        Genotypic and Phenotypic Diversity of PGPR Fluorescent Pseudomonads Isolated from the Rhizosphere of Sugarcane (Saccharum officinarum L.)

        The genetic diversity of plant growth-promoting rhizobacterial (PGPR) fluorescent pseudomonads associated with the sugarcane (Saccharum officinarum L.) rhizosphere was analyzed. Selected isolates were screened for plant growthpromoting properties including production of indole acetic acid, phosphate solubilization, denitrification ability, and production of antifungal metabolites. Furthermore, 16S rDNA sequence analysis was performed to identify and differentiate these isolates. Based on 16S rDNA sequence similarity, the isolates were designated as Pseudomonas plecoglossicida, P. fluorescens, P. libaniensis, and P. aeruginosa. Differentiation of isolates belonging to the same group was achieved through different genomic DNA fingerprinting techniques, including randomly amplified polymorphic DNA (RAPD), amplified ribosomal DNA restriction analysis (ARDRA), repetitive extragenic palindromic (REP), enterobacterial repetitive intergenic consensus (ERIC), and bacterial repetitive BOX elements (BOX) analyses. The genetic diversity observed among the isolates and rep-PCR-generated fingerprinting patterns revealed that PGPR fluorescent pseudomonads are associated with the rhizosphere of sugarcane and that P. plecoglossicida is a dominant species. The knowledge obtained herein regarding the genetic and functional diversity of fluorescent pseudomonads associated with the sugarcane rhizosphere is useful for understanding their ecological role and potential utilization in sustainable agriculture.

        Flipped Reposition Laminoplasty for Excision of Intradural Extramedullary Tumors in the Thoracolumbar Spine: A Case Series of 14 Patients

        Study Design: A retrospective study was done to assess the outcome of the new technique of flipped reposition laminoplasty for excision of intradural extramedullary (IDEM) spinal tumors of the thoracolumbar region.Purpose: To describe flipped reposition laminoplasty technique and evaluate its outcomes. Overview of Literature: Laminectomy has been the conventional approach for the surgical excision of IDEM spinal tumors, but it has potential postoperative complications. Laminoplasty maintains the posterior arch of the spine and avoids complications seen in Laminectomy, such as instability, epidural scarring, and kyphotic deformity.Methods: Fourteen patients (nine females and five males) diagnosed with IDEM tumors of the thoracolumbar region operated between 2016 and 2018 were included in this study. Pathologically, five cases were schwannomas; four cases were meningiomas; two cases were ependymomas; and one case each was lymphoma, neurofibroma, and teratoma. All patients had their neurological deficits documented using the American Spinal Injury Association (ASIA) impairment scale. After completion of all preanesthetic formalities, the patients were operated upon by a single surgeon using the flipped reposition laminoplasty technique. Follow-up was done at 1, 3, 6, and 12 months post operation and yearly thereafter.Results: The mean age of the patients was 35.28 years (14–65 years), and the mean follow-up duration was 17 months (6–26 months). Two patients were assessed with ASIA grade A neurology, one patient improved to ASIA grade B, whereas the other did not improve. Two patients improved from ASIA grade B to ASIA grade D, and seven patients with ASIA grades C and D improved to ASIA grade E. Fusion at the osteotomy site was seen in 92.85% (13 out of 14) cases on one side within 6 months post operation. Fusion was seen in all the cases within 1 year post operation.Conclusions: Flipped reposition laminoplasty is an excellent technique providing adequate exposure and additional stability postoperatively.

        Effects of Surface Profiles on the Joint Formation during Magnetic Pulse Crimping in Tube-to-Rod Configuration

        Magnetic pulse crimping is a solid state, high-speed impact crimping technique. In this paper, copper tube was successfully crimped on the steel 1020 rod. Three types of profiles threaded, knurled, and plain profile were created on the steel rod and then a copper tube was crimped on the rod. The influence of surface profiles on the joint strength, surface roughness and microstructure of the joint were analyzed and compared. Compression-shear tests as well as pull-out tests were carried out to compare the strength of the crimped joint. The result of the pull-out tests as well as compression-shear tests revealed that threaded surface profile on the steel rod gives better strength in comparison to the other two profiles. Along the circumference of the Cu-steel composite rod the microstructure images of the joints were taken and analyzed, it was found that there was no gap along the circumference of the sample crimped at 5.3 kJ of discharge energy with plain profile. For surface roughness, the Ra values of the crimped samples at 5.3 kJ were analyzed and it was found that average Ra value of the sample crimped on the threaded base was highest (0.80 μm) among threaded, knurled, and plain base samples.

