        Consolidation is the time-dependent settlement of soils resulting from the expulsion of water from the soil pores. When a saturated compressible clay layer is subjected to a stress increase, consolidation settlement occurs greatly. Because the hydraulic conductivity of clay is significantly small, the excess pore water pressure generated by loading gradually dissipates over a long period. The settlement caused by consolidation in clay may be several times greater than the immediate settlement. A number of researches are carried out to predict the consolidation behavior in field, however, most of researches show the discrepancies between the prediction and the field settlement data. Therefore, the estimation of consolidation rate is one of the important factors in the construcion on soft clayey deposits. In this paper, laboratory tests have analyzed to apply accurately the soil parameters that executed the plan of stage construction. This study analyzes consolidation behavior of overconsolidated clay with two-dimensional consolidation analysis program by use of the numerical method. By using this program, the time dependent consolidation behavior of soft deposits can be analyzed appropriately. Elastic and elastic-plastic finite element analysis are compared with Terzaghi's theory of one-dimensional consolidation.

      • 다핵균 및 단핵균으로부터 고protease 생산균주의 Screening

        4 multinucleate and 7 mononuclear fungi were investigated for screening of high protease productive strains. The protease activity of mononuclear strains was generally greater than that of multinucleate strains. For acid proteinase, Aspergillus awamori AOK 1007 and Asp. oryzae shoyu 2 as mononuclear strains were selected in glucose-polypeptnoe medium. For acid carboxypeptidase, two mononuclear strains were screened as Asp. awamori AOK 1007 and Asp. usamii mut. shirousami AOK 1011 in glusose-sodium glutanate medium. In this result, mononuclear fungi appeared higher protease production than that of multinuclear fungi. So, mononuclear fungi may gave many merits to improve protease activity by using of mutation or cell fusion technology.

      • 부착성 동물세포의 Scale-up을 위한 방법의 개발

        This study was a protocol of serial propagation for anchorage-dependent animal cell. Vero. by the optimization of trypsin and EDTA concentration in T-25 flask. Commen enzymetic treatment in PBS solution has been modified for a high degree of cell datachment from T-25 flask and for transfer of a maximum number of viable cells from one culture to the next during the scale up process. Culture of Vero cells were washed with Dulbecco's modified Eagle's medium(DMEM) and then exposed to DMEM containing trypsin(0.025%) and EDTA(0.16%). Up to 90% of cells detached from bottom of flask and the viability of cells was greater than 93%. The protocol using the developed method was successfully applied to the next 10 times scale up process. Consequently, the developed method changed multiple steps of processes to one step, so it promised th easy scale-up and low cost demand of animal cell culture.

      • 갓김치 ether 추출물의 발효 과정상의 생리 기능성 변화

        일반 김치의 경우 17±1℃에서 2일 사이에 pH가 4.0에 이르는 것에 비해 갓김치의 경우 18℃에서 저장한 경우에 14일 경에 pH가 4.0(Fig.1)에 이르러 일반 김치 보다 저장성이 3~5배 높은 것으로 나타났으며 총균수에서도 비슷한 양상을 나타내었다. 또한 간암세포에 대한 항암성에서 IC30의 농도가 발효기간별 시료 모두 1000㎍이상으로 나타났으며 발효 0일째 시료가 1000㎍에서 50%의 간암세포 사멸성을 나타내서 가장 항암효과가 우수한 것으로 나타났고, 간정상세포에 대해서는 1000㎍ 이상에서도 28% 이하의 사멸성을 나타내서 정상세포에 대해 큰 영향을 미치지 않는 것으로 나타났다. 또한 항산화 효과에 있어서는 초기(0~2일) 발효시 수소공여능이 53%로 나타나서 다른 식품 추출물의 항산화 효과에 비해서는 비교적 높은 효과를 나타내었다. 그러나 발효 14일에는 15%로 감소되어서, ethyl ether에 의해 추출되어 항산화 효과를 나타내었던 물질들이 발효가 진행됨에 따라 감소된 것으로 사료되었다. The anticancer and antioxidative effects of Mustard Leaf Kimch ether extracts according to various fermentation period were investigated. pH of the Mustard Leaf Kimchi was reached at 4.0 on 14th day, so the period reaching well-fermentating state was up to 3~5 times longer than that of ordinary Kimchi. The anticancer effect on HepG2 by the addition of fermented Mustard Leaf Kimchi ether extracts was higher than that on CCL-13. IC30 of Mustard Leaf Kimchi in HepG2 was showed more than 1000㎍.Especially, 50% of death ratio was showed in HepG2 cells by the addition of ether extracts(0 day). the other side. 28% of death ratio was shown in CCL-13 up to 1000㎍ of ether extracts. As well as the Mustard Leaf Kimchi ether extracts(0-2 days) showed higher antioxidative effect than of others. The hydrogen donor activity was 53% at initial fermentation period, but it was decreased at 14% on 14th day. So, ether soluble antioxidative materials in Mustard Leaf Kimchi was decreased during fermentation.

