Potential Use of Mycobacterium paragordonae for Antimycobacterial Drug Screening Systems

Our recent genome-based study indicated that Mycobacterium paragordonae (Mpg) has evolved to become more adapted to an intracellular lifestyle within free-living environmental amoeba and its enhanced intracellular survival within Acanthamoeba castellanii was also proved. Here, we sought to investigate potential use of Mpg for antimycobacterial drug screening systems. Our data showed that Mpg is more susceptible to various antibiotics compared to the close species M. marinum (Mmar) and M. gordonae, further supporting its intracellular lifestyle in environments, which would explain its protection from environmental insults. In addition, we developed two bacterial whole-cell-based drug screening systems using a recombinant Mpg stain harboring a luciferase reporter vector (rMpg-LuxG13): one for direct application to rMpg-LuxG13 and the other for drug screening via the interaction of rMpg-LuxG13 with A. castellanii. Direct application to rMpg-LuxG13 showed lower inhibitory concentration 50 ( IC50) values of rifampin, isoniazid, clarithromycin, and ciprofloxacin against Mpg compared to Mmar. Application of drug screening system via the interaction of rMpg-LuxG13 with A. castellanii also exhibited lower IC50 values for rifampin against Mpg compared to Mmar. In conclusion, our data indicate that Mpg is more susceptible to various antibiotics than other strains. In addition, our data also demonstrate the feasibility of two whole cellbased drug screening systems using rMpg-LuxG13 strain for the discovery of novel anti-mycobacterial drugs.

As the AI industry evolves, neural network processing units (NPUs) are being developed to deliver AI services faster and more efficiently. One of the most important challenges for these NPUs is task scheduling to minimize off-chip memory accesses, which incur significant performance overhead. In particular, convolutional layers can be fused with multiple layers to reduce the memory accesses, but it is difficult to find the optimal schedule due to the too large exploration space. In this paper, we propose an efficient schedule exploration algorithm to optimize the fusion of multiple convolutional layers in NPUs. The proposed algorithm organizes the fusion group exploration space in the form of a grid to explore the optimal schedule. Experimental results show that the fusion schedule explored by the proposed method reduces the latency by 7.7% and reduces the off-chip memory access by 15% compared to the baseline algorithm.

이 연구의 목적은 소아 환자의 유구치 인접면 우식증 치료에 유동성 복합레진을 이용한 II급 와동 수복물의 생존율을 기성금속관과 비교하여 후향적으로 평가하는 것이다. 2015년 6월부터 2019년 8월까지 인접면 우식증으로 진단되어 유동성 복합레진과 기성금속관 수복 치료를 시행한 590명의 1,504개의 유구치에 대한 전자의무기록과 방사선사진을 조사하였다. 수집된 자료는 Kaplan-Meier method를 이용해 생존 분석을 하였다. 유동성 복합레진 수복된 치아의 1년 생존율은 98.5%, 3년 생존율은 87.7%, 평균 생존기간은 39개월이었으며, 기성금속관과 비교할 때 통계적으로 유의한 차이는 없었다(p = 0.896). 현 연구의 한계 내에서 유동성 복합레진을 이용한 II급 수복은 유구치 인접면 우식증 치료의 예지력 있는 선택지가 될 수 있다고 사료된다. The purpose of this retrospective study was to evaluate the survival rate by comparing Class II restoration using flowable resin composite with stainless steel crown in primary molars. Electronic medical records and radiographs of 1,504 primary molars with proximal caries of 590 patients from June 2015 to August 2019 were analyzed. With the collected data, survival analysis was performed using the Kaplan-Meier method. The 1-year survival rate of flowable resin composite in the primary molar was 98.5%, 3-year survival rate was 87.7%, and mean survival time was 39 months. There was no statistically significant difference between flowable resin composite and stainless steel crown (p = 0.896). Within the limits of this study, Class II restoration using flowable resin composite can be considered a promising option for the treatment of proximal caries in primary molars.
골 성숙도 단계의 골령 평가를 위한 Greulich-Pyle 방법을 이용한 인공지능 프로그램의 활용

The purpose of this study was to measure bone age using an artificial intelligence program based on the Greulich-Pyle (GP) method to find out the bone age corresponding to each stage of cervical vertebral maturation (CVM) and the middle phalanx of the third finger (MP3). This study was conducted on 3,118 patients who visited pediatric dentistry at Kyung Hee University Dental Hospital and Pusan National University Dental Hospital from 2013 to 2021. The CVM stage was divided into 5 stages according to the classification by Baccetti, and the MP3 stage was divided into 5 stages according to the methods of Hägg and Taranger. Based on the GP method, bone age was evaluated using an artificial intelligence program. The pubertal growth spurt in the CVM stage was CVM II and III. The mean bone age in CVM II was 11.00 ± 1.81 years for males and 10.00 ± 1.49 years for females, and in CVM III, 13.00 ± 1.46 years for males and 12.00 ± 1.44 years for females (p < 0.0001). The pubertal growth spurt in the MP3 stage was MP3 - G stage. The bone age at the MP3 - G stage was 13.14 ± 1.07 years for males and 11.40 ± 1.09 years for females (p < 0.0001). Bone age evaluation using artificial intelligence is worth using in clinical practice, and it is expected that a faster and more accurate diagnosis will be possible.
