        Monocytic Differentiation of K562 Cells Induced by Proanthocyanidins from Grape Seeds

        Grape seeds procyanidins can inhibit the proliferation of some cancer cell lines and have strong antioxidant activity. The purpose of this study was to investigate whether grape seeds procyanidins affect the proliferation and redifferentiation in K562 cells. The sulforhodamine B colorimetric assay and trypan blue staining were used to measure cell proliferation and survival. Morphological changes, NBT reductive activity, and surface antigens were used to detect redifferentiation of K562 cells. Intracellular reactive oxygen species (iROS) were detected by a fluorescent probe. Grape seeds procyanidins inhibited cell proliferation but the treatment did not appreciably increase lethality. After treatment with grape seeds procyanidins, a typical differentiated morphology was observed. The positive rate of CD11b and CD14 cells and NBT reductive activities increased significantly. As antioxidants, grape seeds procyanidins can induce arrest in the phase G1 and decrease iROS formation. All results indicate that the antioxidant grape seeds procyanidins are likely to induce monocytic differentiation in leukemia cells, mostly through decreasing iROS formation and inducing phase G1 arrest.

        Influence of Rashba Spin-orbit Coupling on Fano-Kondo Effect in a Parallel Double-quantum-dot Structure

        Electron transport through a parallel double-quantum-dot structure is theoretically studied withone dot in the Kondo region and the other one in the region of local Rashba interaction. Thecoupling between the Kondo dot and the electron reservoirs is found to offer a reference channel forelectron travels, and the connection of the Rashba dot and the leads is found to afford a resonantchannel. The interplay of these two mechanisms gives rise to the Fano effect, which also depends onthe adjustment of the Rashba field. We then conclude that in this structure, the Fano interferenceis more robust because it causes a quenching of the Kondo resonance. In the presence of a localmagnetic flux, the quenched Kondo resonance becomes determined by the electron spin, which ishelpful for spin manipulation.

        Inhibitory effects of piceatannol on human cytomegalovirus (hCMV) in vitro

        Human cytomegalovirus (hCMV) is a ubiquitous herpesvirus, which results in the establishment of a latent infection that persists throughout the life of the host and can be reactivated when the immunity is low. Currently, there is no vaccine for hCMV infection, and the licensed antiviral drugs mainly target the viral enzymes and have obvious adverse reactions. Thus, it is important to search for compounds with antihCMV properties. The present study aimed to investigate the suppressive effects of piceatannol on hCMV Towne strain infection and the putative underlying mechanisms using human diploid fibroblast WI-38 cells. Piceatannol supplementation prevented the lytic changes induced by hCMV infection in WI-38 cells. Furthermore, piceatannol suppressed the expression of hCMV immediate-early (IE) and early (E) proteins as well as the replication of hCMV DNA in a dose-dependent manner. Moreover, hCMV-induced cellular senescence was suppressed by piceatannol, as shown by a decline in the senescence-associated β-galactosidase (SA-β-Gal) activity and decreased production of intracellular reactive oxygen species (ROS). p16INK4a, a major senescence-associated molecule, was dramatically elevated by current hCMV infection that was attenuated by pre-incubation with piceatannol in a dose-dependent manner. These results demonstrated that piceatannol suppressed the hCMV infection via inhibition of the activation of p16INK4a and cellular senescence induced by hCMV. Together, these findings indicate piceatannol as a novel and potent anti-hCMV agent with the potential to be developed as an effective treatment for chronic hCMV infection.

        Hepatoprotective effect of <i>Amomum xanthoides</i> against dimethylnitrosamine-induced sub-chronic liver injury in a rat model

