      • Cellular and ultrastructural changes in the seedling roots of upland rice (Oryza sativa) under the stress of two allelochemicals from Ageratina adenophora

        Two sesquiterpene-derivative compounds, 4,7-dimethyl-1-(propan-2-ylidene)-1,4,4a,8atetrahydronaphthalene-2,6(1H, 7H)-dione (DTD) and 6-hydroxy-5-isopropyl-3,8-dimethyl-4a,5,6,7,8,8a-hexahydronaphthalen-2(1H)-one (HHO), are the major putative allelochemicals of the aqueous leachates of Ageratina adenophora.A laboratory experiment was conducted, using the hydroponic method, to evaluate the cellular and ultrastructural changes in the seedling roots of upland rice under the stress of DTD and HHO. The subsequent changes were observed in the treated upland rice roots in comparison with their controls. The scanning electron microscopy results showed that the DTD-treated root tip cells turned into an irregular arrangement and shape and that most of them were wizened, with a poor cytoplasm. In the HHO treatment, the root tips had many irregularly shaped cells, with a greater number of sloughing cells, as well as short, wide cells that resulted in spherical and wider, but shorter, roots. At the ultrastructural level, DTD and HHO induced irregularly shaped and lobed nuclei, increased cytoplasmic vacuolation, reduced ribosome density and dictyosomes, and a reduced number of mitochondria in the cells, which indicated limited protein transportation and a reduced capability to export substances for cell development and growth in the upland rice seedling roots. The overall effect of HHO on the upland rice seedlings was more pronounced than that of DTD.

        Physicochemical and Sensory Characterization of Ginger-juice Yogurt during Fermentation

        The primary purpose of this study was to investigate the effects of a ginger (Zingiber ofcinale) juice addition to milk on the physicochemical and sensory properties and consumer acceptability of yogurt during fermentation. Under the presence of ginger juice at 2.0 to 10.0%(v/v), there was a significant influence (p<0.05) on the viability of characteristic microorganisms in yogurt,the pH, titratable acidity (ºSH), viscosity, and hardness of the products. The descriptive data showed significant differences (p<0.05) among the samples for all attributes. The consumer results from external preference mapping indicated that samples with the lower ginger juice (≤4%)were liked over the higher samples (≥6%), with a negative impact on the acceptance by the consumers. Therefore,these findings suggest that 4%(v/v) may be the more appropriate content of ginger juice in the preparation of ginger-juice yogurt.

        Synchronous Ectopic Pancreatoblastoma in a Child: A Case Report

        Pancreatoblastoma is a rare primary pancreatic neoplasm of children that may arise in any portion of the pancreas. We report a case of a 3-yr-old boy who presented to with abdominal pain our hospital and a progressive bulge in his right abdomen. Biochemical evaluation and serum levels of tumoral markers were within reference limits. On the computed tomography, two tumors were found. One located in the head of the pancreas;however, a laparotomy revealed that the head of pancreas was compressed but normal. The other was in the left abdomen near the spleen and the tail of the pancreas. The diagnosis of two synchronous pancreatoblastoma originating from the omentum was confirmed by pathology. Therefore, a pancreatoblastoma should be considered when a large well-defined, lobulated, and heterogeneous mass is identified in the pancreas of children. In addition, an ectopic pancreatoblastoma should be considered when identified within or near the ectopic pancreatic tissue.

        A Calculation Method for the Nonlinear Crowbar Circuit of DFIG Wind Generation based on Frequency Domain Analysis

        The ride-through control of a doubly-fed induction generator (DFIG) for the voltage sags on wind farms utilizing crowbar circuits by which the rotor side converter (RSC) is disabled has being reported in many literatures. An analysis and calculation of the transient current when the RSC is switched off are of significance for carrying out the low voltage ride through (LVRT) of a DFIG. The mathematical derivation is highlighted in this paper. The zero-state and zero-input responses of the transient current in the frequency domain through a Laplace transformation are investigated, and the transient components in the time domain are achieved. With the characteristics worked out from the linear resolving without modeling simplification, the selection of the resistance in the linear crowbar circuit and the value conversion from a linear circuit to a nonlinear one is proposed to setup the attenuation rate. In terms of grid code requirements, the theoretical analysis for the time constant of the transient components attenuation insures the controllability when the excitation of the RSC is resumed and it guarantees the reserved time for the response of the reactive power compensation. Simulations are executed in MATLAB/SIMPOWER and experiments are carried out to validate the theoretical analysis. They indicate that the calculation method is effective for selection of the resistance in a crowbar circuit for LVRT operations.

