        Heme oxygenase-1 mediates nicotine- and lipopolysaccharide-induced expression of cyclooxygenase-2 and inducible nitric oxide synthase in human periodontal ligament cells

J Periodont Res 2010; 45: 177–183. © 2010 John Wiley & Sons A/S</I></P><P>Background and Objective: </P><P>Although heme oxygenase-1 (HO-1) plays a key role in inflammation, its anti-inflammatory effects and mechanism of action in periodontitis are still unknown. This study aimed to identify the effects of HO-1 on the proinflammatory mediators activated by nicotine and lipopolysaccharide (LPS) stimulation in human periodontal ligament (PDL) cells.</P><P>Material and Methods: </P><P>The production of nitric oxide (NO) and prostaglandin E<SUB>2</SUB> (PGE<SUB>2</SUB>) was evaluated using Griess reagent and an enzyme immunoassay, respectively. The expression of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) and HO-1 proteins was evaluated by Western blot analysis.</P><P>Results: </P><P>Lipopolysaccharide and nicotine synergistically induced the production of NO and PGE<SUB>2</SUB> and increased the protein expression of iNOS, COX-2 and HO-1. Treatment with an HO-1 inhibitor and HO-1 small interfering RNAs blocked the LPS- and nicotine-stimulated NO and PGE<SUB>2</SUB> release as well as the expression of iNOS and COX-2.</P><P>Conclusion: </P><P>Our data suggest that the nicotine- and LPS-induced inflammatory effects on PDL cells may act through a novel mechanism involving the action of HO-1. Thus, HO-1 may provide a potential therapeutic target for the treatment of periodontal disease associated with smoking and dental plaque.</P>

      • Ceruloplasmin is an endogenous protectant against kainate neurotoxicity

        To determine the role of ceruloplasmin (Cp) in epileptic seizures, we used a kainate (KA) seizure animal model and examined hippocampal samples from epileptic patients. Treatment with KA resulted in a time-dependent decrease in Cp protein expression in the hippocampus of rats. Cp-positive cells were colocalized with neurons or reactive astrocytes in KA-treated rats and epileptic patient samples. KA-induced seizures, initial oxidative stress (i.e., hydroxyl radical formation, lipid peroxidation, protein oxidation, and synaptosomal reactive oxygen species), altered iron status (increasing Fe<SUP>2+</SUP> accumulation and L-ferritin-positive reactive microglial cells and decreasing H-ferritin-positive neurons), and impaired glutathione homeostasis and neurodegeneration (i.e., Fluoro-Nissl and Fluoro-Jade B staining analyses) were more pronounced in Cp antisense oligonucleotide (ASO)- than in Cp sense oligonucleotide-treated rats. Consistently, Cp ASO facilitated KA-induced lactate dehydrogenase (LDH) release, Fe<SUP>2+</SUP> accumulation, and glutathione loss in neuron-rich and mixed cultures. However, Cp ASO did not alter KA-induced LDH release or Fe<SUP>2+</SUP> accumulation in the astroglial culture, but did facilitate impairment in glutathione homeostasis in the same culture. Importantly, treatment with human Cp protein resulted in a significant attenuation against these neurotoxicities induced by Cp ASO. Our results suggest that Cp-mediated neuroprotection occurs via the inhibition of seizure-associated oxidative damage (including impairment in glutathione homeostasis), Fe<SUP>2+</SUP> accumulation, and alterations in ferritin immunoreactivity. Moreover, interactive modulation between neurons and glia was found to be important for Cp upregulation in the attenuation of epileptic damage in both animals and humans.

