        MUC16 facilitates cervical cancer progression via JAK2/STAT3 phosphorylation‑mediated cyclooxygenase‑2 expression

        Objectives MUC16 (mucin 16, also known as CA-125, cancer antigen 125, carcinoma antigen 125, or carbohydrate antigen 125) has been predicted as tumor biomarker for therapy. We determined to investigate effects and regulatory mechanism of MUC16 on cervical tumorigenesis. Methods Expression levels of MUC16 in cervical cancer cell lines was analyzed via qRT-PCR (quantitative real-time polymerase chain reaction). Knockdown of MUC16 was conducted via shRNA (Short hairpin RNA) transfection. MTT and colony formation assays were used to investigate effect of MUC16 on cell proliferation. Wound healing assay was utilized to detect migration and transwell assay to detect invasion. The underlying mechanism was demonstrated via western blot analysis. Results MUC16 was elevated in cervical cancer cell lines. MUC16 knockdown inhibited cell proliferation, invasion and migration. Gain- and loss-of functional assays revealed that over-expression of MUC16 activated Janus Kinase 2 (JAK2)/ signal transducer and activator of transcription 3 (STAT3) via phosphorylation, thus facilitating cyclooxygenase-2 (COX-2) expression, while knockdown of MUC16 demonstrated the reverse effect on JAK2/STAT3 activation and COX-2 expression. Moreover, inhibition of JAK2/STAT3 attenuated the regulation of MUC16 on COX-2. Conclusions MUC16 enhanced proliferation and invasion of cervical cancer cells via JAK2/STAT3 phosphorylation-mediated cyclooxygenase-2 expression, suggesting the potential therapeutic target ability of MUC16 to treat cervical cancer.

      • PLCE1 Gene in Esophageal Cancer and Interaction with Environmental Factors

        Objective: To study the PLCE1 gene rs2274223 polymorphism with regard to esophageal cancer and its interaction with diet, lifestyle, psychological and environmental factors in Southwest Shandong province. Materials and Methods: A case series study (case-case) was conducted. Questionnaire data were collected and 3 ml-5ml venous blood was drawn for DNA extraction among the qualified research subjects. PLCE1 gene polymorphism was detected after PCR amplification of DNA. SPSS 13.0 software was used for statistical analysis of the data. Results: The three genotypes A/A, A/G and G/G PLCE1 gene rs2274223 was 31, 16 and 4 cases, accounting for 60.8%, 31.4%, 0.08% respectively. The difference of three genotypes (AA/GA/GG) proportion between negative and positive family history of patients was statistically significant, ${\chi}^2=6.213$, p=0.045. There was no statistically significant relationship between PLCE1 gene rs2274223 polymorphism and smoking, drinking, ${\chi}^2=0.119$, p=0.998, and ${\chi}^2=1.727$, p=0.786. There was no linkage of the three rs2274223 PLCE1 gene genotypes (AA/GA/GG) proportion with eating fried, pickled, hot, mildew, overnight, smoked, excitant food, eat speed, salt taste or not (p>0.05). or with living environment pollution and nine risk factors of occupational exposure (p>0.05). There was no statistically significant difference in TS scores between different genotype of rs2274223 PLCE1 gene. Conclusions: The PLCE1 rs2274223 polymorphism has a relationship with family history of esophageal cancer, but does not have any significant association with age, gender, smoking, alcohol drinking, food hygiene, eating habits, living around the environment and occupation in cases.

      • Pretreatment anemia is associated with poorer survival in patients with stage I and II gastric cancer

        <B>Background and Objectives</B><P>A negative correlation between anemia and outcome has been demonstrated in various cancers treated with radiotherapy. However, it is rarely studied whether this correlation may exist in surgical setting. Our aim was to investigate the relationship between pretreatment anemia and survival in surgically treated patients with gastric cancer.</P><B>Methods</B><P>A total of 1,688 patients who had undergone curative resection for gastric cancer between 1991 and 1995 were reviewed. Anemia was defined as a hemoglobin level <12.0 g/dl. The influence of anemia on patient overall survival was evaluated by univariate and multivariate analysis.</P><B>Results</B><P>Pretreatment anemia was present in 39.9% of the patients. The 10-year overall survival rate in anemic patients was 48.2% as compared with 62.6% in nonanemic patients (P < 0.001). In subgroup analysis according to the stage, the significant difference in 10-year overall survival rate between anemic and nonanemic patients was found in stage I and II gastric cancer (76.1% vs. 83.5% in stage I, P = 0.030; 55.1% vs. 67.2% in stage II, P = 0.043). On multivariate analysis, anemia was an independent prognostic predictor in patients with stage I and II disease (P = 0.007; RR, 1.466; 95% CI, 1.109–1.937).</P><B>Conclusions</B><P>Pretreatment anemia was found to have an independent relationship to the long-term survival of patients with stage I and II gastric cancer.         Eclosion hormone functions in larva-pupa-adult ecdysis in Leptinotarsa decemlineata