        Two Androstane Derivatives from the Cultures of Fungus Marasmiellus ramealis (Bull.) Singer

        A new androstane derivative, $4{\beta}$-methyl-15-oxa-$14{\beta}$-androstane-7-ene-$4{\alpha}$-carboxylic acid (1) and a known one $4{\beta}$-methyl-15-oxa-$14{\beta}$-androstane-7-ene-$4{\alpha}$-hydroxyl (2) were isolated from the EtOAc extract of the cultures of the fungus Marasmiellus ramealis (Bull.) Singer. Their structures were elucidated on the basis of 1D and 2D NMR as well as MS spectroscopic data analysis. The inhibitory activity of two isolates against acetylcholinesterase (AChE) revealed that compound 1 exhibited definitely inhibitory activity.

        Separation and Purification of Antioxidant Peptide from Fermented Whey Protein by Lactobacillus rhamnosus B2-1

        In this study, a antioxidant activity peptide fraction was separated and purified from metabolites of whey protein fermented by Lactobacillus rhamnosus B2-1. The fermentation sample was separated by macroporous resin D101 and Sephadex G-15. The collected fractions were tested for antioxidant and antitumor activities. In order to test the antioxidant activity of fractions, Hydroxyl (·OH), 2,2'-azino-bis (3-ethylbenzothiazoline-6- sulfonic acid (ABTS), and Oxygen Radical Absorbance Capacity (ORAC) were used. The final purified peptide B11 showed highest ABTS and ·OH radical scavenging rate by 84.36±1.89% and 62.43±2.64%, respectively, and had an ORAC activity of 1,726.44± 2.76 μM Trolox equivalent/g. Further, the inhibitory effect of B11 on the proliferation of LoVo human colon cancer cells, KB and Cal-27 human oral cancer cells were enhanced with increasing concentrations of B11. B11 contains 51.421% amino acids, with Glu and Asp being the major constituents. In this study, we obtained peptide fraction B11 with antioxidant activity, which is promising for development.

        CD64 Expression Is Increased in Patients with Severe Acute Pancreatitis: Clinical Significance

        Background/Aims: Upregulated CD64 expression on neutrophils is the most useful marker for acute bacterial infections and systemic inflammation. However, it is unknown whether CD64 is involved in the pathogenesis of acute pancreatitis (AP). This study was designed to determine whether CD64 is implicated in severe acute pancreatitis (SAP), and thus, is a suitable marker for SAP. Methods: SAP was induced in rats with an intraperitoneal injection of L-arginine. CD64 expression in the rat pancreas was determined by quantitative real-time polymerase chain reaction (qRT-PCR) and immunohistochemistry. Additionally, the CD64 mRNA expression in peripheral blood leukocytes from 21 patients with mild acute pancreatitis (MAP) and 10 patients with SAP was investigated at the time of admission and during remission by qRT-PCR. Results: CD64 mRNA and protein expression in the pancreas was significantly higher in rats with SAP, compared to the controls. The CD64 expression was higher in the patients with SAP than in the patients with MAP. During remission, CD64 mRNA decreased in both the MAP and SAP patients. The area under the curve of CD64 expression for the detection of SAP was superior to both the Ranson and the Acute Physiology and Chronic Health Evaluation II scores. Conclusions: The CD64 level was significantly increased in correlation with the disease severity in SAP and may act as a useful marker for predicting the development of SAP.         An experimental identification model of rotordynamic coefficients of seals using unbalanced synchronous excitation method

        This study presents an improved impedance method based on unbalanced synchronous excitation to identify the rotordynamic coefficients of labyrinth seals. The rotordynamic coefficient test is implemented near the cylinder resonance frequency to enlarge the influence of seal force. The force generated by the rotor unbalance is used to provide synchronous frequency excitation for the rotordynamic coefficient test. Four unique equations are set up under two sets of different rotor unbalance conditions to obtain four unknown complex rotordynamic coefficients. The factors that influence the rotordynamic coefficients of seals, namely, unbalance mass, inlet/outlet pressure ratio, and rotating speed, are considered. The dynamic coefficients are minimally affected by different rotor unbalances. The direct items are nearly equal with same signs, whereas the cross-coupled items are nearly equal with opposite signs. All coefficients increase with increasing inlet/outlet pressure ratio and rotating speed. The direct stiffness coefficients increase more quickly than the cross-coupled items. In addition, the effect stiffness and effect damping coefficients are analyzed; results indicate that both coefficients increase with increasing rotating speed.

