        Silicon-to-silicon anodic bonding was accomplished using lithium doped interlayer. Lithium doped interlayers were deposited on silicon substrates by electron beam evaporation, which has a fast deposition rate compared with sputtering method. Silicon-to-silicon bonding occurs in the range of temperatures from 250℃ to 300℃ with the applied voltages from 70V_(DC) to 100V_(DC). The bonding strength obtained from tensile test was about 5MPa under the condition of 80V_(DC) in 300℃. The surface morphology of lithium doped interlayer was studied with the atomic force microscopy (AFM). Finally, secondary ion mass spectroscopy (SIMS) analysis was carried out in order to investigate the role of the lithium ions in bonding mechanism.

        Multimodal evaluation of an interphotoreceptor retinoid-binding protein-induced mouse model of experimental autoimmune uveitis

        We aimed to characterize the vascular phenotypes of an experimental autoimmune retinal uveitis (EAU) model induced by interphotoreceptor retinoid-binding protein (IRBP) using multimodal imaging techniques. We systemically administered IRBP or vehicle to adult C57BL/6 mice. Fundus photography, optical coherence tomography (OCT), in vivo live confocal imaging using different tracers, OCT angiography (OCTA), and electroretinography (ERG) were performed after IRBP immunization. Hematoxylin and eosin and immunofluorescence staining were performed to characterize the immune response and vascular permeability. Mice with EAU exhibited perivascular inflammation, vitritis, and superficial retinal inflammation on fundus photography and OCT. H&E revealed immune cell infiltration in the perivascular area of the retina and choroid accompanied by a significant degree of perivasculitis that subsequently damaged photoreceptors 3 weeks postimmunization. Immunofluorescence staining showed subsequent transcytosis induction after local microglial activation followed by neutrophil recruitment in the perivascular area. Transcytosis in the superficial and deep vascular areas was improved by immune cell suppression. Intravital in vivo confocal imaging showed signs of neutrophil infiltration and obstructive vasculitis with perivascular leakage 3 weeks postimmunization. OCTA revealed a significant decrease in vascular flow in the deep capillary layer of the retina. Functional analysis showed that scotopic responses were intact at 2 weeks; however, normal photopic and scotopic responses were hardly detected in mice with EAU mice at 3 weeks postimmunization. Our data suggest that inflammatory cell activation and subsequent transcytosis induction in endothelial cells might be a major pathogenic factor for vascular leakage in uveitis, providing new insights into the pathophysiology of retinal vasculitis in noninfectious uveitis.

        Long-term effects of human induced pluripotent stem cell-derived retinal cell transplantation in Pde6b knockout rats

        Retinal degenerative disorders, including age-related macular degeneration and retinitis pigmentosa (RP), are characterized by the irreversible loss of photoreceptor cells and retinal pigment epithelial (RPE) cells; however, the long-term effect of implanting both human induced pluripotent stem cell (hiPSC)-derived RPE and photoreceptor for retinal regeneration has not yet been investigated. In this study, we evaluated the long-term effects of hiPSC-derived RPE and photoreceptor cell transplantation in Pde6b knockout rats to study RP; cells were injected into the subretinal space of the right eyes of rats before the appearance of signs of retinal degeneration at 2–3 weeks of age. Ten months after transplantation, we evaluated the cells using fundus photography, optical coherence tomography, and histological evaluation, and no abnormal cell proliferation was observed. A relatively large number of transplanted cells persisted during the first 4 months; subsequently, the number of these cells decreased gradually. Notably, immunohistochemical analysis revealed that the hiPSC-derived retinal cells showed characteristics of both RPE cells and photoreceptors of human origin after transplantation. Functional analysis of vision by scotopic electroretinogram revealed significant preservation of vision after transplantation. Our study suggests that the transplantation of hiPSC-derived retinal cells, including RPE cells and photoreceptors, has a potential therapeutic effect against irreversible retinal degenerative diseases.         Classical swine fever disease in Cheolwon

        Two cases of classical swine fever(CSF) disease have broken out in Cheolwon (7 April, 2002). The suspected pig herds were huddled together because of high fever (over 40℃) and showed remarkable decrease of the leukocytes. The staggering gait related to posterior weakness, constipation and lethargy, hyperemia, hemorrhagic lesions(on the skin, muzzle, ears, limbs, tail and inner part of legs) and conjunctivitis with dirty streaks below the eyes were observed. The inflammation in the lung, infarction in the spleen, swelling and hemorrhage in lymph nodes, kidney, intestine, heart and cheese like purulent inflammation of the tonsil were observed. The ulcers of the colon were also detected. Several clinical and laboratory techniques including blood test, histo-pathological examinations, indirect fluorescent antibody(IFA) test and RT-PCR test were applied to diagnose the disease. Inoculation test on PK-15 cell was also performed. The necrosis of the lymphatic cells and infiltration of the vessel circumferential cells in the brain and lymph organs were commonly viewed. The proliferation of the glia cell(gliosis) in the lymph was particular. Cytopathogenic effect(CPE) and specific fluorescent-bright-green areas(with IFA) appeared in PK-15 cells inoculated with suspected blood plasma. The IFA test on the epithelial and mucous membrane cells of tonsil was positive. RT-PCR technique required more working hours and labor than other techniques in this examination but it was useful because of the sensitivity to the CSF viral gene.

