      • DJ-1/park7 modulates vasorelaxation and blood pressure via epigenetic modification of endothelial nitric oxide synthase

        <P><B>Aims</B></P><P>DJ-1/park7, a multifunctional protein, may play essential roles in the vascular system. However, the function of DJ-1/park7 in vascular contractility has remained unclear. The present study was designed to investigate whether the DJ-1/park7 is involved in the regulation of vascular contractility and systolic blood pressure (SBP).</P><P><B>Methods and results</B></P><P>Norepinephrine (NE) elevated contraction in endothelium-intact vessels in a dose-dependent manner, to a greater extent in DJ-1/park7 knockout (DJ-1/park7<SUP>−/−</SUP>) mice than in wild-type (DJ-1/park7<SUP>+/+</SUP>) mice. Acetylcholine inhibited NE-evoked contraction in endothelium-intact vessels, and this was markedly impaired in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP>. Nitric oxide (NO) production (82.1 ± 2.8% of control) and endothelial NO synthase (eNOS) expression (61.7 ± 8.9%) were lower, but H<SUB>2</SUB>O<SUB>2</SUB> production (126.4 ± 8.6%) was higher, in endothelial cells from DJ-1/park7<SUP>−/−</SUP> mice than in those from DJ-1/park7<SUP>+/+</SUP> controls; these effects were reversed by DJ-1/park7-overexpressing endothelial cells from DJ-1/park7<SUP>−/−</SUP> mice. Histone deacetylase (HDAC)-1 recruitment and H3 histone acetylation at the eNOS promoter were elevated and diminished, respectively, in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP> controls. Moreover, SBP was significantly elevated in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP> controls, but this elevation was inhibited in mice treated with valproic acid, an inhibitor of Class I HDACs including HDAC-1.</P><P><B>Conclusion</B></P><P>These results demonstrate that DJ-1/park7 protein may be implicated in the regulation of vascular contractility and blood pressure, probably by the impairment of NO production through H<SUB>2</SUB>O<SUB>2</SUB>-mediated epigenetic inhibition of eNOS expression.</P>

      • Microsatellites in the genome of the edible mushrooms, Pleurotus ostreatus cultivars, 'Heuktari' and 'Miso'

        Molecular markers were crucial role in understanding and using genomic information for marker-assisted breeding, mapping genes of interest and cloning genes. Among the molecular markers, Simple Sequence Repeat (SSR) has became increasingly important marker due to their co-dominant and high polymorphic nature and abundant distribution throughout the genome. Simple sequence repeats (SSR), also called “microsatellites” consist of tandemly repeated short DNA sequence motifs and have applied in various marker-based studies. SSRs were isolated and characterized from “Heuktari” and “Miso”, which are one of the major oyster mushroom cultivars in Korea by the genome sequencing and bioinformatic analysis. The genome sizes of “Heuktari” were 40.8 Mb and “Miso” were estimated to be 40.3 Mb, which were larger than the other P.ostreatus species (PC9 and PC10) and less than P.eryngii (KNR2312 P5). A total of 949 and 968 SSRs were found in the “Heuktari” and “Miso” genomes, respectively. A comparative analysis of five mushrooms including P.ostreatus var. florida (PC9 and PC15), P.eryngii, revealed that SSR numbers from “Heuktari” and “Miso” were the highest among them. Studied all mushrooms showed similar pattern of SSR distributions. Tri-, hexa- and octanucleotide motifs accounted for the top three fractions of all SSRs.         Hematopoietic stem cell expansion and generation: the ways to make a breakthrough

        <P>Hematopoietic stem cell transplantation (HSCT) is the first field where human stem cell therapy was successful. Flooding interest on human stem cell therapy to cure previously incurable diseases is largely indebted to HSCT success. Allogeneic HSCT has been an important modality to cure various diseases including hematologic malignancies, various non-malignant hematologic diseases, primary immunodeficiency diseases, and inborn errors of metabolism, while autologous HSCT is generally performed to rescue bone marrow aplasia following high-dose chemotherapy for solid tumors or multiple myeloma. Recently, HSCs are also spotlighted in the field of regenerative medicine for the amelioration of symptoms caused by neurodegenerative diseases, heart diseases, and others. Although the demand for HSCs has been growing, their supply often fails to meet the demand of the patients needing transplant due to a lack of histocompatible donors or a limited cell number. This review focuses on the generation and large-scale expansion of HSCs, which might overcome current limitations in the application of HSCs for clinical use. Furthermore, current proof of concept to replenish hematological homeostasis from non-hematological origin will be covered.</P>

