        ‘제3세대’이후 국내 러시아 연구의 현황과 과제 : 사회과학을 중심으로

        Since the mid-1990s, Russian Studies in Korea have been in the stage of "relative stabilization" after undergoing the ups and downs in the previous years. The rise of the "third generation" scholars, many of which returned to Korea after earning their doctoral degrees in Russia, has contributed to this trend. The current research, based on a comprehensive review of social science works on Russia, published as M. A. and Ph.D theses, in book forms and monographs, and as articles in 12 academic journals during the past decade in Korea, shows the following developmental tendencies in the Russian Studies in Korea since the mid-1990s. First, the number of Korean specialists on Russia has consistently increased. Second, the scope of research has been extended in spatial dimension as well as in research topics. In the former, while CIS countries other than Russia have increasingly became the subject of research, the investigation of local politics and economy in Russia has also grown. In the latter, topics have been more diversified beyond the traditional fields of politics, economy, society, and history into education, law, media, anthropology, and political geography. Third, the cases of field researches and surveys have continued to increase, which can contribute to a qualitative progress in the Russian Studies in Korea. Fourth, more research efforts have been devoted to the analysis of various aspects of Russia from a comparative theoretical perspective. Fifth, new attention has been called to the methodological issues in understanding the Russian experiences. Despite these new developments and achievements, however, Russian Studies in Korea still offer the room for continuing improvement. First, compared with those countries with more advanced Russian Studies, the Russian Studies community in Korea is still much small, and there are very few specialists on other CIS countries than Russia. Second, Russian politics and economy still command a disproportionally high position in terms of research topic. Third, researches are lopsidedly tilting toward policy-oriented studies rather than contributing to strengthening the foundation of Russian Studies. Fourth, the previous tendency to approach the Russian experiences from the viewpoint of break with the past has not been fully overcome. Fifth, although efforts have been made for establishing a "Korean style of studying Russia", the Western approaches to the Russian realities still enjoy a disproportionately high popularity. Sixth, strenuous endeavor among the Russian Studies community is required for implementing interdisciplinary researches of Russia as subject of area studies.

      • 뇌기능 장애 환자에서 뇌 SPECT의 유용성

        Objective: To evaluate usefulness of the brain SPECT in brain dysfunction patients who showed low Mini-MentaI State Examination(MMSE) score. Method: Eighty four patients who were visited to a rehabilitation hospital for their brain dysfunction were retropectively studied. We investigated percent of positive finding in brain SPECT, brain CT, MRI, P-300 studies, and correlation of MMSE scores, brain SPECT, P-300 studies, and correlation of brain SPECT, brain CT and MRI, P-300 studies. Resutts: Percent of positive finding in brain SPECT was high than brain CT, MRI, P-300 studies. MMSE scores were positive correlation with brain SPECT, but no correlation with P-300 study(P>0.05). Brain SPECT were significant correlation with brain CT (P<0.05), but no correlation with brain MRI and P-300 studies (P>0.05). P-300 study was no correlation with brain CT, MRI (P>0.05). Conclusion: We concluded that brain SPECT is useful for reflection of brain dysfunction.

      • 난치성 외측 상과명에서 포동당 용액과 자가혈 주사치료의 효과

        Background and Objectives: To investigate the effect of hyperosmolar dextreose and autologous blood injection in the refractory lateral epicondylitis. Materials and Methods: The sunjects consisted of 39 patients suffering refractory lateral epicondylitis. All patients had failed previous nonsurgical treatments including all or combinations of physical therapy, splinting, nonsteroidal anti-inflammatory medication, and prior steroid injections, Sixteen subjects were injected with 15% dextrose solution under the extensor carpi radialis and tender points. Other subjects were injected with 2mL of autologus blood under same area. ResuKs: In the comparison of the VAS before and after treatment, Prolotherapy was 6.38 and 3.44, autologous blood injects was 6.12 and 2.54 respectively, which demonstrated statistically significant decrease..The effect of autologous blood injections was shown to be more effective than that of prolotherapy. Conclusion: After autologous blood injection therapy in whom nonsurgical modlalities had failed were relieved completely of pain even during strenuous activity. This study offers encouraging results of an alternative minimally invasive treatemnet that addresses the pathophysiology of latera epicondylitis that has failed traditional nonsurgical modalities.

