        Crataegus pinnatifida Bunge root extract induces apoptosis of murine lung carcinoma cells in vitro

        This study sought to evaluate the anticancer effects of Crataegus pinnatifida Bunge root extract (CPE) on murine Lewis lung carcinoma cells (LLC1) in vitro. CPE treatment (2.5, 5, 10 μg/mL, 24 h) of LLC cells led to a dose-dependent decrease in cell viability, while CPE treatment did not have a cytotoxic effect on non-cancer cells (NIH/3T3). CPE affects LLC by flipping the plasma membrane and making the membrane more permeable; by flow cytometry, CPE-induced annexin V and propidium iodide positivity, indicating induction of apoptosis in LLC cells. In addition, CPE enhanced the expression of apoptotic proteins caspase-3 and poly (ADP-ribose) polymerase 1 (PARP-1). CPE upregulated the proapoptotic protein BCL-2-associated X while downregulating the anti-apoptotic protein B-cell lymphoma 2 (BCL-2), suggesting that CPE induces apoptosis via the mitochondrial pathway. Furthermore, CPE upregulated the phosphorylation of the mitogen activated protein kinase p38. In conclusion, the results suggest that CPE has an anticancer effect in LLC cells by inducing apoptosis via p38.

        공감활동이 유아 정서조절능력과 배려행동에 미치는 영향에 대한 연구

        본 연구의 목적은 그림책을 활용한 공감활동이 유아의 정서조절능력과 배려행동에 미치는 효과를 확인하는 것으로 기존 유아 정서조절능력을 측정하기 위한 선행연구에서의 측정도구를 사용하여 만 4세 유아 34명을 대상으로 실험 집단과 통제집단 그룹 17명씩을 배치하여 실험을 진행하였다. 실험은 총 7주간 14회에 거쳐 진행되었으며 그림책 활용이 유아 정서조절과 배려행동에 영향을 미치는지에 대해 사전, 사후 검사를 실시하여 t-test를 실시함으로써 검증하였다. 연구결과 첫째로 그림책 활용 공감활동은 전체적인 정서조절 능력을 향상시키는 효과를 검증할 수 있었지만 하위요인인 적응적 정서조절과 부정 정서조절의 경우 통계적으로 효과를 검증하지 못하였다. 둘째로 그림책 활용은 전체적인 배려행동을 향상시키는 효과뿐만 아니라 배려행동의 하위요인을 향상시키는 영향을 통계적으로 검증하였다. 이를 통해 그림책을 활용한 유아 활동은 유아에게 상대방의 생각과 감정을 인식하고 알맞게 조절하고 유지하며 상대방을 배려하는 행동에 대해 긍정적인 효과가 있다는 것을 검증하였다. 향후 연구에서는 정서조절능력과 배려행동의 하위요인이 보다 더 구체적으로 연결되어 측정할 수 있는 측정도구들이 개발되어 연구에 활용이 필요하다. The purpose of this study is to determine the effect of sympathetic activity using picture books on children's emotional regulation ability and considerate behavior. The experiment was conducted by placing 17 people each in an experimental group and a control group. The experiment was conducted 14 times over 7 weeks, and it was verified by conducting pre- and post-tests on whether the use of picture books had an effect on infant emotional regulation and considerate behavior. As a result of the study, first, the effect of sympathetic activity using picture books to improve overall emotion regulation ability could be verified, but the effects of adaptive emotion regulation and negative emotion regulation, which are sub-factors, could not be verified statistically. Second, the effect of using the picture books on improving not only the overall considerate behavior but also the sub-factors of the considerate behaviors were statistically verified. Through this, it was verified that children's sympathetic activities using picture books have a positive effect on children's behaviors to recognize the thoughts and emotions of the other person, adjust and maintain them appropriately, and be considerate of the other person. In future research, measurement tools that can connect and measure the sub-factors of emotional regulation ability and considerate behavior more specifically are developed and need to be utilized in research.

        Extract of high hydrostatic pressure-treated danshen (Salvia miltiorrhiza) ameliorates atherosclerosis via autophagy induction

        Danshen (Salvia miltiorrhiza) is a traditional medicinal plant widely used in Asian countries for its pharmacological activities (e.g., amelioration of cardiovascular diseases). In this study, we investigated the anti-atherosclerotic activity of raw danshen root extract prepared using high hydrostatic pressure (HHP) at 550 MPa for 5 min and hot water extraction. This method was useful for elimination of bacteria from cultured danshen plants and for better extraction yield of active principles. The HHPtreated danshen extract (HDE) inhibited proliferation of human umbilical vein endothelial cells (HUVECs) and induced autophagy that was assessed by LC3 conversion and p62 degradation. HDE suppressed foam cell formation in oxLDL-induced RAW264.7 macrophages; lysosomal activity simultaneously increased, measured by acridine orange staining. HDE also reduced atherosclerotic plaque development in vivo in apolipoprotein E knock-out (ApoE<sup>-/-</sup>) mice fed a high cholesterol diet. Taken together, these results indicated that HDE exhibited anti-atherosclerotic activity via autophagy induction. [BMB Reports 2020; 53(12): 652-657]

