        Study on Method of Sperm Aspiration and Injection into an Oocyte in Intracytoplasmic Sperm Injection(ICSI) Immobilization of spermatozoa prior to intracytoplasmic sperm iniection(ICSI) sometimes results in crooked tail and this makes it difficult to aspirate sperm into an injection pipette tail first. Head-first sperm aspiration into an injection pipette avoid this problem due to the bigger size of the sperm head. The effect of head or tail-first sperm injection into an oocyte on fertilization cleavage, percentage of grade I embryos and development to blastocyst stage in ICSI program has been studied. A single living immobilized spermatozoa from oligoasthenozoospermic patient was injected into an oocyte head-first or tail-first according to the treatment. Eighteen hours after microinjection, oocytes ware inspected for survival and fertilization Fertilized oocytes with two pronuclei were cultured in 30μl drop of mHTF supplemented with 10% heat-inactivated follicular fluid(FF) at 37℃. On day 2. embryo transfer was performed with cleaved embryos. The remaining 2-8 cell stage embryos were co-cultured with BRL cells in mHTF + 10% FF for 72 hours and the developmental stage was observed. The data were analyzed by Analysis of Variance. A total of 164 oocytes from 36 cycles were assigned to earth treatment and ICSI was performed(88 head-first, tail-first). The rates of normal fertilization were 81.8% and 76.3% for head-first and tail-first, respectively. Of the fertilized oocytes, the percentage of cleaved embryos and the percentage of grade 1 embryo among cleaved embryos were 88.9% and 68.8%, 93.1% and 74.1% for head-first and tail-first, respectively. Of the 2-8 cell embryos cultured, 44.4%(16/36) and 50.0%(10/20) for head first and tail first, respectively developed to blastocyst stage. There were no differences in fertilization, cleavage, rates of grade 1 embryos, and development to blastocyst stage. In conclusion, head-first or tail-first sperm injection into an oocyte in ICSI program does not affect fertilization and subsequent embryo development to blastocyst stage in vitro.

      • 魚類 및 새우 低溫貯藏中의 鮮度 變化

        Saito等(1959)이 提案한 K-値에 依한 鮮度 判定法이 우리나라에서 生産되는 魚類나 甲殼類의 鮮度를 判定하는데 適用할 수 있는가를 檢討하기 위한 資料를 얻기 위하여 Kobayashi and Uchiyama(1970)의 K-値 迅速定量法으로 우선 底棲魚種이며 白色肉이 많은 붕장어, 表層회游魚이고 活動性이 强한 학꽁치 및 海底에 묻혀 사는 보리새우를 試料로 하여 氷藏및 凍結貯藏中의 K-値의 變化를 實驗하여 揮發性鹽基窒素量, 總菌數, pH 및 官能檢査 結果와 比較하였다. 살아 있는 붕장어와 보리새우를 卽殺하여 氷藏하였을 때 貯藏 中의 K-値는 貯藏期間에 따라 거의 직선적으로 增加하였다. 官能檢査 結果와 比較하여 보면 氷藏 8日만에 腐敗初期에 도달하였으며, 이 때 붕장어의 K-値는 26%, 보리 새우의 K-値는 23%였다. 한편 鮮度 좋은 학꽁치도 氷藏 中 K-値가 急激히 增加하였으며 腐敗初期라고 보아 지는 氷藏 7日째의 K-値는 43%였다. K-値 揮發性鹽基窒素量, 總菌數, pH 및 官能檢査 結果를 比較하여 볼 때 K-値는 生鮮의 品質을 評價하는데 있어 有效한 判定方法 中의 하나라고 볼 수 있다. It is so far suggested from the results of investigation on the changes in nucleotides during the storage of fish and shellfish that ATP rapidly breaks down and forms such derivatives as adenylic acid, inosinic acid, inosine and hypoxanthine, and ribose, mainly in the early stage of storage. Therefore K value representing the ratio of optical densities of inosine and hypoxanthine fraction and total nucleotide fraction at 250 nm may be useful as an index to measure the freshness of the fishes in prior to bacterial deterioration. In this paper, the correlation between K value and freshness was discussed to elucidate whether K value is an useful index of freshness. The freshness variations of conger eel, half beak and oriental brown shrimp during icing and cold storage were determined by means of volatile basic nitrogen, pH, total bacterial count, and organoleptic test. The results showed that the K value of the three smaples were rapidly and linearly increased during iced storage being changed 26% for the conger eel, 23% for the oriental brown shrimp and 43% for the half beak respectively, while that was maintained almost consistently during three months cold storage at -20℃. As a conclusion it is noted that the K value could be useful as an index to measure the freshness of fish muscle preserved with ice.