        유전자재조합 pST 투여가 돼지의 성장생산성 및 육질에 미치는 영향

        The current study was undertaken to evaluate the effects of a locally-produced recombinant porcine somatotropin (pST) on growth perfermance and pork quality of finishing pigs. A total of 32 barrows weighing average 61kg were employed for the 6-week growth trial : 8 barrows were allocated into the control group (CONT): 8 barrows into the slow release-low dose pST group (SR-L); 8 barrows into the solve release - high dose pST group (SR-H); 8 barrows into the daily pST group(DAILY). CONT pigs received no pST. SR-L and SR-H pigs were treated with pST twice during 6 week trial(Each dose contained the pST equavalent to 50 ㎍ and 100 ㎍ per day, respectively). DAILY pigs received 100 ㎍ pST/㎏ B W daily. All pigs were fed a commercial feed containing 0.87% lysine ad lib. There was no significant difference in average daily gain among treatments. Feed/gain was reduced by 20% in DAILY (3.09 vs 3.85; p$lt;0.05) and 11% in SR-H (3.43 vs 3.84; p$lt;0.05) compared to the CONT. Backfat of the DAILY group was 20% thinner than that of the CONT and loin eye area was wider by 17% (p$lt;0.05) in DAILY group, compared to the CONT. The extent of the improvement in growth perfermance of SR groups was less (p$lt;0.05) than that of DAILY group. There were no significant differences in the color, juiciness, flavor and texture of pork among treatments, with the tenderness of DAILY group being less desirable compared to that of CONT group. Moisture content of DAILY group was higher (p$lt;0.05) than that of CONT group, while lipid content was reduced in DAILY group. SR group showed the middle values between CONT and DAILY group in most criteria on growth performance and pork quality investigated. These results indicated that the recombinant pST produced by LG Chem., Ltd., Korea stimulated growth performance in growing pigs and did not show any major negative effects on pork quility, suggesting that slow release pST could be used as a growth stimulant for growing pigs.

      • Lipopolysaccharide from <i>Prevotella nigrescens</i> stimulates osteoclastogenesis in cocultures of bone marrow mononuclear cells and primary osteoblasts

        <P>Background and Objective: </P><P>Lipopolysaccharide is thought to be a major virulence factor of pathogens associated with periodontal diseases and is believed to stimulate bone resorption <I>in vivo</I>. Although <I>Prevotella nigrescens</I> has been implicated in periodontitis, its role in osteoclastogenesis has not been reported. In this study, we investigated the effects of lipopolysaccharide from <I>P. nigrescens</I> on the formation of osteoclasts and the production of cytokines related to osteoclast differentiation.</P><P>Material and Methods: </P><P>Mouse bone marrow mononuclear cells were cultured in the presence of macrophage colony-stimulating factor (M-CSF) and receptor activator of nuclear factor &kgr;B ligand (RANKL), with or without lipopolysaccharide. Bone marrow mononuclear cells were also cocultured with calvarial osteoblastic cells in the presence or absence of lipopolysaccharide. Osteoclast formation was determined by tartrate-resistant acid phosphatase cytochemistry. The production of osteoprotegerin (OPG), M-CSF, tumor necrosis factor alpha (TNF-&agr;), transforming growth factor-beta (TGF-&bgr;) and prostaglandin E<SUB>2</SUB> (PGE<SUB>2</SUB>) was determined by enzyme-linked immunosorbent assay (ELISA).</P><P>Results: </P><P><I>P. nigrescens</I> lipopolysaccharide inhibited osteoclast differentiation from bone marrow mononuclear cells cultured in the presence of M-CSF and RANKL. However, in the coculture system, <I>P. nigrescens</I> lipopolysaccharide stimulated osteoclastogenesis. Notably, <I>P. nigrescens</I> lipopolysaccharide decreased OPG production but increased TGF-&bgr; secretion. In addition, treatment with <I>P. nigrescens</I> lipopolysaccharide increased PGE<SUB>2</SUB> production during the late stage of the culture period. There was no difference in M-CSF and TNF-&agr; production.</P><P>Conclusion: </P><P>These results demonstrate that <I>P. nigrescens</I> lipopolysaccharide stimulates osteoclastogenesis in the coculture system by decreasing the production of OPG and increasing the production of TGF-&bgr; and PGE<SUB>2</SUB>. Through the mechanisms involving these factors, <I>P. nigrescens</I> lipopolysaccharide may cause alveolar bone resorption in periodontal diseases.</P>

        The E3 ubiquitin ligase CHIP selectively regulates mutant epidermal growth factor receptor by ubiquitination and degradation

