Toxic hepatitis has been reported as a major cause of acute hepatitis, but its potential induction by herbal remedies and/or health foods is usually neglected. We experienced a case of toxic hepatitis associated with Polygoni multiflori, a Chinese herb commonly known as Ho-Shou-Wu. A 54-year-old woman consumed Ho-Shou-Wu for 1 month, after which she experienced fatigue and overall weakness. A diagnosis of toxic hepatitis was made based on her clinical history, the findings for viral markers and other laboratory data, and ultrasonography. Her condition improved considerably after she stopped taking Ho-Shou-Wu. However, she resumed taking Ho-Shou-Wu immediately after discharge from hospital, which aggravated her symptoms and liver function. She was immediately readmitted and stopped taking Ho-Shou-Wu. Her relapse into hepatitis immediate after resuming consumption of the herb is strongly indicative of the validity of Koch`s postulate in this case.
국내산 전호잎 추출물의 기능성 원료 표준화를 위한 지표성분 Cynaroside의 RP-HPLC 분석법 유효성 검증

본 연구는 항염 효과 및 관절 연골 개선 효과가 있는 전호잎 추출물에 대해 개별인정형 건강기능식품의 기능성 원료 표준화의 일환으로 전호잎 추출물의 지표성분으로 cynaroside를 설정하고 이에 대한 HPLC 분석법 확립을 위한 분석법 유효성의 검증을 실시하였다. Cynaroside의 분석은 0.1% trifluoroacetic acid가 포함된 water와 acetonitrile을 이동상으로 하고 octadecyl-silica의 역상 컬럼을 사용하여 기울기 용리법으로 분리하고 최대흡수파장을 고려하여 345 nm에서 검출하였다. 분석법 유효성 검증은 특이성, 직선성, 정확성, 정밀성 등을 확인하였으며 표준물질과 전호잎 추출물 간의 HPLC 크로마토그램의 머무름시간과 UV 및 ESI-TOF/MS spectrum의 일치 여부 등의 비교를 통해 다른 물질과 간섭 없이 분리된 것으로 특이성을 확인하였다. 표준용액과 시험용액의 농도별 검량선의 상관계수는 0.9999 이상으로 나타나 높은 직선성으로 분석에 적합한 것으로 확인되었으며, 반응의 표준편차와 검량선의 기울기에 근거한 검출 한계는 0.26 mg/L였고 정량한계는 0.78 mg/L로 전호잎 추출물의 cynaroside 함량 분석을 위한 적합한 한계수준으로 확인되었다. 전호잎 추출물에 표준물질을 25, 50, 80 mg/L의 3개 수준으로 첨가하고 분석한 회수율은 98.55±0.81~102.53±1.11%였으며, RSD는 0.25~1.09%로 나타나 RSD 2.0% 이하로서 높은 정확성이 있는 것을 확인할 수 있었다. 전호잎 추출물의 조제농도 1.0~5.0 mg/mL 범위에서 일내 반복성은 RSD 0.86~0.91%으로 나타났고 일간 반복의 실험실 내 정밀성은 RSD 0.81~1.47%로 확인되어 본 분석법은 정밀성이 있음을 확인하였다. 이상의 결과로 전호잎 추출물의 지표성분인 cynaroside의 HPLC 분석법은 국내외 가이드라인에 적합한 시험법으로 검증되었으며, 본 연구에서 사용한 3회 제조 단위의 전호잎 추출물 중 cynaroside는 2.18%(w/w) 수준으로 함유되어 있는 것으로 확인되었다. 본 시험법은 전호잎 추출물의 개별인정형 건강기능식품 기능성 원료 인정을 위한 표준화의 기초자료로 활용될 것으로 기대한다. This study was undertaken to establish an analytical method using RP-HPLC for standard determination of cynaroside in the leaves extract of domestic Anthriscus sylvestris (L.) Hoffm., as a functional ingredient. The analytical method of cynaroside was optimized by HPLC on a reversed-phase octadecyl-silica (ODS) column with a gradient elution of 0.1% (v/v) trifluoroacetic acid (TFA) and methanol at 40°C and flow rate of 1.0 mL/min. The analyte was detected at 345 nm. The specificity was confirmed by the chromatogram obtained from the HPLC analytical method, as well as the results of UV and ESI-TOF mass spectra, and the coefficient of correlation (R²) obtained was 0.9999, which confirmed this as a suitable analysis with high linearity. The limit of detection (LOD) was 0.26 mg/L, and limit of quantitation (LOQ) was 0.78 mg/L, which was confirmed as a suitable limit level for the analysis of cynaroside content in the Anthriscus sylvestris (L.) Hoffm. leaves extract. The recovery was determined to be 98.55±0.81∼102.53±1.11%, and the relative standard deviation (RSD) was 0.25∼1.09%, indicating high accuracy. The intra-day repeatability and the intra-laboratory precision of the daily repetition were confirmed to be 0.86∼0.91% and 0.81∼1.47%, respectively, for the RSD. These results validate the established HPLC analytical method, by confirming the specificity, linearity, precision, and accuracy to be within the ICH guideline.
막 결합형 연속회분식 반응조에서 침지형 막의 여과 특성

The effects of sequencing batch reactor (SM-SBR) process operation methods on membrane separation character and removal efficiency of organics and nutrients were investigated to minimize the membrane contamination and for simultaneous removal. The operation pressure of membrane separation was controlled to be maintained lower than critical permeable flux to minimize the membrane contamination which was defect of membrane bioreactor (MER) process. The contaminated was intermittently fed into membrane to extend operation period. When SM-SBR process operation cycle was 6~12 hr/cycle, the contamination of membrane was decreased and the operation period could be extended. Also the removal efficiencies of total nitrogen and total phosphorus were improved. The removal efficiency of organic matter was over 90% for all operation cycles. From these results, the SM-SBR process, use simultaneous removal of organics and nutrients, developed in this study can be used for small scale water recycle system.