      • 淡水漁의 呈味成分에 관한 硏究 : 第1報 天然産 잉어의 유리아미노산 및 핵산관련물질 Part 1. Contents of Free Amino Acid, Nucleotides and Their Related Compounds in Wild Common Carp

        우리나라 全域에 걸쳐 분포하고 있는 天然産 잉어, Cyprinus corpio의 맛성분으로서의 유리 아미노산 및 핵산관련물질을 分析하였다. 全유리아미노산에 대한 含量이 많은 아미노산이 차지하는 比率은 암수 各各 histidine이 39.3%, 56.1%, lysine이 13.5%, 9.5%, arginine이 10.5%, 6.2%. glycine이 9.8%, 8.2%, taurine이 8.0%, 7.5%로서 이들 5種의 아미노산이 全유리아미노산의 81.1%, 87.5%를 各各 차지하였고 다음이 alanine, threonine, glutamic acid, leucine, serine이었으며, 含量이 적은 것은 isoleucine, methionine, valine, proline, aspartic acid의 順이었다. 핵산관련물질은 암컷과 수컷에 있어서 各各 ATP가 0.8, 0.3 μmole/g, ADP가 0.7, 0.8 μmole/g, AMP가 0.5, 0.5 μmole/g, IMP가 3.0, 1.5 μmole/g, inosine이 0.8, 3.5 μmole/g, hypoxanthine 0.6, 1.3 μmole/g이었다. Common carp, Cyprinus carpio, has been esteemed as one of the most tasty fresh-water fishes in korea. The free amino acids composition of the extracts of wild common carp meat was analysed by the amino acid autoanalyser. In the extract, histidine, lysine, arginine, glycine and taurine were abundant, holding 39.3%, 15.3%, 10.5%, 9.8%, and 8.0% of total free amino acid contents, respectively and then followed by alanine, threonine, gutamic acid, leucine, serine, isoleucine and methionine, in oder. While valine, proline and aspartic acid were poor in content. IMP was dominant in the fresh wild common carp showing 60% of total nucleotides while ATP, ADP, AMP, inosine hypoxanthine were low in content. It would be considered that IMP is quite important as the index flavor quality of the carp muscle, because IMP is the major nucleotide in the carp muscle the flavor enhancing constituent.

      • 인공 건조법에 의한 마른 명태의 품질 개선에 관한 연구 : 1. 열풍 건조중의 명태의 핵산 관련 물질 및 유리 아미노산의 변화 1. Changes of Nucleotides and Their Related Compounds and Free Amino Acid Composition of Alaska Pollack during Hot-air Dehydration

        북양산 냉동명태를 원료로서 열풍건조법으로 마른명태를 가공하였을 때, 건조중의 핵산관련물질 및 유리아미노산의 변화를 실험하였다. 원료육중에는 inosine 함량이 가장 많아 건물증량 기준으로 13.5μmole/g였고, 다음으로 함량이 높은 IMP였는데 5.1μmole/g로서 비교적 낮은 값이었다. 열풍건조중 IMP는 급격히 감소하여 잔존량은 14%에 불과하였으며, inosine은 건조중 50%감소하였다. 한편 hypoxanthine는 약 5배 증가하여 건물중량 기준으로 16.9μmole/g 였다. 건조후 3개월간 저장중에는 건조직후와 비교할 때 핵산관련물질의 변화는 거의 없었다. 그리고 원료 명태 육중의 유리아미노산 조성은 taurine이 가장 많아 전 유리아미노산의 40%를 차지하고 alanine, glycine, glutamic acid가 그 다음이고 비교적 함량이 낮은 것은 lysine, serine, histidine이며, 함량이 극히 낮은 것은 valine, aspartic acid, threonine, isoleucine, leucine등이었다. 열풍 건조중 유리아미노산의 질적 변화는 없었으며. 원료육 유리아미노산에 대하여 건조중 유리아미노산양이 20% 증가하였다. In this paper, the changes of nucleotides and their related compounds and free amino acid composition of Alaska Pollack muscle during hot-air dehydration and storage for three months were determined. The results showed that the contents of inosine and IMP in defrosted sample were 13.5 and 5.1 μmole/g respectively on dry base, and followed by hypoxanthine in order. ATP, ADP and AMP was very low in content. IMP and inosine tended to degrade rapidly during hot-air dehydration while hypoxanthine increased about five times. But the quantitative changes of nucleotides and their related compounds in the muscle of hot-air dehydrated Alaska Pollack during storage were not detected. Taurine was abundant in the muscle extracts, showing 40% of the total amino acid content and followed by alanine, glycine and glutamic acid. Lysine, serine and histidine were low in content respectively, and valine, aspartic acid, threonine and leucine were very poor. The qualitative change in the pattern of free amino acid composition of muscle extracts was not observed during hot-air dehydration. The increase of the total free amino acid during hot-air dehydration was approximately 20%.

