        콜라주 집단미술치료 프로그램이 장애자녀 어머니의 양육스트레스 감소에 미치는 효과

        본 연구는 콜라주 집단미술치료가 장애자녀 어머니의 양육스트레스 감소에 효과가 있는지 를 알아보고자 실시한 연구이다. 연구대상은 경기도 P시의 장애인부모회에 소속되어 있는 선천적인 장애를 지닌 초, 중학교에 재학 중인 장애자녀를 둔 어머니 7명을 대상으로 하였 으며, 콜라주 집단미술치료프로그램을 주 2회, 회기 당 120분씩, 총 10회기를 실시하였다. 프로그램은 콜라주 미술치료의 치료적 요인과 효과성을 근거로 하여 전 회기를 콜라주 기 법만으로 구성하여 적용하였으며, 측정도구는 양육스트레스 척도 검사와 빗속의 사람그림 검사를 사용하였다. 연구결과를 요약하면 다음과 같다. 첫째, 콜라주 집단미술치료는 장애자 녀 어머니의 양육스트레스 전체 점수 감소에 효과적으로 작용하였다. 둘째, 콜라주 집단미 술치료는 장애자녀 어머니의 양육스트레스 척도의 하위영역인 부모의 고통, 부모-자녀 간 역기능적 상호작용, 자녀의 까다로운 특성의 세 가지 영역에서 통계적으로 유의미하게 감소 되는 변화를 보였다. 셋째, 콜라주 집단미술치료는 장애자녀 어머니의 빗속의 사람 그림검 사에서 스트레스 감소와 대처능력을 향상시키는 결과를 보였다. 이러한 결과로 볼 때 콜라 주 집단미술치료가 장애자녀 어머니의 양육스트레스 감소에 효과적임을 입증하였고 장애자 녀 어머니 관련 상담 및 심리치료현장에서 활용 가능성을 시사한다. The purpose of study is to inquire into the effect of collage group art therapy on reducing parenting stress of parents of disabled children. The subjects of this study are 7 parents with children who have disability by nature and also members of disabled children’s parent meeting. The children are attending in middle school and elementary school in P city. Collage group art therapy consists of 10 sessions, twice a week, 120 minutes per each session. All the sessions contain collage art therapy only, based on the techniques that have qualified therapeutic effects. The instruments used in this study are Parenting Stress Assessment and Draw a Person in the Rain assessment, using quantitative analysis to evaluate the effect of Collage group art therapy. The results of this study are summarized as follows. First, Collage group art therapy was effective for reducing parenting stress of parents who have disabled children. That is, the experiment group who attended Collage group art therapy session had statistically significant difference in reducing parenting stress. Second, collage group art therapy has shown the statistically meaningful changes in 3 sub categories of Parenting Stress Assessment Scale; parents' pain, dysfunctional interaction between parents and a child, and child's particular character. Third, Collage group art therapy has shown result of the decrease in stress and the improvement of the ability to cope with in PITR conducted by the parents of disabled children. Thus, Collage group art therapy proves to have effects on reducing parenting stress of disabled children and implies that it can be used in the field of psychotherapy as well as in the consultation with parents of disabled children.

        Phylogenetic relationships of Coreanomecon (Papaveraceae: Papaveroideae), an endemic genus in Korea, using DNA sequences

        Coreanomecon is a monotypic and endemic genus in Korea, distributed mainly in the southern regions. Coreanomecon is morphologically similar to Hylomecon by producing red latex, easily distinguished from Chelidonium, which produces yellow latex. Coreanomecon were merged into Hylomecon or Chelidonium depending on the authors. To understand the phylogenetic relationship of Coreanomecon, DNA sequences of chloroplast rbcL and matK and nuclear Internal Transcribed Spacer (ITS) regions were determined from the spe- cies of Papaveroideae (Papaveraceae) in Korea and analyzed with the Maximum Parsimony and Bayesian meth- ods. Phylogenetic analyses of Papaveroideae suggest that Coreanomecon is sister to the clade of Chelidonium and Stylophorum in the ITS data and that it is sister to Hylomecon in the chloroplast (cpDNA) data. A con- straining analysis using the Shimodaira-Hasegawa test (S-H test) suggested that the ITS data do not reject the sister relationship of Coreanomecon and Hylomecon. The S-H test also suggested that the cpDNA data is com- patible with the placement of Coreanomecon as a sister to the clade of Chelidonium and Stylophorum. Although the conflicting phylogenetic results may stem from insufficient phylogenetic signals, they may also be associ- ated with hybridization between Hylomecon and an ancestor of Stylophorum and Chelidonium. The results of this study suggest that Coreanomecon is a distinct lineage as an endemic genus, supporting the morphological data.

