he current study was carried out to detect metrafenone residues and estimate the half-lives in lettuce grown under greenhouse conditions using gas chromatography with a micro-electron capture detector. Metrafenone was applied in two and three doses (with a 7-day interval) to two different sites in Naju (distance between sites 1 and 2 = 6.5 km) in different seasons (spring and autumn). Samples were extracted with acetone and matrix components were purified using a silica solid-phase extraction (SPE) cartridge. The calibration curve over the concentration range of 0.02–5.0 mg/L was linear with a determination coefficient (R2) = 0.9993. The limits of detection and quantification were 0.003 and 0.01 mg/L, respectively. Recoveries at two fortification levels (0.1 and 0.5 mg/kg) ranged from 78.5 to 113.3 % with a relative standard deviation <8 %. The calculated half-lives were 2.3 and 2.2 days and 4.9 and 5.0 days for the double and triple doses at sites 1 and 2, respectively. These results suggest that the method is effective and sensitive for the determination of metrafenone residues in field-incurred lettuce samples, and the difference in the reported half-lives is due to seasonal variation.

<P><B>Abstract</B></P> <P>The residual behavior of the systemic fungicide, metalaxyl, in Swiss chard cultivated at two different locations under greenhouse conditions was investigated using high-performance liquid chromatography coupled with an ultraviolet detector (HPLC-UVD). Samples were randomly collected over 14 days and extracted using acetonitrile, partitioned using solid sodium chloride, and a solid-phase extraction (SPE) NH<SUB>2</SUB> cartridge was used for cleanup. The linearity over a concentration range 0.05–50 mg/L was excellent with a coefficient of determination (<I>R</I> <SUP> <I>2</I> </SUP>) of 0.9997. The recovery rate ranged from 77.05 to 88.92% with relative standard deviations (RSDs) ≤ 10.74, and the limits of detection (LOD) and quantification (LOQ) were 0.0033 and 0.01 mg/kg, respectively. The initial (2 h after application) deposits were 4.69 and 5.90 mg/kg for sites 1 and 2, respectively, which increased to 4.95 and 6.57 mg/kg, respectively, one day post-application, owing to the systemic properties of the fungicide. The dissipation half-life was 5.3 and 6.0 days for sites 1 and 2, respectively. The pre-harvest residue limit (PHRL) suggested that if 55.38 and 47.23 mg/kg was applied 10 days before harvest or 33.28 and 30.73 mg/kg was applied 5 days before harvest (for sites 1 and 2, respectively) then the concentration will fall below the maximum residue limit (MRL = 20.0 mg/kg) at the time of harvest. The dietary risk assessment, estimated as hazard quotient (RQ%), indicate that metalaxyl can be safely used in/on Swiss chard, with no hazardous effects expected for consumers.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Metalaxyl residual behavior was investigated under greenhouse conditions. </LI> <LI> Swiss chard were extracted with ACN and analyzed using HPLC-UVD. </LI> <LI> The method performance was good with satisfactory parameters. </LI> <LI> Residues were decreased by time, except for 1 day post-application. </LI> <LI> The analyte can be safely used in Swiss chard with no hazardous effects expected for consumers. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

An efficient protocol was established to produce virus free in vitro banana plantlets through meristem culture of virus infected four months old banana suckers cv. Sabri. Murashige and Skoog (MS) medium supplemented with different levels of BAP (0, 3, 4 and 5 ㎎ · ℓ?¹) and IAA (0.0, 1.0, 1.5, 2.0 and 2.5 ㎎ · ℓ?¹) were used for shoot proliferation in Experiment-I. Half strength of MS medium supplemented with different levels of IBA (0, 1, 2, 3 ㎎ · ℓ?¹), NAA (0, 2, 3, 4 ㎎ · ℓ?¹) were treated for root formation in Experiment-II. ELISA test was performed to confirm virus eradication rate of regenerated plants and survival rate of regenerated plants in Experiment-III. The treatment combination of 4 ㎎ · ℓ?¹ BAP + 2.5 ㎎ · ℓ?¹ IAA produced the highest number of shoots (3.4 explant-1), while the longest shoot (3.49 cm), maximum number of leaves (3.37 plantlet-1) and longest leaves (2.22 cm) were obtained in the culture treated with 5 ㎎ · ℓ?¹ BAP. On the contrary, for plantlets cultured on half strength of MS medium supplemented with different levels of IBA and NAA, the highest number of vigorous roots (8.01 explant-1) and maximum root length (4.64 cm) were observed at treatment combinations of 2 ㎎ · ℓ?¹ IBA + 4 ㎎ · ℓ?¹ NAA and 2 ㎎ · ℓ?¹ IBA+ 3 ㎎ · ℓ?¹ NAA, respectively. The combinations of 3 ㎎ · ℓ?¹ IBA + 3 ㎎ · ℓ?¹ NAA produced more vigorous roots while less vigorous roots were observed at respective control. The rate of virus free plant regeneration from banana bunchy top virus (BBTV) infected sources was 57.14%. However, the highest plant survival rate (83.33%) was observed in a mixture of sand, soil and cow dung (1:1:1) and in a mixture of coir and soil (1:2). These results suggested that eradication of BBTV through meristem culture can be used as an effective tool for commercial exploitation of virus free banana production.

The dissipation pattern of metrafenone in green leafy vegetables (lettuce) grown under green house condition were investigated using GC-μECD. Residues were extracted using acetone and samples were taken out by liquid-liquid extraction. Matrix component was cleaned by SPE silica cartridge. Calibration curves were linear over the concentration ranges (0.02-5.0 mg/kg) with a determination co-efficient (r2)≥0.999. The limit of detection (LOD) and quantification (LOQ) were 0.006 and 0.02 mg/kg respectively. Recoveries at two fortification level were ranged between 78.5% and 113.3% with relative standard deviation (RSD)<9%. Residues half-life from dissipation pattern of site 1 and 2 double/triple spray was 2.3/2.2 and 4.9/5.0 days respectively. The method was effective and sensitive enough to determine the residue of metrafenone in field-incurred lettuce sample.
