Histone deacetylase 3 is selectively involved in L3MBTL2-mediated transcriptional repression

<P><B>Abstract</B></P><P>This is the first report that L(3)mbt-like 2 (L3MBTL2) specifically interacts with the histone deacetylase domain of histone deacetylase 3 (HDAC3) via its MBT domain. Here, we show that L3MBTL2 selectively interacts with HDAC3, but not other class I HDACs. An in vitro peptide-binding assay demonstrated the specific association of HDAC3 with methylated histone-K20 tail and L3MBTL2. Furthermore, depletion of HDAC3 resulted in a decrease of methylated K20-H4, as well as an increase in acetylated histone H3. Consequently, HDAC3 knock-down selectively suppressed L3MBTL2-mediated transcriptional repression. Factors influencing choosing teaching as a career: South Korean preservice teachers

This study validated the underlying factor structure of the Factors Influencing Teaching (FIT)-Choice survey in the South Korea context and examined motivations and perceptions of 903 Korean preservice teachers by major demographic characteristics. Secondary preservice teachers reported significantly higher values than elementary preservice teachers for Qualification fit and desire to teach and Task return. Male preservice teachers believed more strongly than female preservice teachers that they chose the teaching profession because of Qualification fit, Influencing others, and Task return. Compared to freshmen/sophomores, juniors/seniors reported significantly higher values for Task demand. Participants with low- to medium-level self-reported income ($12K–$46K) weighted Benefits significantly higher than those with medium-level income ($46K–$88K). STEM majors reported higher values for the Qualification fit and Influencing others than non-STEM majors. As South Korea is one of the few countries which has an oversupply of highly qualified teacher candidates, findings were compared with other countries to provide useful insights to improve recruitment of highly qualified individuals to teaching workforce in many countries where teacher shortage is a chronic problem. Based on these data analyses, we argue that policy makers, politicians, education researchers, and others who care about quality education and student academic achievements need to make efforts to increase task returns and benefits.

Highly pathogenic avian influenza viruses (HPAIV) of the H5N1 subtype have spread since 2003 in poultry and wild birds in Asia, Europe and Africa. In Korea, the highly pathogenic H5N1 avian influenza outbreaks took place in 2003/2004, 2006/2007 and 2008. As the 2006/2007 isolates differ phylogenetically from the 2003/2004 isolates, we assessed the clinical responses of chickens, ducks and quails to intranasal inoculation of the 2006/2007 index case virus, A/chicken/Korea/IS/06. All the chickens and quails died on 3 days and 3-6 days post-inoculation (DPI), respectively, whilst the ducks only showed signs of mild depression. The uninoculated chickens and quails placed soon after with the inoculated flock died on 5.3 and 7.5 DPI, respectively. Both oropharyngeal and cloacal swabs were taken for all three species during various time intervals after inoculation. It was found that oropharyngeal swabs showed higher viral titers than in cloacal swabs applicable to all three avian species. The chickens and quails shed the virus until they died (up to 3 to 6 days after inoculation, respectively) whilst the ducks shed the virus on 2-4 DPI. The postmortem tissues collected from the chickens and quails on day 3 and days 4-5 and from clinically normal ducks that were euthanized on day 4 contained the virus. However, the ducks had significantly lower viral titers than the chickens or quails. Thus, the three avian species varied significantly in their clinical signs, mortality, tissue virus titers, and duration of virus shedding. Our observations suggest that duck and quail farms should be monitored particularly closely for the presence of HPAIV so that further virus transmission to other avian or mammalian hosts can be prevented.

Nano-crystalline ZnS:Mn and its derivatives are promising as a novel luminescent center because of its high quantum eciency atroom temperature. In this work, three dierent kinds of ZnS based nano-crystalline particles were synthesized and characterized inorder to be utilized for polymer light-emitting diodes; ZnS:Mn, ZnS:CuCl, and ZnS:AgCl. Some of the ZnS based nanocrystals wereeither hybridized with poly(acrylic acid)(PAA) or doped to poly(9-vinyl carbazole)(PVCz)). The XRD analysis showed that thediameter of the ZnS based particles was in the range of 11.823.4 nm. In all the ZnS based nanocrystals hybridized with PAA, thephotoluminescence (PL) was enhanced in comparison to the ZnS:Mn regardless of the metal ion. It was found that the ZnS:AgClhybridized with PAA showed the highest PL and the ZnS:CuCl hybridized with PAA showed the lowest PL among the three ZnSbased nanocrystals hybridized with PAA. The PL of PVCz doped with the ZnS based nanocrystals, however, was lower in com-parison to the ZnS:Mn regardless of the metal ion though the dierences were not large
Enhanced Production of Phenolic Compounds from Pumpkin Leaves by Subcritical Water Hydrolysis

Enhanced production of individual phenolic compounds by subcritical water hydrolysis (SWH) of pumpkin leaves was investigated at various temperatures ranging from 100 to 220℃ at 20 min and at various reaction times ranging from 10 to 50 min at 160℃. Caffeic acid, p-coumaric acid, ferulic acid, and gentisic acid were the major phenolic compounds in the hydrolysate of pumpkin leaves. All phenolic compounds except gentisic acid showed the highest yield at 160℃, but gentisic acid showed the highest yield at 180℃. The cumulative amount of individual phenolic compounds gradually increased by 48.1, 52.2, and 78.4 ㎍/g dry matter at 100℃, 120℃, and 140℃, respectively, and then greatly increased by 1,477.1 ㎍/g dry matter at 160℃. The yields of caffeic acid and ferulic acid showed peaks at 20 min, while those of cinnamic acid, p-coumaric acid, p-hydroxybenzoic acid, and procatechuic acid showed peaks at 30 min. Antioxidant activities such as 2,2-diphenyl-1-picrylhydrazyl and ferric reducing antioxidant power values gradually increased with hydrolysis temperature and ranged from 6.77 to 12.42 ㎎ ascorbic acid equivalents/g dry matter and from 4.25 to 8.92 m㏖ Fe<SUP>2+</SUP>/100 g dry matter, respectively. Color L<SUP>*</SUP> and b<SUP>*</SUP> values gradually decreased as hydrolysis temperature increased from 100℃ to 140℃. At high temperatures (160℃ to 220℃), L<SUP>*</SUP> and b<SUP>*</SUP> values decreased suddenly. The a<SUP>*</SUP> value peaked at 160℃ and then decreased as temperature increased from 160℃ to 220℃. These results suggest that SWH of pumpkin leaves was strongly influenced by hydrolysis temperature and may enhanced the production of phenolic compounds and antioxidant activities.
