significant difference (5% range) depending on sex, school year or athletic experience. Second, psychological health resulted in a statistically significant difference (5% range) depending on sex or athletic experience while no difference was noticed regarding school year. Third, happiness in life showed a statistically significant difference (5% range) depending on sex or school year while no difference was noticed with regard to athletic experience.

      • Characterization of endoplasmic reticulum oxidoreductase (ERO1) gene from Bombyx mori

        Disulfide bond formation, reduction and isomerization are important posttranslational modification in proteins that occur in most, if not all, living organisms. In eukatoyes, disulfide bond in substrate proteins are primarily formes by ERO1 and PDI. ERO1, oxidized by molecular oxygen, acts as a specific oxidant of PDI, which then makes disulfide bonds in folding proteins oxidized directly. It means that ERO1 plays an essential role in setting the redox potential in the ER, and the regulation of Ero1p activity is critical to maintain redox homeostasis and proper ER folding activity. We have isolated and analysed a endoplasmic reticulum oxidoreductase (ERO1) from Bombye mori. It apperas that both an N-terminal CxxxxC motif and a C-terminl CxxCxxC motif are necessary for Ero1p fuction. In vivo, the result of the 5day of 5th instar larvae by RT-PCR and Real-Time PCR shows that posterior silkgland, skin and mid silkgland are revealed more than rhose of other tissues. The same result for tissue distribution of transcripts is appeared about ERO1 and PDI. In Bombyx mori, ERO1 is also supposed to correlate with PDI. Afterwards, more experiments are needed to figure out accurate interrelation between ERO1 and PDI.

        Effects of green tea or Sasa quelpaertensis bamboo leaves on plasma and liver lipids, erythrocyte Na efflux, and platelet aggregation in ovariectomized rats

        This study was conducted to investigate the effects of Sasa quelpaertensis bamboo and green tea on plasma and liver lipids, platelet aggregation, and erythrocyte membrane Na channels in ovariectomized (OVX) rats. Thirty female rats were OVX, and ten female rats were sham-operated at the age of 6 weeks. The rats were divided into four groups at the age of 10 weeks and fed the experiment diets: sham-control, OVX-control, OVX-bamboo leaves (10%), or OVX-green tea leaves (10%) for four weeks. Final body weight increased significantly in the OVX groups compared with that in the sham-control, whereas body weight in the OVX-green tea group decreased significantly compared with that in the OVX-control (P < 0.01). High density lipoprotein (HDL)-cholesterol level decreased in all OVX groups compared with that in the sham-control rats (P < 0.05) but without a difference in plasma total cholesterol. Plasma triglycerides in the OVX-green tea group were significantly lower than those in the sham-control or OVX-control group (P < 0.05). Liver triglycerides increased significantly in the OVX-control compared with those in the sham-control (P < 0.01) but decreased significantly in the OVX-green tea group compared with those in the OVX-control or OVX-bamboo group (P < 0.01). Platelet aggregation in both maximum and initial slope tended to be lower in all OVX rats compared with that in the sham-control rats but was not significantly different. Na-K ATPase tended to increase and Na-K cotransport tended to decrease following ovariectomy. Na-K ATPase decreased significantly in the OVX-green tea group compared with that in the OVX-control group (P < 0.01), and Na-K cotransport increased significantly in the OVX-bamboo and OVX-green tea groups compared with that in the OVX-control (P < 0.05). Femoral bone mineral density tended to be lower in OVX rats than that in the sham-control, whereas the green tea and bamboo leaves groups recovered bone density to some extent. The results show that ovariectomy caused an increase in body weight and liver triglycerides, and that green tea was effective for lowering body weight and triglycerides in OVX rats. Ovariectomy induced an increase in Na efflux via Na-K ATPase and a decrease in Na efflux via Na-K cotransport. Furthermore, consumption of green tea and bamboo leaves affected Na efflux channels, controlling electrolyte and body water balance.

      • Molecular characterization of heat shock protein 70 promoter from the silkworm, Bombyx mori

        Recently Transgenesis was achieved in Bombix mori. For stable and effective transgenesis in B.mori, B.mori cytoplasmic actin gene (BmA3) promoter was used to expression of marker gene, the green fluorescent protein(GFP). Green fluorescent protein expression for selection of transformants was visible in all larval, pupal, and adult tissues but, unexpectdly, was not detectable in embryos. So, it spend times and money on rearing of silkworm. Furthermore, the BmA3 promoter is predominantly active in the midgut, which makes it difficult to reliably identify transformants since autofluorescence of many insect foods can mask low-level fluorescence and only allows the detection of strongly expressing individuals with potentially multiple insertions. Therefore, we need more intensely promoter than BmA3 promoter for selected by expression of GFP in embryos and selected by reliable expression of GFP in larvae. We performed dot blot hybridization to develop strong promoter. Nine differentially expressed clones were isolated and we focused one clone of them which has high similarity with heat shock protein 70 gene from D.melanogaster. We named it as bHSP70 (Bombyx mori heat shock protein 70). Expression from the hsp70 promoter was strong and heat shock-dependent. And Drosophila hsp70 promoter appears useful for regulating expression of Exogenous DNA. So, we analyzed transcriptional activity of promoter with bHSP70 gene by using dual luciferase assay system. bHSP70 promoter has about 264 folds more intensely than BmA3 promoter. Also, when bHSP70 promoter treated heat shock(42℃), transcriptional activity incresed 2 times more than normal condition. Therefore, we suggest that bHSP70 promoter is more effective candidate for stable transformation and selection of transformants.

