        Synthesis, antioxidant and anticancer screenings of berberine–indole conjugates

        <P>A variety of heterocyclic nitrogen cores in the form of indole moieties were linked to the natural isoquinoline alkaloid molecule berberine to achieve anticipated antioxidant and anticancer properties. An efficient synthetic pathway afforded final compounds 5a-j, which were tested in vitro for antioxidant potency using 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt radical (ABTS) bioassays, and for anticancer activity using sulforhodamine B (SRB) assay against HeLa and Caski cancer cell lines. Moreover, the toxic nature of the resultant molecules was investigated using Madin-Darby canine kidney cells. The therapeutic indices of 5a-j were more appreciable against the Caski than HeLa cell line, in which compounds with electron-releasing alkyl or alkoxy functional group on indole entity as well as azaindole derivative performed well. In addition, these compounds were well endowed with antioxidant properties, in addition to the equal antioxidant effect of the compound with electron-withdrawing chlorine atom within indole entity. Adequate confirmation of the structure of the final analogues was achieved using Fourier-transform infrared (FT-IR), H-1 nuclear magnetic resonance (NMR), and mass spectroscopy and elemental (CHN) analysis.</P>

        Efficacy of indole-3-acetic acid-producing PGPFs and their consortium on physiological and biochemical parameters of Trigonella foenum-graecum L

        Fungi produce phytohormones like indole-3-acetic acid (IAA), which stimulates the growth of plants. In the present study, Aspergillus niger (AN) and Aspergillus caespitosus (DS-3) were identifi ed in screens for highly effi cient IAA-producing fungi. Initially, IAA production was assessed through UV–Visible spectroscopic analysis using Salkowski’s reagent. HPTLC confi rmed the presence of IAA in all fungal extracts when compared with the standard IAA. HPTLC analysis revealed that AN exhibit the highest IAA production, which was 251.86 ± 0.7 μg ml −1 . AN, DS-3 and a combination of both strains as a growth-promoting consortium (GPC-1) were applied to Trigonella foenum-graecum L seeds for 6 h. After 14 days, the fenugreek plants were used for both the physiological and biochemical analysis. Results indicated that the GPC-1 induced more desirable physiological properties, with higher protein content, carbohydrate content, total phenolic content and antioxidant activity compared to treatment with individual PGPFs or distilled water. The extract from fenugreek plants treated with GPC-1 had the maximum diosgenin content (342.374 ± 0.67 μg ml −1 ), as assessed by HPTLC.

        Behaviour of asymmetric building with double variable frequency pendulum isolator

        Presented in this paper is the behaviour of asymmetric building isolated by the double variable frequency pendulum isolator (DVFPI). The DVFPI is an adoption of single variable frequency pendulum isolator (VFPI). The geometry and coefficient of friction of top and bottom sliding surfaces can be unequal. The governing equations of motion of the building-isolation system are derived and solved in incremental form. The analysis duly considers the interaction of frictional forces in the two principal directions developed at each sliding surface of the DVFPI. In order to investigate the behaviour of the base isolation using the DVFPI, the coupled lateral-torsional response is obtained under different parametric variations for a set of six far-fault earthquake ground motions and criterion to optimize its performance is proposed. Further, influences of the initial time period, coefficient of friction and frequency variation factors at the two sliding surfaces are investigated. The numerical results of the extensive parametric study help in understanding the torsional behaviour of the structure isolated with the double sliding surfaces as in the DVFPI. It is found that the performance of the DVFPI can be optimized by designing the top sliding surface initially softer and smoother relative to the bottom one.

        Therapeutic potential of quercetin as a cardiovascular agent

        <P><B>Abstract</B></P> <P>Flavonoids are integral components of various vegetation and in foods; consequently, they represent an inevitable part of the diet. Historical and epidemiological proof recommend that diet plans consisting of flavonoids such as quercetin have positive health benefits, especially on the heart. Flavonoids have been proven to be active against hypertension, inflammation, diabetes and vascular diseases. Quercetin exhibits significant heart related benefits as inhibition of LDL oxidation, endothelium-independent vasodilator effects, reduction of adhesion molecules and other inflammatory markers, the protective effect on nitric oxide and endothelial function under conditions of oxidative stress, prevention of neuronal oxidative and inflammatory damage and platelet antiaggregant effects. Searching for experimental evidence to validate the cardioprotective effects of quercetin, we review here the recent detailed <I>in vivo</I> studies. Quercetin and its derivatives lead to an enhancement in heart features, indicating the prospective for quercetin to be used therapeutically in the treatment of cardiac diseases. Several evidence-based studies suggest mechanisms to observe cardiovascular diseases such as aging effects, hypertension, angiotensin-converting enzyme activity and endothelial-dependent and independent functions. Different animal models including human are also used to elucidate the <I>in vivo</I> role of quercetin in cardiovascular diseases.         Synthesis and anticancer activities of polymethylenedioxy analogues of combretastatin A‑2

        Combretastatin A-4 is a highly potent natural stilbene that can inhibit cancer cell proliferation. Numerous analogues of combretastatin A-4 have been proposed for clinical applications. However, structural studies of combretastatin A-2, a methylenedioxy derivative of combretastain A-4, are not available. In this study, various analogues of combretastatin A-2 with polymethylenedioxy spacer were prepared and their antiproliferative activities to four human cancer cell lines (HeLa, SK-OV-3, A549, and HT-29) and two normal cells (HaCaT and MDCK) were evaluated. Binding characteristics were evaluated based on computational docking and previously reported experimental data. Results suggest that their binding conformations are highly dependent on steric volume and electrostatic properties of substituents.

