        Design and applications of fluorescent detectors for peroxynitrite

        <P><B>Abstract</B></P> <P>Peroxynitrite (ONOO<SUP>−</SUP>) is one of the endogenous reactive oxygen species (ROS), which causes damage to a wide array of molecular components in the cells, including DNA and proteins, owing to its high oxidizing as well as nitrating properties. However, the precise pathogenic roles played by this substance in biological systems have not yet been elucidated completely owing to its short lifetime, high reactivity, low concentration and elusive nature in the <I>in vivo</I> applications. Thus, the development of more sensitive and selective techniques for detecting ONOO<SUP>−</SUP>, with high biocompatibilities, sensitivities, and site-specificities, is a significant goal.         Epidemiology, Characteristics, and Prognostic Factors of Primary Atypical Teratoid/Rhabdoid Tumors in the Spinal Canal: A Systematic Review

        Primary atypical teratoid/rhabdoid tumors (AT/RTs) in the spinal canal are rare central nervous system (CNS) neoplasms that are challenging to diagnose and treat. To date, there has been no standard treatment regimen for these challenging malignant tumors. Thus, we conducted this research to explore potential prognostic factors and feasible treatment modalities for improving the prognosis of these tumors. Articles were retrieved from the PubMed, MEDLINE, and Embase databases, using the keywords “atypical teratoid/rhabdoid tumor,” “rhabdoid tumor,” “spine,” “spinal,” “spinal neoplasm”, and “spinal cord neoplasm.” All eligible cases demonstrated SMARCB1-deficient expression validated by pathological examination. We collected and analyzed data related to clinical presentation, radiological features, pathological characteristics, treatment modalities and prognosis via Kaplan-Meier and Cox regression analyses. Thirty-six articles comprising 58 spinal AT/RT patients were included in the study. The median progression-free survival (PFS) and overall survival (OS) were 18 and 22 months, respectively. Kaplan-Meier analysis demonstrated significant survival improvements for OS in the nonmetastasis, male, radiotherapy and intrathecal chemotherapy groups as well as for PFS in the chemotherapy and radiotherapy groups. Multivariate analysis revealed that chemotherapy and radiotherapy were prognostic factors for improved PFS, and that intrathecal chemotherapy reduced the risk of mortality. Spinal AT/RTs are uncommon malignant entities with a dismal survival rate. Although our review is limited by variability between cases, there is some evidence revealing potential risk factors and the importance of systematic chemotherapy, intrathecal chemotherapy and radiotherapy in spinal AT/RT treatment modalities.

        Long Noncoding RNA Expression Profiling During the Neuronal Differentiation of Glial Precursor Cells from Rat Dorsal Root Ganglia

        Long noncoding RNAs (lncRNAs) play important roles in the process of cell fate determination. However, their function and expression profiles have not yet been systematically investigated during the transdifferentiation of glial precursor cells derived from dorsal root ganglia (DRG) in the peripheral nervous system. Our results demonstrated significant differences in gene architecture and expression among the three transcript types (lncRNA, mRNA, and TUCP). Distinct differences in transcript length, exon number, and ORF length were identified between lncRNAs and mRNAs after comparative analysis of their structure and sequence conservation. We found that the upregulated lncRNAs outnumbered the downregulated lncRNAs in glial precursor cells cultured with proBDNF antiserum compared with the levels in glial precursor cells cultured without proBDNF antiserum. By a series of GO and KEGG analyses, we found that the effects of some lncRNAs on their target genes in cis were related to nerve growth factor-induced cell cycle, cell phenotype change, and neuronal differentiation. The qRT-PCR verification results of lncRNAs ENSRNOT00000091991, ENSRNOT00000087717, and LNC_000429 were mostly consistent with the sequencing results. The candidate lncRNAs may be associated with the neuronal transdifferentiation of glial precursor cells. Our study provides the first evidence for a remarkably diverse pattern of lncRNA expression during neuronal differentiation of glial precursor cells from rat DRG, and also provides a resource for lncRNA studies in the field of cell differentiation.