        폐쇄성 수면무호흡 환자의 상기도 형태의 특징과 압력강하에 관한 3차원 전산유체역학해석

        폐쇄성 수면무호흡(obstructive sleep apnea, OSA)은 수면 중에 반복적으로 상기도의 완전폐쇄나 부분폐쇄가 일어나는 질환으로서 흡기된 공기는 반드시 상기도라는 연조직 관(tube) 구조를 통과해야 하므로 상기도의 폐쇄경향은 관의 형태 및 관을 통과하는 공기의 유체역학적 특성에 따라서도 큰 영향을 받을 수 있다. 본 연구에서는 OSA 환자 3인의 치료 전 상기도 CT 이미지를 이용하여 개별화된 3차원 유한요소모델 A, B, C를 제작하고, 비공 당 170, 200, 230 ml/s의 흡기유량에 대하여 3차원 전산유체역학 해석을 시행하였다. 상기도의 유속, 음압 그리고 압력강하를 측정한 결과 관찰된 3개의 모델에서 모두 단면적이 가장 작은 부위에서 유속이 증가하였고, 음압이 크게 나타났다. 기도의 형태는 구개인두와 구인두 부위에서 좁아지는 형태를 가지며, 최소 단면적 영역과 하인두 단면적의 차이가 클수록 유속과 음압의 변화가 크게 나타났다. 비강 부위의 최고 압력과 최소 단면적 영역의 최저 압력의 차이를 의미하는 압력강하는 상기도 저항을 종합적으로 판단할 수 있는 유용한 지표이며, 유량에 따라 증가하였다. Objective: Obstructive sleep apnea (OSA) is a common disorder which is characterized by a recurrence of entire or partial collapse of the pharyngeal airway during sleep. A given tidal volume must traverse the soft tissue tube structure of the upper airway, so the tendency for airway obstruction is influenced by the geometries of the duct and characteristics of the airflow in respect to fluid dynamics. Methods: Individualized 3D FEA models were reconstructed from pretreatment computerized tomogram images of three patients with obstructive sleep apnea. 3D computational fluid dynamics analysis was used to observe the effect of airway geometry on the flow velocity, negative pressure and pressure drop in the upper airway at an inspiration flow rate of 170, 200, and 230 ml/s per nostril. Results: In all 3 models, large airflow velocity and negative pressure were observed around the section of minimum area (SMA), the region which narrows around the velopharynx and oropharynx. The bigger the Out-A (outlet area)/ SMA-A (SMA area) ratio, the greater was the change in airflow velocity and negative pressure. Conclusions: Pressure drop meaning the difference between highest pressure at nostril and lowest pressure at SMA, is a good indicator for upper airway resistance which increased more as the airflow volume was increased.

        뇌실외 배액술 시행과 관련된 뇌실염 : 원인균주와 경험적 치료약제의 적절성

        목적 : 본 연구는 뇌실외 배액술을 시행한 신경외과 환자들에서 뇌실염 발생 빈도, 원인균주와 감수성 추이 및 경험적 치료로 사용되는 항생제인 ceftazidime과 vancomycin의 적절성 여부에 대하여 조사하였다. 재료 및 방법 : 2000년 12월부터 2003년 10월까지 EVD를 시행한 경북대학교병원 신경외과 입원환자 340명 중 뇌실염이 발생한 39명을 대상으로 후향적으로 조사하였다. 결과 : 340명 중 39예 (11.5%)에서 뇌실염이 발생하였고 이들 가운데 4명이 뇌실염과 연관하여 사망하였다(10.3%). EVD 시행일로부터 뇌실염 발생까지는 평균 8.4일이었다. 원인균은 33예(87.1%)에서 확인되었으며 Aci-netobacter (19예, 45%), methicillin-resistant coagulase negative Staphylococcus (9예, 22%), methicillin-resistant Staphylococcus aureus (9예, 22%) 등의 순이었다. 가장 많이 동정된 Acinetobacter 19예 중에서 8예(42.1%)가 ceftriaxone에 내성을 보였으며 현재 경험적으로 널리 사용되는 ceftazidime에 대해서는 11예중 3예(27%)가 내성을 보였다. 결론 : 최근 EVD 시행과 관련된 뇌실염 뇌실염 최근 그람음성 non-fermenter 특히 Acinetobacter에 의한 경우가 증가하고 있으며, 이들 그람음성 non-fermenter는 현재까지 경험적으로 사용되는 ceftazidime에 점차 내성을 보이고 있다. 경험적 치료제에 대한 성공률을 높이기 위해서는 각 병원별 내실염의 원인균주의 감수성에 대한 검사가 필요하겠다. Purpose : To investigate the etiologic microorganisms of external ventricular drain (EVD)-related ventriculitis and the appropriateness of using ceftazidime and vancomycin as an empiric therapy in neurosurgical patients with EVD-related ventriculitis. Materials and Methods : Retrospective analysis of 39 patients with EVD-related ventriculitis among 340 neurosurgical patients to whom EVD had been placed during December 2000 and October 2003 at Kyungpook National University Hospital. Results : Thirty-nine EVD-related infections (39/340, 11.5%) occurred and the attributable mortality rate was 10.3% (4/39). The average duration from the ventricular catheter placement to the development of ventriculitis was 8.4 days. All patients with EVD infection had fever and 89.7% (35/39) of the patients showed nuchal rigidity. The positive culture rate in CSF was 87.1% (34/39) and the frequency of individual organism is as follows : Acinetobacter 45% (19 cases), methicillin-resistant coagulase negative Staphylococcus 22% (9 cases), methicillin-resistant Staphylococcus aureus 22% (9 cases), Enterococcus 5% (2 cases), Streptococcus pneumoniae 3% (1 case), non-fermenting gram-negative bacilli 3% (1 case). The polymicrobial infection rate was 15.4% (6/39). Among 19 cases of Acinetobacter infection, 42.1% (8/19) of the strains showed resistance to ceftriaxone and 15.7% (3/11) to ceftazidime. However, all cases were sensitive to meropenem. Conclusion : These findings show that the major etiologic organisms causing EVD-related ventriculitis have recently changed to Gram-negative non-fermenters, especially Acinetobacter. Because Gram-negative non-fermenting rods resistant to ceftazidime are increasing, an immediate change from ceftazidime plus vancomycin, the widely accepted empiric antibiotic therapy, to meropenem plus vancomycin should be considered when clinical symptoms and signs show no improvement or even deterioration.

