Proper wound management is an increasingly important clinical challenge and is a large and growing unmet need. Pressure ulcers, hard-to-heal wounds, and problematic surgical incisions are emerging with increasing frequency. At present, the wound-healing industry is experiencing a paradigm shift toward innovative treatments that exploit nanotechnology, biomaterials, and biologics. Here we determined the effectiveness of a radiation-processed hydrogel patch for the delivery of chestnut honey (CH) for the promotion of cutaneous wound healing in diabetic mice. CH is a natural compound that has antioxidant and bactericidal effects. Two full-thickness wounds were made on the dorsal side of diabetic (db/db) mice and the wounds were covered with hydrogel-incorporated CH. Time course observations revealed that mice treated with CH hydrogel showed accelerated wound closure and formation of granulated tissue, enhanced Ki-67 expression and early upregulated HO-1 proteins in the wound region compared with water hydrogel or non-treated mice. Taken together, these findings indicate that CH hydrogel can promote wound healing in diabetics with early HO-1 protein expression.

Cytotoxicity of 2-chloroethylethyl sulfide (CEES) was investigated in murine spleen lymphocytes. Following exposure of CEES (1 mM) to spleen cell, the level of intracellar ATP and the cell viability decreased to 80% and 81% of control, respectively after 30 min, and 24% and 63%, respectively after 60 min. Thus, the decrease of viability was preceded by the loss of ATP, suggesting that ATP loss may be the cause of cell death. Nicotinamide and 1,5-dihydroxyisoquinoline, inhibitors of poly(ADP-ribose) polymerase (PAD-PRP) effectively protected against the loss of ATP and decrease of viability. The EC_(50) value of nicotinamide was approximately 400-folds greater than that of 1,5-dihydroxyisoquinoline, indicating that 1,5-dihydroxyisoquinoline is more effective than nicotinamide. In the protection effect, a good relationship between an EC_(50) value and a potency (IC_(50)) as inhibitors of PADPRP activity was noticed. Thymine and nicotinamide showed a synergistic effect in preventing both the loss of ATP and the decrease of viability. These results support that the activation of PADPRP might be involved in the cytotoxicity of CEES in the spleen lymphocytes, and PADPRP inhibitors could be used to prevent CEES-induced cytotoxicity.
