<P>In order to isolate grain-specific genes during kernel development, two differential screening methods—suppression subtractive hybridization (SSH) and differential hybridization (DH)—were applied. Two cDNAs encoding hordoindolines known to be related to grain hardness were isolated and designated as <I>Hordeum vulgare</I> indoline a and b (<I>HvIDa</I> and <I>HvIDb</I>). The cDNAs encoding <I>HvIDa</I> and <I>HvIDb</I> contained a 450 bp and a 444 bp open reading frame (ORF) that encoded the putative hordoindoline-a and -b precursors consisting of 150 and 148 amino acids, respectively. The deduced amino acid sequences of both <I>HvIDa</I> and <I>HvIDb</I> contained one tryptophan-rich domain and ten highly conserved cysteine residues. The expression of the <I>HvIDa </I>gene was high at 5 DAF, reached a peak at 8 DAF and decreased slightly until 20 DAF, while the expression of <I>HvIDb</I> began to be detectable at 8 DAF when it was higher than that of other developmental stages and decreased slightly until 20 DAF. The <I>HvIDa</I> and <I>HvIDb</I> genes were predominantly detected in the aleurone cell layers in the late part of kernel development, e.g. at 20 DAF. These findings may provide clues to the molecular mechanisms of kernel development and contribute to the determination of the grain texture in barley.</P>

DNA probes for the l6S rRNA have been designed for the detection of Edwardsiella tarda. In order to accomplish this purpose, the l6S rRNA gene from E. tarda has been cloned and sequenced. Two highly feasible oligonucleotide probe sites have been determined by the database analysis programs presented by PCGENE and BLAST. These two probes have been evaluated by slot blot hybridization analysis. Hetero- and homo-trimeric templates have been synthesized using these two probe sites. The templates have been further multimerized by PCR to generate between 150 and 300 bp long DIG-11-dUTP labeled probes. Unlike 3' end labeled oligonucleotide probes or templates, multimerized probes showed no crosshybridization in the given experimental condition. Furthermore, a significant increase in sensitivity has been observed with these probes. This method, we presented here, may be useful for the designing of probes for the detection of other fish pathogenic microorganisms also.

Background: The purpose of this study is to report a modified transtibial technique to approach the center of anatomical femoral footprint in anterior cruciate ligament (ACL) reconstruction and to investigate the accurate femoral tunnel position with 3-dimensional computed tomography (3D-CT) and radiography after reconstruction. Methods: From December 2010 to October 2011, we evaluated 98 patients who underwent primary ACL reconstruction using a modified transtibial technique to approach the center of anatomical femoral footprint in single bundle ACL reconstruction with hamstring autograft. Their femoral tunnel positions were investigated with 3D-CT and radiography postoperatively. Femoral tunnel angle was measured on the postoperative anteroposterior (AP) radiograph and the center of the femoral tunnel aperture on the lateral femoral condyle was assessed with 3D-CT according to the quadrant method by two orthopedic surgeons. Results: According to the quadrant method with 3D-CT, the femoral tunnel was measured at a mean of 32.94% ± 5.16% from the proximal condylar surface (parallel to the Blumensaat line) and 41.89% ± 5.58% from the notch roof (perpendicular to the Blumensaat line) with good interobserver (intraclass correlation coefficients [ICC], 0.766 and 0.793, respectively) and intraobserver reliability (ICC, 0.875 and 0.893, respectively). According to the radiographic measurement on the AP view, the femoral tunnel angles averaged 50.43° ± 7.04° (ICC, 0.783 and 0.911, respectively). Conclusions: Our modified transtibial technique is anticipated to provide more anatomical placement of the femoral tunnel during ACL reconstruction than the former traditional transtibial techniques.

Background:Intertrochanteric fractures of the femur are the most common type of fracture, and are an increasing occurrence due to the aging of the population. The objectives of our study are to predict the fate of intertrochanteric fractures treated with intramedullary hip nails by assessing the postoperative fracture stability utilizing the newly developed scoring system, and to help rehabilitate these patients. Methods: Eighty-two patients with intertrochanteric fractures that were treated with intramedullary hip nails between December, 2004 and January, 2011 were subjected to this study. The patients who could be followed for a minimum of one year postoperatively were enrolled. The immediate postoperative conditions were determined by radiograms: reduction status (3 parameters/4 points: contact accuracy of posteromedial cortex, severity of angulation, and distraction), fixation status (3 parameters/3 points: tip-apex distance, location of tip of the lag screw, entry point of the intramedullary nail), and fracture type (1 parameter/1 point: stable or unstable type by the Kyle’s classification). Postoperative reduction loss and fixation failure were checked by radiograms taken at a minimum 3 months postoperative. Results: Reduction loss and fixation failure were observed in 14 consecutive patients (17%). The fixation failure rate was 100% (2 patients) in score 1, 60% (3 out of the 5 patients) in score 2, 39% (3 out of the 8 patients) in score 3, and 50% (4 out of the 8 patients) in score 4 groups. There were fixation failures only in 1 out of 13 patients with score 5, and in 1 out of 18 patients with score 6. There was no fixation failure in 17 patients with score 7 and 11 patients with score 8. Conclusions: Maintenance of the fracture reduction by the stable fixation in the patient scores over 5 could be predicted by the postoperative radiograms.
Mediation effect of herpes zoster derived by statin use on cardiovascular disease risk

Background/Aims: Although statins are widely used to reduce the risk of cardiovascular disease (CVD) including stroke and myocardial infarction (MI), it is reported that statin use increases the incidence of herpes zoster (HZ) that is associated with increased risk of CVD. So, we evaluated the mediation effect of HZ caused by statin use on CVD. Methods: We analyzed a prospective cohort from the National Health Insurance Service-database of South Korea. All individuals received a medical check-up and were followed-up from 2002 to 2013. Results: A total of 275,382 individuals > 40 years old were followed up for 11 years from 2003. Of these, 11,415 people (4%) were classified as statin users and 263,967 (96%) as non-statin users. Those who used statins had significantly lower risks of cardiovascular events, stroke, and MI compared with non-statin users; the adjusted hazard ratios in the multivariate analysis were 0.90 (95% confidence interval [CI], 0.82 to 0.98), 0.88 (95% CI, 0.80 to 0.98), and 0.91 (95% CI, 0.79 to 1.07), respectively. When we calculated the mediating effect of cardiovascular events by statin use through HZ, 11.6% of the total beneficial effect of cardiovascular events by statin use was mitigated through the occurrence of HZ caused by statin use. This mediating effect was higher in the younger age group (< 60 years). Conclusions: This study showed that statin use reduced CVD by 10%, but the protective effect of statin use against CVD was mitigated by approximately 10% through the development of HZ caused by statin use.

Fresh and chopped aerial parts of Oryza sativa were extracted in 80% aqueous mehthanol, and the concentrated extract was successively partitioned using n-hexane, ethyl acetate (EtOAc), nbutanol, and H2O. From the EtOAc fraction, a new flavonolignan (2) and a known flavonolignan, salcolin B (1), were isolated through repeated SiO2 and octadecyl silica gel column chromatography. Based on NMR, mass spectrometer, and IR spectroscopic data, the chemical structure of compound (2) was determined to be tricin-4`-O-[erythro-β-guaiacyl-(7``-O-methyl)- glyceryl] ether, which has thus far never been reported and was named salcolin C (2). Salcolin B (1) and salcolin C (2) dose-dependently inhibited nitric oxide production in RAW 264.7 cells with IC50 values of 29.8±0.6 and 10.8±1.0 μM, respectively, without visible toxic effect.
