        Genetic Profiles Associated with Chemoresistance in Patient-Derived Xenograft Models of Ovarian Cancer

        Purpose Recurrence and chemoresistance (CR) are the leading causes of death in patients with highgrade serous carcinoma (HGSC) of the ovary. The aim of this study was to identify genetic changes associated with CR mechanisms using a patient-derived xenograft (PDX) mouse model and genetic sequencing. Materials and Methods To generate a CR HGSC PDX tumor, mice bearing subcutaneously implanted HGSC PDX tumors were treated with paclitaxel and carboplatin. We compared gene expression and mutations between chemosensitive (CS) and CR PDX tumors with whole exome and RNA sequencing and selected candidate genes. Correlations between candidate gene expression and clinicopathological variables were explored using the Cancer Genome Atlas (TCGA) database and the Human Protein Atlas (THPA). Results Three CR and four CS HGSC PDX tumor models were successfully established. RNA sequencing analysis of the PDX tumors revealed that 146 genes were significantly up-regulated and 54 genes down-regulated in the CR group compared with the CS group. Whole exome sequencing analysis showed 39 mutation sites were identified which only occurred in CR group. Differential expression of SAP25, HLA-DPA1, AKT3, and PIK3R5 genes and mutation of TMEM205 and POLR2A may have important functions in the progression of ovarian cancer chemoresistance. According to TCGA data analysis, patients with high HLADPA1 expression were more resistant to initial chemotherapy (p=0.030; odds ratio, 1.845). Conclusion We successfully established CR ovarian cancer PDX mouse models. PDX-based genetic profiling study could be used to select some candidate genes that could be targeted to overcome chemoresistance of ovarian cancer.

        Alternaria yunnanensis sp. nov., a New Alternaria Species Causing Foliage Spot of Rubber Tree in China

        A new species of Alternaria causing leaf spots on the rubber tree (Hevea brasiliensis) in Yunnan, China, was isolated, examined, and illustrated. Morphologically, it belongs to the section Porri of Alternaria, which produces relatively large conidia and a simple or branched, filamentous long beak. It is, however, characterized by conidiophores gradually enlarging near the apex into a clavate conidiogenous cell and long ellipsoid to obclavate, smoothwalled conidia with a long filamentous beak. Molecular phylogenetic analyses based on ITS rDNA, GAPDH, and TEF1-alpha sequences demonstrate that the phytopathogen falls in the clade of the section Porri, being most closely related to A. sidae, A. sennae, A. deseriticola, A. cyamopsidis, A. rostellata, A. nitrimali, A. crassa, and A. thunbergiae.

        The critical external heat source conditions for subcritical ORC system based on thermo-economics

        Considering the thermodynamic and economic constraints simultaneously, the critical external heat source conditions available for sub-ORC systems with pure or zeotropic mixture working fluids are clarified. The critical external heat source conditions include the maximum allowed heat source temperature T gi,max , the minimum allowed heat source temperature T gi,min and the minimum allowed heat source mass flow rate m g,min . The results show that, no matter using pure working fluid or zeotropic mixture working fluid, the maximum allowed heat source temperature T gi,max increases linearly with the increasing of its critical temperature T cr . For the pure working fluid, the minimum allowed heat source temperature T gi,min is not sensitive to the variation of T cr . While, for the zeotropic mixture working fluid, the minimum allowed heat source temperature T gi,min firstly falls down, then rises up along with the increasing of T cr . An increase in mass flow rate of heat source causes enlarged range of heat source temperature available. And the minimum allowed heat source mass flow rate m g,min reduces sharply with the increase of heat source temperature.

        A Potential Role of Group 2 Innate Lymphoid Cells in Eosinophilic Chronic Rhinosinusitis With Nasal Polyps

        Chronic rhinosinusitis with nasal polyps (CRSwNP), a type 2-based upper airway disease, is mainly characterized by high asthma comorbidity and recurrence after surgery. It has been shown that type 2 cytokines, including interleukin (IL)-4, IL-5, and IL-13 released from T helper 2 (Th2) cells as well as group 2 innate lymphoid cells (ILC2s), contribute to chronic inflammation of CRSwNP. This review summarizes recent progresses made in our understanding of ILC2 activity, particularly ILC2 accumulation at airway inflammation sites, cooperation with Th2 cells in aggravating the CRSwNP inflammatory process and interactions with regulatory T cells (Tregs) in resisting Tregs-mediated suppressive function in allergic inflammation. A better understanding of the biology of ILC2s should lay a good foundation in elucidating the pathogenesis of CRSwNP, and subsequently may lead to the development of new therapeutic strategies for the management of CRSwNP.

