In this study, we investigated profiles of the cytokines IFN-${\gamma}$, IL-12, and IL-10 in active pulmonary tuberculosis (EAPTB) patients, HIV-negative patients with multidrug-resistant tuberculosis (MDR-TB) and in healthy tuberculin reactors (HTR). We studied the responses of peripheral blood mononuclear cells (PBMC) from 12 EAPTB patients and 15 MDR-TB patients to stimulation with a purified protein derivatives (PPD) antigen (Ag), and compared them with those from 14 HTR. Using ELISA, IFN-${\gamma}$ production was found to be significantly depressed, while IL-10 was significantly elevated in both MDR-TB and EAPTB after in vitro stimulation with PPD, compared with those in HTR. Although there was no significant difference in IL-12 production among the three groups, mean IL-12 production was highest in patients with MDR-TB. In these patients, IL-12 production was significantly correlated with IL-10 expression, but not IFN-${\gamma}$ production. In addition, neutralization of endogenous IL-10 led to enhanced IFN-${\gamma}$ and IL-$12R{\beta}2$ mRNA expression in TB patients. Our findings suggest that both groups of TB patients may have a similar disregulated pattern of IL-12, IL-10, and IFN-${\gamma}$ production during M. tuberculosis infection. Furthermore, the results suggest a potentially pathogenic role for IL-10 in impaired Th1 immune responses in TB patients.

N/A We extracted beta antigen from M. tuberculosis Aoyama B and purified it from the cell extract of Mycobacterium tuberculosis by affinity chromatography with 5F3F2 monoclonal antibody. We measured the values of IgG, IgM, and IgA antibodies to this beta antigen using ELISA method in patients with tuberculosis and normal healthy controls. The results are summarized as follows: 1) The mean ELISA values of IgG and IgA antibodies to purified beta antigen in the serum of 73 tuberculous patients and 100 normal controls were 0.5560±0.2879 and 0.2177±0.1394, 0.3732±0.2011 and 0.1530±0.1272 (optical density: O.D.), respectively. This difference between the tuberculosis patients and normal controls was statistically significant (p<0,001). 2) On the results of sputum AFB stain, patients with negative AFB, 1+, 2+, and 3+ AFB findings showed significantly elevated levels of IgG antibodies to beta antigen in the value of 0.4930±0.2300, 0.7884±0.3560, 0.5736±0.3820, and 0.5310±0.1680, respectivly, when compared to that of the normal controls. Elevations of IgA antibodies to beta antigen were noted in patients with 1+, 2+, and 3+ AFB on smear staining with the value of 0.2510±0.1550, 0.2540±0.1370, and 0.2440±0.1240 when compared to that of the normal controls. 3) The mean values of IgG and IgA antibodies to purified beta antigen in patients with moderately advanced pulmonary tuberculosis were 0.5425±0.3580, 0.2128±0.1620, and in patients with far advanced pulmonary tuberculosis, 0.5336±0.2793 and 0.2721±0.2210, which were more significantly elevated than in the normal controls of 0.3732±0.2911 and 0.1530±0.1272, respectively(p<0.05). 4) In patients with superficial cervical tuberculous lymphadenitis, the mean values of antibody to purified beta antigen were not significantly different from that of the normal controls. 5) In patients with tuberculous pleurisy, the mean value of IgG antibody to purified beta antigen was elevated to 0.5520±0.4090 compared to 0.3732±0.2011 of the normal controls (p<0.005). 6) The mean values of antibodies to purified beta antigen follwing treatment showed a desreased tendency, but there was no statistical significance. In conclusion, patients with tuberculosis showed statistically significant elevations of IgG and IgA antibodies to purified beta antigen, and the measurement of these values may be useful for the diagnosis of tuberculosis-related diseases.

As a new class of thermally activated actuators based on polymeric fibers, we investigated polyethylene terephthalate (PET) yarns for the development of a twisted-coiled polymer fiber actuator (TCA). The PET yarn TCA exhibited the maximum linear actuation up to 8.9% by external heating at above the glass transition temperature, 160 °C–180 °C. The payload of the actuator was successfully correlated with the preload and training-load conditions by an empirical equation. Furthermore, the PET-based TCA was electrically driven by Joule heating after the PET surface was metallization with silver. For the fast and precise control of PET yarn TCA, electroless silver plating was conducted to form electrical conductive layers on the PET fiber surface. The silver plated PET-based TCA was tested by Joule heating and the tensile actuation was increased up to 12.1% (6 V) due to the enhanced surface hardness and slippage of PET fibers. Overall, silver plating of the polymeric yarn provided a fast actuation speed and enhanced actuation performance of the TCA actuator by Joule heating, providing a great potential for being used in artificial muscle for biomimetic machines including robots, industrial actuators and powered exoskeletons.

Recent research has focused heavily on semantic segmentation using only RGB images. However, in adverse weather conditions, such as nights with low light or heavy rain obscuring most objects, the performance of artificial intelligence models can significantly deteriorate. For example, when a model trained solely on RGB images attempts to detect a river under circumstances involving nighttime and rain, it is highly likely to yield poor results. Therefore, this paper proposes a solution by training models robust to such situations through sensor fusion of long-wave infrared (LWIR) images and RGB images.
