      • Dexamethasone Disrupts Cytoskeleton Organization and Migration of T47D Human Breast Cancer Cells by Modulating the AKT/mTOR/RhoA Pathway

        Background: Glucocorticoids are commonly co-administered with chemotherapy to prevent drug-induced allergic reactions, nausea, and vomiting, and have anti-tumor functions clinically; however, the distinct effects of GC on subtypes of tumor cells, especially in breast cancer cells, are still not well understood. In this study, we aimed to clarify the effect of GC on subtypes of T47D breast cancer cells by focusing on apoptosis, cell organization and migration, and underluing molecular mechanisms. Materials and Methods: The cell scratch test was performed to observe the cell migration rate in T47D cells treated with dexamethasone (Dex). Hoechst and MTT assays were conducted to detect cell survival and rhodamine-labeled phalloidin staining to observe cytoskeleton dynamics. Related factors in the AKT/mTOR pathway were determined by Western blotting. Results: Dex treatment could effectively inhibit T47D breast cancer cell migration with disruption of the cytoskeletal dynamic organization. Moreover, the effect of Dex on cell migration and cytoskeleton may be mediated by AKT/mTOR/RhoA pathway. Although Dex inhibited T47D cell migration, it alone may not induce cell apoptosis in T47D cells. Conclusions: Dex in T47D human breast cancer cells could effectively inhibit cell migration by disrupting the cytoskeletal dynamic organization, which may be mediated by the AKT/mTOR/RhoA pathway. Our work suggests that glucocorticoid/Dex clinical use may prove helpful for the treatment of breast cancer metastasis.

        A Tentative Study on the Practical Changes of the Oral Manchu in Sanjiazi Village

        Though in Sanjiazi Village where Manchu language is best preserved in China now, the language used is in a serious declining state than before as there are only few elder Manchu people who can speak fluently. In less than fifty years from 1960s to early of 21st century, Manchu language used in Sanjiazi has undergone a declining course from a communicative language to an endangered one. Based on the statistics and materials from the fieldwork of other researchers in different times and the author, this paper tends to study the actual changes of the spoken Manchu in Sanjiazi Village and discusses the major causes of change.

        Use of Nitrate-nitrogen as a Sole Dietary Nitrogen Source to Inhibit Ruminal Methanogenesis and to Improve Microbial Nitrogen Synthesis In vitro

        An in vitro study was conducted to determine the effect of nitrate-nitrogen used as a sole dietary nitrogen source on ruminal fermentation characteristics and microbial nitrogen (MN) synthesis. Three treatment diets were formulated with different nitrogen sources to contain 13% CP and termed i) nitrate-N diet (NND), ii) urea-N diet (UND), used as negative control, and iii) tryptone-N diet (TND), used as positive control. The results of 24-h incubations showed that nitrate-N disappeared to background concentrations and was not detectable in microbial cells. The NND treatment decreased net $CH_4$ production, but also decreased net $CO_2$ production and increased net $H_2$ production. Total VFA concentration was lower (p<0.05) for NND than TND. Suppression of $CO_2$ production and total VFA concentration may be linked to increased concentration of $H_2$. The MN synthesis was greater (p<0.001) for NND than UND or TND (5.74 vs. 3.31 or 3.34 mg/40 ml, respectively). Nitrate addition diminished methane production as expected, but also increased MN synthesis.

        High-energy solid-state asymmetric supercapacitor based on nickel vanadium oxide/NG and iron vanadium oxide/NG electrodes

        <P><B>Abstract</B></P> <P>Solid-state supercapacitors (SCs) are well-known as one of the most competitive power sources for modern electronics. However, most of the reported solid-state SCs suffer from low specific capacitance and energy density. Herein, a novel strategy for the synthesis of nickel vanadium oxide (Ni<SUB>3</SUB>V<SUB>2</SUB>O<SUB>8</SUB>) and iron vanadium oxide (Fe<SUB>2</SUB>VO<SUB>4</SUB>) nanoparticles (NPs) anchored nitrogen doped graphene (NG) for high energy solid-state asymmetric SC (ACS) through a simple and cost-effective hydrothermal technique was demonstrated. SEM and TEM analysis reveals that active Ni<SUB>3</SUB>V<SUB>2</SUB>O<SUB>8</SUB> and Fe<SUB>2</SUB>VO<SUB>4</SUB> NPs with uniform size are anchored on NG sheets. Electrochemical performance of Ni<SUB>3</SUB>V<SUB>2</SUB>O<SUB>8</SUB>/NG and Fe<SUB>2</SUB>VO<SUB>4</SUB>/NG electrodes showed that both have ultra-high specific capacitances (∼1898 F g<SUP>−1</SUP> and 590 F g<SUP>−1</SUP> at current density of 1 A g<SUP>−1</SUP>, respectively), tremendous rate capabilities, and superior cycling stabilities. Most significantly, solid-state ASC consisting of Ni<SUB>3</SUB>V<SUB>2</SUB>O<SUB>8</SUB>/NG as a cathode and Fe<SUB>2</SUB>VO<SUB>4</SUB>/NG as an anode which achieves a high energy density of ∼77.2 W h kg<SUP>−1</SUP> at a power density of 863 W kg<SUP>−1</SUP> and an ultra-long cycle life (capacitance retention of ∼83.3% after 20,000 cycles).       • Elevated Expression of Nuclear Protein Kinase CK2α as a Poor Prognosis Indicator in Lymph Node Cancerous Metastases of Human Thyroid Cancers