        <P><I>Context</I>: <I>Amomum xanthioides</I> Wall. ex Baker (Zingiberaceae) is a tropical medicinal plant that is commonly utilized in the treatment of digestive system disorders in Asia for a long time.</P><P><I>Objective</I>: This study aimed to evaluate the hepatoprotective effect and related mechanisms of <I>A. xanthoides</I>.</P><P><I>Materials and methods</I>: Sub-chronic liver injury was induced by dimethylnitrosamine (DMN, 10 mg/kg, three times per week for 3 weeks, i.p.) in rats. Water extract of <I>A. xanthoides</I> (WAX, 50 and 100 mg/kg) was given once a day for 3 weeks.</P><P><I>Results and conclusion</I>: WAX (100 mg/kg) significantly attenuated the DMN-induced excessive release of alanine aminotransferase (123.6 IU/L), aspartate aminotransferase (227.9 IU/L), alkaline phosphatase (820.9 IU/L) and total bilirubin (0.50 g/dL) in serum (<I>p</I> < 0.01), and hydroxyproline (30.5 mg/g tissue) and malondialdehyde (MDA) (53.6 μM/g tissue) contents (<I>p</I> < 0.01) in liver tissue. Furthermore, WAX significantly ameliorated the depletion of total antioxidant capacity (2.54 μM/mg tissue), superoxide dismutase (0.30 U/mg tissue), glutathione (2.10 μM/mg tissue) and catalase (605.0 U/mg tissue) activities (<I>p</I> < 0.05 or <I>p</I> < 0.01) in liver tissue. Histopathological and immunohistochemical analyses indicated that WAX markedly reduced inflammation, necrosis, collagen accumulation and activation of hepatic satellite cells in the liver. Our findings demonstrated that <I>A. xanthoides</I> exerts favorable hepatoprotective effects via positive regulation of the antioxidative system.</P>

        Explicit Solution and Stability of Linear Time-Varying Differential State Space Systems

        Linear time-varying (LTV) systems naturally arise when one linearizes nonlinear systems about a trajectory. In contrast the linear time-invariant (LTI) cases which have been thoroughly understood in the analysisand synthesis technologies, many features of the LTV systems are still limited and not clear. This paper addressesthe problems of solution and stability of a general unforced LTV differential state space system. Unlike most ofthe work based on the Lyapunov theory, numerical simulations, or specific constraint systems, the paper proposesthe spectral decompositions of the LTV systems by employing extended eigenpairs and with simple mathematicalderivation. The spectral decompositions reveal the mechanisms of inherent characterization in general LTVsystems, rather than a particular class. Moreover, a novel set of auxiliary equations is developed for guiding andobtaining the extended eigenpairs of its system matrix which completely characterize the LTV systems. The solutionsto perform the commutative systems and the second-order systems with companion form are straightforward. The proposed innovative thinking provides a novel guided way to analyze the LTV systems. These findings areeasily extended to LTI cases. Examples from the literature demonstrate the effectiveness and the superiority of theproposed approaches when compared with other methods. The proposed results may be of great interest in both forscientific research and application.

        Naturally Occurring Lactic Acid Bacteria Isolated from Tomato Pomace Silage

        Silage making has become a significant method of forage conservation worldwide. To determine how tomato pomace (TP) may be used effectively as animal feed, it was ensilaged for 90 days and microbiology counts, fermentation characteristics and chemical composition of tomato pomace silage (TPS) were evaluated at the 30th, 60th, and 90th days, respectively. In addition, 103 lactic acid bacteria were isolated from TPS. Based on the phenotypic and chemotaxonomic characteristics, 16S rDNA sequence and carbohydrate fermentation tests, the isolates were identified as 17 species namely: Lactobacillus coryniformis subsp. torquens (0.97%), Lactobacillus pontis (0.97%), Lactobacillus hilgardii (0.97%), Lactobacillus pantheris (0.97%), Lactobacillus amylovorus (1.9%), Lactobacillus panis (1.9%), Lactobacillus vaginalis (1.9%), Lactobacillus rapi (1.9%), Lactobacillus buchneri (2.9%), Lactobacillus parafarraginis (2.9%), Lactobacillus helveticus (3.9%), Lactobacillus camelliae (3.9%), Lactobacillus fermentum (5.8%), Lactobacillus manihotivorans (6.8%), Lactobacillus plantarum (10.7%), Lactobacillus harbinensis (16.5%) and Lactobacillus paracasei subsp. paracasei (35.0%). This study has shown that TP can be well preserved for 90 days by ensilaging and that TPS is not only rich in essential nutrients, but that physiological and biochemical properties of the isolates could provide a platform for future design of lactic acid bacteria (LAB) inoculants aimed at improving the fermentation quality of silage.