      • Development and Pilot Application of an Integrated Support Model for Disadvantaged Young Children in Poor Rural China

        As the biggest developing country in the world, China is now tackling the pressing problems of child poverty, whilst facing the increasing aging population at the same time. In August 2009 and April 2010, the China Development Research Foundation (CDRF) officially launched the Project Equal Start: Program of Early Childhood Development in Poor Rural Areas in Qinghai Province and Yunnan Province, which aims at enhancing early childhood development and tackling poverty in poor rural areas. In this article, we described the development the pilot programs’ sites, objects and phases, and also evaluated the significant improvement in multiple dimensions such as children’s early development, education attainments, and parental involvement. At last, it analysed the CDRF’S pilot programs’ policy advocacy, and prospected its influences over how to universalize early childhood development and education in poor rural China.

      • Giant thermal conductivity in diamane and the influence of horizontal reflection symmetry on phonon scattering

        <P>Diamane, a chemically derived two-dimensional material, shows many superior physical and chemical properties similar to diamond thin films. Through the Peierls-Boltzmann transport equation, we reveal giant thermal conductivity in diamane with a stacking order of both AB and AA (respectively, abbreviated as D-AB and D-AA, hereafter) which are both comparable to that of diamond. Like in graphene, the phonon transport falls into the hydrodynamic regime even at room temperature, and the major contribution to the total thermal conductivity comes from the out-of-plane acoustic phonon modes (>40%). In addition, the thermal conductivity shows a dependence on the stacking order, namely, the thermal conductivity of D-AA, ∼2240 W m<SUP>−1</SUP> K<SUP>−1</SUP> at 300 K, is around 15% larger than that of D-AB, which is due to the strong restriction on the phonon scattering phase space induced by the horizontal reflection symmetry in D-AA. Such a kind of restriction, not limited to single atomic plane systems, is a general feature in two-dimensional materials with a horizontal reflection symmetry.</P>

        Green multi-functional monomer based ion imprinted polymers for selective removal of copper ions from aqueous solution

        <P><B>Abstract</B></P> <P>Green ion imprinted polymers (IIPs) were prepared in aqueous phase via the synergy of three functional monomers of low-cost eco-friendly gelatin (G), 8-hydroxyquinoline (HQ) and chitosan (C), namely G-HQ-C IIPs, and were applied as an effective and recyclable adsorbent to remove Cu(II) from aqueous solution. The as-prepared G-HQ-C IIPs were systematically characterized, and several major factors affecting adsorption capacity including solution pH, temperature and contact time were investigated in detail. The adsorption of Cu(II) on G-HQ-C IIPs followed the pseudo-second-order kinetic and Langmuir isotherm models, and the adsorption capacity increased with temperature increase. Moreover, the maximum adsorption capacities of G-HQ-C IIPs toward Cu(II) reached up to 111.81 mg/g at room temperature, much higher than those of most of the reported adsorbents for Cu(II). The G-HQ-C IIPs displayed excellent selectivity against seven common divalent ions with selectivity coefficients above 18.71, as well as high anti-interference ability. Additionally, a good reusability was demonstrated without significant loss in adsorption capacity after at least ten cycles. The IIPs were applied to environmental water samples for selective removal of Cu(II) with satisfactory results. By replacing Cu(II) template by Cd(II), Hg(II) and Pb(II), respectively, the obtained three kinds of IIPs based on G-HQ-C presented convincing imprinting properties, and therefore the work could provide a simple and general imprinting strategy toward various concerned heavy metal ions through multi-point interactions from multiple functional monomers.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Hypoxia-induced LncRNA DACT3-AS1 upregulates PKM2 to promote metastasis in hepatocellular carcinoma through the HDAC2/FOXA3 pathway