        Structural behavior of the stiffened double-skin profiled composite walls under compression

        Steel-concrete composite walls have been proposed and developed for applications in various types of structures. The double-skin profiled composite walls, as a natural development of composite flooring, provide structural and architectural merits. However, adequate intermediate fasteners between profiled steel plates and concrete core are required to fully mobilize the composite action and to improve the structural behavior of the wall. In this research, two new types of fasteners (i.e., threaded rods and vertical plates) were proposed and three specimens with different fastener types or fastener arrangements were tested under axial compression. The experimental results were evaluated in terms of failure modes, axial load versus axial displacement response, strength index, ductility index, and load-strain relationship. It was found that specimen with symmetrically arranged thread rods sustained more stable axial strain than that with staggered arranged threaded rods. Meanwhile, vertical plates are more suitable for practical use since they provide stronger confinement to profiled steel plate and effectively prevent the steel plate from early local buckling, which eventually enhance the composite action and increase the axial compressive capacity of the wall. The calculation methods were then proposed and good agreement was observed between the test results and the predicted results.

        Profiling Gene Expression During Gland Morphogenesis of a Glanded and a Glandless Upland Cotton

        The pigment gland is an important character of the Gossypium plant. With the aim of identifying genes involved in pigment gland morphogenesis in cotton, gene expression during pigment gland morphogenesis in Chuan 2802, which is glanded both in seed and plant, and a glandless line N5 was profiled using Affymetrix Cotton microarray. The results showed that there were 564 differentially expressed genes greater than twofold during gland morphogenesis. About 60.2% of these genes shares similarity with known genes on GenBank and about 39.8% with no functional description in the database. These described genes may play roles in defense response, response to oxidative stress, peroxidase activity, and the other metabolic pathways. The KEGG Orthology-Based Annotation System indicated that these above twofold expressed genes involved seven biochemical pathways on KEGG. These findings suggest that a complicated regulation is associated with pigment gland morphogenesis and the associated defense response including gossypol biosynthesis in cotton.

        Compressive behavior of profiled double skin composite wall

        Profiled composite slab has been widely used in civil engineering due to its structural merits. The extension of this concept to the bearing wall forms the profiled composite wall, which consists of two external profiled steel plates and infill concrete. This paper investigates the structural behavior of this type of wall under axial compression. A series of compression tests on profiled composite walls consisting of varied types of profiled steel plate and edge confinement have been carried out. The test results are evaluated in terms of failure modes, load-axial displacement curves, strength index, ductility ratio, and load-strain response. It is found that the type of profiled steel plate has influence on the axial capacity and strength index, while edge confinement affects the failure mode and ductility. The test data are compared with the predictions by modern codes such as AISC 360, BS EN 1994-1-1, and CECS 159. It shows that BS EN 1994-1-1 and CECS 159 significantly overestimate the actual compressive capacity of profiled composite walls, while AISC 360 offers reasonable predictions. A method is then proposed, which takes into account the local buckling of profiled steel plates and the reduction in the concrete resistance due to profiling. The predictions show good correlation with the test results.

        Ets-1 enhances tumor migration through regulation of CCR7 expression

        Ets-1 is a prototype of the ETS protein family. Members of the ETS protein family contain a unique ETS domain. Ets-1 is associated with cancer progression and metastasis in many types of cancer. Many studies have shown a link between elevated expression of Ets-1 in cancer biopsies and poor survival. CCR7 is a chemokine that binds to specific ligand CCL21/CCL19. CCR7 expression is associated with tumor metastasis and infiltration into lymph nodes. The objective of this study was to test whether Ets-1 could regulate CCR7 expression and enhance tumor metastasis. Our data showed that CCR7 expression was downregulated in Ets-1-deficient T cells upon T-cell stimulation. Overexpression of Ets-1 increased CCR7 expression in breast cancer cell lines. In contrast, knockdown of Ets-1 reduced CCR7 expression. Ets-1 could directly bind to CCR7 promoter and mediate CCR7 expression in luciferase reporter assays and chromatin immunoprecipitation assays. Transactivation activity of Ets-1 was independent of the Pointed domain of Ets-1. Ets-1 could also enhance NF- κB and CBP transactivation of CCR7 promoter. Our results also showed that Ets-1 could modulate cancer cell transmigration by altering CCR7 expression in transwell assay and wound healing assay. Taken together, our data suggest that Ets-1 can enhance CCR7 expression and contribute to tumor cell migration. [BMB Reports 2019; 52(9): 548-553]