      • Quantitative Traits Loci (QTL) Mapping of Pleurotus eryngii

        Pleurotus eryngii, an edible white-rot fungus, is cultivated widespread in Eurasia, northern Africa and China. It occupies the second position in the world mushroom market. Despites the importance, the small numbers of studies have been done. To promote the availability of P. eryngii, it is important to know the genome and organization of the fungus. In this study, the whole genome sequence of P5 monokaryon from P. eryngii KNR2312 strain was sequenced using Next Generation Sequencing (NGS) strategy. Identified 222 SSR markers based on newly known genome information and various type of markers used to construct the map consisted of 12 linkage groups (LGs) spanning 1047.8 cM. Using composite interval mapping, 71 quantitative trait loci (QTL)s were identified on 12 linkage groups (LG) for nine traits such as yield, quality, cap color and earliness in four different populations. Clusters of more than five QTLs for various traits were identified on three genomic regions on LG 1, 7, and 9. The largest cluster was identified on LG 1 in the range from 65.4 to 110.4 cM. We performed the genes prediction analysis responsible for yield in the LG1 genomic region with highest logarithm of the odd (LOD) scores The identified genes were involved in biomass degradation and synthesis and signal transduction. However, the region was wide to identifie the genes crucial role in important trait, we need to narrow down. To improve the resolution of QTL mapping, we enlarge the populations crossing with additional 205 monokaryons. Find mapping of QTLs will increases the accuracy and efficiency of interpret the genomic region and enhance the usefulness of genomic information.       • Variable-number tandem repeat loci-discriminating <i>Pleurotus ostreatus</i> cultivars

        <P><B>Abstract</B></P> <P> <I>Pleurotus ostreatus</I> is one of the most important edible mushrooms. Many cultivars have been bred to meet consumer needs. The identification of cultivars based on the morphological characteristics is restricted because fruiting bodies are frequently capricious due to environmental conditions; accordingly, sequence-based methods are required. A total of 546 simple sequence repeat primers derived from the <I>P. ostreatus</I> genome were screened, and one primer, JHH_SSR-184, was found to show polymorphisms on the major cultivars in Korea. The sequences of the polymorphic loci showed variable-number tandem repeat loci-like features enabling cultivar specificity. Thus, these loci might be applicable to discriminate <I>P. ostreatus</I> cultivars.</P> <P><B>Highlights</B></P> <P> <UL> <LI> The SSR marker discriminated the Korean major <I>Pleurotus ostreatus</I> cultivars. </LI> <LI> The cultivar specific sequences showed variable number tandem repeat loci-like feature. </LI> <LI> Nine loci with variable copy number of the repeat conferred the cultivar specificity. </LI> </UL> </P>

        Pathogen Associated Molecular Pattern (PAMP)-Triggered Immunity Is Compromised under C-Limited Growth

        In the interaction between plants and pathogens, carbon (C) resources provide energy and C skeletons to maintain, among many functions, the plant immune system. However, variations in C availability on pathogen associated molecular pattern (PAMP) triggered immunity (PTI) have not been systematically examined. Here, three types of starch mutants with enhanced susceptibility to Pseudomonas syringae pv. tomato DC3000 hrcC were examined for PTI. In a dark period-dependent manner, the mutants showed compromised induction of a PTI marker, and callose accumulation in response to the bacterial PAMP flagellin, flg22. In combination with weakened PTI responses in wild type by inhibition of the TCA cycle, the experiments determined the necessity of C-derived energy in establishing PTI. Global gene expression analyses identified flg22 responsive genes displaying C supply-dependent patterns. Nutrient recycling-related genes were regulated similarly by C-limitation and flg22, indicating re-arrangements of expression programs to redirect resources that establish or strengthen PTI. Ethylene and NAC transcription factors appear to play roles in these processes. Under C-limitation, PTI appears compromised based on suppression of genes required for continued biosynthetic capacity and defenses through flg22. Our results provide a foundation for the intuitive perception of the interplay between plant nutrition status and pathogen defense.
