Affecting factors on the decline of serum antimullerian hormone after ovarian cystectomy

Objective: To investigate the affecting factors on the decline of serum antimullerian hormone (AMH) in the reproductive aged women with ovarian cystectomy. Methods: This retrospective study included reproductive aged women who underwent ovarian cystectomy for benign disease from January 2019 to March 2020 at the Konkuk University Medical Center. Medical charts were reviewed about clinical characteristics, preoperative/postoperative serum AMH levels, operative records, and histopathologic results. Results: During the study period, 40 patients were included. The mean age of the patients was 33.3 years old. All patients except one underwent minimally invasive surgery including robotic and laparoscopic ovarian cystectomy. Twenty-six patients found to be endometriotic cyst and 14 patients were non-endometriotic cyst. During the surgery, the method of hemostasis after ovarian cystectomy was performed mainly by suture (97.5%) instead of electrocoagulation (2.5%). The rate of decline of serum AMH level showed statistically significant associations with age (r=0.325, p=0.040), duration of surgery (r=0.424, p=0.006), endometriosis (p<0.0001), bilaterality of ovarian cyst (p=0.014), and adhesiolysis during surgery (p=0.012). A logistic regression analysis was performed for affecting factors on the postoperative AMH decline (≥ 50%) compared to preoperative AMH level. Endometriosis was the only statistically associated with postoperative AMH decline (Odd ratio 7.10; 95% confidential interval, 1.06-47.76; p= 0.044). Conclusion: The rate of decline of serum AMH level is significant association with age, duration of surgery, endometriosis, bilaterality of ovarian cyst, and adhesiolysis during surgery. In particular, endometriosis appears to be the main cause of AMH decline.
OCT4 directly regulates stemness and extracellular matrix-related genes in human germ cell tumours

<P><B>Abstract</B></P> <P>Germ cell tumours (GCTs) are one of the most threatening malignancies in young men and women. Although several reports have suggested the importance of OCT4 in human GCTs, its role has not been clearly investigated on a molecular level. In this study, we revealed GCT-specific direct transcriptional target genes of OCT4. Conditional knockdown of OCT4 in GCT cell lines reduced cell proliferation by affecting both cell cycle and death. Knockdown of OCT4 also reduced stemness of GCTs, as assessed by the expression of other stemness factors, alkaline phosphatase staining, and tumour sphere formation ability. Analysis of whole mRNA expression patterns among GCT cells harbouring endogenous, depleted, and rescued OCT4 revealed 1133 OCT4 target genes in GCT. Combined analysis of both the chromatin binding signature of OCT4 and the genes whose expression levels were changed by OCT4 revealed 258 direct target genes of OCT4 in GCTs. In a similar way, 594 direct target genes in normal embryonic stem cells (ESCs) were identified. Among these two sets of OCT4 direct target genes, 38 genes were common between GCTs and ESCs, most of which were related to regulation of pluripotency, and 220 genes were specific to GCTs, most of which were related to focal adhesion and extracellular matrix organisation. These results provide a molecular basis for how OCT4 regulates GCT stemness and will aid our understanding of the role of OCT4 in other cancers.</P>

The purpose of this study was to identify what, if any, are the effects of self-leadership and nursing professionalism on nursing students’behavior in regard to their career preparation. The data was collected from April 1 to April 30, 2018. The participants were 174 nursing students who completed self-reporting questionnaires that covered the topics of self-leadership, nursing professionalism and career preparation behavior. The SPSS/Win 24.0 Program was used for descriptive statistics, the t-test, ANOVA, and the Pearson correlational coefficients. The average scores for self-leadership, nursing professionalism and career preparation behavior were 3.60, 3.72, and 2.34 respectively. This study found a significant positive correlation between self-leadership, nursing professionalism and career preparation behavior in the participants. It is important to provide educational programs focusing on career preparation that take in consideration the academic year of the nursing students. Additionally, it is recommended to develop educational programs that aim to improve the nursing professionalism of nursing students.

Cytochrome b5 reductase 3 (CYB5R3) is involved in various cellular metabolic processes, including fatty acid synthesis and drug metabolism. However, the role of CYB5R3 in cancer development remains poorly understood. Here, we show that CYB5R3 expression is downregulated in human lung cancer cell lines and tissues. Adenoviral overexpression of CYB5R3 suppresses lung cancer cell growth in vitro and in vivo. However, CYB5R3 deficiency promotes tumorigenesis and metastasis in mouse models. Transcriptome analysis revealed that apoptosis- and endoplasmic reticulum (ER) stress-related genes are upregulated in CYB5R3-overexpressing lung cancer cells. Metabolomic analysis revealed that CYB5R3 overexpression increased the production of nicotinamide adenine dinucleotide (NAD+) and oxidized glutathione (GSSG). Ectopic CYB5R3 is mainly localized in the ER, where CYB5R3-dependent ER stress signaling is induced via activation of protein kinase RNA-like ER kinase (PERK) and inositol-requiring enzyme 1 alpha (IRE1α). Moreover, NAD+ activates poly (ADP-ribose) polymerase16 (PARP16), an ER-resident protein, to promote ADP-ribosylation of PERK and IRE1α and induce ER stress. In addition, CYB5R3 induces the generation of reactive oxygen species and caspase-9-dependent intrinsic cell death. Our findings highlight the importance of CYB5R3 as a tumor suppressor for the development of CYB5R3-based therapeutics for lung cancer.
Atypical Scar Patterns after Gastric Endoscopic Submucosal Dissection

Background/Aims: Endoscopic submucosal dissection (ESD) for gastric neoplasms is a widely performed procedure. Local recurrence is rare, but various post-ESD scars are encountered during follow-up endoscopy. Therefore, we investigated atypical scar patterns and evaluated the associated factors. Materials and Methods: Clinicopathologic and endoscopic reviews of gastric neoplasms treated with ESD from January 2009 to December 2015 were conducted. Atypical scar patterns were classified as irregular erythema, nodularity, or mucosal defect. Results: A total of 264 patients with 274 gastric neoplasms, including 201 adenomas and 73 early gastric cancers, were enrolled. The key endoscopic findings at the resection scar were defined on the basis of gross morphology as follows: irregular erythema, mucosal defect (erosion or ulcer), and nodularity. An irregular erythema scar pattern was associated with male sex, a nodularity scar pattern with smoking, and a mucosal defect scar pattern with infra-angle location (angle and antrum) and cancer. An irregular erythema with nodularity scar pattern was also associated with male sex. An irregular erythema with nodularity and mucosal defect scar pattern was associated with liver disease and chronic kidney disease. Conclusions: The atypical scar patterns after gastric ESD are associated with various clinicopathologic factors.
