      • 小豆收量에 미치는 諸形質의 影響

        小豆의 收量에 影響하는 形質을 正確히 알기 爲하여 形質相互間의 相關關係 그리고 經路係數分析과 各形質相互間의 表現型共分散, 遺傳共分散, 環境共分散 등을 計算하고 여러가지 選拔指數를 作成하여 選拔實驗을 한바 그 結果를 要約하면 다음과 같다. (1) 各形質相互間의 表現型共分散의 값보다 遺傳共分散의 값이 大體로 크고 環境共分散은 그 값의 變動이 甚하다. (2) 遺傳相關의 값이 表現型相關보다 높으며 開花日數와 1株重量, 莖長과 1株重量, 莖直徑과 分枝數, 莖直徑과 1株重量, 莖直徑과 100粒重, 分枝數와 1株重量, 1株重量과 100粒重間에는 높은 正의 相關을 보였고 收量과는 莢數만이 높은 正의 相關을 보였다. (3) 收量에 미치는 諸形質의 直接的 影響을 보면 莢數, 莖直徑, 100粒重, 莖長, 開花日數의 順으로 收量에 正으로 影響하며, 間接的 影響을 보이는 形質은 主로 莢數는 分枝數의 增加로 收量에 間接的으로도 影響하는 것을 알수 있다. 그리고 莖直徑은 開花日數, 莢長, 分枝數, 1株重量, 100粒重과 間接的으로 作用하여 收量에 影響하고 1株重量은 莖直徑, 莢數, 100粒重의 3形質과 100粒重은 開花日數, 莖直徑, 分枝數, 1株重量, 100粒重과도 間接的으로 影響을 미치는 것을 알수 있다. (4) 各形質을 選拔對象 形質로 하여 選拔指數를 作成한바 第3表 및 第4表와 같이 形質의 組合에 따라서 選拔에 관여하는 各形質의 加重値(weight)는 다르나 부호는 大體로 同一하다. (5) 選拔指數에 依한 遺傳的 進展과 그의 關係效率을 본바 單一形質에 있어서는 莢數, 1株重量, 100粒重의 效率이 높고 單一形質보다 2個形質을 2個形質보다 3個形質을 選拔對象形質로 하여 選拔하는 것이 效果的이라는 것을 알수 있다. 以上의 結果를 미루어 보아 小豆의 形質中 莖長, 莖直徑, 莢數, 100粒重의 4個形質이 收量에 直接 또는 間接으로 影響하는 重要形質이 될 수 있을 것으로 믿는다. 그리고 小豆에 있어서는 莖長(X₂), 莖直徑(X₃), 莢數(?), 100粒重(??)의 4個形質中에서 2-3形質을 選拔對象形質로 하여 選拔指數를 作成하여 選拔에 應用하는 것이 時間과 勞力 그리고 選拔效果面에 있어서 效果的일 것으로 믿는다. The studies were intended to clarify the correlations between the characters and the influences of each character upon yield of Adzuki beans. The analysis of covariance was conducted to obtain the genotypic correlations among eight characters-days to flowering, stem length, stem diameter, number of branches per plant, number of pods per plant, plant weight, 100 grain weight and grain weight(yield), etc., and the path-coefficients were calculated by Dewey's method. Selection indexes were calculated by phenotypic and genotypic covariances between the characters, and genetic advances and relative efficiency were also calculated for these characters. The results obtained are summarized as follows: (1) The yield of Adzuki bean was mainly associated with the character, number of pods per plant. (2) The values of genotypic covariances were slightly higher than those of phenotypic covariances between the characters. (3) Stem length, stem diameter, number of pods per plant, and 100 grain weight had a direct positive influence upon the yield of Adzuki beans. (4) Number of branches per plant and total plant weight had little or negative influences upon yield but it had rather considerable indirect influence upon yield by increasing the other characters. As a result, it can be concluded that Adzuki bean yield is primarily determined by increasing number of pods per plant, in turn the yield of Adzuki bean is secondarily determined by the number of pods per plant. (5) Many selection indexes which contain 1-7 characters in calculation were obtained as shown in Tables 2 and 3, and they show the genetic advances and relative efficiency which were calculated with various selection indexes. These results suggest that selection index technique is useful in Adzuki bean breeding. In reality, however, as the selection index varies with population and environment in which the population is located. Inspite of the expected usefulness of selection index technique in Adzuki bean breeding, unsolved problems such as the expense, time and labor involved in calculating the selection index remain. For these reasons and from these studies, it was realized that the selection index for the selection should be calculated on the basis of the data of some 3-4 characters, stem length, stem diameter, number of pods per plant, and 100 grain weight, etc. These characters should be very useful agronomic characters in the selection of Korean Adzuki beans, as they could be measured or counted easily thus saving time and expense, and have a greater effect on more Adzuki bean yield than the other agronomic characters.