        Growing evidence has revealed that hypoxia is involved in multiple stages of cancer development. However, there are limited reports on the effects of long noncoding RNAs (lncRNAs) on hepatocellular carcinoma (HCC) progression under hypoxia. The main purposes of this study were to analyze the effect of the novel lncRNA DACT3-AS1 on metastasis in HCC and to elucidate the related molecular mechanism. Bioinformatics tools were employed. RT–qPCR or western blot assays were conducted to detect RNA or protein expression. Clinical samples and in vivo assays were utilized to reveal the role of DACT3-AS1 in HCC. Other mechanism and functional analyses were specifically designed and performed as well. Based on the collected data, this study revealed that HIF-1α transcriptionally activates DACT3-AS1 expression under hypoxia. DACT3-AS1 was verified to promote metastasis in HCC. Mechanistically, DACT3-AS1 promotes the interaction between HDAC2 and FOXA3 to stimulate FOXA3 deacetylation, which consequently downregulates the FOXA3 protein. Furthermore, FOXA3 serves as a transcription factor that can bind to the PKM2 promoter region, thus hindering PKM2 expression. To summarize, this study uncovered that HIF-1α-induced DACT3-AS1 promotes metastasis in HCC and can upregulate PKM2 via the HDAC2/FOXA3 pathway in HCC cells.

        The Regulation of Phosphorus Release by Penicillium chrysogenum in Different Phosphate via the TCA Cycle and Mycelial Morphology

        Phosphate-solubilizing fungi (PSF) efficiently dissolve insoluble phosphates through the production of organic acids. This study investigates the mechanisms of organic acid secretion by PSF, specifically Penicillium chrysogenum, under tricalcium phosphate ( Ca3(PO4)2, Ca–P) and ferric phosphate ( FePO4, Fe–P) conditions. Penicillium chrysogenum exhibited higher phosphorus (P) release efficiency from Ca-P (693.6 mg/L) than from Fe–P (162.6 mg/L). However, Fe–P significantly enhanced oxalic acid (1193.7 mg/L) and citric acid (227.7 mg/L) production by Penicillium chrysogenum compared with Ca–P (905.7 and 3.5 mg/L, respectively). The presence of Fe–P upregulated the expression of genes and activity of enzymes related to the tricarboxylic acid cycle, including pyruvate dehydrogenase and citrate synthase. Additionally, Fe–P upregulated the expression of chitinase and endoglucanase genes, inducing a transformation of Penicillium chrysogenum mycelial morphology from pellet to filamentous. The filamentous morphology exhibited higher efficiency in oxalic acid secretion and P release from Fe–P and Ca–P. Compared with pellet morphology, filamentous morphology enhanced P release capacity by > 40% and > 18% in Ca–P and Fe–P, respectively. This study explored the strategies employed by PSF to improve the dissolution of different insoluble phosphates.

        Characterization of a Strain of Malva Vein Clearing Virus in Alcea rosea via Deep Sequencing

        Malva vein clearing virus (MVCV) is a member of the Potyvirus species, and has a negative impact on the aesthetic development of Alcea rosea. It was first reported in Germany in 1957, but its complete genome sequence data are still scarce. In the present work, A. rosea leaves with vein-clearing and mosaic symptoms were sampled and analyzed with small RNA deep sequencing. By denovo assembly the raw sequences of virus-derived small interfering RNAs (vsiRs) and whole genome amplification of malva vein cleaning virus SX strain (MVCVSX) by specific primers targeting identified contig gaps, the full-length genome sequences (9,645 nucleotides) of MVCV-SX were characterized, constituting of an open reading frame that is long enough to encode 3,096 amino acids. Phylogenetic analysis showed that MVCVSX was clustered with euphorbia ringspot virus and yam mosaic virus. Further analyses of the vsiR profiles revealed that the most abundant MVCV-vsiRs were between 21 and 22 nucleotides in length and a strong bias was found for “A” and “U” at the 5′-terminal residue. The results of polarity assessment indicated that the amount of sense strand was almost equal to that of the antisense strand in MVCV-vsiRs, and the main hot-spot region in MVCV-SX genome was found at cylindrical inclusion. In conclusion, our findings could provide new insights into the RNA silencing-mediated host defence mechanism in A. rosea infected with MVCV-SX, and offer a basis for the prevention and treatment of this virus disease.