      • 들잔디 성숙종자로부터 캘러스배양 및 식물체 재분화에 미치는 몇 가지 요인의 영향

        In an effort to optimize tissue culture responses of zoysiagrass (Zoysia japonica Steud.) for genetic transformation, factors affecting callus induction and plant regeneration were investigated. MS medium containing 3 ㎎/L 2,4-D was optimal for embryogenic callus induction from mature seed. The plant regeneration frequency of 73.3% was observed when embryogenic calli induced in this medium were transferred to N6 medium supplemented with 0.1 ㎎/L 2,4-D and 5 ㎎/L BA. Among several basic media, MS and N6 medium were optimal for callus induction and plant regeneration, respectively. Regenerated plants were grown normally when shoots transplanted to the soil. A rapid and efficient plant regeneration system established in this study will be useful for molecular breeding of turfgrass through genetic transformation.

        Oral Bioavailability and Enterohepatic Recirculation of Otilonium Bromide in Rats

        This study was conducted to examine the oral bioavailability and the possibility of enterohepatic recirculation of otilonium bromide in rats. A sensitive LC/MS/MS assay (LLOQ 0.5 ng/mL) was developed for the determination of otilonium and applied to i.v. and oral administration studies in bile duct cannulated (BDC) and non-BDC rats. After i.v. injection to BDC rats (1 mg/kg as otilonium), average $t_{1/2}$, CL, $V_z$ and AUC were $7.9\;{\pm}\;1.9\;h,\;8.7\;{\pm}\;3.1$ mL/min/kg, $5.7\;{\pm}\;1.4$ L/kg and $2,088\;{\pm}\;676\;ng{\cdot}h/mL$, respectively, and these values were comparable to those found in non-BDC rats. The percentages of i.v. dose excreted unchanged in bile and urine in BDC rats were $11.6\;{\pm}\;3.0$ and $3.1\;{\pm}\;0.7$%, respectively. Upon oral administration to non-BDC rats (20 mg/kg as otilonium), $t_{1/2},\;C_{max},\; T_{max}$ and AUC were $6.4\;{\pm}\;1.3\;h,\;182.8\;{\pm}\;44.6\;ng/mL,\; 1.9\;{\pm}\;1.6\;h$ and $579\;{\pm}\;113\;ng{\cdot}h/mL$, respectively. The absolute oral bioavailability was low (1.1%), while the drug was preferentially distributed to gastrointestinal tissues. A secondary peak was observed in the serum concentration-time profiles in non-BDC rats following both i.v. and oral administration, indicating that otilonium bromide was subject to enterohepatic recirculation.

        Pharmacokinetics of GST-TatdMt, a Recombinant Fusion Protein Possessing Potent Anti-obesity Activity, in Mice

        This study examined the absorption and pharmacokinetic disposition of $^{125}I-GST-TatdMt$, a recombinant Tat protein possessing potent anti-obesity activity, in mice after vascular and extravascular administration. GST-TatdMt was over-expressed in E. coli, purified, and radioiodinated using the IODO-GEN method. $^{125}I-GST-TatdMt$ was administered to mice by i.v., i.p. and oral administration at doses of 652.7 nCi (102.3 ${\mu}g$). Upon i.v. injection, the average terminal elimination half-life ($t_{1/2,{\lambda}z}$), AUC and AUMC were 6.4 h, 318.2 $nCi{\cdot}h/mL$ and 2518 $nCi{\cdot}h^{2}/mL$, respectively. The highest radioactivity was observed in lung followed by liver, spleen, heart and kidney. The $t_{1/2,{\lambda}z}$ values obtained from i.v., i.p., and oral administration were comparable from each other (range 5.8-6.4 h). The absolute bioavailability of $^{125}I-GST-TatdMt$ was 42.8% and 60.5% after p.o. and i.p. administration, respectively. Given the cell-penetrating nature, $^{125}I-GST-TatdMt$ may be absorbed into the systemic circulation to a relatively high extent after extravascular administration.

