Transcriptional silencing of the inhibin-α gene in human gastric carcinoma cells.

<P>Although inhibin was first identified as a hormone regulating pituitary FSH secretion, it was later recognized to act as a tumor suppressor in the gonad and adrenal glands. Recently, the alpha subunit of this dimeric hormone (inhibin?α) was reported to be involved in prostate tumorigenesis. To identify additional roles outside the reproductive axis, we investigated inhibin?α gene activity and subsequent cell fate in human gastric cancer cells. The results were as follows: all the gastric cancer cells had at least one of a set of abnormalities including hypermethylation of the promoter, mutation of the 5'?UTR or allelic imbalance including LOH in the inhibin?α gene. Hypermethylation of the promoter and mutation of the 5'?UTR in inhibin?α were observed in SNU?1, SNU?5 and SNU?484 cells. LOH was observed in AGS, KATO III, SNU?5, SNU?484 and SNU?668 cells. Treatment with 5?AzaC, a demethylating agent, induced demethylation of the inhibin?α promoter in the SNU?1, SNU?5 and SNU?484 cell lines, with the CpG5 site being strongly influenced by 5?AzaC. In addition, inhibin?α mRNA and protein were maintained at low levels in most of the gastric cancer cell lines. These low levels of mRNA and protein expression could be increased in most lines by treatment with 5?AzaC. These increased inhibin?α expression levels seemed to be closely associated with apoptosis and suppression of cell growth. Taken together, our results reveal that the inhibin?α gene is transcriptionally silenced in human gastric cancer cells, and that reactivation of the gene suppresses their growth characteristics. This suggests that inhibin?α may have a more general tumor suppressor activity outside the reproductive axis.</P>
워드 임베딩을 활용한 한국어 가짜뉴스 탐지 모델에 관한 연구

본 논문에서는 가짜뉴스 탐지 모델에 워드 임베딩 기법을 접목하여 성능을 향상시키는 방법을 제안한다. 기존의 한국어 가짜뉴스 탐지 연구는 희소 표현인 빈도-역문서 빈도(TF-IDF)를 활용한 탐지 모델들이 주를 이루었다. 하지만 이는 가짜뉴스 탐지의 관점에서 뉴스의 언어적 특성을 파악하는 데 한계가 존재하는데, 특히 문맥에서 드러나는 언어적 특성을 구조적으로 반영하지 못한다. 이에 밀집 표현 기반의 워드 임베딩 기법인 Word2vec을 활용한 텍스트 전처리를 통해 문맥 정보까지 반영한 가짜뉴스 탐지 모델을 본 연구의 제안모델로 생성한 후 TF-IDF 기반의 가짜뉴스 탐지 모델을 비교 모델로 생성하여 두 모델 간의 비교를 통한 성능 검증을 수행하였다. 그 결과 Word2vec 기반의 제안모형이 더욱 우수하였음을 확인하였다.

저자들은 특발성 혈소판감소성 자반병을 가진 환자에서 전형적인 일차성 원발성 담즙성 간경변증의 양상이 나타나 혈청검사, 자가면역항체, 간조직 검사 등을 통해 특발성 혈소판감소성 자반병과 쇼그렌 증후군이 동반된 원발성 담즙성 간경변증 1예를 경험하였기에 문헌고찰과 함께 보고하는 바이다. Primary biliary cirrhosis (PBC) is rare in Korea. Although its pathogenesis is not fully understood, immunologic dysregulation is suspected because it is frequently associated with other immunologic disorders. A few cases of primary biliary cirrhosis associated with idiopathic thrombocytopenic purpura (ITP) has previously been reported. However, PBC associated with ITP and Sjo¨gren's syndrome simultaenously has not been reported yet. We experienced a 72-year-old woman had been finally diagnosed as PBC, ITP and Sjo¨gren's syndrome. To the best of our knowledge, this is the first case in the world. We report this case with review of related literature.

Background/Aims: The aim of this study was to evaluate the change in circulating levels of adipokines, including leptin, adiponectin, resistin, and visfatin, after induction therapy of patients with active inflammatory bowel disease (IBD). Methods: We prospectively evaluated 20 patients with Crohn’s disease (CD) and 14 patients with ulcerative colitis (UC) who received induction therapy. The disease activity, body mass index (BMI), and C-reactive protein (CRP) and serum adipolines levels were checked before treatment and 10 weeks after treatment. Results: After induction therapy, significant reduction of disease activity was noted in patients with CD (CD activity index, 267.9±73.7 vs. 50.8±36.5, P<0.001) and patients with UC (Mayo score, 9.8±2.2 vs. 1.4±1.6, P<0.001). The mean serum leptin, adiponectin, resistin, and visfatin levels were 4.0±2.7 ng/mL and 4.4±2.3 ng/mL (P=0.323), 27.0±20.5 μg/mL and 33.8±26.5μg/mL (P=0.084), 9.0±4.0 ng/mL and 10.2±3.2 ng/mL (P=0.108), and 4.6±3.7 ng/mL and 2.5±4.1 ng/mL (P=0.046) before and after infliximab treatment, respectively. No significant correlation between the changes in BMI, CRP level, or the clinical indices of activity and alterations of the measured adipokines was demonstrated. Conclusions: The serum levels of leptin, adiponectin, and resistin showed no significant alterations, whereas the serum visfatin level decreased significantly after induction therapy, suggesting a possible pro-inflammatory property of visfatin and a role as a marker of successful therapy of IBD. Drug rash with eosinophilia and systemic symptoms (DRESS) is a life-threatening systemic drug reaction characterized by fever, rash, hematological abnormalities, lymphadenopathy, and multiple internal organ involvement. Unfortunately, a long latency period as well as clinicians` unawareness of the disease entity often results in a delay of prompt diagnosis and treatment in clinical practice. A search of the literature revealed only few reports on DRESS in patients with inflammatory bowel diseases. The pathogenesis of the disease is not clearly understood, although several possible mechanisms, such as drug detoxification, slow acetylation, and reactivation of human herpes viruses, have been proposed in its development. Here, we present a rare case of DRESS associated with viral reactivation and defects in drug metabolism in a 22-year-old man who had been on sulfasalazine for 6 weeks to treat ulcerative colitis. The etiology of Crohn`s disease, which is a chronic inflammatory condition that potentially involves any location of the alimentary tract from the mouth to the anus, is unknown. However, there is strong evidence that vascular damage could play a role in the pathogenesis of Crohn`s disease. Crohn`s disease is mediated by multifocal gastrointestinal infarctions, which occur at an early stage in the disease process. Persistent activation of coagulation in patients with Crohn`s disease has been shown. In contrast, hemophilia is an inherited disorder of coagulation. The deficiencies of clotting factors usually involve occult or overt bleeding. The pathogenic mechanisms of Crohn`s disease and hemophilia are incompatible. An association between Crohn`s disease and hemophilia has not been reported in Korea. We managed 21- and 33-year-old men with Crohn`s disease associated with hemophilia, who presented with hematochezia. (Intest Res 2011;9:9-39)
