        Molecular and cytological analyses of A and C genomes at meiosis in synthetic allotriploid Brassica hybrids (ACC) between B.napus (AACC) and B.oleracea (CC)

        Success of interspecific hybridization relies mostly on the adequate similarity between the implicated genomes to ensure synapsis, pairing and recombination between appropriate chromosomes during meiosis in allopolyploid species. Allotetraploid Brassica napus (AACC) is a model of natural hybridization between Brassica rapa (AA) and Brassica oleracea (CC), which are originally derived from a common ancestor, but genomic constitution of the same chromosomes probably varied among these species through time after establishment, giving rise to cytogenetic difference in the synthetic hybrids. Herein we investigated meiotic behaviors of A and C chromosomes of synthetic allotriploid Brassica hybrids (ACC) at molecular and cytological levels, which result from the interspecific cross between natural B. napus (AACC) and B.oleracea (CC), and the results showed that meiosis course was significantly aberrant in allotriploid Brassica hybrids, and chromosomes aligned chaotically at metaphase I, chromosome bridges and lags were frequently observed from later metaphase I to anaphase II during meiosis. Simultaneously, we also noticed that meiosis-related genes were abruptly down-regulated in allotriploid Brassica hybrids, which likely accounted for irregular scenario of meiosis observed in these synthetic hybrids. Therefore, these results indicated that inter-genomic exchanges of A and C chromosomes could occur frequently in synthetic Brassica hybrids, and provided an efficient approach for genetic changes of homeologous chromosomes during meiosis in polyploid B.napus breeding program.

        Dynamic performance of a rotor system with an initial bow and coupling faults of imbalance-rub during whirling motion

        The dynamic characteristics of a rotor system with an initial bow and coupling fault of imbalance-rub are investigated in this work. The geometrical nonlinearity of shafts becomes significant due to the large-amplitude whirling motion. Then, the influences of the initial bow and geometrical nonlinearity on the natural frequencies corresponding to the linear part of the rotor system are studied. Moreover, the coupling faults of imbalance-rub are introduced to the rotor system. Complicated nonlinear phenomena are revealed by bifurcation diagrams, time histories, Poincaré sections, and spectra. The influences of several key design parameters, such as initial bow, shaft radius, and casing stiffness, are analyzed. One of the main findings of this investigation is that when initial bow and geometrical nonlinearity coexist in a system, the resonance characteristics are obviously affected by the initial bow’s degree. This coexistence can result in the jump phenomenon, which rapidly increases the amplitude of whirling motion. These findings are useful in the fault diagnosis and feature recognition of rotating machines.

        Research on Characteristic Model-based Adaptive Control of High-speed Permanent Magnet Synchronous Motor With Time Delay

        In a high-speed operation, the iron loss of motor cannot be neglected since it may not be small enough. The existence of iron loss will lead to unknown nonlinearities and model uncertainties. Additionally, the existence of output delay in the system will bring speed fluctuation, which may degrade the control efficiency and even cause system instability. Hence, with the aid of continuous form characteristic model, this article provides the adaptive controller to address the control problem for high-speed permanent magnet synchronous motor (HSPMSM) system subject to time delay and plant uncertainties, simultaneously. Specifically, by employing characteristic model is more conducive to the design of controller, that is, the characteristic model only includes the input and output of the system, and the current is not required for parameter estimation. In addition, the proposed controller can deal with time delay, parametric uncertainties and uncertain nonlinearities at the same time. Furthermore, the numerical simulation results demonstrate the effectiveness and robustness of the proposed control strategy.

      • Research on the application of visual symbols in recording APP

        Objective: To prove that the rational use of visual symbols based on human cognitive characteristics can effectively improve the efficiency of daily rapid recording. Background: Smart phones and today’s human beings are almost inseparable. The task of recording trivial information in daily life is naturally transferred to mobile phones. How to quickly complete the recording task and provide good user experience has become a problem faced by many application developers. Method: The characteristics and principles of human cognition were analyzed, the advantages of perceptual design in cognitive process were explained, and a visual model based on user experience was proposed. The model mainly consisted of four levels, namely, strategic layer, range layer, structure layer and presentation layer. Results: Visual symbols have a positive impact on user’s dual experience of recording operation, and different aspects have improved user experience. Conclusion: The case shows that the visual symbol model method based on household experience can be applied to the visual recording of relevant information and improve the efficiency of recording. Application: The proposed visual symbol recording model may provide new help for the development of mobile phone applications with recording function.