      • Quantitative Determination of Absorption and First-Pass Metabolism of Apicidin, a Potent Histone Deacetylase Inhibitor

        <P>Apicidin, a potential oral chemotherapeutic agent, possesses potent anti-histone-deacetylase activity. After oral administration, the total bioavailability of apicidin is known to be low (14.2%–19.3%). In the present study, we evaluated the factors contributing to the low bioavailability of apicidin by means of quantitative determination of absorption fraction and first-pass metabolism after oral administration. Apicidin was given to rats by five different routes: into the femoral vein, duodenum, superior mesenteric artery, portal vein, and carotid artery. Especially, the fraction absorbed (<I>F</I><SUB>X</SUB>) and the fraction that is not metabolized in the gut wall (<I>F</I><SUB>G</SUB>) were separated by injection of apicidin via superior mesenteric artery, which enables bypassing the permeability barrier. The <I>F</I><SUB>X</SUB> was 45.9% ± 9.7%, the <I>F</I><SUB>G</SUB> was 70.9% ± 8.1% and the hepatic bioavailability (<I>F</I><SUB>H</SUB>) was 70.6% ± 12.3%, while the pulmonary first-pass metabolism was minimal (<I>F</I><SUB>L</SUB> = 102.8% ± 7.4%), indicating that intestinal absorption was the rate-determining step for oral absorption of apicidin. The low <I>F</I><SUB>X</SUB> was further examined in terms of passive diffusion and transporter-mediated efflux by in vitro immobilized artificial membrane (IAM) chromatographic assay and in situ single-pass perfusion method, respectively. Although the passive diffusion potential of apicidin was high (98.01%) by the IAM assay, the in situ permeability was significantly enhanced by the presence of the P-glycoprotein (P-gp) inhibitor elacrider. These data suggest that the low bioavailability of apicidin was mainly attributed to the P-gp efflux consistent with the limited <I>F</I><SUB>X</SUB> measured in vivo experiment.</P>

        Orally administrated drugs permeate the biological membrane by various transport mechanisms. The oral absorption potential is closely related to the physicochemical properties of the drug and interaction with the physiological factors surrounding the site of absorption. Assessment of the drug membrane permeability is an integral part of the early stage drug developmental process. Appropriate selection of the permeability screening method at the right stage of drug development process is important in achieving successful developmental outcomes. This review aims at introducing currently available in vitro and in vivo screening methods for the membrane permeability assessment.

        Rapid Quantification of Topotecan in Biological Samples by Liquid Chromatography/Tandem Mass Spectrometry

        A rapid liquid chromatography/tandem mass spectrometry (LC/MS/MS) assay method was developed for the determination of topotecan levels in rat serum. The assay utilized a single liquid-liquid extraction with a mixture of ethy l acetate and acetonitrile (6:1 v/v) and isocratic elution. The multiple reaction monitoring was based on the transition of m/z 422.0$\rightarrow$376.5 for topotecan and 315.1$\rightarrow$226.6 for clomipramine (internal standard). The developed assay was validated to demonstrate the specificity, recovery, lower limit of quantification (LLOQ), accuracy and precision. The assay was linear over a concentration range from 0.5-100 ng/mL, with LLOQ being 0.5 ng/mL using a small volume of rat serum (0.1 mL). The mean intra- and inter-day assay accuracy was 87.7-111.0% and 97.8-108.3, respectively, and the mean intra- and interday precision was between 1.6-4.3% and 3.8-10.3, respectively. The developed assay was applied to a pharmacokinetic study after a bolus i.v. injection of topotecan in rats.