      • Sectional arch technique 및 Continuous arch technique에 의한 치료후 악궁 형태의 변화에 대한 연구

        This study was carried out in order to ocompare the amount of tooth movement and the arch size and shape following sectional and continuous arch technique treatment. The sample group for this study consists of 24 persons (older than Hellman stage Ⅲc, Angle's classificationⅠ), who received orthodontic treatment with first premolar extraction in both jaw at Orthodontic Department of Dankook Univ. Dental Hospital. They are classified into Sectional arch technique treatment group (GroupⅠ), Continuous arch technique treatment group (Group Ⅱ) according to methods of treatment. Following conclusions were obtained by cephalometric linear measurements and dental arch measurements using pre- and post- treatment lateral cephalograms and plaster study models. 1. Intercanine widths were increased in both jaw of Group Ⅰ and Group Ⅱ after treatment. The amount of increase in Group Ⅰ was sigmificantly larger than that of Group Ⅱ. 2. Intermolar widths were reduced in both jaw of each groups after treatment. The amount of decrease in Group Ⅱ was significantly larger than that of GroupⅠ. 3. Arch lengths were decreased in both jaw in each group after treatment. Between two groups, no difference was noted before and after treatment. 4. Considering the antero-posterior movement of canine, canine was moved posterioly in both jaw in each group. The amount of canine movement was significantly different in maxilla between two groups. But in mandible, no difference was found between them. 5. Considering the antero-posterior movement of the first molar, the first molar was moved anteriorly in both jaw in each group. But no difference was found between two group.

      • 고구마 收量에 미치는 諸形質의 直接效果 및 間接效果

        고구마의 수량에 影響하는 形質을 보다 正確하게 알기 위하여 形質相互間의 相關關係 그리고 經路係數를 分析한바 그 結果를 要約하면 다음과 같다. 1. 고구마의 收量과 關係있는 形質은莖長, 莖數, 直徑, 節數, 莖重, 地下重, 塊根數의 各形質이었다. 2. 고구마의 收量에 直接的으로 영향하는 形質은 直徑節數, 莖重, 地下重이었다. 3. 間接效果는 莖長과 地下重, 莖數와 地下重, 直徑과 地下重, 節數와 地下重, 莖重과 地下重 間에 높은 값을 보였다. 4. 고구마의 收量은 直徑이 크고 節數가 많고 莖重과 地下重이 무거운 것일수록 收量이 많다는 사실을 알 수 있다. The results obtained are summerized as follows; 1. The yield of sweet potatoes was mainly associated with the characters such as stem-length, stem-number, node-number, stem-weight, undergroundweight, etc. 2. Node-number, stem-weight and underground-weight have positively influenced directly upon the yield of sweet potatoes. 3. Stem-length, stem number and root number also had little or negative influences upon yield but it had rather considerable indirect influence upon yield by increasing of sweet potatoes. 4. As a result, it can be concluded that the yield of sweet potatoes is determined underground weight, node number and stem number etc.

      • 대장균에서 대량 생산한 B형 간염바이러스 X 단백질의 발현 특성

        The X protein of human hepatitis B virus (HBx) consists of 154 amino acids and transactivates various cellular and viral promoters and enhancers. This protein has been also implicated in virus-mediated oncogenesis of hepatocytes. However, there are still controversial reports on its mechanism related to its oncogenicity. To elucidate the presence of any correlation between immune responses against HBx protein and hepatitis pathogenicity, convenient sources for recombinant HBx protein and anti-HBx antibodies are necessary. Three recombinant vectors were constructed to express the X open reading frame (ORF) of hepatitis B virus in E. coli. Recombinant E. coli transformed with one of these plasmid, pET- AX plasmid, which containd X ORF deleted in N-terminus, did not produce HBx protein. However, when X ORF containing 154 amino acids was cloned in pET, the HBx protein was produced. A GST fusion protein containing 146 amino acids encoded by ORF X was also successfully expressed. This protein was produced in large quantities of more than-60% of total proteins However, nonfused HBx protein was produced in small quantities. As both proteins were produced in inclusion bodies, HBx recombinant GST-fusion protein was further proccessed for purification. GST-X fusion protein was solubilized by sarkosyl. The solublized protein was purified by affinity to glutathione sepharose bead. The purified HBx protein was fairly good immunogen eliciting good humoral immune responses in mice. We produced polyclonal antibodies against HBx recombinant proteins and confirmed the specificity of the antibodies.