        In Drosophila melanogaster, ecdysis-triggering hormone (ETH) and eclosion hormone (EH) mutually enhance the release of each other through a positive feedback loop. Both are involved in the stimulation of ecdysis behavior series and molting-related physiological preparations. However, whether 20-hydroxyecdysone (20E) controls eh expression and whether EH plays a similar role in non-Drosophilid insects remain to be determined. In Lep tinotarsa decemlineata, we found that Ldeh mRNA levels were high in the early and/or late stages and low in the intermediate period within each larval instar, indicating that the mRNA peaks positively correlate with 20E pulses. Consistent with the indication, we confirmed that 20E signaling stimulated the transcription of Ldeh. RNA interference (RNAi)-aided knockdown of Ldeh at the final instar stage slightly impaired pupation and signifi cantly affected wing expansion. Around 20% of the Ldeh RNAi larvae remained as prepupae, completely wrapped in the old larval cuticles. These prepupae gradually darkened, dried and withered, and finally died. The remaining (about 80%) Ldeh hypomorphs became pupae and emerged as abnormal adults, bearing smaller and wrinkle elytra and hindwings. The adult tracheae in the Ldeh hypomorphs were full of liquid. In addition, the low pupation rate and high deformed adult proportion cannot be rescued by ingestion of either 20E or juvenile hormone. Thus, EH is critical for ecdysis and trachea clearance in L. decemlineata. The failure of trachea clearance may disenable air-swallowing during larva-pupa-adult transition and thus impacts wing growth and expansion.

        Prediction of Failure Mode in Hole Clinching of Al Alloy and Advanced High-Strength Steel

        <P>Common joining methods used in automotive industry are welding, adhesive bonding, friction stir welding, mechanical fastening, self-piercing rivets, mechanical clinching and so on, for multi-material designed automotive bodies. Among different joining methods, mechanical clinching which achieves geometrical interlocking by plastic deformation has several advantages such as no need of additional joining elements and fast joining. But mechanical clinching is difficult to join a ductile material with a high-strength or low ductility material. Therefore the hole clinching as a new mechanical clinching process has been proposed to join these material combinations. In the hole clinching process, as punch force is applied to a upper sheet (a ductile material), it is indented into hole of lower sheet (low ductile material) on die and then interlocked by plastic deformation. It is very important for a successful design of hole clinching to predict the failure mode such as neck fracture and button separation and the strength of hole-clinched joint. For this an analytical approach was carried out for the hole clinching process of Al6061 and DP980. Tool geometry used in hole clinching was designed by the predicted failure mode. Preliminary finite element simulation was performed to validate the geometrical interlocking and joinability. The predicted failure mode and strength were verified by the results of cross tension test.</P>

        Dietary supplementation of Eucommia leaf extract to growing-finishing pigs alters muscle metabolism and improves meat quality