        Employee engagement as an antecedent of organizational commitment – A study on Indian seafaring officers

        Employee engagement has been the most widely researched area of human resource in recent times because of its practical importance in business organizations today. Engagement plays a major role as an antecedent in many organizational outputs like employee performance, commitment, and competitive advantage, etc. This research paper is aimed at identifying the factors contributing to employee engagement and its association with the affective, continuance and normative modules of organizational commitment among Indian ship officers. This research work attempts to explore the correlation existing if any, between work and organizational engagement variables with three aspects of organizational commitment. The whole study is mainly focused on seafarer’s perspective and so few alterations in questionnaires of engagement and commitments were made in discussion with experts from the shipping industry. This study confirms that employee engagement is positively associated with affective and normative components whereas, engagement is not significantly associated with continuance component of organizational commitment which is a contradicting result from existing studies where continuance commitment has reported a significant negative relationship with engagement. This study also discloses the fact that the engagement and commitment levels of officers working with deck and engine department did not contrast significantly, nor did they significantly differ between officers working in different types of ships, different designations and between officers with a different experience.

        Real Time Implementation and Analysis of Enhanced Artifcial Bee Colony Algorithm Optimized PI Control algorithm for Single Phase Shunt Active Power Filter

        This paper proposes a new hybrid novel optimization approach, called Enhanced Artifcial Bee Colony Algorithm (EABC) for designing an optimal PI controller for single-phase Shunt Active Power Filter (SAPF). The proposed EABC algorithm optimizes the gain values of the PI controller to improve the dynamic performance of SAPF. In this EABC, the adaptive real coded genetic algorithm (ARGA) is integrated with the Artifcial Bee Colony (ABC) algorithm and this integration improves the exploration and exploitation ABC and speed up the convergence rate. The minimization of integral square error (ISE) is considered as an objective function to manipulate the gain values of the PI controller. The system tested with MATLAB simulation results are implemented in the hardware circuit with the same set of parameters. The proposed hardware system is designed with the CycloneIV EP4CE30F484 FPGA controller and the gain value for this proposed controller is fed from the simulation results tested with ABC and EABC algorithm. The results obtained from the hardware setup is compared with simulation results. The experimental result enhanced the performance of THD of source current, settling time and percentage peak overshoot of DC Link voltage

        Humpback Whale Assisted Hybrid Maximum Power Point Tracking Algorithm for Partially Shaded Solar Photovoltaic Systems

        This paper proposes a novel hybrid maximum power point tracking (MPPT) algorithm combining a Whale Optimization Algorithm (WOA) and the conventional Perturb & Observation (P&O) to track/extract the highest amount of power from a solar photovoltaic (SPV) system working under partial shading conditions (PSCs). The proposed hybrid algorithm is based on a WOA which predicts the initial global peak (GP) and is followed by P&O in the final stage to achieve a quicker convergence to a GP. Thus, this hybrid algorithm overcomes the computational burden encountered in a standalone WOA, grey wolf optimization (GWO) and hybrid GWO reported in the literature. The conventional algorithm searches for the maximum power point (MPP) in the predicted region by the WOA. The proposed MPPT technique is modelled and simulated using MATLAB/Simulink for simulating an environment to check its effectiveness in accurately tracking the MPP during the GP region. This hybrid algorithm is compared with a standalone WOA, GWO and hybrid GWO. From the simulating results, it is shown that the proposed algorithm offers high tracking performance and that it increases the output power level of a SPV system under partial shading. The algorithm also verified experimentally on various PSCs.

        Importance of Expression System in the Production of Unnatural Recombinant Proteins in Escherichia coli

        In this study, we investigated the efficiencies by which the pET and pQE expression systems produce unnatural recombinant proteins by residue-specific incorporation of unnatural amino acids, a method through which it was found that type of gene expression system tremendously influences the production yield of unnatural proteins in Escherichia coli. Green fluorescent protein (GFP) and a single-chain Fv antibody against c-Met were utilized as model recombinant proteins while Lhomopropargylglycine (Hpg), a methionine analogue that incorporates into the methionine residues of a recombinant protein, was used as model unnatural amino acid. The pET system produced an almost negligible amount of Hpg-incorporated unnatural protein compared to the amount of methionine-incorporated natural protein. However, comparable amounts of unnatural and natural protein were produced by the pQE expression system. The amount of unnatural GFP protein produced through pET expression was not increased despite the over-expression of methionyl tRNA synthetase, which can enhance the activation rate of methionyl-tRNA with a methionine analogue. Incorporation of Hpg decreased the productivity of active GFP by approximately 2.5 fold, possibly caused by the inefficient folding of Hpg-incorporated GFP. Conversely, the productivity of functional anti-c-Met sc-Fv was not influenced by incorporation of Hpg.