      • Culex pipiens pallens 모기의 Trypsin과 Chymotrypsin에 관한 연구

        Culex pipiens pallens 암모기의 Trypsin과 chymotrypsin에 관하여 pH Ca^++ 이온 농도, 그리고 온도가 효소의 활성에 미치는 영향과 흡혈 후 중장 내 효소 활성의 변화를 조사하여 이를 Aedes aegypti 크기의 효소와 비교하였다. Culex pipiens 모기의 chymotrypsin은 Ca^++ 이온 존재시에 최적 pH가 8.5에서 9.5호 상승하였고 Trypsin 양은 흡혈 후 30시간에 최대에 달했으며, 온도에 대한 안정성에 있어서는 50℃에서 반감 시간이 21초였다. 그러나 이와는 달리 A. aegypi에 있어서는 Chymotrypsin의 최적 pH에 대한 Ca^++ 이온의 영향이 없었고 Trypsin 양은 흡혈 후 24시간에 최대에 이르렀으며 50℃에서의 안정성에 있어서 반감 시간이 100초였다는 점이 상이함을 관찰하였다. 이상의 관찰 결과는 두 종의 흡혈성 모기가 소지하는 Trypsin과 Chymotrypsin 분자 구조 결정의 기본이 되는 아미노산의 조성 및 서열이 서로 다르다고 지적하는 증거가 된다. Trypsin and chymotrypsin like enzymes from the midguts of larvae and the blood fed females of the mosquito, C. pipiens pallens, have been compared with those of A. aegypti. The optimum pH for the chymotrypsin activity from C. pipiens was found to be at 8.5, which rose to 9.5 when 0.03 M Calcium chloride was present in the reaction mixture. Calcium ion had no effects on A. aegypti chymotrypsin activity. Trypsin from A. aegypti was more stable to temperature than that of C. pipiens. Trypsin activity in both species of mosquitoes began to rise by 6 hours post blood metal and reached at the maximum level by 30 and 24 hours post blood meal for C. pipiens and A. aegypti, respectively. The results seem to suggest that trypsin and chymotrypsin like enzymes from these species of mosquitoes may differ in their primary structures.

        디지털 및 일반 측방두부규격방사선사진에서 측정 방법에 따른 계측치의 비교

        Purpose : To compare cephalometric measurement between measuring methods in digital and conventional lateral cephalometric radiograph. Materials and Methods : Twenty digital and conventional lateral cephalometric radiographs were selected. In digital group, cephalometric measurements were performed manually using hardcopies and automatically using VCeph^(TM) program on the monitor. In conventional group, the same measurements were performed manually on conventional films, and for automatic measurement conventional films were digitized by scanner. All measurements were performed twice by 4 observers, and 24 cephalometric variables were calculated and the time spent for each measurement was recorded. The differences in measurements data and the time spent for each measurement were compared within each group. Intra-observer and inter-observer comparisons were performed. Results : In both groups, no statistically significant difference between manual and automatic measurements was observed and most of the variables didn’t show statistically significant differences between methods. The observer with less experience tended to show statistically significant differences of measurements between methods, and differences from other observers. The differences of measurements between methods in digital group were lesser than those of conventional group with statistical significance in 8 variables out of 24. With automatic method and in digital group, the spent time was shorter. Conclusion : With direct digital radiograph, automatic method using manually idenitified landmarks can be preferable in cephalometric analysis.

      • 청가시덩굴(Smilax sieboldii Miq.)의 잎 및 지하부의 성분과 항고지혈작용에 관한 연구

        Nutritional value of the leaf and antihyperlipidemic effect of the subterranean part of Smilax siebolidd Miq.(Liliaceae) were investigated in this experiment. 8 minerals, 17 amino acids and 5 fatty acids were quantitatively analyzed in the leaf. In comparison with other vegetables, the content of amino acids was about twice as much as those and a lot of essential amino acids were found. The content of fatty acids was similar with that of other vegetables. In addition, various trace elements such as Mg, Zn and Mn were contained. The chloroform extract of the subterranean part of this plant was effective to decrease total cholesterol and triglyceride level and decrease HDL-cholesterol level in plasma of mouse.