        Risk Factors for Anxiety in Major Depressive Disorder Patients

        Objective: To analyze the sociodemographic and clinical factors related to anxiety in patients with major depressive disorder (MDD). Methods: This study involved a secondary analysis of data obtained from the Diagnostic Assessment Service for People with Bipolar Disorders in China (DASP), which was initiated by the Chinese Society of Psychiatry (CSP) and conducted from September 1, 2010 to February 28, 2011. Based on the presence or absence of anxiety-related characteristics, 1,178 MDD patients were classified as suffering from anxious depression (n=915) or non-anxious depression (n=263), respectively. Results: Compared with the non-anxious group, the anxious-depression group had an older age at onset (t=−4.39, p<0.001), were older (t=−4.69, p<0.001), reported more lifetime depressive episodes (z=−3.24, p=0.001), were more likely to experience seasonal depressive episodes (χ2=6.896, p=0.009) and depressive episodes following stressful life events (χ2=59.350, p <0.001), and were more likely to have a family history of psychiatric disorders (χ2=6.091, p=0.014). Their positive and total scores on the Mood Disorder Questionnaire (MDQ) and the 32-item Hypomania Checklist (HCL-32) (p<0.05) were also lower. The logistic regression analysis indicated that age (odds ratio [OR]=1.03, p<0.001), a lower total MDQ score (OR=0.94, p=0.011), depressive episodes following stressful life events (OR=3.04, p<0.001), and seasonal depressive episodes (OR=1.75, p=0.039) were significantly associated with anxious depression. Conclusion: These findings indicate that older age, fewer subclinical bipolar features, an increased number of depressive episodes following stressful life events, and seasonal depressive episodes may be risk factors for anxiety-related characteristics in patients with MDD.

        Effects of repeated tuberculin skin testings on immune responses in experimental monkeys

        Though many alternative methods to tuberculin skin testing (TST) have been established and evaluated in recent years, sensitivities and specificities of most methods could not meet the requirements of golden standards. In this study, we sought to identify whether repeated TSTs could affect the immune responses in experimental monkeys. Nine natural tuberculosis (TB) monkeys receiving repeated TSTs biweekly were used to demonstrate the effect on TST responsiveness. Two healthy monkeys were administrated with repeated TSTs to analyze the immune response profiling. Intrapalpebral reactions in TB infections gradually weakened or presented intermittent positive reactions. The leukocyte counts, cytokine responses, and antibody responses to all antigens except Old tuberculin (OT) and MPT64L showed no specific changes for TB in healthy monkeys. Positive antibody responses to OT and MPT64L emerged during the first half experimental period, which may cause by their cross-reactivity with mycobacterial species. Results showed that repeated TSTs had no significant effects on immune responses in healthy monkeys but a progressive reduction in TST responsiveness in TB infections.

        A Double Flux Magnetic Field Modulation Motor with Ventilation Slot and Uneven Air Gap

        Magnetic fi eld-modulated motors have the strengths of low speed, high torque, and high power density. However, the dual fl ux magnetic fi eld modulation motor (DFMM) structure is complex and not conducive to heat dissipation. To reduce losses and reduce the risk of permanent magnets (PMs) demagnetization, this paper proposes a DFMM with ventilation slot and uneven air gaps. The three symmetry slots of the outer stator are not connected to the winding for ventilation, which can eff ectively dissipate heat during motor operation. The proposed DFMM adopts a single-layer winding, which can reduce the complexity of wiring. The inner rotor permanent magnet adopts Halbach structure and uneven air gap structure to reduce the risk of PMs demagnetization. Finally, the temperature fi eld of the motor was calculated using fi nite element method. It was found that the output torque of the proposed DFMM increased by 16.92N m and the loss decreased by 19.52W. The proposed DFMM can increase the output torque and reduce the risk of PMs demagnetization.

        Correlation Between Bone Marrow Blasts Counts With Flow Cytometry and Morphological Analysis in Myelodysplastic Syndromes

        Dear Editor, Determination of the percentage of blasts in the bone marrow (BM) is one of the critical factors in diagnosing MDS. Flow cytometry (FCM) of BM cells has been introduced as an important co-criterion for diagnosing MDS [1-3]. However, FCM has not been well accepted because of the lack of consensus of the criteria to define a phenotypic myeloblast and the appropriate denominator for calculation. Moreover, BM samples contain variable amounts of peripheral blood mixed with immature cells, thereby complicating interpretation. The present study was designed to choose reagent combinations to identify blasts by FCM in MDS patients, to determine which cell mass as the denominator in the process of counting the percentages of blasts, and to reveal whether the aspirates with high proportions of mature neutrophils should be normalized based on the proportion of dim CD16 maturing myeloid cells.