        Penicillium sp. KJ81이 생산하는 Erythritol 4-Phosphate Dehydrogenase의 특성

        Erythritol 생합성에 중요한 효소인 erythritol 4-phosphate dehydrogenase를 Penicillium sp. KJ81로부터 분리 정제하여 효소의 특성을 조사한 결과 다음과 같은 결론을 얻었다. Erythritol 4-phosphate dehydrogenase의 최적 생산 조건은 30% sucrose, 0.5% yeast extract, 0.5% $(NH_4)_2SO_4$, 0.1% $KH_2PO_4$ 그리고 0.05% $MgCl_2$ (pH 7.0) 배지에서 1 vvm aeration, 교반속도 200 rpm, $37^{\circ}C$로 Penicillium sp. KJ81을 배양한 결과 8일 배양 시 최대의 생산량을 보였다. Penicillium sp. KJ81주의 균체 추출액으로부터 ultrafiltration, 조제용 disc gel electrophoresis를 이용하여 erythritol 4-phosphate dehydrogenase를 분리 정제하였다. 최종 수율은 33%이었으며 정제배수는 39.5였다. 정제된 효소의 pI값은 4.6으로, erythrose 4-phosphate에 특이적으로 반응하였으며, Km값은 1.07mM이었다. Native-PAGE에서 single band를 보인 효소의 분자량은 약 1,500 kDa이었다. 효소활성의 최적 pH와 온도는 pH 7.0, $30^{\circ}C$였으며, 효소는 pH 4.0~9.0, 그리고 $30^{\circ}C$까지 안정하였다. 다양한 금속이온 중 $Cu^{2+}$, $Zn^{2+}$에 의하여 효소의 활성이 저해되었다. 다양한 아미노산 반응 잔기 변형시약 중 iodine과 NBS에 의해 효소의 활성이 저해되는 것을 확인하였다. In this study, the characterization of purified erythritol 4-phosphate dehydrogenase, key enzyme of erythritol biosynthesis, produced by Penicillium sp. KJ81 was investigated. Optimum production conditions of erythritol 4-phosphate dehydrogenase was 1 vvm areration, 200 rpm agitation, at $37^{\circ}C$ for 8 days in the medium containing 30% sucrose, 0.5% yeast extract, 0.5% $(NH_4)_2SO_4$, 0.1% $KH_2PO_4$, and 0.05%$MgCl_2$. Erythritol 4-phosphate dehydrogenase was purified through ultrafiltration and preparative gel electrophoresis from cell extract of Penicillium sp. KJ81. This enzyme was especially active on erythrose 4-phosphate with 1.07 mM of Km value. It gave a single band on native polyacrylamide gel electrophoresis and an isoelectric point of 4.6. The enzyme had an optimal activity at pH 7.0 and $30^{\circ}C$. It was stable between pH 4.0 and 9.0, and also below $30^{\circ}C$. The enzyme activity was completely inhibited by 1mM $Cu^{2+}$ and 1 mM $Zn^{2+}$, but was not significantly affected by other cations tested. This enzyme was inactivated by treatment of tyrosine specific reagent, iodine and tryptophan specific reagent, N-bromosuccinimide. The substrate of the enzyme, erythrose 4-phosphate showed protective effect on the inactivation of the enzyme by both reagents. These results suggest that tryptophan and tyrosine residues are probably located at or near active site of the enzyme.

        Sexuality of Aruncus aethusifolius (Rosaceae)

        Aruncus L. is a small genus which is distributed in the circumpolar regions of the Northern Hemisphere, including Europe, Asia, and North America. In Korea, there are two taxa, A. dioicus var. kamtschaticus and A. aethusifolius. Aruncus aethusifolius is a species endemic to Korea, occurring only on Jeju Island. An important characteristic of Aruncus is dioecy. However, there is some controversy regarding the sexuality of this genus, and little is known about A. aethusifolius. To characterize the sexuality of A. aethusifolius and to provide insight into the evolution of its sexual system, we investigated natural populations and herbarium specimens of A. aethusifolius. The results indicate that the species has carpellate, staminate, and bisexual flowers. Carpellate flowers are always borne on separate individuals, and staminate flowers are borne either on separate individuals or mixed with bisexual flowers on the same individuals. The sexuality of A. aethusifolius is defined as polygamo-dioecious. The polygamo-dioecious type is also found in A. dioicus var. dioicus and A. dioicus var. kamtschaticus and thus may be a general condition in Aruncus.

        Genetic variation of the endangered species Halenia coreana (Gentianaceae)

        Halenia coreana is an endangered, endemic species that is distributed in only a few locations in Korea, such as Mts. Hwaaksan and Daeamsan. It has been recently segregated from H. corniculata, broadly distributed in cold temperate regions that include northern Japan, the Russian Far East, northeastern China, Mongolia, and eastern Europe, where population sizes are usually large. To examine the genetic diversity of H. coreana and evaluate the level of genetic differentiation of the species compared with that of H. corniculata, we surveyed 183 candidate simple sequence repeats (SSR) motif markers for H. coreana and H. corniculata from sequence data of amplified fragments of a specific length in the genome. A total of 17 genomic-SSR markers were selected to examine the levels of genetic diversity and differentiation using 17 samples of H. coreana and 60 samples of three populations of H. corniculata. The results here suggest that the genetic diversity of H. coreana is very low with a high frequency of inbreeding within its population. We found that H. coreana is genetically differentiated from H. corniculata, supporting the recognition of the geographically isolated H. coreana as a distinct species.
