The tobacco whitefly, Bemisia tabaci (Hemiptera: Aleyrodidae), is a serious pest that transmits tomato yellow leaf curl virus (TYLCV) in tomato. The tobacco whitefly exhibits morphological similarity with the greenhouse whitefly, Trialeurodes vaporariorum (Hemiptera: Aleyrodidae) which also damages various plants in the green house. Therefore, it is very important to quickly and accurately diagnose whiteflies for applying pest manage ment strategies. In this study, we used a loop-mediated isothermal amplification (LAMP) assay to quickly and effectively detect B. tabaci. Primer sets were investigated for the specificity of B. tabaci by visible detection of the target DNA fragment. Two primer sets for pheromone biosynthesis activating neuropeptide (PBAN) gene were obtained from transcriptomic analysis of B. tabaci biotype Q. The PBAN-based LAMP primer set showed specific amplification of the target region. The optimal conditions for B. tabaci detection were 60 ◦ C for 60 min with four LAMP primers of PBAN. The minimum amount of genomic DNA required for visible detection was 100 ng. These results suggest that the PBAN-based LAMP assay can be applicable for field monitoring of B. tabaci.

Recently the necessity of study on optical measuring method using laser to detect the pipeline's defect in nuclear facilities, chemical industries and power plants has been increased. Because laser light can be delivered to a remote area without any difficulties, the application of laser in many industries can solve several difficulties from the limitation of access in danger area and reduce the risks of workers. Therefore, we applied a new experimental technique to the measurement of internal defects in weld zone with the combination of ESPI and image processing technique and detected the internal cracks of weld zone in the former paper.

The performance of engine cooling system is influenced by various factors which are bumper, opening area of grill, radiator, condenser, cooling fan etc. To optimize the engine cooling system is important as well as improvement of component of engine cooling module. To predict performance of engine cooling system using the 1D simulation code is very helpful at early development stage because the simulation can reduce the time and cost of test. This study focuses on prediction of performance of engine cooling system using the 1D simulation code and optimization of engine cooling module using the Taguchi method.