      • AOPWIN을 이용한 Acetanilide의 광분해 특성 평가

        Acetanilide is a High Production Volume Chemical, which is produced about 2,300 tons/year in Korea as of 1998 survey. Most is used as an intermediate for synthesis of pharmaceuticals and dyes, and the chemical is one of seven chemicals of which human and environmental risk are being assessed by National Institute of Environmental Research under the frame of OECD SIDS program. The Atmospheric Oxidation Program for Microsoft Windows (AOPWIN) is used to estimates the rate constant for the atmospheric, gas-phase reaction between photochemically produced hydroxyl radicals and organic chemicals. It is also used to estimates the rate constant for the gas-phase reaction between ozone and olefinic/acetylenic compounds. The rate constants estimated by the program are then used to calculate atmospheric half-lives for organic compounds based upon average atmospheric concentrations of hydroxyl radicals and ozone. AOPWIN requires only a chemical structure to make these predictions. Structures are entered into AOPWIN by SMILES (Simplified Molecular Input Line Entry System) notations. In this study, one of environmental fate/distribution of the chemical elements, photodegradation of acetanilide was estimated using AOPWIN model based on SMILES notation and chemical name data.

        Emergence of synthetic mRNA: <i>In vitro</i> synthesis of mRNA and its applications in regenerative medicine

        <P><B>Abstract</B></P> <P>The field of gene therapy has evolved over the past two decades after the first introduction of nucleic acid drugs, such as plasmid DNA (pDNA). With the development of <I>in vitro</I> transcription (IVT) methods, synthetic mRNA has become an emerging class of gene therapy. IVT mRNA has several advantages over conventional pDNA for the expression of target proteins. mRNA does not require nuclear localization to mediate protein translation. The intracellular process for protein expression is much simpler and there is no potential risk of insertion mutagenesis. Having these advantages, the level of protein expression is far enhanced as comparable to that of viral expression systems. This makes IVT mRNA a powerful alternative gene expression system for various applications in regenerative medicine. In this review, we highlight the synthesis and preparation of IVT mRNA and its therapeutic applications. The article includes the design and preparation of IVT mRNA, chemical modification of IVT mRNA, and therapeutic applications of IVT mRNA in cellular reprogramming, stem cell engineering, and protein replacement therapy. Finally, future perspectives and challenges of IVT mRNA are discussed.</P>

        Enrichment of Short-Chain Ceramides and Free Fatty Acids in the Skin Epidermis, Liver, and Kidneys of db/db Mice, a Type 2 Diabetes Mellitus Model

        Patients with diabetes mellitus (DM) often suffer from diverse skin disorders, which might be attributable to skin barrier dysfunction. To explore the role of lipid alterations in the epidermis in DM skin disorders, we quantitated 49 lipids (34 ceramides, 14 free fatty acids (FFAs), and cholesterol) in the skin epidermis, liver, and kidneys of db/db mice, a Type 2 DM model, using UPLC-MS/MS. The expression of genes involved in lipid synthesis was also evaluated. With the full establishment of hyperglycemia at the age of 20 weeks, remarkable lipid enrichment was noted in the skin of the db/db mice, especially at the epidermis and subcutaneous fat bed. Prominent increases in the ceramides and FFAs (>3 fold) with short or medium chains (<C26) occurred in the skin epidermis (16NS, 18NS, 24NS, 16NDS, 18NDS, 20NDS, 22NDS, 24NDS, C16:1FA, C18:2FA, and C18:1FA) and the liver (16NS, 18NS, 20NS, 24:1NS, 18NDS, 20NDS, 22NDS, C16:1FA, C18:2FA, C18:1FA), whereas those with very long chains were not affected. In the kidney, only slight increases (<3 fold) were observed for 16NS, 18NS, 20NS, 26NDS, C26FA, and C22:1FA. Consistently, LXRα/β and PPARγ, nuclear receptors promoting lipid synthesis, lipid synthesis enzymes such as elongases 1, 4, and 6, and fatty acid synthase and stearoyl-CoA desaturase were highly expressed in the skin and livers of the db/db mice. Collectively, our study demonstrates an extensive alteration in the skin and systemic lipid profiles of db/db mice, which could contribute to the development of skin disorders in DM.

The chemical is one of seven chemicals, which are under the frame of OECD SIDS program sponsored by National Institute of Environmental Research of Korea, Regarding the information on the environmental fate, bioconcentration is one of important factor to estimate the environmental tranfer. However, measurement of bioconcentration needs high expense and time. For this reason, OECD recommends to use BCFWIN model to estimate bioconcentration of organic chemicals. BCFWIN estimates the bioconcentration factor(BCF) of an organic compound using the log octanol-water partition coefficient (Kow) of the compound. Structures are entered into BCFWIN through SMILES (Simplified Molecular Input Line Entry System) notations, The BCFWIN method classifies a compound as either ionic or non-ionic. Ionic compounds include carboxylic acids, sulfonic acids and salts of sulfonic acids, and charged nitrogen compounds (nitrogen with a +5 valence such as quaternary ammonium compounds). All other compounds are classified as non-ionic. In this study, bioaccumulation of acetanilide was estimated using BCFWIN model based on SMILES notation, chemical name data and partition coefficient as one of environmental fate/distribution of the chemical elements.