      • The Impact of Line Resistance on Load Sharing and An Improved Droop Control of DC Microgrid

        This paper proposes an effective DC voltage and power sharing droop control structure for DC microgrid based on public bus voltage feedback signal. Firstly, the impact of the line resistance on power sharing among droop controlled distributed generators is analyzed and the reason why the line resistance cannot be ignored is presented. Then, an improved droop control is proposed. The simulation test based on MATLAB/Simulink was implemented to demonstrate the proposed approach.

        Expression profiles of circular RNAs in sheep skeletal muscle

        Objective: Circular RNAs (circRNAs) are a newfound class of non-coding RNA in animals and plants. Recent studies have revealed that circRNAs play important roles in cell proliferation, differentiation, autophagy and apoptosis during development. However, there are few reports about muscle development-related circRNAs in livestock. Methods: RNA sequencing analysis was employed to identify and annotate circRNAs from longissimus dorsi of sheep. Reverse transcription followed by real-time quantitative (q) polymerase chain reaction (PCR) analysis verified the presence of these circRNAs. Targetscan7.0 and miRanda were used to analyse the interaction of circRNA-microRNA (miRNA). To investigate the function of circRNAs, an experiment was conducted to perform enrichment analysis hosting genes of circRNAs using gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) pathways. Results: About 75.5 million sequences were obtained from RNA libraries of sheep skeletal muscle. These sequences were mapped to 729 genes in the sheep reference genome. We identified 886 circRNAs, including numerous circular intronic RNAs and exonic circRNAs. Reverse transcription PCR (RT-PCR) and DNA sequencing analysis confirmed the presence of several circRNAs. Real-Time RT-PCR analysis exhibited resistance of sheep circRNAs to RNase R digestion. We found that many circRNAs interacted with muscle-specific miRNAs involved in growth and development of muscle, especially circ776. The GO and KEGG enrichment analysis showed that hosting genes of circRNAs was involved in muscle cell development and signaling pathway. Conclusion: The study provides comprehensive expression profiles of circRNAs in sheep skeletal muscle. Our study offers a large number of circRNAs to facilitate a better understanding of their roles in muscle growth. Meanwhile, we suggested that circ776 could be analyzed in future study.

        <P>Environment-related parameters, including viscosity, polarity, temperature, hypoxia, and pH, play pivotal roles in controlling the physical or chemical behaviors of local molecules. In particular, in a biological environment, such factors predominantly determine the biological properties of the local environment or reflect corresponding status alterations. Abnormal changes in these factors would cause cellular malfunction or become a hallmark of the occurrence of severe diseases. Therefore, in recent years, they have increasingly attracted research interest from the fields of chemistry and biological chemistry. With the emergence of fluorescence sensing and imaging technology, several fluorescent chemosensors have been designed to respond to such parameters and to further map their distributions and variations <I>in vitro</I>/<I>in vivo</I>.         Establishment and application of a solid-phase blocking ELISA method for detection of antibodies against classical swine fever virus

        Background: Classical swine fever (CSF) is a severe infectious disease of pigs that causes significant economic losses to the swine industry. Objectives: This study developed a solid-phase blocking enzyme-linked immunosorbent assay (spbELISA) method for the specific detection of antibodies against the CSF virus (CSFV) in porcine serum samples. Methods: A spbELISA method was developed based on the recombinant E2 expressed in Escherichia coli. The specificity of this established spbELISA method was evaluated using reference serum samples positive for antibodies against other common infectious diseases. The stability and sensitivity were evaluated using an accelerated thermostability test. Results: The spbELISA successfully detected the antibody levels in swine vaccinated with the C-strain of CSFV. In addition, the detection ability of spbELISA for CSFV antibodies was compared with that of other commercial ELISA kits and validated using an indirect immunofluorescence assay. The results suggested that the spbELISA provides an alternative, stable, and rapid serological detection method suitable for the large-scale screening of CSFV serum antibodies. Conclusions: The spbELISA has practical applications in assessing the vaccination status of large pig herds.
