        LncRNA TMPO-AS1 promotes esophageal squamous cell carcinoma progression by forming biomolecular condensates with FUS and p300 to regulate TMPO transcription

        Esophageal squamous cell carcinoma (ESCC) is one of the most life- and health-threatening malignant diseases worldwide, especially in China. Long noncoding RNAs (lncRNAs) have emerged as important regulators of tumorigenesis and tumor progression. However, the roles and mechanisms of lncRNAs in ESCC require further exploration. Here, in combination with a small interfering RNA (siRNA) library targeting specific lncRNAs, we performed MTS and Transwell assays to screen functional lncRNAs that were overexpressed in ESCC. TMPO-AS1 expression was significantly upregulated in ESCC tumor samples, with higher TMPOAS1 expression positively correlated with shorter overall survival times. In vitro and in vivo functional experiments revealed that TMPO-AS1 promotes the proliferation and metastasis of ESCC cells. Mechanistically, TMPO-AS1 bound to fused in sarcoma (FUS) and recruited p300 to the TMPO promoter, forming biomolecular condensates in situ to activate TMPO transcription in cis by increasing the acetylation of histone H3 lysine 27 (H3K27ac). Targeting TMPO-AS1 led to impaired ESCC tumor growth in a patient-derived xenograft (PDX) model. We found that TMPO-AS1 is required for cell proliferation and metastasis in ESCC by promoting the expression of TMPO, and both TMPO-AS1 and TMPO might be potential biomarkers and therapeutic targets in ESCC.

        Deep Learning in Drebin: Android malware Image Texture Median Filter Analysis and Detection

        This paper proposes an Image Texture Median Filter (ITMF) to analyze and detect Android malware on Drebin datasetsMedian Filter (MF) to reflect the similarity of the malware binary file block. At the same time, using the MAEVS (Malware Activity Embedding in Vector Space) to reflect the potential dynamic activity of malware. In order to ensure the improvement of the classification accuracy, the above-mentioned features(ITMF feature and MAEVS feature)are studied to train Restricted Boltzmann Machine (RBM) and Back Propagation (BP). The experimental results show that the model has an average accuracy rate of 95.43% 1. We design a model of “ITMF” combined with Image Processing of with few false alarms. to Android malicious code, which is significantly higher than 95.2% of without ITMF, 93.8% of shallow machine learning model SVM, 94.8% of KNN, 94.6% of ANN.

        ≪最高人民法院<關于民事訴訟證據的若幹規定>≫的頒行,在中國民事訴訟實踐中確認了相當程度的蓋然性證明標准,成爲我們民事訴訟法制化進程中一個重要的裏程碑. 但該規則由于具備不確定性與模糊性等令人遺憾之處,其同時拉開了中國學者對民事訴訟證明標准激烈論爭的序幕. 本文通過對證明標准問題的分析與探討,旨在能九將該理論運用于具體訴訟,從而使民事訴訟證明標准在民事審判實務中能九發揮應有的作用. The enforcement of “Some Provisions of the Supreme People``s Court on Evidence in Civil Procedures” confirmed the standard of high probability for the burden of proof in the practice of civil procedure in China, and became an important milestone in the course of juridification of civil procedure. But there are imperfect features such as indetermination and faintness in this rule, which brought on the hot controversy of standard of the burden of proof in civil procedure. Through the analysis and research of standard of the burden of proof, the author tried to make the civil procedure standard of the burden of proof effective in civil trial, by applying this theory in specific cases.

        Research progress and treatment of radiation enteritis and gut microbiota

        Radiation enteritis is a kind of intestinal radiation injury in patients with pelvic and retroperitoneal malignancies after radiotherapy, and its occurrence and development process are very complicated. At present, studies have confirmed that intestinal microecological imbalance is an important factor in the formation of this disease. Abdominal radiation causes changes in the composition of the flora and a decrease in its diversity, which is mainly manifested by a decrease in beneficial bacterial species such as Lactobacilli and Bifidobacteria. Intestinal dysbacteriosis aggravates radiation enteritis, weakens the function of the intestinal epithelial barrier, and promotes the expression of inflammatory factors, thereby aggravating the occurrence of enteritis. Given the role of the microbiome in radiation enteritis, we suggest that the gut microbiota may be a potential biomarker for the disease. Treatment methods such as probiotics, antibiotics, and fecal microbiota transplantation are ways to correct the microbiota and may be an effective way to prevent and treat radiation enteritis. Based on a review of the relevant literature, this paper reviews the mechanism and treatment of intestinal microbes in radiation enteritis.