      • 인공 건조법에 의한 마른 명태의 품질 개선에 관한 연구 : 2. Some Methods for Hot-air Dehydration of Alaska Pollack 2. 열풍 건조법에 의한 마른 명태 제조

        북양명태를 원료로서 마른명태를 가공하는 실험을 하여 다음과 같은 결과를 얻었다. 원료를 해동하여 내장을 제거하고, 그대로 29시간 열풍건조하여 마른명태를 가공할 수 있었으며, 제품의 수율은 22% 수분은 18%였다. 또한 원료어를 등갈이한 다음 머리, 내장, 흑막을 제거하고, 염장한 다음 22시간 열풍건조하여 건조시간도 단축할 수 있고, 색택도 좋은 제품을 만들 수 있고, 수율은 약 12%였다. 또한 원료어를 fillet로 처리하여 염장한 다음 열풍건조하므로서 건조시간을 더욱 단축할 수 있었고, 포장 및 조리하기에 편리한 제품을 가공할 수 있었다. 제품의 수율은 어체의 크기에 따라 차이가 있어 10∼22%였다. 그리고 채육기로 육을 채취한 다음 과산화수소를 써서 거품을 일어켜 새로운 형태의 다공성인 박층건제품을 가공할 수 있었고, 이 다공성 박층건제품은 흡수성이 좋아 instatant 식품으로 이용 가능하다는 결론을 얻었다. In this paper, some dehydration methods for frozen Alaska Pollack Theragra chalcogramma were studied. The cabinet type drier was used for the study. Samples were dehydrated for appropriate time at the air temperature of 38 to 40℃ under the air stream of 3m/sec. The frozen samples were defrosted, eviscerated, and hot-air dehydrated for 29 hours. The yield of dehydrated product was about 22% and moisture content was 18%. The brine salted samples which were headed, split and skinned were also hot-air dehydrated under the same conditions. This dehydration process could reduce dehydration time. The yield of dehydrated product was about 12% and moisture content was about 20%. For another method, frozen samples were first defrosted, washed thoroughly to remove dirt, slime and scales, and then beheaded, skinned and filleted. The fillet was dehydrated after brine salting. The yield was 10 to 22% and moisture content was 12%. The tissue of these hot-air dehydrated products such as round, split and fillet were very firm and difficult to reconstitute. In order to obtain dehydrated product of agreeable texture, another dehydration process was examined. Chopped meat was dipped in alkaline solution of 0.4% hydrogen peroxide for 30 minutes and then agitated for 10 minutes. By these treatment the foamy and emulsified minced meat was spread on a nylon net seated on metal tray, drained, and dehydrated. A better result was obtained by this process comparing with other's. This could be explained for the reason that the process resulted in the meat flake porous by which the meat could be rapidly reconstituted. The texture of the porous meat flake was good for a convenience food.

        Case Report : Metastatic malignant melanoma with peritoneal seeding in a young woman: A case report

        Malignant melanomas of the uterus, either primary or metastatic, are extremely rare. They can be mistaken as other tumors, such as uterine sarcomas during diagnosis. We describe here the first case of a metastatic melanoma of the uterus with peritoneal seeding in a young woman. It was first diagnosed as a uterine sarcoma from a frozen-section biopsy.
