      • 한강 하류 김포평야의 최종빙기 이래 퇴적환경변화

        한강하류부에 위치한 김포시 고촌면 김포평야의 충적충과 사면퇴적충에서 퇴적상 분석 및 화분 분석을 실시하여 식생환경, 해면변동, 기후변화와 같은 자연환경인화를 검토하고 이것을 선사시대 인간활동과의 관계에서 살펴보았다. 최종빙기 이래 형성된 연구지역의 퇴적상은 빙기의 선상지성사면퇴적층과 흘로세 충적충으로 구분되며, 화분분석은 흘로세 충적충의 유기질이 다소 포함된 회색실트충, 분해되지 않은 토탄충, 분해된 토탄충에서 이루어졌다. 화분조성은 흘로세 중기 이후 해면미변동을 반영하며, 화분대GP-Ⅰ은 흘로세 해진의 영향으로 수심이 있는 저습지 환경에서 Alnus를 중심으로 한 목본 우점기에 퇴적되었고, 화분대GP-ll시기는 해면의 지속적인 정체와 하강에 따른 지하수위 저하 그리고 활발해진 인간활동의 영향으로 목본이 크게 줄고, 포자와 초본류가 우점하게 되었다. The environmental change such as vegetation environment, sea-level change, climate change and human impact was restored at Gimpo alluvial plain on the upper Han river according to the results of sedimentary facies and pollen analysis. The sedimentary facies which has been constructed since the last glacial stage is composed of the hillslope deposits as the alluvial fan during the glacial stage and Holocene alluvium. Materials for pollen analyses were sampled from gray organic silt layer, peat I layer(abundant fibroid material included the branches of trees and roots of grasses) and peat Ⅱ layer(decomposed peat including organic clay) in the alluvium of the Holocene. The change of the pollen composition reflected sea-level change and vegetation environment during the Holocene. Pollen zone GP-Ⅰ was the period of AP-dominance, including mostly Alnus. Study area was under the lagoon or swamp environment influenced by Flandrian transgression. Pollenzone Ⅱ was the period of Spore & NAP-dominance. It represents that vegetation environment was changed to drier condition by falling the surface of ground-water caused by sea-level regression after long-term stability, and influenced of human activity.

        ORiginal Article : Gastroprotective Effects of PMK-S005 against Ethanol-Induced Acute Gastric Damage in Rats

        Background/Aims: This study aimed to examine the gastroprotective effects of PMK-S005, which is a synthetic S-allyl-Lcysteine (SAC; a sulfur-containing amino acid), against acute ethanol-induced gastric damage in rats. Methods: Sprague- Dawley rats were divided into six groups, including a nonethanol group, groups treated with absolute ethanol 1 hour after pretreatment with various doses of PMK-S005 (1, 5, and 10 mg/kg) or rebamipide (50 mg/kg), and an absolute ethanolonly group. Ethanol-induced gross ulcer and mucus levels were measured. Myeloperoxidase, tumor necrosis factor a, interleukin 1β, PGE2, LTB4, cPLA2, COX-1, and COX-2 levels were estimated by enzyme-linked immunosorbent assay or Western blot analysis. Furthermore, the protein expression levels of antioxidant enzymes, including heme oxygenase-1 (HO-1), NAD(P)H:quinine oxidoreductase 1 (NQO-1), GCLC, and GCLM, were assessed. Results: PMK-S005 significantly attenuated the ethanol-induced gastric damage; it reduced mucosal inflammatory cytokine production and increased mucus levels. The expression levels of cPLA2, COX-1, and COX-2 were decreased by PMK-S005. PMK-S005 did not affect PGE2 synthesis, but LTB4 production was significantly suppressed. In addition, long-term administration of PMKS005 significantly increased the expression of HO-1, NQO-1, GCLC, and GCLM. Conclusions: These results strongly suggest that PMK-S005 prevents gastric mucosal damage and that these gastroprotective activities are due to anti-inflammatory effects and enhancement of the gastric defense system, including antioxidant enzymes.       • Preparation of resistant starch/poly(vinyl alcohol) blend films with added plasticizer and crosslinking agents

        <P>We report on the physical properties of films synthesized with native corn starch (NCS) and resistant starch (RS4) prepared with NCS. NCS and RS4/poly(vinyl alcohol) (PVA) blend films were synthesized with a mixing process and casting method. Glycerol (GL) and citric acid (CA) were used as additives. Glutaraldehyde (GLU) was used as a crosslinking agent of the films. RS4 was synthesized with sodium trimetaphosphate and sodium tripolyphosphate as a crosslinker. Then, the RS4 thus synthesized was confirmed by the pancreatin–gravimetry method, swelling power, differential scanning calorimetry, and X-ray diffraction. The tensile strength, elongation, swelling behavior, and solubility of the films were measured. The results of the measurements indicated that the RS4-added film was better than the NCS-added film. In particular, the RS4/PVA blend film with CA as an additive showed physical properties superior to those of the other films. Also, the physical properties with GLU added as a crosslinking agent to the films were investigated. With increasing GLU contents, the tensile strength increased but the elongation, swelling behavior, and solubility values of the GL-added and CA-added films decreased. © 2007 Wiley Periodicals, Inc.         Acai Berries Inhibit Colon Tumorigenesis in Azoxymethane/Dextran Sulfate Sodium-Treated Mice