        사회치리(社會治理)로 방향전환을 모색하는 광둥성의 사회관리 정책

        개혁개방이 낳은 많은 사회 문제에 대응하기 위해 최근 중국에서는 사회건설과 사회관리가 주목받고 있다. 광둥성은 이런 변화의 최전선에 있다. 이 논문은 선전시와 광저우시의 사례를 중심으로 사회통제에서 사회치리로 방향 전환을 모색하는 광둥성의 사회관리 정책의 변화를 살펴본다. 중요한 변화는 세 가지 영역에서 관찰된다. 첫째는 사구를 세분된 격자로 나누어 정보수집과 즉시개입을 결합하는 “격자망화 관리”가 등장한것이다. 둘째는 정부 역할을 민간으로 이전하는데, 특히 ‘사회공작’ 업무를 사회에서 구매하는 방식으로 발전시키는 것이다. 셋째는 이런 업무를 민간에서 맡도록 사회조직을 지원ㆍ육성하는 것이다. 광둥성의 사회관리정책의 변화는 두 가지 서로 모순되는 목표를 결합하고 있다. 한편에서는 ‘사회치리’라는 용어가 강조하듯이, 사회치리의 주체를 다원화하고자 하는 것이다. 다른 한편에서는 ‘안정유지’(維穩)의 사고를 우위에 두어 기층사회의 안정성을 확보하고자 하는 것이다. 선전시의 ‘타오위안 모델’이나 광저우시의 ‘가종’ 중심 모델 모두 이런 특징을 공유한다. 전체적으로 보자면, 광둥성의 변화는 아직까지는 ‘안정유지’ 우위의 제약을 크게 벗어나지 못하고 있다. Recently social management and social construction have been emphasized in China since China faces big social problems resulted from the legacy of economic reforms. Guangdong is one of the most outstanding provinces that lead new changes in social policies to solve those problems. This paper focuses on the experiments of Shenzhen and Guangzhou, displaying rapid changes in Guangdong province from social control to social governance. Three important changes are witnessed. First, “grid network management” is introduced that divides social communities into many small units to collect information on the communities and to allow local government to intervene in bottomlevel conflicts. Second, social work is systemically emphasized to transfer the role of the local governments to the civil society. Local governments become agencies to buy social services. Three, social organizations are encouraged to take the roles in this outsourcing of social services. Guangdong’s social management policies combines two contradictory aims together: on the one hand, the multiplication of social subjects are emphasized and confirmed by the concept of ‘social governance’; on the other hand, it aims to maintain social orders at grass roots level, that is ‘weiwen’. ‘Taoyuan model’ of Shenzhen and ‘Jiazhong’ model of Guangzhou share common characteristics of these contradictory aims. Guangdong is still trapped in the limitation of ‘maintenance of social order’.

        Migraine Susceptibility Genes in Han Chinese of Fujian Province

        Background and Purpose Five single-nucleotide polymorphisms (SNPs) (rs4379368, rs10504861, rs10915437, rs12134493 and rs13208321) were recently identified in a Western population with migraine. These migraine-associated SNPs have not been evaluated in a Han Chinese population. This study investigated the associations of specific SNPs with migraine in a Han population. Methods This was a case-control study of Han Chinese residing in Fujian Province. Polymerase chain reaction—restriction-fragment-length polymorphism analysis and direct sequencing were used to characterize the relationships of SNPs in a control group of 200 subjects and in a migraine group of 201 patients. Results The frequencies of the five SNPs did not differ between patients with migraine and healthy non migraine controls. However, subgroup analysis indicated certain SNPs were more strongly associated with migraine with aura or migraine without aura than with controls. The CT genotype of rs4379368 was more common in migraine patients with aura (75%) than in migraine patients without aura (47.9%) and controls (48.5%) (p<0.05), and the TT genotype of rs10504861 was more common in migraine patients with aura than in controls (8.3% vs. 0.5%) (p<0.05). Meanwhile, the CC genotype of rs12134493 was less common in migraine patients without aura than in controls (80.6% vs. 88%) (p<0.05). Conclusions Our findings suggest that the rs4379368 and rs10504861 SNPs are markers for susceptibility to migraine with aura and that rs12134493 is a marker for the risk of migraine without aura in this Han population. Future studies should further explore if these associations vary by ethnicity.

        Optimization of Medium Composition for Lipopeptide Production from Bacillus subtilis N7 using Response Surface Methodology

        The nutritional requirements for the maximum production of lipopeptides by Bacillus subtilis N7 (B. subtilis N7) were investigated and optimized using response surface methodology (RSM) under shake flask fermentation. A one-factor-at-a time experimental setup was used to screen carbon and nitrogen sources. A Plackett?Burman design (PBD) was employed to screen the most critical variables for lipopeptides production amongst ten nutritional elements. The central composite experimental design (CCD) was finally adopted to elucidate the composition of the fermentation medium. Statistical analyses (analysis of variance, ANOVA) of the results showed that KCl, MnSO4 and FeSO4·6H2O were important components and that their interactions were strong. Lipopeptide production was predicted to reach 709.87 mg/L after a 60 h incubation using an optimum fermentation medium composed of glucose 7.5 g/L, peanut oil 1.25 g/L, MgSO4 0.37 g/L, KH2PO4 0.75 g/L, monosodium glutamate 6.75 g/L, yeast extract and NH4Cl (5:3 w/w) 10 g/L, KCl 0.16 g/L, FeSO4·6H2O 0.24 mg/L, MnSO4 0.76 mg/L, and an initial pH of 7.0. Lipopeptide production (706.57 ± 3.70 mg/L) in the optimized medium confirmed the validity of the predicted model.

        Post-Transcriptional Control of Tropoelastin in Aortic Smooth Muscle Cells Affects Aortic Dissection Onset

        Aortic dissection (AD) is a catastrophic disease with high mortality and morbidity, characterized with fragmentation of elastin and loss of smooth muscle cells. Although AD has been largely attributable to polymorphisms defect in the elastin-coding gene, tropoelastin (TE), other undermined factors also appear to play roles in AD onset. Here, we investigated the effects of post-transcriptional control of TE by microRNAs (miRNAs) on elastin levels in aortic smooth muscle cells (ASMC). We found that miR-144-3p is a miRNA that targets TE mRNA in both human and mouse. Bioinformatics analyses and dual luciferase reporter assay showed that miR-144-3p inhibited protein translation of TE, through binding to the 3'-UTR of the TE mRNA. Interestingly, higher miR-144-3p levels and lower TE were detected in the ASMC obtained from AD patients, compared to those from non-AD controls. In a mouse model for human AD, infusion of adeno-associated viruses (serotype 6) carrying antisense for miR-144-3p (asmiR-144-3p) under CAG promoter significantly reduced the incidence and severity of AD, seemingly through enhancement of TE levels in ASMC. Thus, our data suggest an essential role of miR-144-3p on the pathogenesis of AD.