      • Intracellular Protein Delivery System Using a Target-Specific Repebody and Translocation Domain of Bacterial Exotoxin

        <P>With the high efficacy of protein-based therapeutics and plenty of intracellular drug targets, cytosolic protein delivery in a cell-specific manner has attracted considerable attention in the field of precision medicine. Herein, we present an intracellular protein delivery system based on a target-specific repebody and the translocation domain of <I>Pseudomonas aeruginosa</I> exotoxin A. The delivery platform was constructed by genetically fusing an EGFR-specific repebody as a targeting moiety to the translocation domain, while a protein cargo was fused to the C-terminal end of the delivery platform. The delivery platform was revealed to efficiently translocate a protein cargo to the cytosol in a target-specific manner. We demonstrate the utility and potential of the delivery platform by showing a remarkable tumor regression with negligible toxicity in a xenograft mice model when gelonin was used as the cytotoxic protein cargo.       • BBT-877 is a potent autotaxin inhibitor in clinical development to treat idiopathic pulmonary fibrosis

        Background: Idiopathic Pulmonary Fibrosis (IPF) is a progressive, irreversible and fatal lung disease with unmet medical needs. Autotaxin (ATX) is an extracellular enzyme involved in the generation of lysophosphatidic acid (LPA). Preclinical and clinical data have suggested the ATX - LPA - LPA receptor axis plays a pivotal role in the pathogenesis and the progression of IPF. Methods: The inhibitory potency of BBT-877, an orally available small molecule inhibitor of ATX, was measured by FS-3 and plasma lysophosphatidylcholine (ex vivo) assays. Transwell cell migration assay was used to quantify in vitro anti-migratory efficacy of BBT-877. In mouse disease model, bleomycin was intra-nasally administrated to induce pulmonary fibrosis at day 0, and BBT-877 was given orally twice-a-day from day 7 to 21. In phase 1 clinical trial, there were 80 healthy volunteers, in which 50 - 800 mg (SAD) and 200 - 800 mg QD or 100 - 200 mg BID for two weeks (MAD) doses were administrated. Results: In ex vivo enzymatic assays using human plasma, IC50 of BBT-877 was 6.5 - 6.9 nM (LPA 18:2) whereas that of GLPG1690 was 75 - 132 nM. In the mouse model, BBT-877 treatment showed anti-fibrotic efficacy as revealed by significantly reduced lung weight and Ashcroft score as well as collagen content compared to the vehicle-treated group (Figure 1). During phase 1 clinical trial, only mild adverse events (headache, stomach ache, rash, or etc.) were noted. Pharmacokinetic analysis revealed the dose-proportional increase in systemic exposure with elimination half-life of 12hr. The decrease of plasma LPA level was maintained at 80% or higher for 24hr when 400 mg BBT-877 was administrated. Conclusions: Taken together, nonclinical data suggest BBT-877 is a potent, selective, and potentially best-in-class ATX inhibitor. Phase 1 clinical data demonstrate BBT-877 is a safe and well-tolerated drug with excellent pharmacokinetic-pharmacodynamic profiles.

        화장품 OEM/ODM 산업에서의 한국콜마 성장사례

        [연구목적] 본 연구의 목적은 국내 최초의 화장품 OEM 전문업체로 시작하여 세계 화장품 ODM 업계 2위로 도약한 한국콜마의 성장 과정을 통해 시사점을 도출하는 것이다.[연구방법] 본 연구는 시장 선도자의 우위에 관한 연구와 사업 다각화 전략, 그리고 글로벌화 전략 등에 대한 이론적 고찰을 바탕으로 진행되었다. 한국콜마와 화장품 OEM/ODM 산업에 관한 선행 연구들을 충분히 검토하였고, 한국콜마 윤상현 대표와 실무진들과의 심층적인 면담을 통해 이 기업의 성장을 견인한 전략적 선택과 그 이면의 시사점을 연구하였다.[연구결과] 본 연구를 통해 발견된 한국콜마의 성장전략은 다음과 같다. 첫째, CGMP의 선구적 도입으로 국내 화장품 업계의 생산 표준을 확립하여 글로벌 메이저 고객의 확보 및 해외 진출의 기초를 닦았다. 둘째, 지속적인 연구개발 노력으로 화장품 OEM 산업을 ODM으로 한 단계 업그레이드시키는 데에 이바지하였다. 셋째, 기술 정보의 공유를 통해 시장의 확대와 후발기업의 동반성장을 도모하였다. 넷째, 혁신적 발상으로 새로운 시장을 개척하였다.[연구의 시사점] 한국콜마의 성장전략은 이 기업이 화장품이라는 레드오션 시장에 후발주자로 진입하여 ‘OEM/ODM’이라는 새로운 시장을 개척하여 ‘시장 선도자’의 위치를 점하였다는 데에 시사점이 있다. [Purpose] This paper reviews and foresees the case of a successful global cosmetic company, Kolma Korea to provide findings for following companies.[Methodology] For this purpose, various thesis on ‘first mover advantages’, ‘diversifications’ and ‘globalizations’ were reviewed. Also prior researches on Kolmar Korea were examined. To provide understandings of strategies that Kolmar Korea pursued, consecutive in-depth interviews with CEO and employees of Kolmar Korea were conducted.[Findings] Four strategies were found in the success of Kolmar Korea. First, Kolmar Korea introduced CGMP to establish global standard for manufacturing. Second, Kolmar Korea made a significant contribution to cosmetic market by shifting OEM industry to ODM industry. Third, Kolmar Korea shared technological know-hows with the followers who entered the market later. Fourth, they developed new markets with their inverse thinking.[Implications] Kolmar Korea provides a successful model to companies through globalization and diversification without their own ‘brand.’