        Background/Aims: The aim of this study was to investigate the protective effect of acai against azoxymethane (AOM)/dextran sulfate sodium (DSS)-induced colorectal cancer development. Methods: The effect of acai on tumorigenesis was assessed by evaluating tumor incidence, multiplicity and invasiveness in the mouse colon. The levels of myeloperoxidase (MPO) and proinflammatory cytokines (tumor necrosis factor α [TNF-α], interleukin [IL]-1β, and IL-6) were measured via enzyme-linked immunosorbent assay. Protein levels of cyclooxygenase 2 (COX-2), proliferating cell nuclear antigen (PCNA), B-cell lymphoma 2 (Bcl-2), Bcl-2-associated death promoter (Bad) and cleaved-caspase-3 were assessed by immunoblotting. Results: Administration of pellets containing 5% acai powder reduced the incidences of both colonic adenoma and cancer (adenoma, 23.1% vs 76.9%, respectively, p=0.006; cancer, 15.4% vs 76.9%, respectively, p=0.002). In the acai-treated mice, the MPO, TNF-α, IL-1β and IL-6 levels in the colon were significantly down-regulated. Acai inhibited PCNA and Bcl-2 expression and increased Bad and cleaved-caspase-3 expression. In vitro studies demonstrated that acai treatment reduced lipopolysaccharide-induced expression of TNF-α, IL-1β, IL-6 and COX-2 in murine macrophage RAW 264.7 cells. Conclusions: Acai demonstrated protective effects against AOM/DSS-induced colon carcinogenesis, which suggests that the intake of acai may be beneficial for the prevention of human colon cancer.         Identification of a picornavirus from chickens with transmissible viral proventriculitis using metagenomic analysis.

        <P>Transmissible viral proventriculitis (TVP), an infectious disease in chickens, is responsible for economic losses in the commercial poultry industry. The major etiologic agent, however, is unknown. Using metagenomics, we compared the diversity of viruses present in proventriculus samples from flocks diagnosed with TVP to those of healthy flocks in South Korea between 2003 and 2012. Each sample had a mean of 21,538,726 sequence reads generated by high-throughput sequencing, with a mean length of 160 nt. Enrichment in viral sequences suggested that at least three viruses were present in each TVP sample. Although we could not determine a pathogen of TVP that matched the known morphology, picornavirus sequences were present in all five disease samples, suggesting an association with TVP. The five samples yielded 1,045-1,720 bp contigs with 81-84 % nt sequence identity to turkey hepatitis virus (accession number: HM751199). Whole-genome analysis indicated that the QIA01 strain of the novel picornavirus was similar to turkey hepatitis virus in the P2 and P3 regions (82.7 % nt and 95.5 % aa sequence identity), but different in the structural region and partial 2A peptides (56.2 % nt and 23.9 % aa sequence identity). In addition, the QIA01 virus was similar (87.0 % nt and 95.6 % aa sequence identity) to chicken megrivirus, recently detected in chickens with malabsorption syndrome in Hungary. Our results are useful for understanding the genetic diversity of avian picornaviruses and for classifying chicken megrivirus as a pathogen affecting the digestive tract of chickens.</P>

        Anti-inflammatory activity of doxorubicin on lipopolysaccharide-induced hyperactivation of spleen cells

        Doxorubicin has been used to treating cancers, including breast cancer, bladder cancer, and acute lymphocytic leukemia, however, few studies have investigated its anti-inflammatory activity. In this study, we used mouse spleen cells treated with lipopolysaccharide (LPS), a representative inflammatory agent to investigate the effects of doxorubicin. Specially, we investigated the effects of doxorubicin on metabolic activity, cell size, cell death, and cytokine production of LPS-treated spleen cells. Doxorubicin significantly decreased the metabolic activity, even when applied at relatively low concentrations (1.6-8 ng/mL). To investigate the potential mechanism, we measured the mitochondrial membrane potential (MMP) of the LPS-treated spleen cells using Rhodamine 123. Doxorubicin decreased MMP and cell size, and induced cell death. Furthermore, doxorubicin suppressed the production of tumor necrosis factor (TNF)-alpha, a representative cytokine, in LPS-treated spleen cells. Taken together, doxorubicin decreased metabolic activity and the production of inflammatory cytokines, while increasing the death of LPS-induced hyperactivated spleen cells. This results will enable broader application of doxorubicin, as an anti-inflammatory agent, in clinical and research fields.