        Aim: To investigate the expression of protein kinase $CK2{\alpha}$ ($CK2{\alpha}$) in human thyroid disease and its relationship with thyroid cancer metastasis. Materials and Methods: Using immunohistochemistry we measured the expression of $CK2{\alpha}$ in 76 benign and malignant human thyroid cancer tissues, including 10 pairs of papillary carcinoma tissues with or without lymph node cancerous metastasis and similarly 10 pairs of lymph nodes. Results: The expression of $CK2{\alpha}$ was found to be higher in thyroid carcinoma cases (papillary carcinoma, follicular carcinoma, anaplastic carcinoma and medullary carcinoma) than in ones such as chronic lymphocytic thyroiditis, nodular goiter and adenoma. These findings were also confirmed by RT-PCR and Western blotting. More strikingly, elevated expression of $CK2{\alpha}$ in thyroid papillary carcinoma tissues was not only significantly associated with lymph node cancerous metastasis and clinical stage of thyroid cancers; but also correlated with epithelial-mesenchymal transition (EMT) and high tenascin C (TNC) expression. In addition, EMT and high TNC expression in thyroid carcinoma tissues was significantly associated with lymph node cancerous metastasis. Conclusions: Elevated expression of nuclear $CK2{\alpha}$ is a poor prognosis indicator in lymph node cancerous metastasis of human thyroid cancers.

        Chinoketides A and B, Two New Antimicrobial Polyketides from the Endophytes of Distylium chinense with the "Black-Box" Co-culture Method

        Two new polyketides, chinoketides A and B (1 - 2) with a known compound xylarphthalide A (3), were isolated from the solid medium of the endophytes from the leaves of the relic plant Distylium chinense with the "black-box" co-culture method, and the structures of two new compounds were elucidated by NMR, MS and CD spectra. And the absolute configurations of chinoketides A (1) and B (2) were determined as 2R,3R,8S and 5R,6S by calculating their ECD spectra to compare with the experimental CD spectra. Finally, the antimicrobial activities were evaluated to Erwinia carotovora sub sp. Carotovora (Jones) Bersey et al, and the results showed that compounds 1 - 3 displayed the antimicrobial activities with MIC value at 20.5, 30.4 and 10.2 mg/mL.

        Polyethylene-Based Dielectric Composites Containing Polyhedral Oligomeric SilSesquioxanes Obtained by Ball Milling

        High-energy ball milling was tested as a method for producing Ultra High Molecular Weight Polyethylene (UHMWPE)- based nanodielectrics containing 1 wt% and 5 wt% OctaIsoButylPOSS (OibPOSS). Qualitative and quantitative evaluations were used to explore the compatibility between OibPOSS and PE. Several ball milling variables were optimized in a bid to achieve UHMWPE/OibPOSS nanodielectrics. The morphology, as well as the thermal and the dielectric properties of the samples, were characterized by scanning electron microscopy, thermogravimetric analysis, broadband dielectric spectroscopy, and progressive-stress breakdown tests. The results showed that (i) ball milling was an effective method for producing UHMWPE/OibPOSS dielectric composites, but appeared ineffective in dispersing OibPOSS at the nanoscale, and (ii) the resulting UHMWPE/OibPOSS dielectric composites presented thermal and dielectric properties similar to those of neat UHMWPE.

        Changing Process and Causes of Manchu Language in Heihe District

        Heihe District used to be an important area where most Manchu people live and thus the fluent use of Manchu language. However, as the time passes, Manchu language is on the process of decline. This paper explores the changing and causes of Manchu language declination by summarizing fieldwork materials and authors observation. Manchu language used to be widely used especially in villages such as Dawujiazi, Xiamachang and Siji of Sunwu of Heihe District where Manchu was preserved fairly completely before mid-1960s, and at that time Manchu was a live language used for communication among the Manchu people. However, between mid-1960s and mid-1980s, the Manchu language was in the state of declining, and from mid-1980s till today, the Manchu language is in the state of extinction. The authors of the paper believe that though there are many causes contributing to the declining of the language, the most important ones are the following three aspects: the political pressure, the immigration of the nonManchu language speakers and the dispersed Manchu resident situation.
