        Nitric Oxide (NO), derived from L-arginine, is produced by two types (constitutive and inducible) of nitric oxide synthase (NOS: cNOS and iNOS). The NO produced in large amounts by the iNOS is known to be responsible for the vasodilation and hypotension observed in septic shock and inflammation. The inhibitors of iNOS, thus, may be useful candidate for the treatment of inflammatory diseases accompanied by the overproduction of NO. We prepared alcoholic extracts of herbal drugs which have been used for the treatment of inflammation in oriental medicine. We have screened the inhibitory activity of NO production in lipopolysaccharide (LPS)-activated macrophages after the treatment of these extracts. Among the 81 kinds of extracts of herbal drugs, 34 extracts showed potent inhibitory activity of NO production above 50% at the concentration of 50 (μg/ml. The inhibitory activities of NO production were also evaluated for several solvent fractions at three different concentrations. Especially, hexane soluble fractions of Agrimonia pilosa, Hydrangea serrata, Machilus thunbergii, Prunella vulgaris, Saussurea lappa, Tussilago farfara, and ethyl acetate soluble fractions of Angelica gigas, Ostericum koreanum, Torilis japonica, Perilla frutescence showed moderate activity at 10 and/ or 25 (μg/ml. These fractions are promising candidates for the study of the activity-guided chromatographic purification of active compounds.

        The study of p53 tumor suppressor protein is one of most important subjects to an environmental toxicology as well as in cancer biology Generally, p53 has been known to Involve the cell cycle regulation and apoptosis by the activation of its target genes such as p2l and bax in a number of cellular stress responses In addition, associations of p53 with cellular proteins presumably reflect the involvement of p53 in critical cellular processes such as DNA repair The complex formation of p53 and exogenous proteins such as viral or cellular proteins has been shown in many cases to play important roles in carcinogenic processes against environmental mutagen Recently the disruption of p53 protein by oxidative stress has been also reported to have relevance to carcinogenesis These findings suggested that the maintaining of stability and functional activity of p53 protein was also important aspect to play as a tumor suppressor protein Therefore, the detection of functional status of p53 proteins might be an effective biomarker for the cancer and human diseases under the environmental toxicologic carcinogen.

      • 양강의 nitric oxide synthase 저해활성

The activity guided column chromatography of the chloroform soluble fraction obtained from the methanol extract of Alpinia officinarum gave an inhibitory active compound of NO synthesis in LPS-activated macrophages (IC_(50) value; 0.6 μM). The structure of active compound was elucidated as 7-(4-hydoxy-3-methoxyphenyl)-1-phenylhept-4-en-3-one by spectral analysis. This compound can be an useful candidate for the development of new drug to treat endotoxemia and inflammation accompanied by the overproduction of NO.

      • 봉출 추출물의 nitric oxide synthase 저해활성

        Nicotinamide Adenine Dinucleotide Phosphate (NADPH) has antioxidant effect in the body and thus decreases the symptoms of senility. The development of a transdermal delivery formulation for NADPH is a very difficult task, due to its rapid decomposition by oxidation, low partition coefficient into stratum corneum, high molecular weight and electrical charge at neutral pH in aqueous solution. The purpose of this work is to test several anti-oxidants as the stabilizing agent for NADPH in the aqueous formulation, and to study the effect of these agents on the in-vitro iontophoretic transport through hairless mouse skin. The stability of NADPH in aqueous solution with/without various antioxidants such as butylated hydroxy toluene (BHT), resveratrol, a-tocopherol and Vitamine C was studied. BHT an a-tocopherol (0.01 % w/w) exhibited minimal stabilizing effect, however resveratrol and vitamine C showed significant stabilizing effect. Stabilizing effect of vitamine C was proportional to the concentration in aqueous solution. Although vitamine C increased the stability ofNADPH significantly, it decreased flux of NADPH in a concentration dependent fashion, therapeutic amount ofNADPH can be delivered through skin using iontophoresis and These results suggest that, in order to develope a efficient iontophoresis formulation, it is important to choose a stabilizing agents which are not only potent in stabilizing effect but also not competitive for the delivery of NADPH.