      • Structure and Vibration Analyses of Low Speed Contra-Rotating Fan Stage with High Aspect Ratio

        Contra-rotating fan is comprised of two rotors which are rotating in the opposite direction. The fan stages are named rotor-1 and rotor-2. Benefits from the use of contra rotation are in terms of better efficiency and improved thrust to weight ratio. Failure of contra-rotating fan stage blade in-service results in safety risks, repair costs, and revenue losses. This paper focuses on the vibration analysis and one way fluid-structure interaction of high aspect ratio, low speed contrarotating fan rotors. Modal analysis and modal pre-stress analysis of contra-rotating fan rotors were carried out to calculate the natural frequencies, One way fluid-structure interaction (FSI) was carried out where the computational analysis of the blades was performed using ANSYS CFX. The boundary conditions for CFD analysis were considered from the actual experimental velocity flow field at the inlet and pressure outlet. Based on the results obtained from the CFD analysis, the structural analysis such as deformation and Von-Misses stresses was carried out by using the finite element method (FEM) with ANSYS. The results provide necessary guidelines for the safe running of the contra-rotating fan. The analysis also will be helpful to understand the change of flow behavior due to a rotor deformation.

        Effect of axial extension on parameterized endwall contour with incidence change for low-pressure turbine linear cascade

        Non-axisymmetric endwall (NAEW) contouring method has shown promising results to reduce secondary losses using both experimental and numerical studies. The parameterized endwall contour shaping redistributes the local static pressure resulting in a reduced effect of loss generating elements. Present study discusses the effect of the fore extension of NAEW shape on secondary flow field at design and off-design incidences numerically for low-pressure (LP) linear turbine cascade. The steady Reynolds averaged Navier-Stokes (RANS) simulations are performed using shear stress transport (SST) based γ − θ turbulence model in commercial solver. The simulations are performed for a low Re = 1.6×10 5based on blade chord and isentropic exit velocity. The two-part study discusses the effect of upstream extension of NAEW profile at design and off-design incidence cases, respectively. The study observed reduction in total pressure loss coefficient ( ω ) by implementing NAEW contour profiling. The NAEW with 40 % C ax extension is the most favorable case.

        Adsorptive remediation of cobalt oxide nanoparticles by magnetized α-cellulose fibers from waste paper biomass

        <P><B>Abstract</B></P> <P>Remediation of engineered-nanomaterials is an up-coming major environmental concern. This study demonstrates adsorptive-remediation of cobalt oxide nanoparticles (CoO NPs) from the water. The α-cellulose-fibers were extracted from waste-paper biomass (WP-αCFs) and magnetized with Fe<SUB>3</SUB>O<SUB>4</SUB> NPs (M-WP-αCFs). The XRD, FT-IR, and TGA were performed for detailed characterization of the newly developed bioadsorbent. The M-WP-αCFs was then applied for adsorptive remediation of CoO NPs. The adsorptive kinetics of CoO NPs adsorption onto the M-WP-αCFs reveals the pseudo-second-order model. The various adsorption isotherm studies revealed Langmuir is a best-fit isotherm. A prominently high adsorption capacity <I>q<SUB>m</SUB> </I> (1567 mg/g) corroborated extraordinary adsorptive potential of M-WP-αCFs. Furthermore, CoO NPs were adsorbed onto M-WP-αCFs were analyzed by the XPS, VSM, and TEM.         Individual exome analysis in diagnosis and management of paediatric liver failure of indeterminate aetiology

        Background & Aims: In children with liver failure, as many as half remain of indeterminate aetiology. This hinders timely consideration of optimal treatment options. We posit that a significant subset of these children harbour known inherited metabolic liver diseases with atypical presentation or novel inborn errors of metabolism. We investigated the utility of whole-exome sequencing in three children with advanced liver disease of indeterminate aetiology. Methods: Patient 1 was a 10year-old female diagnosed with Wilson disease but no detectable ATP7B mutations, and decompensated liver cirrhosis who underwent liver transplant and subsequently developed onset of neurodegenerative disease. Patient 2 was a full-term 2day-old male with fatal acute liver failure of indeterminate aetiology. Patient 3 was an 8year-old female with progressive syndromic cholestasis of unknown aetiology since age 3months. Results: Unbiased whole-exome sequencing of germline DNA revealed homozygous mutations in MPV17 and SERAC1 as the disease causing genes in patient 1 and 2, respectively. This is the first demonstration of SERAC1 loss-of-function associated fatal acute liver failure. Patient 1 expands the phenotypic spectrum of the MPV17-related hepatocerebral mitochondrial DNA depletion syndrome. Patient 3 was found to have syndromic cholestasis due to bi-allelic NOTCH2 mutations. Conclusions: Our findings validate the application of whole-exome sequencing in the diagnosis and management of children with advanced liver disease of indeterminate aetiology, with the potential to enhance optimal selection of treatment options and adequate counselling of families. Moreover, whole-exome sequencing revealed a hitherto unrecognized phenotypic spectrum of inherited metabolic liver diseases.