        Objective: The objective of this study was to investigate the influence of dietary supplementation of Eucommia ulmoides leaf extract (ELE) on muscle metabolism and meat quality of pigs with and without pre-slaughter transportation. Methods: In a 43-day feeding experiment, a total of 160 pigs with an initial body weight 60.00±2.00 kg were randomly assigned into four groups in a completely randomized design with 10 replicates. Pigs in groups A and C were fed a basal diet and pigs in groups B and D were fed a basal diet supplemented with 0.5% ELE. Pigs were slaughtered with (group B and D) or without (group A and C) pre-slaughter transport. Muscle chemical composition, postmortem glycolysis, meat quality and muscle metabolome were analyzed. Results: Dietary ELE supplementation had no effect on the proximate composition of porcine muscle, but increased free phenylalanine, proline, citruline, norvaline, and the total free amino acids in muscle. In addition, dietary ELE increased decanoic acid and eicosapentaenoic acid, but decreased heptadecanoic acid, oleic acid, trans-oleic acid, and monounsaturated fatty acids in muscle. Meat quality measurement demonstrated that ELE improved meat water holding capacity and eliminated the negative effects of pre-slaughter transport on meat cooking yield and tenderness. Dietary ELE reduced muscle glycolytic potential, inhibited glycolysis and muscle pH decline in the postmortem conversion of muscle to meat and increased the activity of citrate synthase in muscle. Metabolomics analysis by liquid chromatographic tandem mass spectrometric showed that ELE enhanced muscle energy level, regulated AMP-activated protein kinase (AMPK) signaling, modulated glycogenolysis/glycolysis, and altered the metabolism of carbohydrate, fatty acids, ketone bodies, amino acids, purine, and pyrimidine. Conclusion: Dietary ELE improved meat quality and alleviated the negative effect of preslaughter transport on meat quality by enhancing muscle oxidative metabolism capacity and inhibiting glycolysis in postmortem muscle, which is probably involved its regulation of AMPK.

        Effect of Heat Treatment on Microstructure and Mechanical Properties of Mg-10Li-3Al-3Zn-0.22Si Alloy

        The microstructure evolution and mechanical properties of as-extruded Mg-10Li-3Al-3Zn-0.22Si during heat treatment from473 to 673 K were investigated in this paper, and then the strengthening mechanism and fracture behavior of the studied alloywas also analyzed. Experimental results show that the matrix of as-extruded Mg-10Li-3Al-3Zn-0.22Si alloy mainly consistsof α-Mg and β-Li, and dispersive granular phases are AlLi and Li2MgAl. Mg2Siphase shows banded distribution alongextrusion direction. The morphology of α-Mg is from long strip to nearly spherical block at 573 K. The α-Mg reprecipitatesfrom β-Li matrix in the orientation relationship: (0001) α-Mg // (110) β-Li, [11 −2 0] α-Mg // [1 −1 0] β-Li at 623 K. While theAl-rich granular phase is decomposed and dissolve into the gradually, and then AlLi increase again at 673 K, which may becaused by the dissolution at high temperature and precipitation during air cooling in heat treatment. Besides, the solid solutionand dispersion strengthening enhances the strength of alloy after heat treatment. After heat-treated at 573 K for 1 h and thenair cooling, alloy presents excellent mechanical properties with the ultimate tensile strength of 253 ± 3 MPa and elongationof 27 ± 3%, respectively. Compared with the extruded state, ultimate tensile strength of alloy can be increased by about 49%.

        Development of a position sensitive CsI(Tl) crystal array

        A position-sensitive CsI(Tl) crystal array coupled with the multi-anode position sensitive photomultiplier tube (PS-PMT), Hamamatsu H8500C, has been developed at the Institute of Modern Physics. An effective, fast, and economical readout circuit based on discretized positioning circuit (DPC) bridge was designed for the 64-channel multi-anode flat panel PSPMT. The horizontal and vertical position resolutions are 0.58 mm and 0.63 mm respectively for the 1.0 × 1.0 × 5.0 ㎣ CsI(Tl) array, and the horizontal and vertical position resolutions are 0.86 mm and 0.80 mm respectively for the 2.0 × 2.0 × 10.0 ㎣ CsI(Tl) array. These results show that the CsI(Tl) crystal array with low cost could be applied in the fields of medical imaging and high-resolution gamma camera.

        Multivariate analysis of original identification and chemical markers exploration of Chinese ginger

        Ginger (Zingiber officinale) is one of the most widely consumed dietary supplements. However, the content of active ingredients varied greatly from place to place. In this study, we first identified and compared the compositions of ginger samples from six different origins. Then, we evaluated the anti-inflammatory activity of different samples in LPS-stimulated RAW264.7 cells. The results indicated that highly variable in chemical composition and activity for ginger from different origin. Further, correlation analysis showed that isoborneol, terpineol, α-curcumene, germacrene D, α-elemol and 8-shogaol exhibited a strong correlation with inflammatory factors, which could be used as potential chemical markers to evaluate quality and distinguish source of ginger. Finally, comprehensive evaluation found that the ginger from Sichuan exerts stronger anti-inflammatory properties. This study will help to select ginger varieties with excellent characteristics, provide theoretical basis for the development and utilization of ginger.