      • The Korea <i>Brassica</i> Genome Project: a Glimpse of the <i> Brassica </i> Genome Based on Comparative Genome Analysis With <i>Arabidopsis</i>

        <P>A complete genome sequence provides unlimited information in the sequenced organism as well as in related taxa. According to the guidance of the Multinational Brassica Genome Project (MBGP), the Korea Brassica Genome Project (KBGP) is sequencing chromosome 1 (cytogenetically oriented chromosome #1) of <I>Brassica rapa</I>. We have selected 48 seed BACs on chromosome 1 using EST genetic markers and FISH analyses. Among them, 30 BAC clones have been sequenced and 18 are on the way. Comparative genome analyses of the EST sequences and sequenced BAC clones from <I>Brassica</I> chromosome 1 revealed their homeologous partner regions on the <I>Arabidopsis</I> genome and a syntenic comparative map between <I>Brassica</I> chromosome 1 and <I>Arabidopsis</I> chromosomes. <I>In silico</I> chromosome walking and clone validation have been successfully applied to extending sequence contigs based on the comparative map and BAC end sequences. In addition, we have defined the (peri)centromeric heterochromatin blocks with centromeric tandem repeats, rDNA and centromeric retrotransposons. In-depth sequence analyses of five homeologous BAC clones and an <I>Arabidopsis</I> chromosomal region reveal overall co-linearity, with 82% sequence similarity. The data indicate that the <I>Brassica</I> genome has undergone triplication and subsequent gene losses after the divergence of <I>Arabidopsis</I> and <I>Brassica</I>. Based on in-depth comparative genome analyses, we propose a comparative genomics approach for conquering the <I>Brassica</I> genome. In 2005 we intend to construct an integrated physical map, including sequence information from 500 BAC clones and integration of fingerprinting data and end sequence data of more than 100 000 BAC clones.         Zinc Sulfadiazine 연고제의 제제학적 연구

        Silver sulfadiazine has been introduced to replace silver nitrate in the topical treatment of extensive burns and this drug exerts a prominent antibacterial action against Pseudomonas aeruginosa. The compound is painless upon application and insufficient sulfadiazine is absorbed to cause crystalluria. The primary purpose of the study was to clarify the antimicrobial action of zinc sulfadiazine ointment in comparison with silver sulfadiazine ointment as well as the pharmaceutical properties of the zinc sulfadiazine preparations. The results are summerized as followings: 1) The optimum ratio of two substrate compounds for the synthesis of zinc sulfadiazine are 2 moles of sulfadiazine and 1 mole of zinc sulfate at pH 6.0. 2) The stability of zinc sulfadiazine ointment preparation by using polyethylene glycol base, Beeler's base or polyoxyl 40 stearate base was more stable than that of silver sulfadiazine preparations. 3) The antimicrobial action of zinc sulfadiazine exhibits a stronger antimicrobial activity than that of sulfadiazine against Staphylococcus aureus but the opposite is true against Pseudomonas aeruginosa.

        초음파로 페놀 분해 시 염소계화합물의 첨가와 음향 강도의 영향

        Degradations of phenol and chlorinated compounds mixtures were studied with ultrasound of 20 kHz and 0.57, 1.14 W/mL. In presence of carbon tetrachloride (CT), degradation rate of phenol is faster than chloroform (CF), dichloromethane (DCM) and phenol solution. It is due to that CT generates of free chlorine (HOCl and OCl-) from the sonochemical degradation and plays a role of hydrogen atom scavenger. CF and DCM are react with free chlorine, so amount of free chlorine is smaller than CT solution. The degradation rates of chlorinated compounds decreased with co-presence of phenol in the solution due to the distribution ultrasonic energy to both compounds. The measured chloride ion was lower than the theoretical concentration assuming complete degradation. This means not all the contaminants destructed went through complete degradation.

        전립선 암에서 Bicalutamide 사용으로 생긴 간질성 폐질환

        Androgen deprivation therapy, which is the standard treatment for metastatic prostate cancer, includes nonsteroidal antiandrogenic drugs, such as flutamide, nilutamide and bicalutamide. Of them, bicalutamide rarely induces interstitial pneumonia. We report a case of bicalutamide-induced interstitial pneumonia. A 68-year old male diagnosed with prostate cancer and multiple bone metastases presented with dry cough and low grade fever for 3 days. He had taken bicalutamide (50 mg/day) for 13 months. High resolution computed tomography revealed ground glass opacity in his right upper lung. The laboratory studies showed no eosinophilia in the serum and bronchoalveolar lavage fluid. Despite the use of antimicrobial agents for 2 weeks, the extent of the lung lesions increased to the left upper and right lower lung. He had no environmental exposure, collagen vascular disease and microbiological causes. Under the suspicion of bicalutamide-induced interstitial pneumonia, bicalutamide was stopped and prednisolone (1 mg/kg/ day) was initiated. The symptoms and radiologic abnormalities were resolved with residual minimal fibrosis.