        Somatic mutation in the tyrosine kinase domain of epidermal growth factor receptor (EGFR) is a decisive factor for the therapeutic response to EGFR tyrosine kinase inhibitors (EGFR-TKIs) in lung adenocarcinoma. The stability of mutant EGFR is maintained by various regulators, including heat shock protein 90 (Hsp90). The C terminus of Hsc70-interacting protein (CHIP) is a Hsp70/Hsp90 co-chaperone and exhibits E3 ubiquitin ligase activity. The high-affinity Hsp90-CHIP complex recognizes and selectively regulates their client proteins. CHIP also works with its own E3 ligase activity independently of Hsp70/Hsp90. Here, we investigated the role of CHIP in regulating EGFR in lung adenocarcinoma and also evaluated the specificity of CHIP's effects on mutant EGFR. In HEK 293T cells transfected with either WT EGFR or EGFR mutants, the overexpression of CHIP selectively decreased the expression of certain EGFR mutants (G719S, L747_E749del A750P and L858R) but not WT EGFR. In a pull-down assay, CHIP selectively interacted with EGFR mutants and simultaneously induced their ubiquitination and proteasomal degradation. The expressions of mutant EGFR in PC9 and H1975 were diminished by CHIP, while the expression of WT EGFR in A549 was nearly not affected. In addition, CHIP overexpression inhibited cell proliferation and xenograft's tumor growth of EGFR mutant cell lines, but not WT EGFR cell lines. EGFR mutant specific ubiquitination by CHIP may provide a crucial regulating mechanism for EGFR in lung adenocarcinoma. Our results suggest that CHIP can be novel therapeutic target for overcoming the EGFR TKI resistance.

        Continuous lithium mining from aqueous resources by an adsorbent filter with a 3D polymeric nanofiber network infused with ion sieves

        Electrospun composite nanofiber (NF) was fabricated and employed as an adsorbent membrane filter in a continuous Li<SUP>+</SUP> mining process from seawater. The filter was composed of a hydrophilic polyacrylonitrile (PAN) matrix infused with lithium ion sieves (LIS) H<SUB>1.6</SUB>Mn<SUB>1.6</SUB>O<SUB>4</SUB>. Characterization of the LIS/PAN NF confirmed its favorable structural and surface properties for effective Li<SUP>+</SUP> adsorption. The LIS/PAN NF was mechanically suitable as a microfiltration membrane with high water flux and low pressure requirement. Breakthrough experiments at varied feed concentrations (C<SUB>f</SUB>), seawater flowrates (F), and NF thicknesses (Z) revealed the dynamic adsorption behavior of the filter. The seawater residence time was most critical and must be kept ≥0.12min at any given C<SUB>f</SUB> and Z to maximize the Li<SUP>+</SUP> capacity of the filter. This can be conveniently achieved by adjusting the F of the process. Analogous to a packed bed system, the predictive power of nine breakthrough models were determined through non-linear regression analyses. Results reveal that bed-depth-space-time, Bohart-Adams (BA) and Thomas models adequately predicted the performance of the filter albeit BA exhibited the best agreement. Meanwhile, Wolborska failed to converge with any of the experimental results while Yoon-Nelson, Wang, Clark, dose-response, and modified dose-response were too simple to provide any meaningful information. Cycled Li<SUP>+</SUP> adsorption-desorption runs successfully collected and concentrated Li<SUP>+</SUP> in a mild acid stripping solution. After ten cycles, Li<SUP>+</SUP> was separated 155-1552 times more efficiently than Na<SUP>+</SUP>, K<SUP>+</SUP>, Mg<SUP>2+</SUP> and Ca<SUP>2+</SUP>. Overall results demonstrate the potential of LIS/PAN NF as an adsorbent membrane filter for continuous Li<SUP>+</SUP> mining from aqueous resources.

        Temporal and Spatial PFC Temperature Profiles in KSTAR 2010

        <P>In and outboard plasma facing components (PFCs) of Korea Superconducting Tokamak Advanced Research (KSTAR) have been fully installed in 2010 for D-shaped diverted plasmas. Before the start of plasma operation, the PFCs were baked up to 200 by hot nitrogen gas circulation system to remove impurities including water. The surface temperature of the PFC tiles was monitored by 200 thermocouple sensors during the plasma operation [plasma shot or glow discharge cleaning (GDC)], and the temporal and spatial (poloidal) temperature profiles are obtained. Depending on the heat flux on each tile, the surface temperature shows time-dependent behavior. After 1-h morning He GDC, the temperature of the PFCs at the inboard side has reached to 40 . After an H-mode shot, the temperature of divertor tiles around the striking points was substantially increased. The time-averaged total heat flux after a few of L- and H-modes in 2010 was estimated to be approximately 0.010 .</P>