      • 수종 한약제의 위암세포에 대한 항암작용 효능 검색 및 약리작용에 관한 분자생물학적 연구

        Traditional oriental medicines have ken used for tmament of various kinds of human cancers for long times and some of mem proven to k effective clinical-. However, the pItarmacologica1 actions ad mechanisms related to cancer treatment are generally unknown. In an effort to clarify the action mechanisms of several oriental medicines used for cancer treatments. we planned this experimental praedura. We selected Cadyceps sinensis (동충하초), Punellae Herba (夏柱草), Rehmanniae Radix (熟地黃), Paeoniae Radix (白均案), Oldedamar Herba (白花批告草), Partulaceae Herba (馬首算), Scdopendrasubspinipes mudans (蜈松), Mylabris Phalerara (班藝), Phellinus igniarius(蔡黃), Ganodermae Lignum(靈芝) for evaluation, which have ken used for patients of gastric cancers. The twenty grams of medicines were boiled in 100㎖ of water for 1 hour and filtered with O.2㎛ pore-sized filter unit to remove insoluble particles. Initially we evaluated the effects oriental medicines on growth inhibition in stomach cancer cells. The gastric cancer cell line, AGS, was cultured in RPMI l640 supplemented with lO% heat-inactivated fetal bovine serum and Heated with lO㎕ oriental medicines per l㎖ of medium up to 48 hours. The specimens were subjected to MTT assay for evaluation of growth inhibition. We found mat Mylabris piulema (班 察) markedly supmessed the growth of cancer cells as shown in results. Next, we checked the effects of orielntl medicines on cancer on cell cycles and apMonsis. Mylabrls phalerata (반모) inhibited cell cycle progression of Canea cells a compared with connol cells and cells treated with other medicines. In addition, Mylabri phalerata (반모) induced apptosis in 30-40% of stomch cancer cells within 24 hours. Other oriental medicines used for this experiments did not show apoptosis-inducing effects on cancer cells. Finally, we determined the expmSSion levels of genes associated with cell cycle Md apoptosis. The expressions of bcl-2 and bcl-XL were downregulated by the treamerm of Mylabris phalerata (반모). However, the expression levels of genes related to cell cycles were not altered significantly. In conclusion, we found that Mylams phalerata (반모) has in vivo gowth-inhibiting ad apptosis- inducing effects on stomach cancer cells. However, we think that at least animal experiments are necessay for evduations.

        Yomogin, an Inhibitor of Nitric Oxide Production in LPS-Activated Macrophages

        In activated macrophages the inducible form of nitric oxide synthase (i-NOS) generates high amounts of toxic mediator, nitric oxide (NO) which contributes to the circulatory failure associated with septic shock. A sesquiterpene lactone compound (yomogin) isolated from medicinal plant Artemisia princeps Pampan inhibited the production of NO in LPS-activated RAW 264.7 cells by suppressing i-NOS enzyme expression. Thus, yomogin may be a useful candidate for the development of new drugs to treat endotoxemia and inflammation accompanied by the overproduction of NO.

      • Experimental observation and modified driving method to improve the high-temperature misfiring in AC PDP

        Misfiring is often observed during the high-temperature quality assurance test of plasma display panels. This limits the productivity of the plasma display panel industry. In this paper, experimental observations on the misfiring at high-panel temperatures have been performed through time-dependent discharge light output and static margin measurement. For the high-temperature condition, the firing voltage increment is found in both surface and facing discharges. This in turn increases time lag in address discharge, and results in increment of misfiring probability. In order to reduce this kind of misfiring, a modified method that applies automatically to a different slope of ramp erase pulse on the sustain electrode according to temperature variation is proposed. The experimental results show that controlling the slope of the ramp erase pulse is quite effective for compensating the temperature-dependent variation of reset and address discharge.

        Effect of Working Gas Pressure on Misfirng of ac PDP at High Ambient Temperature

        One of the important problems in ac PDP in recent years is the misfiring of ac PDP at high ambient temperatures which consequently degrades the image quality of the ac PDP. This may be due to the change of working gas pressure and/or MgO surface characteristics at high ambient temperatures. This paper deals with the effect of working gas pressure on the misfiring of ac PDP at high ambient temperature. From this study, we found that the main cause of the misfiring at high ambient temperature is the increase in discharge firing voltage induced by increased working gas pressure