        Human umbilical cord mesenchymal stem cells overexpressing RUNX1 promote tendon-bone healing by inhibiting osteolysis, enhancing osteogenesis and promoting angiogenesis

        Background Rotator cuff injury (RCI) is a common shoulder injury, which is difficult to be completely repaired by surgery. Hence, new strategies are needed to promote the healing of tendon-bone. Objective We aimed to investigate the effect of human umbilical cord mesenchymal stem cells (hUC-MSCs) overexpressing RUNX1 on the tendon-bone healing after RCI, and to further explore its mechanism. Methods Lentiviral vector was used to mediate the overexpression of RUNX1. RUNX1-overexpressed UCB-MSCs (referred to as MSC-RUNX1) were co-cultured with osteoclasts, and TRAP staining was performed to observe the formation of osteoclasts. Then MSC-RUNX1 was cultured in osteogenic differentiation medium, Alizarin red staining was conducted to detect osteogenic differentiation. The expression of markers of osteogenesis and osteoclast was detected by RT-qPCR. EA. hy926 cells were co-cultured with MSC-RUNX1. Transwell assay was used to detect the migration, and the expression of angiogenesis related-genes VEGF and TGF-β was detected by RT-qPCR. The rat rotator cuff reconstruction model was established and MSCs were injected at the tendon-bone junction. Biomechanical test and micro-CT scanning were performed, and HE, Masson and Alcian Blue staining were used for histological evaluation of tendon-bone healing. TUNEL and PCNA immunofluorescence (IF) staining were performed to evaluate apoptosis and proliferation at the tendon-bone healing site. The levels of TNF-α, IL-6 and IL-8 in serum were detected by ELISA. The expression of CD31 and Endomucin that related to angiogenesis was detected by IF. Safranin O-fast and TRAP/CD40L immunohistochemical staining were used to assess the levels of osteoclasts and osteoblasts at the tendon-bone healing site. Results hUC-MSCs overexpressing RUNX1 inhibited osteoclast formation and promoted osteogenic differentiation. MSC-RUNX1 could promote the migration and tube formation of EA. hy926 cells, and up-regulate the levels of VEGF and TGF-β. Model mice treated with MSC-RUNX1 partially restored the biomechanical indexes. Treatment of MSC-RUNX1 obviously increased the bone density, accompanied by the formation of new bone. In vivo experiments showed that MSC-RUNX1 treatment could promote tendon-bone healing and inhibit inflammatory response in rats. MSC-RUNX1 treatment also promoted angiogenesis at the tendon-bone healing site, while inhibiting osteoclast formation and promoting osteogenic differentiation.         Comparison of Apixaban and Low Molecular Weight Heparin in Preventing Deep Venous Thrombosis after Total Knee Arthroplasty in Older Adults

        Purpose: To compare the effect of apixaban and low molecular weight heparin (LMWH) in the prevention and treatment of deepvenous thrombosis (DVT) after total knee arthroplasty in older adult patients. Materials and Methods: A total of 220 patients (average age of 67.8±6.4 years) undergoing total knee arthroplasty were randomlyselected as research subjects and were divided into apixaban and LMWH groups (110 in each group). Results: The incidence of DVT was lower in the apixaban group than in the LMWH group (5.5% vs. 20.0%, p=0.001). Activatedpartial thromboplastin times (35.2±3.6 sec vs. 33.7±2.2 sec, p=0.010; 37.8±4.6 sec vs. 34.1±3.2 sec, p<0.001; 39.6±5.1 sec vs. 35.7±3.0sec, p=0.032) and prothrombin times (14.0±1.0 sec vs. 12.8±0.9 sec, p<0.001; 14.5±1.2 sec vs. 13.0±1.1 sec, p<0.001; 15.3±1.4 sec vs. 13.2±1.3 sec, p=0.009) in the apixaban group at 1 week after surgery, 3 weeks after surgery, and the end of treatment were higherthan those in the LMWH group. Platelet and fibrinogen levels in the apixaban group were lower than those of the LMWH group. Also, capillary plasma viscosity and erythrocyte aggregation in the apixaban group at 1 week after surgery, 3 weeks after surgery,and the end of treatment were lower than those in the LMWH group. Conclusion: Apixaban, which elicits fewer adverse reactions and is safer than LMWH, exhibited better effects in the preventionand treatment of DVT after total knee arthroplasty in older adults.
