가스크로마토그라피-염광광도검출기를 이용한 지방산의 신속간편한 정량

A new method of determination of fatty acid using gas chromatography-flame photometric detector (GC-FPD) is described. Fatty acid was methylthiomethyl-esterified with methylthiomethyl chloride in 1,8-diazabicyclo[5,4,O]-undec-7-ene catalyst and its concentration measured by GC-FPD with 3% OV-1 or 3% FS-1 column. The FPD responses of thirteen fatty acid methylthiomethyl esters were examined and were proportional to the concentration of the esters without regard to their chemical structures. Consequently it was possible to determine various fatty acids using one standard calibration curve by GC-FPD. We could rapidly and conveniently determine 13 fatty acids in Ginseng Radix alba by this method.
人蔘屬과 吾加皮屬의 Nucleobases 및 Nucleosides의 分析

人蔘과 五加皮中의 核酸系 成分을 HPLC로 分離 定量하여 다음과 같은 결과를 얻었다.<br/> 1. 동일한 원료수삼에서 제조한 白蔘과 紅蔘의 核酸遊離鹽基와 nucleoside의 조성이 거의 같았다.<br/> 2. 人蔘과 五加皮중의 nucleosides 함량에서 adenosine, guanosine, cytidine 등이 特徵的인 差異를 나타냈으며 人蔘屬이 約 3倍이상 많게 나타났다. 그러나 tymidine과 inosine은 五加皮屬이 다소 많았다.<br/> 3. nucleobase 함량면에서는 人蔘이나 五加皮間에 비슷한 함량으로 1~2㎎/100g를 함유했다. Nucleobases and nucleosides in Panax and Acanthopanax genus were determined by high-performance liguid chromatography. Chromatomatography was performed on a reversed-phase system with μ Bondapak C_(18) column using phosphate buffer and 80% methanol gradient. Content of each nucleobase in two genera was about 0-2㎎/100g. Panax was contained guanosine and/or adenosine ca. 15-22㎎/100 g;and Acanthopanax guanosine ca. 3-8㎎/100g and adenosine ca. 2-7㎎/100g. Considerable amounts of cytidine, uridine, inosine, and thymidine were also detected in two genera.

For gas chromatographic determination with the sulfur-specific flame photometric detector, nine ${\beta}$-lactam antibiotics without ${\alpha}$-amino group were esterified with borontrifluoride-methanol complex and then N-benzoylated with benzoyl chloride. The gas chromatographic separation of these products was successfully carried out on various silicon polymers (OV-1, OV-101, OV-17, OV-225, and QF-1) coated on the acid washed, silanized diatomite. The structure of the esterified and N-benzoylated product was confirmed by mass spectromer.

A gas chromatographic procedure is described for the quantitative analysis of ephedrine hydrochloride, pseudoephedrine hydrochloride and norephedrine hydrochloride in pharmaceutical preparations. The procedure was based upon the determination of thioureas formed from amines and phenylisothiocyanate. The recoveries of ephedrine, pseudoephedrine and norephedrine from pharmaceutical preparations were 100.7 (S.E. 0.9), 99.5 (0.8), 99.1 (0.8) and 99.1% (0.9), respectively. The high accuracy and precision of this assay were inherent in the flame photometric detectors normally used in gas chromatography.

Such carboxyl drugs as mefenamic acid, alclofenac, ketoprofen, cicloxilic acid and tolfenamic acid in rat plasma were determined by the gas chromatography flame photometric detector (GC-FPD). After methylthiomethyl (MTM) esterification with MTM-chloride in 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU) catalyst, determination of these drugs by this method was tried and compared with that by the GC-flame ionization detector (FID) method in respect to sensitivity and effect of interferences. The results showed it was possible to analyze with accuracy by this method because of specificity of the FPD, although these drugs were not separated from interferences in plasma on GC column. The GC-FPD method was more sensitive than GC-FID method and the minimum detectable amount of monocarboxylic drugs on 3% QF-1 column was about 15fmol/injection.

Cornelia de Lange syndrome (Amsterdam dwarfism) is a congenital disease characterized by mental retardation associated with multiple malformation. A genetic etiology has been proposed, with suggestions of autosomal dominant and recessive inheritance. It affects one in 30,000 to 60,000 live births. Two-thirds of patients with the condition die before the end of their first year; death occurs from pulmonary aspiration in infancy, and from infections and bowel obstruction in later life. One reference to the syndrome highlights problems with seizures, cardiac abnormalities and difficult tracheal intubation but apart from this paper, there is little published information about the anesthetic management. Authors developed the computer application program (language: APPLE SOFT BASIC) for derivative spectrophotometry. By means of this program, derivative of spectral absorbance with respect to wavelength is recorded versus wavelength. To try this program in connection with spectrophotometer system, the authors have done the simultaneous quantitation of pyridoxine.HCl and nicotinamide in the mixture, and the result was compared with that of absorbance method.