      • 국균세포벽을 모델로 한 새로운 고정화담체의 개발

        The purpose of this investigation was to develop a new carrier using chitin and glucan as the model for Aspergillus oryzae cell wall. In case of the ratio of chitin and glucan(3/1), the activity of the immobilized protease had higher than that of model(chitin/glucan=1/3) and carriers were formed gel. In case of soluble chitin and glucan treated with calcium carbonate, there were a 1.8-fold increase in activity of immobilized protease compared with untreated chitin. The scanning electron microscope image showed that a pore size of this carrier had a homogenized pore size and a smooth surface.

        The effect of aquarobics exercise on the physical strength, BMI and WHR of elderly women

        The purpose of this study is the effects of aquarobics exercise on physical strength, BMI, and WHR of elderly women and to provide basic data of fitness program for elderly women. For statistical analysis, we used SPSS 20 to calculate the mean and standard deviation, and the two-way ANOVA was performed by repeated measures in order to verify the mean difference by group and time. For interaction effect analysis, we performed one-way ANOVA by time and group and correspondence t-test by time and group. The significance level was set at p<.05. The result obtain. First, Significant differences were found in upper extremity muscle function(p<.05), lower extremity muscle function(p<.05), and cardiopulmonary endurance(p<.01). second, There was a significant difference in body fat percentage(p<.01). third, There was a significant difference in waist to height ratio(p<.001). Through this study, aquarobics exercise will bring about positive changes in the physical strength, body fat precentage and WHR of the elderly.

        Solid Oxide Fuel Cells (SOFCs) have received considerable attention because of the advantages of high efficiency, low pollution, cogeneration application and excellent integration with simplified reformer.<br/> In this paper, we reported development of anode-tubular SOFC by wet process. For making tubular cell, Ni-cermet YSZ anode tube was fabricated using extrusion process, and YSZ electrolyte layer and LSM-YSZ composite, LSM, LSCF cathode layer were coated onto the anode supported tube using slurry dipping process and sintered by co-firing process. By using this tubular cell, we fabricated single cell consisted of the various cathode layers and 4 cell stack with an effective area of 75 ㎠ per single cell, and evaluated their performance characteristics.

        Fabrication and Characterization of Composite LSCF-Ag Cathode for Solid Oxide Fuel Cells using Electron Beam Irradiation Process

        A new process to fabricate a composite LSCF-Ag cathode material for SOFCs by electron beam (e-beam) irradiation process has been suggested for operation under intermediate temperature range of $600-700^{\circ}C$. A composite LSCF-Ag cathode with uniformly coated Ag nanoparticles on the surface of the LSCF material was prepared by a facile e-beam irradiation method at room temperature. The morphology of the composite LSCF-Ag material was analyzed using a TEM, FE-SEM, and EDS. The prepared composite LSCF-Ag material can play a significant role in increasing the electro-catalytic activities and reducing the operating temperature of SOFCs. The performance of a tubular single cell prepared using the composite LSCF-Ag cathode, YSZ electrolyte and a Ni/YSZ anode was evaluated at reduced operating temperature of $600-700^{\circ}C$. The micro-structure and chemical composition of the single cell were investigated using a FE-SEM and EDS.
